Abstract
Description
本発明は、生存細胞におけるアポトーシスのような特異的分子事象を証明する方法に関する。
生存細胞におけるアポトーシスのような特異的分子事象を証明するには、通常はBaxタンパク質または活性化カスパーゼのようなアポトーシス現象に伴うマーカータンパク質のウェスタンブロット法または免疫蛍光法による検出のような若干数の手法が必要である。
特異的分子事象の発生についての直接的または間接的マーカーである「結合した」(bound)マーカータンパク質の「可溶化」(solubilization)を(「可溶化した」(solubilized)マーカータンパク質の「結合」(bindig)とは別に)検出し、
上記マーカータンパク質が、上記検出の前に細胞中に存在し、
検出前に、細胞の「原形質膜」(plasma membrane)の透過性(permeabilization)を上昇させて、可溶化したタンパク質を細胞外媒質中に放出させ、
次に、可溶化(あるいは、結合(respectirely bindig))が起こったかどうか、従って相当する分子事象を決定することができる適当な手段によって、マーカータンパク質の存在を、細胞中または細胞外媒質中に検出する
ことを特徴とする。
分子事象の発生の際に可溶化あるいは結合をそれ自身が行う感受性タンパク質、
蛍光性断片、特に蛍光タンパク質、例えば緑色蛍光タンパク質、赤色蛍光タンパク質、またはそれから誘導されるまたは同様の蛍光特性を有する任意のタンパク質
を含む融合タンパク質から構成される。
この方法の実行に有用なタンパク質マーカーであって、可溶化(結合)を行う感受性成分と、検出を行うことができるようにする標識成分を含むもの
にも関する。
適当な細胞環境でマーカータンパク質を発現するベクター、
安定的または一過的にマーカータンパク質を腫瘍細胞、好ましくはヒト腫瘍細胞であることができる上記細胞で発現することができる形質転換細胞
にも関する。
上記のような
形質転換細胞、および/または
ベクター、および/または
トランスジェニック動物
を含んでなる、上記態様の一つに記載の方法を実行するためのキットにも関する。
下記の実施例は、アポトーシスの誘導の際のBaxタンパク質のコンホメーション変化を検出することができる簡単な試験を説明する。
この手法により、Baxのミトコンドリアへの再局在化が起こった細胞の比率を測定することによってBaxの誘導を評価することができる。
遺伝子の発現がBax活性化を誘導するかどうかを分析し、
薬剤のプロ−アポトーシスまたは細胞保護能力を定量的に評価する
ことができる。
アポトーシスの主要なエフェクターはカスパーゼであり、これは開裂部位におけるP1位のアスパルテートに対する絶対的特異性を特徴とするシステインプロテアーゼである。これらの酵素はいずれも、その活性部位に同一のペンタペプチド配列を含んでおり、カルパインのような他のプロテアーゼと共にアポトーシスの際の細胞で起こる多くのタンパク質分解事象に関与しており、正常な細胞機能に重要な役割を果たしているタンパク質基質の開裂を生じる(細胞骨格タンパク質、核タンパク質、またはDNA修復酵素)。
遺伝子の発現がカスパーゼ3の活性化を誘導するかどうかを分析し、
薬剤のプロ−アポトーシスまたは細胞保護能力を定量的に評価する
ことができる。
上記のカスパーゼ3プローブは、外部ミトコンドリア膜に埋設されている短い膜貫通ドメインからなる膜アンカーリングを基剤としている(突然変異チトクロームb5 C末端セグメント)。従って、このタンパク質ドメインは融合タンパク質にミトコンドリア分布と、原形質膜の選択的透過性化に関して抵抗する特性を付与する。この実施例では、試験の原理を、原形質膜の透過性化に対する蛍光シグナルの抵抗性を含む他の細胞内アンカーリング法に拡張することができることが示されている。アンカーリングの種類は、必ずしも膜貫通ドメイン自身によって表すことはできないが、その分子相互作用またはその翻訳後修飾によって、融合している蛍光タンパク質に目的のプロテアーゼによる開裂の後に細胞質ゾルまたは細胞外環境で「アンカーリング」しているが潜在的に拡散可能な状態を付与するタンパク質ドメインによって極めて単純に表すことができる。更に、特異的な細胞内局在化(原形質膜、核、膜間ミトコンドリア空間)は、プロテアーゼに対する基質の接近可能性についての追加の情報、および従ってタンパク質分解活性の分子内局在化についての情報を与えることができる。
この融合タンパク質では、細胞内アンカーリングドメインはネズミSNAP−25タンパク質の一部から構成されている。SNAP−25は分泌小胞融合過程に含まれるタンパク質であり、3個のシステイン残基のパルミトイル化によって原形質膜の細胞質ゾル表面に配置される。
この第二の実施例では、プローブのアンカーリングに用いたタンパク質ドメインは、ミトコンドリアアデニントランスロケーター(ANT2)(配列番号8)の全配列によって表される。これは、N末端が膜間空間に露出している内部ミトコンドリア膜の内在性タンパク質である。開裂性リンカーを有する蛍光タンパク質は、この末端で融合した。
(i) この融合タンパク質はミトコンドリアに適性に配置されており、
(ii) 蛍光タンパク質はカスパーゼ3によって開裂可能であり、
(iii) 蛍光シグナルは、カスパーゼ3の開裂の後は透過性化に対して感受性になる
ことが示された(図8)。
この第三の実施例は、細胞内アンカーリングは、高度に安定なタンパク質−タンパク質、およびタンパク質−核酸相互作用の効果によって得ることができることを示している。
カスパーゼ3について開発した測定法を、プログラミングされた細胞死に含まれる2種類の他のプロテアーゼに拡張した。
この実施例では、本発明のプローブおよび方法の2つの他の応用を示し、第一のものは、連結した二重パラメーターフローサイトメトリー試験の開発に関し、第二のものは、動物モデルでの抗癌活性の評価におけるバイオセンサーの応用を記載している。
特定のプローブによって明らかにされる分子事象の測定を、細胞サイクルのような別の細胞パラメーターの測定と連結した二重試験における手法を応用することができる。細胞サイクルの測定は、透過性化細胞懸濁液に十分な濃度のヨウ化プロピジウム(0.4−0.8mg/ml)を加えることによって伝統的な方法で行うことができる。PIをゲノムDNAに挿入させるため細胞を30分間インキュベーションした後、同時に「緑色」蛍光(FL1=緑色範囲に放射するGFPまたは別の蛍光タンパク質に基づく組換えバイオセンサーシグナル)および「赤色」蛍光(FL3=細胞サイクルの読み取りをできるようにするそれぞれの細胞のクロマチン含量に相当するPIの強度)を測定することによるフローサイトメトリーを用いて分析した。
検討した分子過程の活性化と細胞サイクルの所定の相の間の可能な関係を検出することができ、
抗癌分子の候補のスクリーニングに用いると、細胞増殖抑制活性のみを有する化合物、プロ−アポトーシス活性のみを有する化合物、および細胞増殖抑制およびプロ−アポトーシス活性を両方とも有する化合物を同時に同定することができる
という2つの主要な利点を有する。
イン・ビボでの「ヌード」マウス(細胞性免疫を欠く)におけるヒト腫瘍細胞系の皮下異種移植は、抗癌性の潜在能力を有する新たな分子の有効性の評価のための古典的前臨床モデルである。しかしながら、この評価は、一般的に確立した腫瘍の大きさと成長の単なる測定に基づいており、従って正確な分子的目的で試験した分子の「巨視的」効果を関連付けることはできない。

Claims (25)
- 細胞における特異的分子事象の発生を証明する方法であって、
特異的分子事象の発生についての直接的または間接的マーカーである「結合した」マーカータンパク質の「可溶化」(あるいは「可溶化」マーカータンパク質の「結合」)を検出し、
上記マーカータンパク質が、上記検出の前に細胞中に存在し、
検出前に、細胞の原形質膜の透過性を上昇させて、可溶化したタンパク質を細胞外媒質中に放出させ、
次に、可溶化(あるいは、結合)が起こったかどうか、従って相当する分子事象を決定することができる適当な手段によって、マーカータンパク質の存在を、細胞中または細胞外媒質中に検出する
ことを特徴とする、方法。 - タンパク質の細胞内アンカーリング、好ましくは膜アンカーリングによって、または、細胞内レベルでのタンパク質の区画化によって、タンパク質の細胞「結合」を行う、請求項1に記載の方法。
- タンパク質の細胞可溶化を、細胞質ゾル中のマーカータンパク質の放出によって行う、請求項1または2に記載の方法。
- マーカータンパク質が蛍光断片を含む融合タンパク質である、請求項1〜3のいずれか一項に記載の方法。
- マーカータンパク質が発現ベクターによって細胞中に産生される、請求項1〜4のいずれか一項に記載の方法。
- マーカータンパク質が細胞によって構成的に産生する、請求項1〜5のいずれか一項に記載の方法。
- 膜の透過性を上昇させた後、フローサイトメトリーまたは蛍光顕微鏡法によって、細胞における蛍光タンパク質の可溶化あるいは結合を検出する、請求項4〜6のいずれか一項に記載の方法。
- 分子事象の発生によりマーカータンパク質が開裂または修飾され且つそれが可溶化される、請求項1〜6のいずれか一項に記載の方法。
- 分子事象の発生により、マーカータンパク質の細胞内アンカーリング断片、好ましくは膜アンカーリング断片が出現し、マーカータンパク質が結合し、またはマーカータンパク質が区画化される、請求項1〜6のいずれか一項に記載の方法。
- 検出する分子事象がBax活性化であり、マーカータンパク質がBax−蛍光タンパク質融合タンパク質であり、蛍光タンパク質がBaxのN−末端に融合しており且つBax活性化の事象においてBaxが結合している、請求項1〜9のいずれか一項に記載の方法。
- 検出する分子事象がプロテアーゼの活性化であり、マーカータンパク質がプロテアーゼ開裂部位と、いずれかの側に細胞内アンカーリング部位、好ましくは膜アンカーリング部位と蛍光タンパク質を含む融合タンパク質であり、プロテアーゼが発現するときにマーカー分子が開裂によって可溶化し、蛍光タンパク質が放出される、請求項1〜9のいずれか一項に記載の方法。
- プロテアーゼがカスパーゼである、請求項11に記載の方法。
- 分子事象の発生の証明を、細胞サイクルの測定、特に細胞サイクルの様々な段階における細胞個体群の分布の測定と結びつけて考える、請求項1〜12のいずれか一項に記載の方法。
- 分子事象がBaxの活性化またはカスパーゼの活性化である、請求項13に記載の方法。
- 可溶化(結合)を行う感受性成分と検出できるようにする標識成分を含む、請求項1〜14のいずれか一項に記載の方法で用いるマーカータンパク質。
- 融合タンパク質であって、その標識成分が蛍光タンパク質である、請求項15に記載のタンパク質。
- 感受性成分の配列が、配列番号1、3、5、7、9、11および13の配列から選択される配列を含んでなる核酸によってコードされる、請求項15または16に記載のマーカータンパク質。
- 感受性成分の配列が、配列番号2、4、6、8、10、12および14の配列から選択される配列を含んでなる、請求項15〜17のいずれか一項に記載のマーカータンパク質。
- 細胞環境において、請求項15〜18のいずれか一項に記載のマーカータンパク質を発現する、ベクター。
- 請求項15〜18のいずれか一項に記載のマーカータンパク質を発現する、形質転換細胞。
- マーカータンパク質の発現が安定である、請求項20に記載の細胞。
