Description
一つには、非特許文献1では酵母でのオートファジーを観察しているが、酵母の液胞はおよそpH5.5~6.0とそれほど強い酸性の状態ではない点が挙げられる。しかし、哺乳動物細胞のリソソームはpH4.0~5.0と、酵母に比べて酸性が強い環境となっている。Rosellaを構成する2つの蛍光タンパク質は、ともに酸性条件および酸性プロテアーゼに弱い性質を有している。そのため、これらは哺乳動物細胞に即した条件下では不可逆的に消光してしまうか、または分解されてしまうという課題があった。また、哺乳動物細胞におけるオートファジーの検出において、2つの蛍光タンパク質部分の細胞内のフォールディング速度または退色による消光特性などのpHに依存しない蛍光特性の差異によって、レシオの値が実験条件で変化する可能性が挙げられる。加えて、レシオの変化がsuper ecliptic pHluorinの蛍光強度の変化のみに依存しているため、レシオ変化の振幅が小さいことも課題であった。
(2)上記蛍光タンパク質はオワンクラゲ由来黄色蛍光タンパク質である、(1)に記載の一分子FRETプローブ。
(3)上記蛍光タンパク質は、YFP、EYFP、Ypet、Topaz、Citrine、mCitrine、mEYFP、Venus、mVenusおよびTagYFPからなる群から選択される、(1)または(2)に記載の一分子FRETプローブ。

本発明の一分子FRETプローブは、リソソームまたは液胞において酵素分解を受ける蛍光タンパク質からなるアクセプタ、および配列番号1によって表されるアミノ酸配列と95%以上の配列同一性のアミノ酸配列を有しているドナーを含んでいる。
オートファジー(自食作用とも称される)は、細胞が自らのタンパク質、細胞内小器官(例えば、ミトコンドリアまたは小胞体)などの構成要素を分解する過程である。オートファジーは、生じる過程の機序に基づいて、主にマクロファジー、ミクロオートファジーまたはシャペロン介在性オートファジーに分類される。
上記ドナーは、酸性プロテアーゼに対する高い分解耐性、高い蛍光強度および酸性条件における蛍光強度の安定性を有している。酸性条件においてポジティブシグナルを発するために、上記ドナーには、酸性プロテアーゼに対する高い分解耐性を有していることが必須に求められる。そして、ネガティブシグナルに対するポジティブシグナルのレシオ値を極大化するために、上記ドナーには、高い蛍光強度および酸性条件における蛍光強度の安定性が求められる。このような本発明に係るドナーの詳細について以下に説明する。
上記アクセプタは、上述の通り、リソソームまたは液胞において酵素分解を受ける蛍光タンパク質である。したがって、当該アクセプタは、リソソームまたは液胞の内部における酵素分解を、上記ドナーと比べて、より受けやすい蛍光タンパク質とも呼ばれ得る。上記一分子FRETプローブがリソソームまたは液胞の内部に輸送されると、当該アクセプタは酵素分解される。酵素分解の後に一分子FRETプローブのうち残っているのは、上記ドナーである。したがって、上記一分子FRETプローブがリソソームまたは液胞の内部に輸送されると、上記アクセプタに基づく第1の波長の蛍光は消失する。この結果として、上記ドナーの励起にともなう第2の波長の蛍光のみが、当該ドナーから発せられる。このような機序にしたがって、上記一分子FRETプローブは上述の作用を発揮する。
本発明に係る一分子FRETプローブは、必要に応じてリンカーを含み得る。リンカーは、1つ以上のアミノ酸残基からなり、上記アクセプタとドナーとを結合するペプチド配列である。リンカーの長さは好ましくは、2アミノ酸残基~100アミノ酸残基、より好ましくは2アミノ酸残基~50アミノ酸残基の範囲内である。
上記一分子FRETプローブは、リソソームまたは液胞の内部において分解を受ける任意の生体分子を含み得る。当該生体分子は、オートファジーの活性化に関与すると知られているか、もしくは予想されている分子、またはこれまでにオートファジーの活性化との関与について示唆されていない分子であり得る。当該生体分子は、タンパク質であり得ることが好ましい。後述する通り、ポリヌクレオチドまたは当該ポリヌクレオチドを含んでいるキットを用いて、オートファジーの活性化を測定し得るからである。
上記一分子FRETプローブは、ミトコンドリア局在化配列をさらに含み得る。ミトコンドリア局在化配列を含んでいるプローブは、細胞内全体における一様な局在ではなく、ミトコンドリア内に主に集積する。したがって、当該プローブを使用すると、リソソームまたは液胞によるミトコンドリアの特異的な分解を定量化可能である。
ミトコンドリア局在化配列に代えて、上記一分子FRETプローブは、他の局在化配列またはシグナル配列を含み得る。他の局在化配列またはシグナル配列の例としては、液胞局在化配列、核移行シグナル配列、ならびにリソソーム、液胞および核以外の細胞内器官への移行シグナル配列が挙げられる。これらの配列を上記一分子FRETプローブに導入すれば、例えば、核の特異的なオートファジーを定量化することによって、有核細胞の無核細胞への成熟の定量化などが可能になる。
本発明に係るポリヌクレオチドは、上記一分子FRETプローブをコードしている。したがって、当該ポリヌクレオチドを、種々のオートファジーについて分析する細胞に導入することによって、上記一分子FRETプローブは、細胞内に発現させられ得る。
(発現コンストラクト)
本発明は、本発明に係る一分子FRETプローブを産生するために使用される発現コンストラクトを提供する。「発現コンストラクト」は、発現宿主内において機能的な発現制御領域、および当該発現制御領域に対して作動可能に連結されているポリヌクレオチドを含んでいる発現単位を指す。発現コンストラクトの一例は、上記発現制御領域および上記ポリヌクレオチドを遺伝子工学的に連結した核酸構築物である。「作動可能に連結」は、ポリヌクレオチドの発現が発現制御配列によって制御されている状態を指す。発現コンストラクトは発現ベクターの形態であり得、発現ベクターは、宿主細胞において一分子FRETプローブを組換え発現させるためのベクターであるか、または一分子FRETプローブのインビトロ生成に用いるベクターであり得る。
本発明に係るポリヌクレオチドは、適当なベクターに挿入して使用され得る。ベクターの種類は、(1)プラスミドのような自律的に複製するベクター、または(2)宿主細胞に導入されると宿主細胞のゲノムに組み込まれ、宿主細胞の染色体と共に複製されるベクターである。なお、単にベクターという場合、当該用語は、上述の発現ベクターのみならず、例えばクローニング用ベクター等も包含している。
また、本発明は、本発明に係るポリヌクレオチドを含んでいるキットを提供する。このキットは、本発明に係るポリヌクレオチドに加え、当該ポリヌクレオチドを挿入するためのベクター、およびベクターを用いて形質転換するための宿主細胞等から選択される少なくとも1つをさらに含み得る。また、本発明に係るキットにおいて、本発明に係るポリヌクレオチドは、当該ポリヌクレオチドが挿入されているベクター、または当該ポリヌクレオチドを含んでいる形質転換体として含まれ得る。
細胞へのベクターの導入(すなわち、形質転換)は、リン酸カルシウム法、リポフェクション法、エレクトロポーレーション法などの手法によって実施され得る。
(オートファジーを定量化する方法)
上記一分子FRETプローブを用いて、オートファジーの活性を定量化する方法は、細胞からの蛍光シグナルを検出することを包含している。上述の通り、上記一分子FRETプローブを用いれば、検出される蛍光シグナルに基づいて、容易かつ簡便に細胞におけるオートファジーの活性化を定量化し得る。当該方法の詳細を、具体例を用いて以下に説明する。なお、本項目の説明は、項目(オートファジーについて分析される対象)に記載の対象が上記一分子FRETプローブに含まれている実施形態に対応している。
マイトファジーを定量化する方法は、ミトコンドリア局在化配列を含んでいる一分子FRETプローブを利用する点、およびミトコンドリアのみを観察の対象としている点において、項目(オートファジーを定量化する方法)と異なる。よって、本項目における記載されるべき事項は、「オートファジー」を「マイトファジー」に、「(オートファジーについて分析される対象)」を「ミトコンドリア局在化配列」に置き換えた(オートファジーを定量化する方法)の記載と同一である。よって、(オートファジーを定量化する方法)を引用することによって、詳述を省略する。
上記一分子FRETプローブを含んでいる試料は、透明化処理に供され得る。試料を透明化処理することによって、試料の深部観察を実施可能である。透明化処理としては、例えば、国際公開公報WO2011/111876A1(米国出願番号13/583,548)、国際公開公報WO2012/147965A1(米国出願番号14/113,639)、または国際公開公報WO2012/161143A1(米国出願番号14/118,150)に記載のような、尿素および尿素誘導体からなる群から選択される少なくとも1種の化合物、好ましくは尿素を有効成分として含んでいる溶液(以下、単に生物材料用透明化試薬と呼ぶ)を用いて、試料を透明化処理することが挙げられる。生物材料用透明化試薬は、水溶液であることが好ましい。
(ドナー候補物の作製)
ドナー候補物を作製する材料として、イシサンゴに属するアザミサンゴから単離された、緑色の蛍光を発する蛍光タンパク質であるAzamiGreenの単量体化改変体mAG(Karasawa et al., (2003) J Biol Chem. 278, 34167-34171)を選択した。mAGに、以下の手順にしたがって変異を導入することによって、ドナー候補物を作製した。
作製したドナー候補物が、本発明に係るドナーに求められる性質を満たしているか否かについて、以下のように確認した。
AFFPおよびmAG407を、大腸菌(18℃)および哺乳類細胞(Mouse embryonic fibloblast(MEF)細胞)(37℃)に発現させて、蛍光強度を比較した。大腸菌において発現させたAFFPおよびmAG407の蛍光強度の比較は、既報(Katayama, H, et al., (2008) Cell Struct. Funct. 33, 1-12)にしたがって行った。精製組換えタンパク質の濃度を合わせて、pH7.0のバッファーにおいて各蛍光タンパク質の蛍光強度を測定した。上述のKatayama, H, et al.,にしたがう、大腸菌発現させたタンパク質に対する試験を表すとき、以下では単に「大腸菌試験」と記載する。
次に、酸性条件下におけるAFFPの蛍光強度の安定性を調べるために、AFFPが中性~酸性において示す蛍光強度を測定した。AFFPの比較対象として、緑色蛍光タンパク質(EGFPおよびSapphire)、黄色蛍光タンパク質(Ypet)、ならびに赤色蛍光タンパク質(mCherryおよびTag-RFP)から適宜選択して使用した。
3つの反応バッファー(pH7.0/ペプシンなし、pH4.0/ペプシンなし、pH4.0/0.05%のペプシン)において、精製組換えタンパク質をインキュベートする処理を含む大腸菌試験を行った。上記反応バッファーは、25mMのHEPESバッファー(pH7.0)または酢酸バッファー(pH4.0)に、119mMのNaCl、2.5mMのKCl、2mMのCaCl2、および2mMのMgCl2、30mMのグルコースとなるよう調製されたものである。上記反応バッファーに精製した各タンパク質を加えた混合物を、2時間にわたって37℃においてインキュベートした。当該混合物を2つに分けて、それぞれを蛍光強度測定およびウエスタンブロットに供した。蛍光強度の測定は、pH7.0のバッファーによって上記混合物を200倍に希釈してpHを調製した後に行った。ウエスタンブロットによるタンパク質の検出には、以下の抗体を使用した。AFFP:ウサギ抗Azami-Green抗体(MBL、PM052M);ならびにEGFPおよびYpet:ウサギ抗GFP抗体(CST、#2555)。
図2に示す通り、従来の蛍光タンパク質のうちでは、mCherryが酸性条件における蛍光強度が高かった。そこで、AFFPおよびmCherryのいずれが酸性プロテアーゼによる分解に対してより高い耐性を示すかを以下のように評価した。
(オートファジープローブ“Signal Retaining Autophagy Indicator”(SRAI)の作製)
AFFPの特性を利用して、酸性条件下でのプロテアーゼ感受性の差の大きい2つの蛍光タンパク質を組み合せたFRETプローブのSRAI(Signal Retaining Autophagy Indicator)を、以下のように設計した。SRAIの模式的な構成を図5の上段に示す。
SRAI/pcDNA3をMEF細胞に導入することによってSRAIを発現させ、オートファジーの観察に適しているかを評価した。比較対照としてmCherry-EGFPを用いた。mCherry-EGFPが、既報(Elsa-Noah N'Diaye, et al., (2009) EMBO reports 10, 173-179)に示されているアミノ酸配列と同等になるように、終止コドンを含んでいないmCherryのBamHI/HindIII断片と、N末端にリンカー(SGLRSAGPGTSLYKKAGFPVAT)をコードするcDNAを付加したEGFPのHindIII/EcoRI断片と、pcDNA3のBamHI/EcoRI断片とをライゲーションすることによって、mCherry-EGFP/pcDNA3を作製した。Lipofectamine(登録商標) 2000を用いてSRAI/pcDNA3またはmCherry-EGFP/pcDNA3をMEF細胞に導入した。1日後、培地を5%ウシ胎児血清を含んでいるDMEM培地(栄養培地)からHBSSに置換して37℃において2時間培養し、オートファジーを誘導した。対照群を栄養培地のまま培養した。
続いて、固定後の細胞においても、SRAIがオートファジーを検出できるかを調べた。SRAIのみを観察した点、および生細胞の観察後に、固定した同視野の細胞をふたたび観察した点を除いて、上述のオートファジーの検出と同じ操作を行った。4%PFAを含むPBSで生細胞を室温において15分固定し、HBSSを用いて5分×3回の洗浄し、2度目の観察に固定細胞を供した。
次に、SRAIが、動物個体内におけるオートファジーを検出可能であるかを調べた。1mg/mlのSRAI/pcDNA3とTransIT(登録商標)-QR Hydrodynamic Delivery Solution(Mirus Bio LLC)とを1:209の比率に混合した溶液を、マウスの体重のグラム数の1/10±0.1mL量において7週齢のオスのC57BL6Jマウスに尾静脈注射した。注射の24時間後から、食餌有り(control)または水のみ(starvation)の条件において、マウスを24時間飼育した。それから、マウスの体重のグラム数の1/100ml量におけるソムノペンチルによってマウスを麻酔した後、4%PFAを含むPBSを用いて灌流固定した。このマウスの肝臓から薄切切片を作製し、イメージングに供した。イメージングを、対物レンズにUplanApo 20x N.A.0.70を用いた点を除いて、細胞のイメージングと同様に実施した。細胞の場合と同様に画像の取得および解析を行った。レシオ画像を、0~0.4の範囲におけるIMD表示によって生成した。
(マイトファジー検出用のミトコンドリア局在型SRAIコンストラクトの作製)
次に、マイトファジー(ミトコンドリアのオートファジー)を検出するため、ミトコンドリア局在性を向上させたプローブを発現するコンストラクトを設計した。当該プローブとして、ミトコンドリア局在化シグナル配列がSRAIに付加された形態を想定した。
(1)5μgずつのmt-SRAI/pMCSTRE3GおよびTet-On調節ベクター(pEF1α-Tet3G(Takara))、
(2)5μgずつのmt-mKeima/pMCSTRE3GおよびTet-On調節ベクター。
導入から6時間後に、1μg/mLのdoxycyclineを含む栄養培地に置換した。24時間後に発現誘導を停止させ、doxycyclineなしの栄養培地に培地を置き換えて細胞質に残存するプローブの局在化を促してから、18時間後にイメージングに供した。しかし、mt-SRAIは、mt-mKeimaに比べて低いミトコンドリア局在効率を示した(図示せず)。これは、mt-SRAIの分子量が大きいために生じたと考えられた。細胞質に残存するプローブはマイトファジーの正確な検出を妨げるため、より良好な局在を示すプローブを発現するコンストラクトの構成を検討した。
mt-SRAI’を発現するコンストラクトをMEF細胞に導入して、マイトファジーの可視化および検出を試みた。0.5μgずつのmt-SRAI/pMCSTRE3G、pEF1α-Tet3Gおよびマイトファジーに必要な因子であるmCherry-Parkin/pcDNA3を、Lipofectamine(登録商標) 2000を用いてMEF細胞に導入した。導入から6時間後に1μg/mlのdoxycyclineを含む栄養培地に置換した。24時間後に、発現誘導を停止させ、doxycyclineなしの栄養培地に置換して細胞質に残存するプローブの局在化および分解を促した。18時間後に、30μMのCarbonyl cyanide m-chlorophenylhydrazone(CCCP)を用いてマイトファジーを誘導し、さらに18時間後に細胞をイメージングに供した。さらに、細胞内酸性環境が中性化してもシグナルが消失しないことを示すため、50mMのNH4Clを加えて画像を再取得した。イメージング、ならびに画像の取得および解析には、固定後の哺乳類細胞におけるオートファジーの検出と同じ条件を適用した。レシオ画像を、0~0.4の範囲におけるIMD表示によって生成した。
Rosadoらが報告したプローブ(Rosella)が、SRAIと比較してオートファジーの測定に適しているかを検討した。Rosellaのコンストラクトを大腸菌にて発現し、精製した組換え蛍光タンパク質を用い、既報(Katayama, H,他(2008) Cell Struct. Funct. 33, 1-12)にならい、pH7.0、pH4.0(0.05%ペプシンあり、なし)の反応バッファー中、37℃で2時間インキュベートし、蛍光測定に供した。蛍光測定の際は、各サンプルをpH7.0のバッファーで200倍希釈してpHをあわせてから測定を行った。反応バッファーは、25mM HEPESバッファー(pH7.0)または酢酸バッファー(pH4.0)に119mM NaCl、2.5mM KCl、2mM CaCl2、2mM MgCl2、30mM Glucoseとなるよう調製されたものである。大腸菌試験を実施した結果、酸性条件(pH4.0)、酸性プロテアーゼ処理により、Rosellaの構成成分のDsRed.T3、SEPともに蛍光強度が低下していた(図12のグラフを参照)。
Claims (9)
- リソソームまたは液胞の内部において酵素分解を受ける蛍光タンパク質からなるアクセプタ、および配列番号1によって表されるアミノ酸配列と98%以上の配列同一性のアミノ酸配列を有しているドナーを含んでおり、
上記ドナーは、配列番号1の2位を先頭として数える表記方法にしたがったときの、Y63およびG64を有しており、
上記ドナーは、配列番号1によって表されるアミノ酸配列のドナーと実質的に同等、または当該ドナー以上の、酸性プロテアーゼに対する分解耐性、蛍光強度および蛍光強度の安定性を有している、一分子FRETプローブ。 - リソソームまたは液胞の内部において酵素分解を受ける蛍光タンパク質からなるアクセプタ、および配列番号1によって表されるアミノ酸配列と98%以上の配列同一性のアミノ酸配列を有しているドナーを含んでおり、

上記ドナーは、配列番号1の2位を先頭として数える表記方法にしたがったときの、S58、S60、A82、S184、E199、C210およびA217を有している、一分子FRETプローブ。 - 上記蛍光タンパク質はオワンクラゲ由来黄色蛍光タンパク質である、請求項1または2に記載の一分子FRETプローブ。
- 上記蛍光タンパク質は、YFP、EYFP、Ypet、Topaz、Citrine、mCitrine、mEYFP、Venus、mVenusおよびTagYFPからなる群から選択される、請求項1~3のいずれか1項に記載の一分子FRETプローブ。
- ミトコンドリア局在化配列をさらに含んでいる、請求項1~4のいずれか1項に記載の一分子FRETプローブ。
- 請求項1~5のいずれか1項に記載の一分子FRETプローブをコードしている、ポリヌクレオチド。
- 請求項6に記載のポリヌクレオチドを含んでいる、キット。
- 請求項1~5のいずれか1項に記載の一分子FRETプローブを含んでいる細胞からの蛍光シグナルを検出することを包含している、オートファジーの活性を定量化する方法。
- 上記細胞が固定されている、請求項8に記載の方法。
