Abstract
Description
(発明の技術分野)
本発明は、診断剤、治療剤、薬剤、薬物開発、および免疫療法の分野に関連している。とりわけ、本発明は、上記Zot/ゾニューリン(Zonulin)/レセプターシステムを操作および使用することによって、健康を改善することに関係している。
腸の接着結合機能障害は、食物アレルギー、胃腸管の感染症、自己免疫疾患、および炎症性腸疾患を含む、多様な臨床状態において発生する(42)。インタクトな接着結合を備えた健常な、成熟した腸粘膜は、高分子の通過に対する中心的な関門として作用する。健常な状態の間に、少量の免疫学的に活性な抗原が、腸の関門を通過する。これらの抗原は、少なくとも2つの経路を通して、上記粘膜の中に吸収される。吸収されたタンパク質の圧倒的多数(90%まで)は、経細胞経路を経て腸の関門を通過し、続いてリソソームによる分解により、もっと小さな非免疫原性のペプチドへと、タンパク質を変換する。これら残余のペプチドは、インタクトなタンパク質として、傍細胞経路を経て輸送される。これは、細胞間接着結合の微妙であるが精巧な調節を伴い、結果として抗原寛容をもたらす。未熟であること、または放射線、化学療法、および/もしくは毒素にさらされることにより、上記接着結合システムの統合性が損なわれた場合、環境性の抗原に対する有害な免疫応答(自己免疫疾患および食物アレルギーを含む)を誘発し得る。このような疾患および状態を、診断および処置することが、当該分野において継続して必要である。このような疾患の処置のための新しい薬物を同定することが、当該分野において継続して必要である。
本発明の第一の実施形態は、ゾニューリンとコレラ菌(Vibrio cholerae)ファージのCTXφ ZOTタンパク質とに対するヒトレセプターの、アゴニストペプチドである。上記アゴニストポリペプチドは、アミノ酸配列FCIGRL(配列番号4)を含む。上記ポリペプチドは、長さが100アミノ酸残基未満である。
本発明者は、ゾニューリンとコレラ菌ファージのCTXφ ZOTタンパク質とに対するヒトレセプターのアゴニストペプチドを、開発した。上記ポリペプチドは、アミノ酸配列FCIGRL(配列番号4)を含む。上記ポリペプチドは、長さが100アミノ酸残基未満であるか、もしくは、50アミノ酸残基未満、40アミノ酸残基未満、30アミノ酸残基未満、20アミノ酸残基未満、10アミノ酸残基未満、または8アミノ酸残基未満である。上記ポリペプチドは、上記6アミノ酸FCIGRL(配列番号4)のみを含み得るか、上記ポリペプチドは付加的なアミノ酸を有し得る。上記他のアミノ酸は、例えば抗原タグといった、精製を容易にする、他の機能を提供し得る。
ラットの小腸組織を、ゲル濾過クロマトグラフィーおよびゾニューリンELISAの組み合わせによって、分析した。ラット腸ホモジネートを、セファクリルカラム(長さ90cm、直径2.6cm、標準分子量マーカーを用いて校正したもの)上にロードし、そして画分を収集し、そしてゾニューリン濃度を定量するためにゾニューリンELISAによって分析した。試験した6画分(F1〜F6)のうち、F5画分にもっとも高い濃度のゾニューリンが含有されていた。各画分はSDS−PAGEによって分離し、転移し、そしてゾニューリン免疫反応性、抗Zot抗体により免疫ブロットした(図1B)。ウェスタン分析により、ゾニューリン陽性画分であるF5において、見かけの分子量約24,000および分子量約23,000に移動した、2つの主要なバンドを明らかにした。一方、ゾニューリン陰性画分であるF1〜F4、F6では、各々ただ1つの免疫活性なバンド(約24kDa)のみが明らかになった。それゆえ、F5画分からの上記約23kDaのバンド(図1B矢印参照)を、クマシーブルー染色されたゲルより切り出し、そしてマトリックス支援レーザー脱離イオン化(MALDI)質量分析法に供した。タンパク質マッチについてProfound検索エンジン(ドメイン129.85.19.192、ディレクトリprofound_bin;プログラムWebProFound.exe?FORM=1)を使用した検索によって、このタンパク質と、ラットマスト細胞プロテアーゼII1との高い類似性(推定Zスコア1.58)を明らかにした。マスト細胞プロテアーゼは、トリプシン様(トリプターゼ)およびキモトリプシン様(カイメース)特性を有する、マスト細胞顆粒内に含まれるセリンプロテアーゼ特性である(46)。粘膜性マスト細胞(MMC)は主にプロテアーゼII(MCP−II)を含有するのに対し、結合組織マスト細胞は主にプロテアーゼIを含有する(46)。MCP−IIは、肺の粘膜および胃腸の粘膜(47)に特に大量に存在する。胃腸管における、MCP−IIの非常に良く特徴づけられた生理活性の一つは、線虫外寄生後の粘膜上皮透過率の調節である(48)。インビトロでの研究は、MCP−IIが接着結合複合体を分断させることにより、上皮関門を開けることを示唆する。それゆえ、本発明者らの提唱する上記ゾニューリンシステムの仮説モデル、およびMCP−IIの確立された機能には、著しい互換性がある。しかしながら、本発明者らは、ゾニューリンとMCP−IIとの間の主要な相違を認めた。それは、それらの起源(ゾニューリンは腸細胞(49)およびマクロファージに存在している)およびそれらの放出のための刺激(MCP−IIについては腸内寄生虫の外寄生[48]、およびゾニューリンについては細菌およびグリアジン[49])が、挙げられる。
本発明者らは、Zotがウサギ腸管上皮の表面に結合すること、そして上記腸の異なる領域に沿ってこの結合が変動することを、以前に明らかにしていた(44)。この結合の分布は、腸透過性に対するZotの局所的な効果、および絨毛の成熟細胞においてZotより誘導される優先的なF−アクチンの再分布と一致する(38、44)。上記Zotレセプターをさらに特徴付けるため、本発明者らは以下の実験を実施した。
結合実験を、(IEC6(ラット、腸)細胞、CaCo2(ヒト、絨毛様腸細胞)細胞、T84(ヒト、陰窩様腸細胞)細胞、MDCK(イヌ、腎臓)細胞、およびウシ肺動脈(BPA)内皮細胞が挙げられる)いくつかの上皮細胞系を用いて実施した。免疫蛍光分析のため、スライドガラス上のコンフルエントな単層(2.0×105)を、増加する時間間隔(5分、30分、60分)の間、5×10−9M Zotもしくは陰性コントロールとともに、4℃もしくは37℃で、インキュベートした。単層をZot(0.2μM)とともに37℃にて15分間インキュベートした後、細胞を冷たいPBSで10回洗浄し、懸濁および溶解した。細胞溶解物をSDS−PAGEによって分離し、PVDF膜へ転移し、そして抗Zot抗体でプローブした。Zot結合の特異性を確立するため、放射標識されたZotを使用した。これらの実験は、10倍もしくは50倍モル濃度過剰な非放射性の非標識Zotの非存在下もしくは存在下において、実施した。増加する時間間隔の間Zotタンパク質とともにインキュベートした場合、陰性コントロールに曝露された細胞と比較して、CaCo2細胞およびIEC6腸上皮細胞ならびに内皮細胞は、細胞表面において結合を表した。対照的に、His−Zotとともに最大で60分間までインキュベーションした後のT84細胞でもMDCK細胞でも、染色は観察されなかった。Zot結合の細胞特異性が、免疫ブロッティング分析において確認された。Zotは、IEC6細胞、CaCo2細胞、およびBPA細胞へは結合するが、T84細胞およびMDCK細胞へは結合しない。
His−Zot親和性カラムを、1.0mgの精製されたHis−Zotを前活性化ゲル(Aminolink、Pierce)に対して室温で終夜固定化することによって、調製した。上記カラムをPBSで洗浄し、次いで106個のIEC6細胞もしくはCaCo2細胞のどちらかを使用して獲得した粗製細胞溶解物を充填した。室温での90分間のインキュベーションの後、上記カラムは14mlのPBSで5回洗浄し、そしてHis−Zotへ結合した上記タンパク質を、50mMグリシン(pH2.5)、150mM塩化ナトリウム、および0.1%(v/v)Triton X−100からなる溶液4.0mlを用いて、上記カラムから溶出した。その1.0ml溶出画分のpHを、1.0N水酸化ナトリウムを用いて直ちに中和した。集めた画分を、還元条件下で6.0%〜15.0%(w/v)勾配SDS−PAGEに供した。分離した上記タンパク質を、ニトロセルロース膜へ転移し、そしてPerkin−Elmer Applied Biosystems Apparatus Model 494を使用してNH2末端微量配列決定に供した。IEC6細胞およびCaCo2細胞の両方から得た溶出画分は、還元条件下でSDS−PAGEによって観察された、見かけ上の分子量66kDaの単一タンパク質バンドを含んだ。ノイラミニダーゼ処置により、推定Zotレセプターのサイズが35kDaまで減少した。このことは、このレセプターがシアル酸付加されていることを示唆している(51)。
ゾニューリンは構造的にマスト細胞プロテアーゼ(MCP)−IIと類似していることを示唆している本発明者らの最近のデータによって、上記Zot/ゾニューリンレセプターが、上記MCP−IIプロテイナーゼ活性化レセプター(PAR−2)と同一ではない場合にしても類似し得ると、本発明者らは仮説した。PAR−2は、Zot/ゾニューリンレセプターについて、本発明者らが記述した特徴と共通したいくつかの特性を持っている。具体的には、成熟PAR−2は、68kDa〜80kDaの糖タンパク質であり、それは脱グリコシル化することにより36kDa〜40kDaへ減少する(47)。同様に、上記Zot/ゾニューリンレセプターは、66kDaの分子量を有しており、それは脱グリコシル化することにより35kDaに減少する(51)。胃腸管におけるPAR−2の分布(47)は、腸におけるZot/ゾニューリンレセプターの分布(44)と、一致する。PAR−2細胞内シグナル伝達には、ホスホリパーゼC(PLC)の活性化、プロテインキナーゼC(PKC)(52)の活性化、および細胞骨格再配列を導くアクチン重合(53)の活性化が関与する。Zotおよびゾニューリンは、共通の腸表面レセプターを介して、これらの同じ細胞内シグナル伝達経路を活性化する(38)。腸における上記Zot/ゾニューリンの効果と同様に、腸のPAR−2の活性化は、腸透過性増加をもたらす(54)。最終的に、PAR−2は、トリプシンによるその細胞外ドメインの切断によって活性化され、「係留リガンド(tethered ligand)」として作用する新規N末端を生成する。タンパク質分解的に活性化された、新しく生成されたN末端に対応するペプチドSLIGRL(PAR−2 AP)の外因性付加もまた、レセプター切断と独立的にPAR−2を活性化する(52)。12kDaの生理的活性Zotフラグメント(ΔG)のN末端は、宿主の腸管内でコレラ菌によりタンパク質分解的に切断されるZot細胞外ドメイン(アミノ酸残基288〜399)を含む。このΔG N末端は、PAR−2のアゴニストリガンドモチーフと構造的に類似したペプチド(FCIGRLアミノ酸残基288〜293)を含む。標的レセプターの関与および活性化に構造上要求されるものをより正確に定義するために、2つのΔG改変体が、推定PAR−2結合モチーフ(ΔG291)またはリガンドモチーフのすぐ下流領域(ΔG298)のいずれかを変異することにより、合成された。これらのペプチドを、それらがIEC−6細胞へ結合する能力、ならびにUssingチャンバーに装着されたラット小腸での生理的活性について、ΔGと比較した。ΔG291(G291V)とともにインキュベートされたIEC6細胞は、ΔGとインキュベートしたときと比較して、IEC6細胞への結合の減少を示した。一方、ΔG298ペプチド(G298V)とともにインキュベートされたIEC6細胞に関して、結合は観察されなかった。Ussingチャンバーにおける生理的アッセイは、ΔG291は、tj分解に対して、残存するが統計学的には有意ではない効果しか有さないことを示した。一方、ΔG298は、いかなる検出可能な浸透効果も誘発しなかった。これらの結果は、この結合アッセイにおいてこれら2つの改変体を用いて観察された効果と平行しており、そして291位におけるG残基および、もっとも重要なことは、298位におけるG残基が、おそらくタンパク質立体構造の変化を通じ、ΔG結合およびその標的レセプターの活性化において重大な役割を果たし得ることを、示唆した。現在、生理的環境下および病理的環境下の両方でのPAR−2機能の研究において、主要な制限のうちの1つは、特異的PAR−2インヒビターを欠くことである(52)。本発明者らの構造−機能分析に基づいて、本発明者らが変異誘発のための標的としていた上記2つのG残基を含むが、上記推定リガンドモチーフの初めの3アミノ酸残基(288〜290)を欠いた合成オクタペプチド(Zotアミノ酸残基291〜298に対応する)を、本発明者らは設計した。この合成ペプチドFZI/0を、単独でかまたはZot、ΔGもしくはゾニューリンと組み合わせてかのいずれかで、Ussingチャンバーに装着された回腸組織に関して、試験した。FZI/0または混合オクタペプチド(FZI/1)のいずれかに曝された組織のRtでは、何の変化も観察されなかった。FZI/0による回腸組織の、実験期間の前、およびその間全体にわたる処理は、Zot、ΔG、およびゾニューリンに対して応答するRtの変化を防いだ。一方、上記の3つのタンパク質の浸透効果は、FZI/1の前処置によって影響されなかった。これらのデータは、Zotおよびゾニューリンが同一のレセプターを標的とするという本発明者らの仮説を補強し、そしてFZI/0はこのレセプターを活性化することによってではなく、このレセプターに結合することによって、その抑制的な効果を発揮し得ることを示唆する。この最後の仮説を試験するため、本発明者らは、フルオレセイン標識されたFZI/0またはフルオレセイン標識されたFZI/1のどちらかとともにインキュベートされたラット小腸を使用して、インサイチュ結合実験を実施した。FZI/0に曝露された組織では、多数の蛍光粒子が示された。一方、FZI/1とともにインキュベートされた組織では、シグナルは検出されなかった。
上記Zot/ゾニューリン合成インヒビターFZI/0は、PAR−2へ結合する。FZI/0がPAR−2へ結合するかどうかを証明するため、二重標識共局在化免疫蛍光顕微鏡検査法を、Caco2細胞の単層において実施した。手短に言えば、細胞は、FITC−FZI/0またはマウスモノクローナル抗ヒトPAR−2抗体のいずれかとともに、増加する時間間隔でインキュベートした後、ローダミン標識された抗マウスIgG抗体とともにインキュベーションした。その後、細胞をPBSで3回洗浄し、PBS中の3.7%パラホルムアルデヒド(pH7.4)において15分間室温にて細胞を固定し、pH8.0でグリセロール−PBS(1:1)とともにカバーガラスを装着し、そして蛍光顕微鏡(ZEISS)により分析した。免疫蛍光粒子は、FITC−FZI/0曝露細胞および抗PAR−2抗体曝露細胞の両方において、可視化した(図3)。2つのイメージを重ねることによって、上記PAR−2レセプターとFZI/0との共局在化が明白であることが、示された(図3)。
(FZI/0−PAR−2 AP競合結合実験)
PAR−2の活性化には、トリプターゼにより生成されたその切断N末端部分もしくは上記合成ペプチド等価物PAR−2 APのいずれかと、上記レセプターの細胞外ループ2(ECL2)との結合が、必要とされる(47)。FZI/0が同じレセプタードメインへ結合するかどうかを証明するため、競合結合実験をCaco2細胞において行った。細胞単層を、過剰のPAR−2 AP(10−6M)もしくは混合ペプチドのいずれかの存在下で、FITC−FZI/0(2×10−8M)とともにインキュベーションし、次いで蛍光顕微鏡により分析した。過剰のPAR−2 APに曝露された細胞では、上記混合ペプチドに曝露された単層と比較し、有意なFZI/0免疫蛍光染色粒子の減少が示され(図4)、FZI/0はPAR−2へ結合すること、およびPAR−2 APによって競合的に置換され得ることが、示唆された。
PAR−2 APによるPAR−2レセプターの活性化が、細胞骨格の再構築を促進することが、最近報告されていた(53)。この効果が、上記合成的Zot/ゾニューリンペプチドインヒビターFZI/1によって阻害され得るかどうかを証明するため、Caco2細胞単層において免疫蛍光研究を行った。10−6MのPAR−2 APに曝露された細胞(図5A)では、張線維の分解を表したのに対し、BSA処理された単層では、張線維の分解を表さなかった(図5B)。2×10−6MのFZI/0とともにプレインキュベートすることによって、これらの細胞骨格の変化はブロックされたが(図5C)、混合ペプチドFZI/0とのプレインキュベーションによっては、細胞骨格の変化はブロックされなかった(図5D)。したがって、PAR−2 APよびFZI/0は、腸細胞の同一構造へ結合するようにみえる。
(腸透過性に対するPAR−2 APおよびMCP−IIの効果)
PAR−2は、腸細胞の頂端膜において高度に発現され、そしておそらく、1つ以上の腸細胞機能を調節している(52)。本発明者らは、これらの機能のうちの1つは、細菌定着に応答する腸透過性のゾニューリン仲介性調節であり得るかどうかを求めた。この仮説を検討するため、本発明者らは、マイクロスナップウェルアッセイにおいて、マウス腸の小腸に対する、MCP−II処理およびPAR−2 AP処理の両方の効果を試験した。10−6MのPAR−2 APもしくはMCP−II(10−8M)を腸の管腔側面へ添加することにより、無処理の組織と比較してTEERが減少し、そしてこのPAR−2依存的減少は、FZI/0で前処理することによって、完全に阻害された(図6)。これらの結果は、PAR−2はZotおよびゾニューリンの両方の標的レセプターであるという上記仮説を支持するための一連の証明をまた1つ提供し、そしてこのレセプターもまた、細胞間接着結合の調節に関与することを、示唆している。
(PAR−2シグナル伝達におけるMyD88の関与)
多くの微生物の構造(例えば、細菌性リポ多糖類もしくはRSウィルス(Respriatory Syncytial Virus)に由来する融合タンパク質)ならびに特定の内在性タンパク質は、Toll様レセプター(TLR;55)によって開始される細胞質内シグナル伝達を通じ、先天免疫系細胞を活性化する。現在までに、10種の哺乳動物のTLRが同定されている。TLRの細胞質内ドメイン内、ならびにインターロイキン−1レセプターおよびインターロイキン−18レセプターの細胞質内ドメイン内に、「Toll−IL−1レセプター」ドメインもしくは「TIR」ドメインと呼ばれる、相同領域がある。上記TIRドメインは、骨髄性分化因子88(MyD88)のような重要アダプター分子の結合を担う。PAR−2活性化に関与するシグナル伝達経路、およびTLR(例えば、NF−κB、など;52)に関与するシグナル伝達経路の著しい類似性によって、上記TLRファミリーのメンバーもしくは近接して関連するタンパク質をzot/ゾニューリンが関与し得ると、本発明者らは仮説した。したがって、本発明者らは、野生型マウスおよび上記MyD88遺伝子において標的化変異(ノックアウト、KO)を有するマウスにおいて試験された腸経上皮電気抵抗(TEER)において、ZotおよびPAR−2 APが変化を誘導する能力について試験した(図7Aおよび図7B)。図7Aの中のデータは、野生型の組織では、PAR−2 APおよびΔGの両方が、腸の抵抗において時間をかけ同等の低下を誘導することを示す。その低下は、阻害性zotペプチドFZI/0とともにプレインキュベーションすることにより、逆転した。対照的に、MyD88ノックアウトマウスに由来する腸組織は、TEERにおける減少を示すいずれの刺激にも応答できなかった。

を包含する、方法。 - 請求項51に記載の方法であって、該方法は、
前記レセプターが濃縮された調製物で哺乳動物を免疫する工程、
該哺乳動物から抗体を採取して、前記レセプターに結合する抗体調製物を形成する工程
をさらに包含する、方法。 - 請求項51に記載の方法であって、該サンプルは、腸管の細胞、腸細胞、内皮細胞、マクロファージ、リンパ球、およびその溶解物からなる群より選択される、サンプル。

を含む、ワクチン組成物。 - 患者における自己免疫疾患を診断する方法であって、該方法は、
該患者由来の第一身体サンプルを、配列番号5にて示されたアミノ酸SLIGKVDGTSHVTGに対して惹起された抗体と接触させる工程、
該第一身体サンプルに結合している該抗体の量と、自己免疫疾患を有さない健常な対照の第二身体サンプルに結合している該抗体の量とを比較する工程、
該第二身体サンプルが結合するよりも多くの該抗体に該第一身体サンプルが結合する場合に、自己免疫疾患を同定する工程、
を包含する、方法。 - 健常な対照の個体に対してゾニューリンの発現が増加した患者を処置する方法であって、該方法は、
該患者に対して、配列番号5にて示されたアミノ酸SLIGKVDGTSHVTGに対して惹起された抗体を投与することによって疾患の症状が軽減する工程、
を包含する、方法。 - 請求項61に記載の方法であって、前記疾患が癌である、方法。

- CaCo2細胞にて発現されるタンパク質と結合する抗体であって、CaCo2細胞にて発現される該タンパク質は、合成インヒビターペプチドFZ1/0(配列番号3にて表示される)と結合したタンパク質とともに局在化し、該抗体は組換えヒトPAR−2を発現するヒト細胞にもラット細胞にも結合せず、該抗体はSAM11ではない、抗体。
