Abstract
【解決手段】本創傷ドレッシング材料は、ポリマー基質、銀塩およびこの銀塩を光に対し安定化する染料を含有する。本基質は、Ag+と複合化したコラーゲンおよび/または酸化再生セルロースを含み得、本染料は、たとえば、アニリンまたはアクリジン染料であり得る。このような材料を作製する方法およびこのような材料を含む創傷ドレッシングも提供される。

Description
コラーゲン/ORC凍結乾燥スポンジ材料に基づく、抗酸化性および抗菌性を有する創傷ドレッシング材料を次のようにして作製する。
メチレンブルー染料をクリスタルバイオレット(塩基性染料)で置き換えた以外は実施例1の手順を採用した。クリスタルバイオレットは、以下の濃度でスラリー中に組み入れられた:0%(参照サンプル)、1mg/mL、0.5mg/mLおよび0.1mg/mL。
メチレンブルー染料をフラビン3,6−ジアミノアクリジン・ヘミスルフェート(3,6-Diaminoacridine hemisulfate)(塩基性染料)で置き換えた以外は実施例1の手順を採用した。このフラビンは、以下の濃度でスラリー中に組み入れられた:0%(参照サンプル)、1mg/mL、0.5mg/mLおよび0.1mg/mL。
メチレンブルー染料をフラビン3,6−ジアミノアクリジン・ヘミスルフェート(塩基性染料)で置き換えた以外は実施例1の手順を採用した。このフラビンは、以下の濃度でスラリー中に組み入れられた:0%(参照サンプル)、1mg/mL、0.5mg/mLおよび0.1mg/mL。
メチレンブルー染料をメチレンブルーとフラビン3,6−ジアミノアクリジン・ヘミスルフェートとの混合物で置き換えた以外は実施例1の手順を採用した。それぞれの染料は、スラリー中に0.5mg/mLの濃度で組み入れられた。
メチレンブルー染料をクリスタルバイオレットとフラビン3,6−ジアミノアクリジン・ヘミスルフェートとの混合物で置き換えた以外は実施例1の手順を採用した。それぞれの染料は、スラリー中に0.5mg/mLの濃度で組み入れられた。
メチレンブルー染料をクリスタルバイオレットとメチレンブルーとの混合物で置き換えた以外は実施例1の手順を採用した。それぞれの染料は、スラリー中に0.5mg/mLの濃度で組み入れられた。
フリーラジカルを含む酸素と反応して、これを除去する創傷ドレッシング材料の能力は、WO94/13333に記述されたDPPH検査によって評価される(本文献は、参照によりその全体が明示的に本明細書に包含される。)。前記検査は、Blois M.S.、”ネイチャー(Nature)”、181:1199(1958年)およびBanda P.W.等、”Analytical Letters”、7:41(1974年)に記載された検査に適合するようにしたものである。
実施例1〜7で作製されたスポンジの殺菌作用は、緑膿菌および黄色ブドウ球菌上で阻止帯を見ることにより検査する。
本発明の具体的な実施態様は、次の通りである。
(1)創傷ドレッシング材料において、
ポリマー基質、銀塩、および当該銀塩を光に対し安定化するための染料を含む、創傷ドレッシング材料。
(2)実施態様(1)に記載の創傷ドレッシング材料において、
前記基質が、固形の生体吸収性材料、好ましくは、コラーゲン、酸化セルロース、キトサン、ガラクトマンナン、グリコサミノグリカン、およびこれらの混合物からなる群から選ばれた固形の生体吸収性材料を含む、創傷ドレッシング材料。
(3)実施態様(2)に記載の創傷ドレッシング材料において、
前記基質が、コラーゲン、酸化セルロース、キトサン、およびこれらの混合物からなる群から選ばれた固形の生体吸収性材料を含む、創傷ドレッシング材料。
(4)実施態様(1)に記載の創傷ドレッシング材料において、
前記基質が、セルロース、アルジネート、ポリアクリレート、ポリウレタン、ポリアミド、およびこれらの混合物からなる群から選ばれたものである、創傷ドレッシング材料。
(5)前記実施態様のいずれかに記載の創傷ドレッシング材料において、
前記創傷ドレッシング材料が、連続固形シート、孔あき固形シート、ウェブ、織布、編み布、不織布、凍結乾燥スポンジ、または溶媒乾燥スポンジの形態を有する、創傷ドレッシング材料。
(6)前記実施態様のいずれかに記載の創傷ドレッシング材料において、
前記ポリマー基質がアニオン性ポリマーを含み、前記銀塩がAg+と当該アニオン性ポリマーとの塩を含む、創傷ドレッシング材料。
(7)前記実施態様のいずれかに記載の創傷ドレッシング材料において、
前記組成物が、当該組成物の乾燥重量に基づいて、0.01重量%〜5重量%の銀を含む、創傷ドレッシング材料。
(8)前記実施態様のいずれかに記載の創傷ドレッシング材料において、
前記染料が、抗酸化性の染料、たとえば、アニリン染料、アクリジン染料、チオニン染料、ビス−ナフタレン染料、チアジン染料、アゾ染料、アントラキノン、およびこれらの混合物からなる群から選ばれた抗酸化性の染料を含む、創傷ドレッシング材料。
(9)実施態様(8)に記載の創傷ドレッシング材料において、
前記抗酸化性の染料が、ゲンチアナバイオレット、アニリンブルー、メチレンブルー、クリスタルバイオレット、アクリフラビン、9−アミノアクリジン、アクリジンイエロー、アクリジンオレンジ、プロフラビン、キナクリン、ブリリアントグリーン、トリパンブルー、トリパンレッド、マラカイトグリーン、アザクリン、メチルバイオレット、メチルオレンジ、メチルイエロー、エチルバイオレット、アシッドオレンジ、アシッドイエロー、アシッドブルー、アシッドレッド、チオフラビン、アルファズリン、インディゴブルー、メチレングリーン、およびこれらの混合物からなる群から選ばれたものである、創傷ドレッシング材料。
(10)前記実施態様のいずれかに記載の創傷ドレッシング材料において、
前記染料が、前記材料の乾燥重量に基づいて0.2〜2重量%の量で存在する、創傷ドレッシング材料。
(11)前記実施態様のいずれかに記載の創傷ドレッシング材料において、
前記ポリマー基質が、本質的に、酸化セルロースとコラーゲンおよび/またはキトサンとの混合物からなる、創傷ドレッシング材料。
(12)前記実施態様のいずれかに記載の創傷ドレッシング材料において、
前記材料が、殺菌され、微生物不透過性の容器で包装されている、創傷ドレッシング材料。
(13)前記実施態様のいずれかに記載の創傷ドレッシング材料において、
前記材料が、ここに明示された抗酸化活性についてのジフェニルピクリルヒドラジル(DPPH)検査で少なくともほぼ15%のフリーラジカル活性を有する、創傷ドレッシング材料。
(14)創傷の治療薬剤の調剤のための、請求項1〜13のいずれかに記載の材料の使用。
(15)実施態様(14)に記載の使用において、
前記創傷が、慢性的創傷、好ましくは、静脈潰瘍、褥瘡性潰瘍、および糖尿病性潰瘍からなる群から選ばれた慢性的創傷である、使用。
(16)抗酸化性の創傷ドレッシング材料の製造方法において、
光化学還元作用に対して銀塩を安定化させる染料でポリマー基質材料を染色するステップと、
水中または有機溶媒中に溶解しまたは分散した銀塩で当該基質材料を処理するステップと、
を含む、製造方法。
(17)実施態様(16)に記載の方法において、
実施態様(1)〜(13)のいずれかに記載の創傷ドレッシング材料の製造のための、方法。
(18)創傷ドレッシングにおいて、
実施態様(1)〜(13)のいずれかに記載の抗酸化性の創傷ドレッシング材料を含む、創傷ドレッシング。
(19)実施態様(18)に記載の創傷ドレッシングにおいて、
前記材料が、殺菌され、微生物不透過性の容器で包装されている、創傷ドレッシング。
Claims (19)
- 創傷ドレッシング材料において、
ポリマー基質、銀塩、および当該銀塩を光に対し安定化するための染料を含む、創傷ドレッシング材料。 - 請求項1に記載の創傷ドレッシング材料において、
前記基質が、固形の生体吸収性材料、好ましくは、コラーゲン、酸化セルロース、キトサン、ガラクトマンナン、グリコサミノグリカン、およびこれらの混合物からなる群から選ばれた固形の生体吸収性材料を含む、創傷ドレッシング材料。 - 請求項2に記載の創傷ドレッシング材料において、
前記基質が、コラーゲン、酸化セルロース、キトサン、およびこれらの混合物からなる群から選ばれた固形の生体吸収性材料を含む、創傷ドレッシング材料。 - 請求項1に記載の創傷ドレッシング材料において、
前記基質が、セルロース、アルジネート、ポリアクリレート、ポリウレタン、ポリアミド、およびこれらの混合物からなる群から選ばれたものである、創傷ドレッシング材料。 - 請求項1〜4のいずれかに記載の創傷ドレッシング材料において、
前記創傷ドレッシング材料が、連続固形シート、孔あき固形シート、ウェブ、織布、編み布、不織布、凍結乾燥スポンジ、または溶媒乾燥スポンジの形態を有する、創傷ドレッシング材料。 - 請求項1〜5のいずれかに記載の創傷ドレッシング材料において、
前記ポリマー基質がアニオン性ポリマーを含み、前記銀塩がAg+と当該アニオン性ポリマーとの塩を含む、創傷ドレッシング材料。 - 請求項1〜6のいずれかに記載の創傷ドレッシング材料において、
前記組成物が、当該組成物の乾燥重量に基づいて、0.01重量%〜5重量%の銀を含む、創傷ドレッシング材料。 - 請求項1〜7のいずれかに記載の創傷ドレッシング材料において、
前記染料が、抗酸化性の染料、たとえば、アニリン染料、アクリジン染料、チオニン染料、ビス−ナフタレン染料、チアジン染料、アゾ染料、アントラキノン、およびこれらの混合物からなる群から選ばれた抗酸化性の染料を含む、創傷ドレッシング材料。 - 請求項8に記載の創傷ドレッシング材料において、
前記抗酸化性の染料が、ゲンチアナバイオレット、アニリンブルー、メチレンブルー、クリスタルバイオレット、アクリフラビン、9−アミノアクリジン、アクリジンイエロー、アクリジンオレンジ、プロフラビン、キナクリン、ブリリアントグリーン、トリパンブルー、トリパンレッド、マラカイトグリーン、アザクリン、メチルバイオレット、メチルオレンジ、メチルイエロー、エチルバイオレット、アシッドオレンジ、アシッドイエロー、アシッドブルー、アシッドレッド、チオフラビン、アルファズリン、インディゴブルー、メチレングリーン、およびこれらの混合物からなる群から選ばれたものである、創傷ドレッシング材料。 - 請求項1〜9のいずれかに記載の創傷ドレッシング材料において、
前記染料が、前記材料の乾燥重量に基づいて0.2〜2重量%の量で存在する、創傷ドレッシング材料。 - 請求項1〜10のいずれかに記載の創傷ドレッシング材料において、
前記ポリマー基質が、本質的に、酸化セルロースとコラーゲンおよび/またはキトサンとの混合物からなる、創傷ドレッシング材料。 - 請求項1〜11のいずれかに記載の創傷ドレッシング材料において、
前記材料が、殺菌され、微生物不透過性の容器で包装されている、創傷ドレッシング材料。 - 請求項1〜12のいずれかに記載の創傷ドレッシング材料において、
前記材料が、ここに明示された抗酸化活性についてのジフェニルピクリルヒドラジル(DPPH)検査で少なくともほぼ15%のフリーラジカル活性を有する、創傷ドレッシング材料。 - 創傷の治療薬剤の調剤のための、請求項1〜13のいずれかに記載の材料の使用。
