ウエスタン免疫ブロット法による結合タンパク質の同定
抗体7BD−33−1のアイソレーション増幅器によって認められる抗原(s)を特定するために、細胞膜標本はドデシル硫酸ナトリウム・ポリアクリルアミドゲル電気泳動法(SDS−PAGE)に従属して、膜に移った。後者は、タンパク質がこの抗体によって見つけられるのを視覚化するために、抗体7BD−33−1のアイソレーション増幅器で探索された。
1.1.全細胞可溶化物

7BD−33−11Aによる抗原結合の同定
1.MB−231完全膜画分からの抗原の免疫沈降
全膜抽出物(5mgの総タンパク質量)は適当なIx細胞溶解バッファ(50mMのトリスpH 7.4、150mMのNaCl、1%のトリトンX−100、0.02%のアジ化ナトリウム、2mMのオルトバナジウム酸ナトリウム、50mMのフッ化ナトリウムとプロテアーゼ阻害薬カクテル(ロシュDiagnostics、マンハイム、ドイツ))量で1mg/mlの最終的なタンパク質集中に希釈された、そして、適当な二倍数RIPAバッファ(50mMのトリスpH 7.4、150mMのNaCl、1.0%のナトリウム・コール酸塩、0.2%のSDS、1%のトリトンX−100と0.02%のアジ化ナトリウム)量で、最終的なIxを得るために、RIPAは集中を緩衝する。抜粋は抜粋がそうであった4°C. IHC検査は、以前、ヒト(S.N. 10/603,006)で7BD−33−11A抗原分布を特徴づけるために実行された。2つの抗体に対する現在の学業比較された7BD−33−11Aは、15からCD63(RF AC4とH5C6)に対して、以前に生化学方法で測定されるにつれて、7BD−33−11A抗原がCD63であると指令した。24の正常な人体組織に対する抗体の結合は、人間の正常な器官組織配列(傾斜マイク、Watervliet、NY)を使用して実行された。AU第一抗体(7BD−33−11A;RFAC4(Cymbus Biotechnology社、Hants、英国)とH5C6抗CD63(ビジネスデザインPharMingen、Oakville、ON);そして、20が制御する(ダコ、トロント、ON)マウスIgGi否定)5つのμg/ml(前の最適化ステップの最適集中であることを分かる)の集中に対する抗体希釈剤バッファ(ダコ、トロント、ON)で希釈した。負の制御抗体は、製造業者によって全ての哺乳類の組織に負であることが示された。IHCのための処置は、以下の通りである。断面が1時間の間58℃で乾燥器で乾燥してdeparaffinizedされて、コプリンのそれぞれ4分の間5回をキシレンに浸漬することによってdewaxedした25のTissueは、揺れる。一連の段階的なエタノール洗浄(100%−75%)を通しての処置の後で、断面は水の中で再水和された。スライドはそれからそれぞれ5分の間高くて、中間で、低い30の推力設定で電子レンジで調理されるpH 6(ダコ、トロント、オンタリオ)で10mMのクエン酸のバッファに没頭していて、最終的に冷たいPBSに浸漬された。スライドはそれから6分の間3%の過酸化水素水に浸漬された。そして、それぞれ5分の間3回、PBSで洗われて、乾燥して、室温で5分のUniversal遮断解決(ダコ、トロント、オンタリオ)で暖められた。7BD−33−11A、単クローン系マウス抗CD63(Cymbus Biotechnology社、Hants、英国またはダコ、トロント、オンタリオ)またはアイソタイプ制御抗体‖(黒色アスペルギルス・グルコースオキシダーゼ(存在しもしなく哺乳類の組織で誘導性でもない酵素)の方へ指示した;ダコ、トロント、オンタリオ)その働く集中(各々の抗体のための5つのμg/mL)に対する抗体希釈剤バッファ(ダコ、トロント、オンタリオ)の希釈されて、そして、室温の1時間の間、終夜孵化した。スライドをPBSそれぞれ5分の間の3回で洗浄した。室温で30分に供給される(ダコEnvisionシステム、トロント、オンタリオ)につれて、主要な抗体の免疫反応性はHRP抱合型二次抗体で発見されていて/視覚化した。これがスライドに段をつけることになることをPBSそれぞれ5分と室温で10分の間免疫ペルオキシダーゼ染色のDAB(3,3’−ジアミノ・ベンジジンtetrahydrachloride、ダコ、トロント、オンタリオ)色原体基質解決を加えることによって開発される呈色反応のための3回で洗浄した。スライドを水道水で洗うことは、色素反応を終了した。マイヤーのHematoxylin(シグマDiagnostics、Oakville、ON)による対比染色の後で、滑り面は段階的なエタノール(75−100%)でdehyrdatedされて、キシレンで掃除された。マウンティング・メディア(ダコFaramount、トロント、オンタリオ)を使用して、スライドはcoverslippedされた。スライドはAxiovert 200(ツァイス・カナダ、トロント、ON)を使用して顕微鏡的に調べられた、そして、ディジタル画像はノーザンブロットのEclipse Imaging Software(ミシソーガ、ON)を使用することを得て、保存した。結果は読み込まれて、勝ち取られて、病理学者によって解釈された。
ヒト胸部腫瘍組織染色
前のIHC検査は、ヒト乳癌で7BD−33−11A抗原の癌共同を決定するために行われた、そして、7BD−33−11A抗体がヒト癌(S.N. 10/603,006)を認識するかどうかを調べた。現在では、比較はRFAC4とH5C6抗CD63とc−erbB−2抗Her2抗体を使用して行われた。乳癌組織配列は50人の乳癌患者に由来した、そして、乳癌患者で非腫瘍性の胸部組織に由来する10の試料が使われた(Imgenex社、サンディエゴ、CA)。以下の情報は、各々の患者に対して用意されていた:年齢(Cancer(AJCC)腫瘍の病期、リンパ節、エストロゲン受容体(ER)とprojesteroneレセプター(PR)状態に関する性的、アメリカのJoint委員会)。実施例4からのIHCのための処置は、続かれた。AU抗体が、1.5のμg/mLの集中が使われた抗Her2抗体を除いて、5つのμg/mLの働く集中で使われた。
ヒト前立腺の組織染色
BD−33−11A抗原が乳癌に加えて他のヒト癌組織の上で表されたかどうか決定する、複数のヒト腫瘍組織配列は、7BD−33−11Aで探索された(S.N. 10/603,006;Imgenex、サンディエゴ、CA)。それらの研究を進める際に、染色パターンof7BD−33−lアイソレーション増幅器は、人間の前立腺腫瘍組織配列(Imgenex社、サンディエゴ、CA)の上で決定された。処置が使用した染色は、実施例4で概説されるものと同様だった。AU抗体が、5つのμg/mLの働く集中で使われた。
インビボのMDA−MB−231予防的用量反応の腫瘍実験
図15と16に示されるデータに関して、6〜8週間目、雌の重篤複合免疫不全マウスは、襟首で皮下に100マイクロリットルの食塩注入されたで500万のMDA−MB−231ヒト乳癌細胞で入れられた。マウスは、10の4つの投与群に、ランダムに分けられた。その翌日に、着床0.2、2.0または20mg/kgの7BD−33−11Aまたは20mg/kgの免疫グロブリンGアイソタイプ制御抗体は、2.7mMのKCl、1mMのKH2PO4、137mMのNaClと20mMのNa2HPO4を含んだ賦形薬で、家畜濃度から希釈の後、300マイクロリットル量で、腹腔内に投与された。同じ様式の7週の期間の間の1週当たり、抗体はそれから投与された。腫瘍成長はカリパス副木で第7の日ごと頃に測定されたか、個々の動物までCCACエンドポイントに着いた。動物の体重は、本研究の期間の間原建築費だった。
インビボのMDA−MB−231確立した化学療法複合腫瘍実験
図17と18に関して、6〜8週間目の雌の重篤複合免疫不全マウスは、襟首で皮下に100マイクロリットルの食塩注入されたで500万のMDA−MB−231ヒト乳癌細胞で入れられた。腫瘍成長は、毎週、カリパス副木で測定された。大部分のコホートが41日ポスト着床で100のmm3(範囲48−122 mm3)の腫瘍容積に達したとき、8匹のマウスはランダムに4つの投与群の各々への隷下のであった。7BD−33−11A抗体、化学療法剤シスプラチン、7BD−33−1でアイソレーション増幅器とシスプラチンの組合せまたはバッファ制御は、2.7mMのKCl、1mMのKH2PO4、137niM NaClと20mMのNa2HPO4を含んだ賦形薬で、家畜濃度から希釈の後、300マイクロリットル量で、それぞれ10または9mg/kgの抗体またはシスプラチンで腹腔内に投与された。7BD−33−1でアイソレーション増幅器またはバッファ制御は、それから日64 post−着床まで同じ様式で全体で10の用量のために1週につき3回投与された。シスプラチンは、治療期間の日1、3と9に投与された。腫瘍成長は移植後の日125までカリパス副木で第7の日ごと頃に測定された、または、個々の動物がCCACに着くまで、end−は指す。動物の体重は、本研究の期間の間原建築費だった。本研究終了後、全ての動物は、CCACガイドラインに従って安楽死させられた。対のt検定を使用して、処置と関連する後処理全身腫瘍組織量減少(図16)が、いずれの7BD−33−11Aも、シスプラチンまたは2つの組合せであった。日69(5日後処理)で、両方の7BD−33−11A、シスプラチンと抗体−複合製剤は、バッファ制御処置と比較して平均の腫瘍容積を減少させた;76(p<0.001)、79(p<0.001)と86パーセント(p<0.001)それぞれ。体重が、幸福の代用物として使われた。シスプラチンと7BD−33−11Aが類似の腫瘍抑制を示したにもかかわらず、7BD−33−1でアイソレーション増幅器抗体で処置にシスプラチン treatmentin比較で見られる重量減少の同じ程度がなかった。バッファ制御と7BD−33−11A処置群間のほとんど差が、時点常時監視の上になかった。実際に、バッファ制御と7BD−33−1でアイソレーション増幅器で処置される群は、治療期間の後、わずかな重量増加を示した。対照的に、シスプラチンで処置される群は、特に最終的な服用の明白な投与後であった重量減少を経験した。移植後の(シスプラチンの最終的な服用の後の4日)日55に、シスプラチン処置群は、体重の24−30パーセントの損失を示した。従って、7BD−33−1でアイソレーション増幅器とシスプラチンは、ヒト乳癌疾患のよく認識されたモデルで、割にバッファ制御まで全身腫瘍組織量を降ろした。しかしながら、体重で測定されるにつれて、7BD−33−11A治療をうけている動物はシスプラチン投与群より良好な幸福を経験した。これらの結果は薬理学で示唆する、医薬とクオリティオブライフは人を含む他の哺乳類でこの抗体fortherapyの利益を得る。
インビボのMDA−MB−468確立した化学療法複合腫瘍実験
図19と20に関して、6〜8週間目の雌の重篤複合免疫不全マウスは、襟首で皮下に100マイクロリットルの食塩注入されたで200万のMDA−MB−468ヒト乳癌細胞で入れられた。腫瘍成長は、毎週、カリパス副木で測定された。大部分のコホートが27日ポスト着床で100のmm3(範囲11−119 mm3)の腫瘍容積に達したとき、8匹のマウスはランダムに4つの投与群の各々への隷下のであった。7BD−33−11A抗体、化学療法剤シスプラチン、7BD−33−1でアイソレーション増幅器とシスプラチンの組合せまたはバッファ制御は、2.7mMのKCl、1mMのKH2PO4、137niM NaClと20mMのNa2HPO4を含んだ賦形薬で、家畜濃度から希釈の後、300マイクロリットル量で、それぞれ10または6mg/kgの抗体またはシスプラチンで腹腔内に投与された。7BD−33−11でAまたはバッファ制御は、それから、移植後の日50まで同じ様式で全体で11の用量のために1週につき3回が続く第1の週の間、1週につき4回投与された。シスプラチンは、治療期間の日1、6、11と16に投与された。腫瘍成長は移植後の日66までカリパス副木で第7の日ごと頃に測定されたか、個々の動物までCCACエンドポイントに着いた。動物の体重は、本研究の期間の間原建築費だった。本研究終了後、全ての動物は、CCACガイドラインに従って安楽死させられた。
Claims (40)
- 癌疾患に罹った患者を処置する方法であって、前記患者に癌疾患の処置に有用(ガン組織の細胞に対して細胞毒性にしながら特徴づけられる前記抗体またはそのフラグメント)である抗癌抗体を生産するための方法に従って発生される抗癌抗体またはそのフラグメントを投与して、非癌細胞に基本的に良性であること; そこで、前記抗体またはそのフラグメントは医薬的に許容されるアジュバントで混合物に置かれて、前記癌疾患の処置を調整するのに有効な量で投与される; 前記癌組織によって発現される抗原性部位に結合する単離モノクローナル抗体またはその結合フラグメント(モノクローナル抗体の同定された特徴をPTA−4890としてATCCに寄託されたクローンによってコードしておいている抗体に束縛されながら特徴づけられる前記抗原性部位)である前記抗体、を含む方法。
- 前記抗体またはそのフラグメントがヒト化またはキメラ化である請求項1に従って癌疾患に罹った患者を処置する方法。
- 請求項1に従って癌疾患に罹った患者を処置する方法であって、毒素、酵素、放射活性化合物、および造血細胞からなる群から選択されるメンバーでそれの前記抗体または抗原結合フラグメントを活用させること;(それによって抗体複合体を形成する)、そして、前記患者に前記抗体複合体またはそれの抱合型のフラグメントを投与すること;そこで前記抗体複合体または抱合型のフラグメントは、医薬的に許容されるアジュバントで混合物に置かれて、前記癌疾患の処置を調整するのに有効な量で投与される、方法。
- 前記抗体またはそのフラグメントがヒト化またはキメラ化である請求項3の方法。
- 請求項1に従って癌疾患に罹った患者を処置する方法であって、前記抗体またはそのフラグメントの細胞毒性は、抗体依存性細胞毒性を通して媒介となる、方法。
- 請求項1に従って癌疾患に罹った患者を処置する方法であって、前記抗体またはそのフラグメントの細胞毒性は、補体依存性細胞毒性を通して媒介となる、方法。
- 請求項1に従って癌疾患に罹った患者を処置する方法であって、前記抗体またはそのフラグメントの細胞毒性は、細胞化学結合の加水分解の触媒作用を及ぼすことを通して媒介となる、方法。

- 前記抗体またはそのフラグメントがヒト化またはキメラ化である請求項12に従って癌疾患に罹った患者を処置する方法。
- 請求項12に従って癌疾患に罹った患者を処置する方法であって、抗体複合体が形成されるそれによって毒素、酵素、放射活性化合物、および造血細胞からなる群から選択されるメンバーで前記抗体またはそのフラグメントを活用させること;そして、前記患者に前記抗体複合体sまたはそれのフラグメントを投与することを含み、そこで前記抱合型の抗体は医薬的に許容されるアジュバントで混合物に置かれて、前記癌疾患の処置を調整するのに有効な量で投与される、方法。

- ATCCでPTA−4890として寄託されるクローンでコードされる単離モノクローナル抗体またはそれの抗原結合フラグメントと特異的に結合するCD63抗原性部位を発現する細胞の存在を決定する結合アッセイであって、細胞試料を提供すること、単離モノクローナル抗体またはその結合フラグメント、前記抗体またはそれの抗原結合フラグメントに前記表されたCD63抗原性部位と結合する単離モノクローナル抗体またはその結合フラグメントであることを提供して、モノクローナル抗体の同定された特徴をPTA−4890としてATCCで寄託したクローンでコードしておいている抗体に束縛されながら特徴づけられる前記抗原性部位であり、前記単離モノクローナル抗体またはその結合フラグメント細胞試料と接触させ、さらに前記細胞試料で前記単離モノクローナル抗体またはその結合フラグメントの結合を決定すること、を含み、それによって、前記単離モノクローナル抗体またはその結合フラグメントを特異的に結合するCD63抗原性部位を表す細胞の存在は、決定される、結合アッセイ。
- 細胞試料が大腸、卵巣、肺、精巣、および乳房組織からなる群から選択される組織から生じている腫瘍から得られる請求項29の結合アッセイ。
- 単離モノクローナル抗体またはその結合フラグメントと特異的に結合するCD63抗原性部位を表す試料の細胞のための分離またはスクリーニングするための方法であり、前記抗原性部位がモノクローナル抗体の同定された特徴を持つ抗体に結合していることを特徴とし、該モノクローナル抗体はPTA−4890としてATCCに寄託されたクローンによってコードされている方法であって、細胞試料を提供すること;それについて、前記表されたCD63抗原性部位(43でありながら特徴づけられる前記抗原性部位)と結合する単離モノクローナル抗体またはその結合フラグメントである単離モノクローナル抗体またはその結合フラグメント、前記抗体または抗原結合フラグメントを提供すること、PTA−4890としてATCCに寄託されたクローンによってコードしたモノクローナル抗体の同定された特徴を持っている抗体が結合しているという特徴を持ち、前記細胞試料で前記単離モノクローナル抗体またはその結合フラグメントを接触させること、そして、前記細胞試料で前記単離モノクローナル抗体またはその結合フラグメントの結合を決定すること、PTA−4890としてのATCCに寄託されたクローンまたはそれの抗原結合フラグメントによってコードされる単離モノクローナル抗体を特異的に結合するCD63抗原性部位を表すそれによって前記細胞は前記結合によって分離され、前記細胞試料のそれらの存在は確認される、方法。
- 細胞試料が大腸、卵巣、肺、精巣、および乳房組織からなる群から選択される組織から生じている腫瘍から得られる請求項31の方法。
- 前記腫瘍がモノクローナル抗体を特異的に結合する抗原を表す、または、前記ほ乳類の全身腫瘍組織量を減らすために効果的量、それによって進行が遅延する疾患および/または生存で前記ほ乳類に前記モノクローナル抗体を投与することを成立している寄託番号PTA−4890が延長されるにつれて、モノクローナル抗体の同定された特徴をクローンでコードしておく抗原結合フラグメントがそれについてATCCで寄託した哺乳類でヒト腫瘍を処置することによって生存および/または遅延疾患進行を延長する方法。
- 前記抗体が細胞毒性部位に抱合型である請求項33の方法。
- 前記細胞毒性部位が放射性同位元素である請求項33の方法。
- 前記抗体が補体を活性化する請求項33の方法。
- 前記抗体が抗体依存性細胞障害作用の媒介となる請求項33の方法。
