本出願は、米国出願番号第60/510,086号(2003年10月10日出願)、同第60/526,571号(2003年4月12日出願)および同第60/567,771号(2004年5月5日出願)[これらはすべて参照により本明細書に組み入れられる]に対して35 USC 119(e)に基づく利益を主張するものである。
本発明は免疫応答の惹起方法に関する。
ワクチン接種方法は当技術分野において記載されている。例えば、Prayagaら (1997) Vaccine 15(12-13): 1349-1352、Kilpatrickら (1997) Hybridoma 16: 381-389、Kilpatrickら (1998) Hybridoma 17: 569-576、Pertmerら (1995) Vaccine 13; 1427-1430およびOlsenら (1997) Vaccine 15; 1149-1156を参照されたい。しかし、核酸投与スケジュール(例えば、増強された細胞性免疫(CMI)応答を特異的に誘導するもの)の最適化が依然として必要とされている。これは、多種多様な免疫、炎症ならびに感染性の疾患および障害の予防および治療に非常に有益であろう。
本発明は、in vivoでCMI応答を惹起(または増強)するための方法を提供する。特に、該方法はin vivoでT細胞応答を惹起(または増強)する。
(i)該T細胞エピトープをコードする目的とするヌクレオチド(NOI)を投与することを各投与が含む、該被験体への1〜14日間隔の少なくとも2回の投与を含む第1の免疫化、および所望により、
(ii)該被験体への(a)該T細胞エピトープをコードするNOIまたは(b)該T細胞エピトープを含むタンパク質の少なくとも1回の投与を含む第2の免疫化を含んでなり、ここで、第1の免疫化の第1の投与と第2の免疫化の第1の投与との間の時間は21〜365日間である。
以下の開示は、投与するNOIによりコードされる配列(例えば、エピトープまたは抗原)について記載する。第2の免疫化およびその後の免疫化の場合には、そのようなNOIの代わりに、同じエピトープまたは抗原を含むタンパク質を投与することが可能であると理解される。
本発明の方法は、本明細書中では「免疫化」と称される一連の投与を含む。したがって、該方法は、本明細書中では「第1の免疫化」、「第2の免疫化」などと称される1、2、3、4、5またはそれ以上(例えば、10(10を含む)まで)のそのような一連の免疫化を含みうる。
本発明は、特に例示されている分子またはプロセスパラメーターに限定されるものではなく、それらはもちろん様々に変化しうると理解されるべきである。また、本発明で用いる用語は、専ら本発明の特定の実施形態を説明することを目的としたものであり、限定的なものではないと理解されるべきである。さらに、本発明の実施は、特に示さない限り、ウイルス学、微生物学、分子生物学、組換えDNA技術および免疫学の通常の方法(それらのすべては当技術分野の通常の技量の範囲内である)を用いる。そのような技術は文献に十分に説明されている。例えば、Sambrookら,Molecular Cloning:A Laboratory Manual(2nd Edition,1989);DNA Cloning:A Practical Approach,vol.I & II(D.Glover編);Oligonucleotide Synthesis(N.Gait編,1984);A Practical Guide to Molecular Cloning(1984)およびFundamental Virology,2nd Edition,vol.I & II(B.N.FieldsおよびD.M.Knipe編)を参照されたい。
免疫系が疾患を抑制するメカニズムは、体液性免疫による中和抗体の誘導、および細胞性免疫によるT細胞応答の生成を含む。本明細書中で用いる、標的抗原(TA)(EOIを含む)に対する「免疫応答」なる語は、宿主哺乳類被験体における、そのTAに対する体液性および/または細胞性免疫応答の発生を意味する。
CMIおよび体液性応答を開始および/または増強するためには、少なくとも2つの特別な型のT細胞が必要になる。CD4補助受容体を発現するT細胞の特定のサブセット上の抗原受容体はTヘルパー(Th)細胞またはCD4 T細胞(本明細書中では以下、Tヘルパー細胞と称される)であってよく、それらは、MHCクラスII分子に結合した抗原ぺプチドを認識する。一方、CD8補助受容体を発現するT細胞の特定のサブセット上の抗原受容体は細胞傷害性Tリンパ球(CTL)またはCD8+T細胞(本明細書中では以下、CD8+T細胞と称される)と称され、それらは、MHCクラスI分子上に提示された抗原と反応する。
ヘルパーT細胞またはCD4+細胞は更に、2つの機能的に異なるサブセット(すなわち、それらのサイトカインおよびエフェクター機能において異なるTh1およびTh2)に分類されうる。Th1およびTh2応答は正のみならず負の様態でも調節されて、Th1細胞性応答はIL-2、IL-12およびIFN-γのようなTh1サイトカインにより増強され、IL-4およびIL-10のようなTh2サイトカインにより減弱される。一方、抗体応答はIL-4およびIL-10のようなTh2サイトカインにより増強されるが、IFN-γ、およびIFN-γを増強し単球により産生される別のサイトカインであるIL-12のようなTh1サイトカインによりダウンレギュレーションされる。したがって、IFN-γ、IL-2およびIL-12のような古典的なTh1サイトカインは、炎症応答を誘導する免疫補助因子とみなされうる。一方、IL-4およびIL-10のような古典的なTh2サイトカインは、いくつかの状況において、重篤な炎症応答を抑制するサイトカインとみなされうる。
CD8+T細胞は2以上の様態で機能しうる。CD8+T細胞の最もよく知られた機能は、MHCクラスI分子の環境においてペプチド抗原を保持する標的細胞の殺細胞または細胞溶解である。したがって、そのような理由により、これらの細胞は細胞傷害性Tリンパ球(CTL)と称されることが多いのである。しかし、おそらく、ある種の感染においてより強力な防御的で重要なもう1つの機能は、インターフェロンγ(IFN-γ)を分泌するCD8+T細胞の能力である。したがって、細胞溶解活性のアッセイおよびIFN-γ放出のアッセイは共に、CD8+T細胞免疫応答の測定(例えば、後記のELISPOTアッセイ)において重要である。感染症においては、疾患の症状が現われる前の疾患の初期段階で、感染性抗原を含む感染因子を殺すことによりCD8+T細胞が防御しうることを示唆する証拠が存在する。
本発明は、宿主被験体における標的抗原に対するCMI応答を増強および/またはモジュレーションしうる方法に関する。本明細書中で用いる「増強」なる語は、CMI応答のすべての態様における向上を包含し、TAのEOIをコードするNOIの反復投与に対するCMI応答の大きさおよび/または持続時間および/または質の刺激および/または増大および/または強化および/またはアップレギュレーションを包含するが、これらに限定されるものではない。例えば、CMI応答は、(i)標的抗原を認識するCD8+T細胞の活性化および/または産生および/または増殖を増強することにより、および/または(ii)CMI応答をTh2型応答からTh1型応答へ変化させることにより、増強されうる。Th1関連応答のこの増強は、細胞内感染に対する応答において特に重要である。なぜなら、前記のとおり、CMI応答は、活性化Th1(例えば、IFN-γ誘導性)細胞により増強されるからである。
本明細書中の開示においては、応答の惹起は、しばしば、CMI応答の惹起として説明される。CMI応答の惹起はT細胞応答の惹起を含むと理解される。本発明の方法により惹起された応答は、「増強」された応答でありうる。本発明の方法において投与したのと同じ量のNOI(また、適用可能な場合には同じ量のタンパク質)を投与した対照方法において惹起される応答より、本発明の方法により惹起された応答のほうが大きい場合には、「増強」された応答が生じたと判定されうる。そのような対照方法は、例えば、1回の投与におけるNOI(また、適用可能な場合にはタンパク質)の投与よりなるものでありうる。あるいは、対照方法は、28日間隔の2回の別々の投与でのNOI(また、適用可能な場合にはタンパク質)の投与よりなるものでありうる。
各病原因子または疾病状態は、免疫認識および宿主における病原因子または疾病状態の最終的な排除または抑制に決定的に重要な抗原または抗原上の免疫優性エピトープに関連づけられる。特定の疾患に対する体液性および/または細胞性免疫応答を開始させるためには、宿主免疫系が、その疾病状態に関連した抗原または抗原上の免疫優性エピトープと接触しなければならない。
本明細書中で用いる「標的抗原(TA)」なる語は、感染性病原体(例えば、限定的なものではないが細菌、ウイルス、真菌、酵母、寄生虫、および哺乳類生物種に感染しうる他の微生物)に対するCMI応答を誘導しうる1以上のエピトープを含む目的とする免疫原性ペプチドまたはタンパク質を意味する。標的抗原には、自己抗原、自家抗原、交差反応性抗原、アロ抗体、寛容原、アレルゲン、ハプテン、免疫原またはそれらの一部およびそれらの任意の組合せが含まれうるが、これらに限定されるものではない。したがって、EOIは、本明細書に記載の抗原またはタンパク質の任意のタイプ、あるいは本明細書に記載の特異的抗原またはタンパク質の任意のものに由来しうる。
本明細書中で用いる「エピトープ」なる語は、一般には、T細胞受容体および/または抗体により認識される標的抗原上の部位を意味する。好ましくは、それは、タンパク質抗原に由来するまたはタンパク質抗原の一部としての短いペプチドである。しかし、この用語は、糖ペプチドおよび糖鎖エピトープを有するペプチドをも包含すると意図される。単一の抗原分子はいくつかの異なるエピトープを含みうる。「エピトープ」なる語は、生物全体を認識する応答を刺激する、アミノ酸または糖の修飾配列をも含む。選択されるエピトープが、感染症を引き起こす感染因子(例えば、細菌またはウイルス)のエピトープである場合に、それは有利である。
EOIは、ペプチドまたはポリペプチドのアミノ酸および対応DNA配列の知見から、ならびに個々のアミノ酸の性質(例えば、サイズ、電荷など)およびコドン表から、過度な実験を伴うことなく作製されうる。例えば、Ivan Roitt, Essential Immunology, 1988; Kendrew, 前掲; Janis Kuby, Immunology, 1992 例えば p. 79-81を参照されたい。目的とするタンパク質またはエピトープが応答を刺激するかどうかを判定するためのいくつかの指針には、ペプチドの長さが含まれる。ペプチドは、MHCクラスI複合体に適合するためには少なくとも8または9アミノ酸長であるべきであり、クラスII MHC複合体に適合するためには少なくとも8〜25アミノ酸長、例えば少なくとも13〜25アミノ酸長であるべきである。この長さは、ペプチドがMHC複合体に結合するための最小の長さである。細胞がペプチドを切断する可能性があるため、ペプチドはこれらの長さより長いことが好ましい。ペプチドは、免疫応答を生成するのに十分に高い特異性でそれが種々のクラスIまたはクラスII分子に結合するのを可能にする適当なアンカーモチーフを含有すべきである(Bocchia, M.ら, Specific Binding of Leukemia Oncogene Fusion Protein Pentides to HLA Class I Molecules, Blood 85:2680-2684; Englehard, VH, Structure of peptides associated with class I and class II MHC molecules Ann. Rev. Immunol. 12:181 (1994)を参照されたい)。これは、目的とするタンパク質の配列を、MHC分子と結合したペプチドの報告のある構造と比較することにより、過度の実験を伴うことなく行うことが可能である。したがって、当業者は、タンパク質配列を、タンパク質データベース中に挙げられている配列と比較することにより、目的とするエピトープを確認することが可能である。
本発明の方法またはプロセスにおいては、TAのEOIは1以上のT細胞エピトープを含有しうる。本明細書中で用いる「T細胞エピトープ」なる語は、一般には、T細胞応答を誘導しうる、ペプチド構造体の特徴を意味する。これに関しては、T細胞エピトープは、MHC分子のペプチド結合間隙内で伸長コンホメーションをとる直鎖状ペプチド決定基を含むと当技術分野においてみなされている(Unanueら (1987) Science 236: 551-557)。本明細書中で用いるように、T細胞エピトープは、一般には、少なくとも約3〜5アミノ酸残基、好ましくは少なくとも5〜10またはそれ以上のアミノ酸残基、例えば8〜25アミノ酸残基を有するペプチドである。しかし、本明細書中で用いる「T細胞エピトープ」なる語は、任意のMHCクラスIまたはMHCクラスII拘束性ペプチドを包含する。CMI応答を個々のT細胞エピトープが刺激/増強する能力は、多数のよく知られたアッセイ、例えばリンパ増殖(リンパ球活性化)アッセイ、CD8+T細胞細胞傷害性細胞アッセイにより、あるいは感作被験体におけるエピトープに特異的なTリンパ球に関するアッセイ(例えば、Ericksonら (1993) J. Immunol. 151: 4189-4199; およびDoeら (1994) Eur. J. Immunol. 24: 2369-2376を参照されたい)またはインターフェロンγ産生の測定のためのCD8+T細胞ELISPOTアッセイ(Miyaharaら PNAS(USA) (1998) 95: 3954-3959)により、測定することが可能である。
好ましくは、EOIはCD8+T細胞EOIである。CD8+T細胞誘導性EOIは、宿主被験体へのその投与の後に特異的CD8+T細胞の形成を刺激しうるまたはその活性を増強しうるエピトープである。CD8+T細胞エピトープは、1または2またはそれ以上のエピトープの一つながりの組換え体のような種々の異なる形態で提供されうる。CD8+T細胞エピトープは多種多様な疾患に関して既に同定されており文献中に見出されうる。そのようなCD8+T細胞EOIを含有する任意の選択されたTAに対するCD8+T細胞応答を生成させるために一連のエピトープを設計することが可能である。有利には、NOIにおいては、CD8+T細胞EOIは、不必要な核酸物質が回避されるよう介在配列を含まずに1つに連結された一つながりの複数のEOIとして提供されうる。1つの実施形態においては、NOIは、発現されたタンパク質からのエピトープの切断を可能にするプロテアーゼ切断部位として作用しうる配列をもコードしている。
好ましくは、EOIはヘルパーTリンパ球EOIである。本発明での使用に適したTヘルパー細胞EOIを同定するためには、種々の方法が利用可能である。例えば、ペプチド配列の両親媒性は、Tヘルパー細胞誘導因子としての機能性をもたらすことが公知である。Tヘルパー細胞誘導性エピトープの詳しい考察は、参照により本明細書に組み入れる米国特許第5,128,319号に記載されている。
好ましくは、EOIはCD8+T細胞EOIとB細胞EOIとの混合物である。本明細書中で用いる「B細胞エピトープ」なる語は、一般には、特異的抗体分子が結合するTA上の部位を意味する。抗体応答を惹起しうるエピトープの同定は、当技術分野でよく知られた技術を用いて容易に達成される。例えば、Geysenら (1984) Proc. Natl. Acad. Sci. USA 81: 3998-4002(所与の抗原中の免疫原性エピトープの位置を決定するためにペプチドを迅速に合成するための一般的方法); 米国特許第4,708,871号(抗原のエピトープを同定し化学合成するための方法); およびGeysenら(1986) Molecular Immunology 23: 709-715(所与の抗体に対する高い親和性を有するペプチドを同定するための技術)を参照されたい。
本発明の好ましい実施形態においては、EOIはCD8+T細胞誘導性EOIとTヘルパー細胞誘導性EOIとの混合物である。
TAの複数のEOIをコードするNOIで個体を免疫すると、多くの場合、応答性Tリンパ球の大多数はそのTA由来の1以上の直鎖状EOIに特異的となり、および/または、応答性Bリンパ球の大多数はそのTA由来の1以上の直鎖状または立体構造EOIに特異的となるであろう。したがって、本発明の目的においては、そのようなEOIは「免疫優性エピトープ」と称される。いくつかの免疫優性EOIを有する抗原においては、単独のEOIが、特異的TまたはB細胞応答の統制に関して、最も優性でありうる。
本発明の好ましい実施形態においては、NOIのコード配列は、哺乳類細胞において高発現される遺伝子のコドン使用頻度に類似するように最適化される。DNA暗号は4つの文字(A、T、CおよびG)を有し、これらにより3文字の「コドン」が形成され、これは、生物の遺伝子においてコードされるタンパク質中のアミノ酸を表す。DNA分子に沿ったコドンの直鎖配列は、それらの遺伝子によりコードされるタンパク質中のアミノ酸の直鎖配列に翻訳される。該コードは高度に縮重性であり、61種のコドンが20種の天然アミノ酸をコードしており、3種のコドンが「終止」シグナルを表す。したがって、ほとんどのアミノ酸は2以上のコドンによりコードされており、実際には、いくつかは4以上の異なるコドンによりコードされている。
本発明の方法またはプロセスは、CMI応答の増強を示すためにはアジュバントの存在を要しない。しかし、アジュバントおよび特に遺伝的アジュバントの含有は、CMI応答を更に増強またはモジュレートするのに有用でありうる。アジュバントは、免疫化被験体において共投与した抗原の免疫原性を増強することにより、およびワクチン製品において有益な共投与抗原に対するTh1様免疫応答を誘導することにより、CMI応答を増強しうる。
本発明のEOIは、EOIをコードするヌクレオチド配列として投与することが可能である。本明細書中で用いる目的とするヌクレオチド配列(NOI)なる語は、本発明の方法において使用する1以上のEOIをコードする1以上のNOIを意味する。「目的とするヌクレオチド配列(nucleotide sequence of interest; NOI)」なる語は「ポリヌクレオチド」なる語と同義である。NOIは、ゲノム由来、合成由来または組換え由来のDNAまたはRNAでありうる。NOIは、センス鎖またはアンチセンス鎖またはそれらの組合せにかかわらず、二本鎖または一本鎖でありうる。いくつかの用途には、好ましくは、NOIはDNAである。いくつかの用途には、好ましくは、NOIは組換えDNA技術(例えば、組換えDNA)の使用により製造される。いくつかの用途には、好ましくは、NOIはcDNAである。いくつかの用途には、好ましくは、NOIは、天然に存在する形態と同じでありうる。NOIは、単離または精製された形態(例えば、非細胞形態)でありうる。
本発明の1つの実施形態においては、NOIを宿主被験体に直接投与する。本発明のもう1つの実施形態においては、NOIを含むベクターを宿主被験体に投与する。好ましくは、NOIは、遺伝的ベクターを使用して製造および/または投与される。当技術分野においてはよく知られているとおり、ベクターは、ある実体(entity)を1つの環境から別の環境へ導入するのを可能にしまたは容易にする手段である。本発明においては、本発明のNOIを含むベクターを複製させ、NOIによりコードされる本発明のEOIを発現させるために、例えば、組換えDNA技術において使用するいくつかのベクターが宿主および/または標的細胞内へのDNAセグメント(例えば、異種DNAセグメント、例えば、異種cDNAセグメント)のような実体の導入を可能にする。組換えDNA技術において使用するベクターの具体例には、プラスミド、染色体、人工染色体またはウイルスが含まれるが、これらに限定されるものではない。「ベクター」なる語は発現ベクターおよび/または形質転換ベクターを包含する。「発現ベクター」なる語は、in vivoまたはin vitro/ex vivo発現を可能にする構築物を意味する。「形質転換ベクター」なる語は、1つの種から別の種へ導入されうる構築物を意味する。
本発明のNOIを含むベクターは、「裸核酸構築物」(好ましくは、宿主細胞ゲノムに相同なフランキング配列を更に含むもの)として直接投与することが可能である。本明細書中で用いる「裸DNA」なる語は、本発明のNOIを、その産生を制御する短いプロモーター領域と共に含むプラスミドを意味する。「裸DNA」と称されるのは、そのプラスミドがいかなる送達運搬体にも担持されていないからである。そのようなDNAプラスミドが真核細胞のような宿主細胞に進入すると、それがコードするタンパク質が該細胞内で転写され翻訳される。
あるいは、本発明のNOIを含むベクターは、当技術分野で公知の種々のウイルス技術(例えば、レトロウイルス、単純ヘルペスウイルスおよびアデノウイルスのような組換えウイルスベクターによる感染)を用いて、適当な宿主細胞内に導入することが可能である。該ベクターは組換えウイルスベクターでありうる。適当な組換えウイルスベクターには、アデノウイルスベクター、アデノ随伴ウイルス(AAV)ベクター、ヘルペスウイルスベクター、レトロウイルスベクター、レンチウイルスベクター、バキュロウイルスベクター、ポックスウイルスベクターまたはパルボウイルスベクターが含まれるが、これらに限定されるものではない(Kestlerら 1999 Human Gene Ther 10(10):1619-32を参照されたい)。ウイルスベクターの場合には、NOIの投与は標的細胞のウイルス感染によりもたらされる。
「標的化ベクター」なる語は、細胞に感染またはトランスフェクトまたは形質導入する能力あるいは宿主および/または標的細胞内で発現される能力が宿主被験体内の特定の細胞型(通常は、共通のまたは類似した表現型を有する細胞)に限定されているベクターを意味する。
好ましくは、ベクター内に挿入された本発明のNOIは、宿主細胞によるEOIの発現をもたらしうる制御配列に機能しうる形で連結されている。すなわち、該ベクターは発現ベクターである。宿主細胞により産生された物質は、使用したNOIおよび/またはベクターに応じて、分泌されうるかまたは細胞内に含有されうる。当業者により理解されるとおり、NOIを含有する発現ベクターは、特定の原核細胞膜または真核細胞膜を介したEOIの分泌を導くシグナル配列を伴ったものとして設計されうる。
得られるCMI応答を更に増強および/または強化するために、本発明のNOIは、EOIに融合したアジュバントおよび/または生物応答修飾物質および/または免疫調節物質を含む融合タンパク質として発現されうる。生物応答修飾物質は、CMI応答の普遍的促進をもたらすという意味でアジュバントとして作用しうる。EOIは生物応答修飾物質のアミノまたはカルボキシ末端に結合されうる。
該タンパク質配列はエピトープ配列と同じでありうる(すなわち、NまたはC末端にいずれの付加配列をも伴わない)。該タンパク質は、典型的には、単離または精製された形態(例えば、非細胞形態)である。該タンパク質は、典型的には、8〜400アミノ酸、例えば10〜300または15〜150アミノ酸の長さを有する。
NOIまたはタンパク質は、単独でまたは組成物の一部として、種々の異なる経路により投与することが可能である。特定の組成物には特定の経路が好ましいかもしれない。なぜなら、それは、より有効なCMI応答の生成をもたらしたり、あるいは副作用を誘発する可能性がより低かったり、あるいは投与がより容易であるからである。ワクチン組成物の投与経路は、予防または治療すべき病原体または感染の種類に応じて様々となりうる。
NOIまたはタンパク質製剤を送達するための粒子媒介方法は当技術分野で公知である。したがって、前記NOIを調製し適切に精製したら、それを当技術分野において公知の種々の技術を用いてコア担体粒子上にコーティング(コート化)することが可能である。担体粒子は、遺伝子銃装置からの細胞内送達に典型的に使用される粒径の範囲内で適当な密度を有する物質から選択される。もちろん、最適な担体粒径は標的細胞の直径に左右される。
本明細書中で用いる「宿主哺乳類被験体」は、脊索動物亜門(subphylum cordata)の任意のメンバー、例えば、限定的なものではないがヒトおよび他の霊長類、例えば非ヒト霊長類、例えばチンパンジーおよび他の類人猿およびサル種;農場動物、例えばウシ、ヒツジ、ブタ、ヤギおよびウマ;家庭哺乳動物、例えばイヌおよびネコ;実験用動物、例えばげっ歯類、例えばマウス、ラットおよびモルモット;鳥類、例えば家禽、野生および競技用鳥類、例えばニワトリ、シチメンチョウおよび他の家禽鳥類、アヒル、ガチョウなどを意味する。好ましい動物には、スポーツにおいて用いられる動物、例えば競争馬または障害飛越馬が含まれる。
本発明のこの方法またはプロセスはワクチン接種方法に広く適用可能であり、予防用および/または治療用ワクチン(免疫治療用ワクチンを含む)の開発に重要である。本明細書における治療に対する全ての言及は治療的、緩和的および予防的処置を含むと理解されるべきである。
本発明において宿主被験体に投与されるNOIまたはタンパク質の用量は、やがて該被験体において有益な予防的または治療的CMI応答をもたらすのに十分なものであるべきである。
予防または治療は、複数の時点におけるまたは単独の時点における1回の直接投与により達成されうる。また、投与は単一または複数の部位に対して行われうる。点眼剤による粘膜投与のようないくつかの投与経路はより高い用量を要しうる。当業者は、個々の送達経路に適合するように投与量および濃度を調節することが可能である。好都合にも、実施例は、NOIまたはNOIを含む組成物の1回の投与が、通常は、増強されたCMI応答を達成するのに十分であることを示している。
本発明の方法は増強されたCMI応答を惹起するため、該方法は、ウイルス、細菌、寄生虫または他の感染因子のような病原体による後続の感染を防御するために使用することが可能である。好ましくは、標的抗原は病原体、または感染症、アレルゲンもしくは癌に関連した抗原である。感染症の具体例には、ウイルス、細菌、放線菌および寄生虫による疾患が含まれるが、これらに限定されるものではない。アレルゲンの具体例には、植物花粉、塵埃ダニタンパク質、動物の鱗せつ、唾液および真菌胞子が含まれるが、これらに限定されるものではない。腫瘍関連抗原(TAA)の具体例には、生腫瘍細胞または照射された腫瘍細胞、腫瘍細胞抽出物、および腫瘍抗原のタンパク質サブユニットが含まれるが、これらに限定されるものではない。また、該抗原は、避妊において使用するための精子タンパク質でありうる。いくつかの実施形態においては、該抗原は環境抗原である。環境抗原の具体例には、呼吸器合胞体ウイルス(「RSV」)、インフルエンザウイルスおよび風邪ウイルスが含まれるが、これらに限定されるものではない。粘膜を介して侵入する病原体には、呼吸器合胞体ウイルス、インフルエンザその他による上気道の病的状態を引き起こすもの、および腸感染症を引き起こす因子も含まれる。
1つの好ましい実施形態においては、本発明の方法における、腫瘍関連標的抗原(TAA)由来のEOI(またはエピトープを含むタンパク質)をコードするNOIの使用は、癌治療に対する標的化抗原特異的ワクチンの開発を可能にする。TAA由来のEOIをコードするNOIおよび場合によっては免疫調節分子をコードするNOIの投与は、癌発生のリスクが増加している宿主被験体における予防(予防的免疫化)、初回手術後の疾患再発の予防(抗転移ワクチン接種)の点で、またはin vivoでT細胞数を増大させて散在性腫瘍の根治におけるそれらの有効性を向上させるための手段として(確立した疾患の治療)、特異的に増強されたCMI応答を惹起するための強力な系を与える。さらに、本発明の方法は、腫瘍保持者に再導入する前のex vivoでの細胞の処理(養子免疫療法としても知られる)により、宿主被験体における増強されたCMI応答を惹起するために使用することが可能である。本発明の方法またはプロセスを使用して、黒色腫のような癌のいずれかの証拠が現れる前に宿主被験体にNOIを投与することが可能であり(= 予防的ワクチン接種)、あるいは黒色腫のような癌に罹患した哺乳動物における疾患の退縮をもたらすことが可能である(治療的または免疫治療的ワクチン接種)。
他の態様においては、本発明は活性化T細胞の製造方法に関する。その最も一般的な意味においては、この方法は、T細胞応答の増強の点でCMI応答を増強しうる標的抗原の予め選択されたEOIをコードするNOIを宿主被験体に投与する(好ましくは経皮的に投与する)ことを含む。本発明のこの態様においては、将来の使用のために、宿主のリンパ節からT細胞を回収する。
本発明で使用するタンパク質(タンパク質抗原を含む)、例えばGag、nefおよび/またはRT(NOIによりコードされている)は、天然に存在する形態に対する相同性および/または配列同一性を有しうる。同様に、そのようなタンパク質を発現しうるNOIコード配列は、一般には、天然に存在する配列に対する相同性および/または配列同一性を有するであろう。核酸およびアミノ酸の「配列同一性」を決定するための技術も当技術分野で公知である。典型的には、そのような技術は、遺伝子のmRNAのヌクレオチド配列を決定しおよび/またはそれによりコードされるアミノ酸配列を決定し、これらの配列を第2のヌクレオチドまたはアミノ酸配列と比較することを含む。
本発明の方法の有効性は、(i)本明細書に記載のNOIを宿主被験体(例えば、ヒト被験体または実験動物、例えば、マウス、ラット、ウサギ、モルモット、ヤギ、アカゲザルまたはチンパンジー)に投与し、(ii)ついで該宿主被験体からの血液、脾臓または他のリンパ系組織から細胞を集め、(iii)活性化T細胞(例えば、感染因子の成分を産生する細胞を殺すかまたは細胞溶解させるよう感作されたT細胞)の存在に関して該組織を試験する工程により、試験することが可能である。
もう1つの態様においては、CMI応答の付随的増強を必ずしも惹起することなく、増強された体液性応答を選択的に惹起する方法であって、TAの1以上のEOIをコードするNOIを宿主被験体に少なくとも3回投与することを含み、ここで、各NOI投与間の時間間隔が約48時間であり、さらに、該宿主哺乳動物被験体における該または各発現されたEOIに対する増強された体液性免疫応答をもたらすのに有効なものである方法を、提供する。
本発明は、T細胞媒介免疫障害の予防および/または治療に有用な組成物を提供する。1つの実施形態においては、該組成物は医薬組成物である。もう1つの好ましい実施形態においては、該組成物は免疫治療用組成物である。より一層好ましい実施形態においては、該組成物はワクチン組成物である。該組成物は、前記の本発明のTAのEOIをコードするNOIの治療的または予防的に有効な量を含みうる。該組成物は、製薬上または免疫学的に許容される担体のような担体をも含みうる。製薬上許容される担体または免疫学的に許容される担体は、1つには、投与される個々の組成物によりおよび該組成物を投与するために用いる個々の方法により決定される。したがって、本発明の医薬組成物またはワクチン組成物または免疫治療用組成物の多種多様な適当な製剤が存在する。
NOIまたはタンパク質は医薬組成物または免疫治療用組成物またはワクチン組成物に製剤化されうる。そのような製剤は、製薬上許容される担体(例えば、無菌水または無菌等張食塩水)と組み合わされたNOIまたはタンパク質を含む。そのような製剤は、ボーラス投与または連続投与に適した形態で製造され包装されまたは販売されうる。注射剤は、保存剤を含有する単位剤形で、例えばアンプルまたは複数用量容器中に製造され、包装されまたは販売されうる。製剤には、懸濁剤、水剤(溶液剤)、乳剤(油性または水性ビヒクル中のもの)、ペースト剤および移植可能な徐放性または生分解性製剤が含まれるが、これらに限定されるものではない。そのような製剤は更に、限定的なものではないが懸濁化剤、安定剤または分散剤を包含する1以上の追加的な成分を含みうる。非経口投与用製剤の1つの実施形態においては、その組成物の非経口投与の前に適当なビヒクル(例えば、発熱物質非含有無菌水)で再構成するための乾燥(例えば、散剤または顆粒剤)形態で有効成分を提供する。医薬組成物は、無菌の注射可能な水性または油性の懸濁液または溶液の形態で製造され包装されまたは販売されうる。この懸濁液または溶液は公知技術に従い製造することが可能であり、有効成分に加えて、本明細書中に記載の分散剤、湿潤剤または懸濁化剤のような追加的な成分を含みうる。そのような無菌注射剤は、無毒性の非経口的に許容される希釈剤または溶媒、例えば水または1,3-ブタンジオールを使用して製造されうる。他の許容される希釈剤および溶媒には、リンゲル液、等張食塩水および不揮発性油、例えば合成モノ-またはジ-グリセリドが含まれるが、これらに限定されるものではない。

Associates and Wiley-Interscience (1988, 現在までの全ての改訂を含む)のような文献の全体にわたって記載され説明されており、それらを参照により本明細書に組み入れることとする。本発明の方法は、TAの少なくとも1以上のEOIをコードするNOIを被験体の組織内に直接的にin vivo導入して、該被験体組織の細胞により該EOIを発現させることを含む。
適当な重量の金粒子を1.5mL エッペンドルフチューブ内に直接的に計り取った。ついで約300μLの0.05M スペルミジン溶液を加え、該金を分散させるためにソニケーターを使用して該金を懸濁させた。ついで、当該DNAプラスミドを含有する溶液(約50μL)を該金/スペルミジン溶液に2μg DNA/mg金の濃度で加えた。該DNA溶液は1つのタイプのプラスミドを含有しうる。あるいは、特定の実験では、2以上のプラスミド(例えば、遺伝的アジュバント)を1つに混合した後、該金溶液と混合することが可能である。該DNA/金混合物を穏やかな速度でボルテックスし、300μLの10% CaCl2溶液を、ボルテックスしながら滴下した。該DNA/金粒子を室温で沈殿させ、ついで短時間(10〜15秒間)遠心分離して該金をペレット化した。該ペレットを約800μLのEtOHで3回洗浄した。ついで該DNA/金粒子を、EtOH中に調製された0.03mg/mL PVP(ポリビニルピロリドン)溶液に、3mLのPVP溶液中約1mgのDNA/金にて懸濁させた。ついで、既に記載されているとおりに、この溶液をTefzelチューブ上にコーティングした。例えば、PCT特許出願PCT/US95/00780ならびに米国特許第5,733,600号、第5,780,100号、第5,865,796号および第5,584,807号(それらの開示を参照により本明細書に組み入れることとする)を参照されたい。
プラスミド(PWRG7128構築物)の構築
B型肝炎表面抗原(HBsAg)ベクタープラスミドを以下のとおりに構築した。HbsAgコード領域を作製するために、pAM6構築物(American Type Culture Collection「ATCC」から入手)をNcoIで切断し、マング・ビーンヌクレアーゼで処理してX抗原の開始コドンを除去した。ついで、得られたDNAをBamHIで切断し、T4 DNAポリメラーゼで処理して該DNAを平滑末端化し、HBsAg発現カセットを得た。該HBsAg発現カセットは1.2kBの断片として存在する。完全長ヒトCMV(Towne株)極初期プロモーター(エンハンサーを伴う)を含有するプラスミド構築物pPJV7077(Schmaljohnら (1997) J. Virol. 71:9563-9569)をHindIIIおよびBglIIで切断し、ついでT4 DNAポリメラーゼおよび子ウシアルカリホスファターゼで処理して平滑末端化DNAを得、該HBsAg発現カセットを該プラスミド内に連結してpWRG7128構築物を得た。
ELISAアッセイを用いて、個々のマウスの血清サンプルを、HBsAgに特異的な抗体に関して試験した。ELISAのために、Falcon Pro Bindマイクロタイタープレートを、PBS(リン酸緩衝食塩水, BioWhittaker)中の0.1μg/ウェルの精製HBsAg(BioDesign)で4℃で一晩コーティングした。該プレートを5% ドライミルク/PBSで室温(RT)で一晩ブロッキングし、ついで洗浄バッファー(10 mM Tris緩衝食塩水, 0.1% Brij-35)で3回洗浄した。希釈バッファー(2% ドライミルク/PBS/0.05% Tween 20)で希釈された血清サンプルを該プレートに加え、室温で2時間インキュベートした。該プレートを3回洗浄し、希釈バッファー中で1:8000希釈されたビオチン化ヤギ抗マウス抗体(Southern Biotechnology)を該プレートに加え、室温で1時間インキュベートした。インキュベーション後、プレートを3回洗浄し、ついで、PBSで1:8000希釈されたストレプトアビジン-ホースラディッシュペルオキシダーゼコンジュゲート(Southern Biotechnology)を加え、該プレートを室温で更に1時間インキュベートした。さらに3回の洗浄後、プレートを3回洗浄し、ついでTMB基質溶液(BioRad)を加え、30分後に反応を1N H2SO4で停止させた。光学濃度を450nmで読み取った。該サンプルと既知力価の標準物との比較により、終点力価を計算した。
HIV-1 GP120抗原をコードするプラスミドの構築
HIV-1 gp120をコードするプラスミドベクターを以下のとおりに構築した。該ベクターは、Bluescript(Stratagene, La Jolla, CA)プラスミドバックボーン、ヒトサイトメガロウイルス(hCMV)極初期プロモーター(Fullerら (1994) Aids Res. Hum Retroviruses 10:1433)およびSV40ウイルス後期ポリアデニル化部位から構築した。hCMVプロモーターは、極初期転写開始部位の522bp上流から96bp下流までの619塩基対(bp)のAccII断片内に含有されている。SV40ウイルス後期ポリアデニル化配列は、pSV2dhfr(もともとBethesda Research Laboratories (カタログ番号5369 SS) から入手可能であった)から誘導される約800bpのBamHI-BglII断片内に含有されている。まず、「pC-Env」と称されるHIV-1 gp160をコードするプラスミドを構築した。このプラスミドは、成熟gp160アミノ末端の4位アミノ酸をコードする配列から開始する、LAV-1BRU(ATCC受託番号53069, GenBankアクセッション番号K02013)からの2565bpのKpnI-XhoI断片を含有する。該envコード配列断片を、単純ヘルペスウイルス糖タンパク質D(gD)シグナルペプチドと成熟gDアミノ末端の非アミノ酸とをコードする160bpの合成断片のすぐ下流に配置し、既に記載されているとおりに(Fullerら (1994) Aids Res. Hum Retroviruses 10:1433)、該合成断片にインフレームで融合させた。
第5週および第6.5週(それぞれ初回免疫後および追加免疫後)に集めた試料にELISAアッセイを用いて、HIV gp120抗原に対する血清抗体応答を試験した。ELISAのために、Costar高結合性EIAプレートを、4℃で一晩のインキュベーションにより、50μlのPBS中0.3μg/ウェルの組換えHIV p120(Intracel)でコーティングした。プレートを3回洗浄し、PBS中の2% BSAで室温で2時間ブロッキングした。血清の系列希釈物を該コート化プレートに加え、37℃で1時間インキュベートした。洗浄後、該プレートをアルカリホスファターゼ結合ヤギ抗マウスIgG(H+L)(BioRad)の1:1500希釈物と共にインキュベートし、ついでp-ニトロフェニルホスフェート(PNPP)(BioRad)で発色させ、405nmでODを読み取った。
HSV-2 ICP27をコードするプラスミドの構築
ICP27をコードするDNAワクチンを構築し、ついで本アジュバントプラスミドベクターの種々の組合せと一緒にしてワクチン組成物を得た。免疫化後、免疫化動物をHSV-2ウイルスでチャレンジし、種々のワクチン組成物の防御効果を判定した。
pPJV7630の抗原遺伝子の起源は、コスミド23と称されるコスミドベクター内にクローニングされたHSV-2株MSのゲノム断片である。コスミド23は、公開配列(HG52株)に基づけばヌクレオチド110,931-147,530に及ぶHSV-2からの3個のEcoRI断片から構成されていた。コスミド23をEcoRIで部分消化し、再連結し、約28,000bpの断片(110,931-139,697)のみを有する構築物を選択した。この分子をOP23と命名した。OP23内の配列を改変するために、この分子に6つの修飾を施した。これらは、HSV-2配列からの非極初期遺伝子およびバックボーンDNAを除去するように設計した。1つの最終的な修飾は、該バックボーン配列を、臨床用の適当な抗生物質耐性遺伝子で置換することであった。この工程を以下に説明する。
マウス
単細胞浮遊液をマウス脾臓から得た。脾臓をメッシュを通して搾り出して単細胞浮遊液を得、ついで細胞を沈降させ、ACKバッファー(Bio Whittaker, Walkersville MD)で処理して赤血球を細胞溶解した。ついで該細胞を、HEPES、1% グルタミン(Bio Whittaker)および5% 熱不活化ウシ胎児血清(FCS, Harlan, Indianapolis IN)を補充したRPMI 1640培地内で2回洗浄した。細胞を計数し、5% 熱不活化FCS、50μM メルカプトエタノール(Gibco-BRL, Long Island NY)、ゲンタマイシン(Gibco-BRL)、1mM MEM ピルビン酸ナトリウム(Gibco-BRL)およびMEM非必須アミノ酸(Sigma, St. Louis MO)を補充した、HEPESおよび1% グルタミンを含有するRPMI 1640よりなる「全」培地内に、適当な濃度となるよう再懸濁させた。CD8特異的アッセイのために、既知CD8エピトープに対応するペプチドの存在下、細胞をin vitroで培養した。BALB/CマウスにおけるICP27の場合には、該ペプチドの配列はHGPSLYRTF(QCB Inc)であった。ペプチドをDMSO中に調製し(10mg/ml)、培地中に10μg/mlに希釈した。
マウス
種々の免疫化スケジュールの、HSV-2での致死性チャレンジからマウスを防御する能力を試験するために、感染チャレンジモデルを使用した。チャレンジ前にマウスを免疫化し、感染のために、該マウスを麻酔し、30μLのPBS中の致死量のHSV-2を該マウスの鼻腔内に投与した。感染後の20日間にわたってマウスを追跡し、病状および死亡率に関して得点化した。
家畜ブタ
末梢血単核細胞(PBMC)を単離するために、全血を、Histopaque-1077クッションを通して室温で遠心し(3000rpmで30分間)、その勾配からバンドとしてPBMCを回収した。PBMCを全培地中で3回洗浄し、25mLの全培地に再懸濁させて計数し、全培地内に1×107細胞/mlとなるよう再懸濁させた。ELISPOTアッセイを、マウスに関して記載されているとおりに行った。ただし、この場合には、抗原は、HSV-2抗原の重複ペプチドライブラリーから誘導されたペプチドのプールであり、検出には、家畜ブタIFN-γに特異的な抗IFN抗体ペア(R&D systems)を使用した。
家畜ブタ
家畜ブタにおける免疫応答を調べるために用いたもう1つのアッセイは遅延型過敏症反応(DTH)アッセイであった。このアッセイでは、それぞれXR1装置および針注射を使用してブタの皮膚に送達される抗原源としてDNAプラスミドおよびタンパク質抽出物の両方を使用した。投与の48時間後、DTH反応の指標として、該抗原の送達部位の周辺の発赤(紅斑)の面積を測定した。皮膚内への抗原送達は、免疫化の完了の7日後に行った。
マウスを免疫化するために、NOIを含む2つの異なるプラスミドを使用した。これらは、HSV-2臨床ワクチン複数遺伝子プラスミド(PJV7630)、およびHCMVプロモーターに機能しうる形で連結されたICP27 NOIを含む単一遺伝子プラスミドであった。ICP27 NOIは、PJV7630内に見出される優性エピトープをコードしており、グラフは、このタンパク質に特異的なCD8応答を表す。
NOIを、1週間に1、2または4回、すなわちそれぞれ第7日(D 7)、第0および7日(D 0, 7)または第0、2、4および7日(D 0, 2, 4, 7)に、投与した。NOIの1投与当たり1回または2回の適用を行い、1つの群においては、LTアジュバントをNOIと共に投与した。
ICP27遺伝子は、PJV7630内に見出される優性抗原であり、図1Aおよび1Bは、この標的抗原に特異的なCD8応答を表す。典型的には、プラスミドPJV7630(図1B)およびICP27(図1A)は共に、1回のみのNOI投与の後には細胞100万個当たり500 ELISPOTを与え、2回のNOI投与の後には細胞100万個当たり1500 ELISPOTを与えている。2回のNOI投与をLT遺伝的アジュバントと共に行った場合には、約3500 ELISPOTが認められる。4回のNOI投与の後には、LTアジュバントの非存在下でさえも、細胞100万個当たり3500を超えるELISPOTが得られたが、LT遺伝的アジュバントはまた、該応答を増強するであろう。図1Aにおいては、NOIのLT共投与における応答は、その結果が尺度外(off-scale)であったため、測定されなかった。
PJV7630で免疫化したマウスにおいて測定したICP27特異的CD8 IFN-γ ELISPOTに基づけば、集積化(clustering)NOI投与は細胞性免疫応答の増強をもたらすことが判明した。
以下の3つの実験の目的は、単一遺伝子プラスミドの集積化(clustered)NOI投与により生じたCMI応答の増強が致死性チャレンジからの防御の増強と相関するかどうかを評価することにあった。
プラスミドPJV7630を1週間にわたってマウスに投与した。1回(第7日)、2回(第0および7日)または4回(第0、2、4および7日)のいずれかのNOI投与を1週間かけて行った。1つの群では各免疫化において2用量のPJV7630を投与したが、大部分のマウスには各免疫化において単回用量のPJV7630を投与した。
グラフには、「投与回数×投与当たりの用量数」が表示されており、4×1は、各投与において単回用量で4回のNOI投与がなされた動物を表す。1週間のNOI投与の後、該動物を1週間または2週間休ませ、ついで該動物にウイルスを感染させた。ウイルスの用量はLD50の約5倍であった。
4回の投与×1用量/投与の1週間内の集積化(clustering)は、単回投与よりも、または1投与当たりより多用量を用いた場合よりも、より強力な強防御性CMI応答を迅速に生成する。
この実験の目的は、単一遺伝子プラスミドのDNA投与間の時間間隔を様々に変化させることにあった。
各投与の投与日および投与間の時間間隔が異なる合計4回の投与をすべてのマウスに行った。6、4、2、1または0日の間隔での投与をマウスに与えた(すなわち、0日の場合は、1日に4回の投与を行った)。各スケジュールの最終投与は、同じ暦日に行い、最終投与の7日後にすべての動物を犠死させた。
図3Aは、ICP27単一遺伝子プラスミドの集積化投与間の0、1、2、4および6日の時間間隔に関するCD8 ELISPOTの結果を示す。この結果は、投与間の時間間隔の増加がCD8 ELISPOTの結果を向上させることを示しており、NOI投与間で4日(すなわち、96時間)の間隔を与えた場合に、最大の結果が得られたことを示している。
マウスにおける集積化NOI投与間の時間間隔に関する試験から、4回のNOI投与を行う場合には、NOI投与を4または6日間隔で行う場合にCD8+T細胞応答に関する最適応答が得られることが、示された。
この実験の目的は、複数遺伝子プラスミド(PJV7630)の集積化NOI投与に対するCD8+T細胞応答に関するCMI応答を評価することにあった。
合計4回のNOI投与を動物に行ったが、但しそのNOI投与間の時間間隔は様々に変化させた。各群における最終投与は同一日に行い(NOI投与の開始時は様々であった)、該投与の完了の1週間後および3週間後に応答を測定した。この3週サンプリングを加えたのは、その異なるスケジュールが投与のタイミングに及ぼした影響を最小にするためであった。例えば、6日の投与間隔で4回のNOI投与を受けた動物は第1の投与と4回目の投与との間に18日の時間間隔を有し、投与間に0の時間間隔を有する動物は第18日にすべての投与を受けた。
図4Aおよび4Bの結果は、ICP27特異的CD8 IFN-γ ELISPOTにより測定した細胞性応答を示している。NOI投与間の時間がグラフに表示されている。すべての動物に合計4回の投与を行った。複数遺伝子プラスミド(PJV7630)の結果は、単一遺伝子プラスミド(ICP27)を使用した初期実験の結果とよく似ていることが明らかである。これに関しては、投与間の4日および6日の時間間隔がELISPOT測定において最適応答を与え(図3Cを参照されたい)、これは3週間の時点では維持されたが、応答は、その時点までに約3倍低下していた(図3Dを参照されたい)。
この実験の目的は、HIV-1gp120のような比較的弱い抗原に対する体液性および細胞性免疫(CMI)応答の増強において、遺伝的アジュバントの使用と集積化NOI投与スケジュールとの間に相乗作用が存在するかどうかを判定することにあった。
この実験では、マウスにおけるgp120抗原の少なくとも2つの投与を含み、ここで、その各「投与」は、集積化した1〜4 XRIの投与から構成される。後述の図5Aの概要図を参照されたい。また、投与間の休止期間は1週間である。LT A+B遺伝的アジュバント(pPJV2012)を使用した。pPJV2012プラスミドの地図を図10に示す。pPJV2012プラスミドは、LTAおよびLTBサブユニットタンパク質をコードするLT遺伝子をWO 03/004055に記載のプラスミドpPJV-2004およびpPJV-2005のそれぞれにクローニングすることにより調製した。ついで、LTAおよびLTBサブユニットタンパク質をコードする遺伝子を元のプラスミドから切り出し、単一プラスミド内に挿入して、pPJV2012プラスミドを得た。
図5Bは、NOI投与間の時間間隔が7日である動物群から得たデータを示す。各集積化におけるXR1投与の回数を、LT A+B遺伝的アジュバント(pPJV2012)の存在または非存在と同様に、様々に変化させた。得られた結果は、遺伝的アジュバントの存在または非存在が細胞応答(IFN-γ産生)に対し最も強い影響を及ぼしたことを示している。NOI投与スケジュールが2回の投与に集積化された場合に、LTにより増強される応答が最強であったことを、図5Bは明らかに示している。これらの結果は、集積化投与スケジュールが、遺伝的LTアジュバントにより得られる全体的な細胞性応答に大きく寄与することを示している。
これらの結果は、弱い抗原をコードするNOIをアジュバントをコードするNOIと共に投与した場合には、2回のNOI投与の後に、インターフェロンγ放出に関して最大CMI応答が得られることを、示している。これとは対照的に、約48時間の投与間隔を伴う集積化された4回のNOI投与によれば、アジュバントを用いても用いなくても、強力な体液性免疫応答が得られる。
この実験の目的は、pPJV7630での家畜ブタの免疫化が、IFN-γ ELISPOTおよびDTH応答により判定した場合に細胞性免疫応答を生成するかどうかを判定することにあった。
この実験のために、pPJV7630またはプラセボ(金のみ)をXR1装置により家畜ブタに投与した。ブタに、各免疫化当たり2用量を投与し、4回の免疫化の集積化スケジュールを1週間かけて施した。したがって、各ブタには1集積化(cluster)において合計8用量のワクチンを投与した。第2の集積免疫化を最初の集積化の最後から28日後に開始した。
図6Aは、最初の集積免疫化後の動物から得たIFN-γ ELISPOTデータを示す。値は8頭の免疫化動物および8頭の対照動物からの平均である。このデータは、免疫原性を測定するのが困難なモデルであると考えられている家畜ブタにおいて集積免疫化スケジュールが細胞性免疫応答を生成し得たことを示している。対照ブタはすべて、ELISPOTのバックグラウンドレベルを示した。
家畜ブタモデルにおいて、集積免疫化は該ワクチン抗原に対する細胞性免疫応答を誘導しうることが判明した。家畜ブタは、免疫応答惹起の良好なモデルであるとは考えられていないが、該集積免疫化は、細胞性免疫応答を惹起する能力を有していた。
以下の表1に詳しく示すとおり、pPJV1671から発現されるhaタンパク質に対する抗体応答に対する用量および装置の効果を調べるために、家畜ブタでの研究を行った(XR粒子加速装置は前記で説明されている)。
リン酸緩衝食塩水中に希釈された200赤血球凝集素単位/ウェルのサルコシル(Sarkosyl)破壊精製Sw/INウイルスを使用して、標準的な方法に従うELISAにより、抗体力価を測定した。ブタ抗体は、ヤギ抗ブタ免疫グロブリンGアルカリホスファターゼコンジュゲートを使用することにより直接的に測定した。
プラスミドpPJV1671は、図8に示すとおり、インフルエンザA/パナマ/2007/99(H3N2)の赤血球凝集素(HA)抗原をコードするヒトDNAワクチンベクターである。HAコード配列は、鋳型RNA源としてCDCから得たA/パナマ/2007/99ウイルスのサンプルを使用する標準的な逆転写酵素/ポリメラーゼ連鎖反応(RT-PCR)クローニング技術により得た。最終的なpPJV1671 HA DNAワクチンベクターの作製においては、以下の工程を用いた。
この研究は、集積免疫化が家畜ブタモデルにおける抗体応答を有意に向上させたことを示している。

pWRG7074内のウシ成長ホルモンポリアデニル化シグナル(BGHpA)をウサギβ-グロブリンポリアデニル化シグナル(RBGpA)により置換してpWRG7284を得た。CMVプロモーターおよびエキソン1/2融合体を含有するpWRG7128由来PCR断片によりCMV配列全てを置換することにより、CMVのイントロンAをpWRG7284から除去した。これによりpWRG7293を得た。
標準的プラスミドバックボーンpWRG7074を作製した。このバックボーンは、いくつかの臨床試験で使用されるHBsAg発現ベクターであるpWRG7128を作製するための前駆体プラスミドとして使用した。この節においては、このバックボーンの誘導を、図15および16(それぞれ「プラスミドPJV7074およびPJV7128の誘導を示す流れ図」および「主要プラスミド特徴マップ」)にて説明し、図示する。簡潔に説明すると、ヒトCMV極初期プロモーターおよびウシ成長ホルモンポリアデニル化配列を含有する単一断片を、標準的な良く特徴づけられているpUC19細菌プラスミドベクター内に挿入することにより、pWRG7074を誘導した。いくつかの後続の操作を行って、アンピシリンをカナマイシン(KanR)耐性マーカーにより置換し、いくつかの制限部位を改変した。CMVプロモーターおよびbGHポリA領域の起源であるDNA断片は、プラスミドpJW4303(当時はスタンフォード大学にいたJim Mullinsから供与されたもの)から得た。
ATCCから得たHPV16ゲノムクローンから、PCRにより、E6およびE7のコピーを調製した。該完全長プラスミドは完全ウイルスゲノムを含有するため、それをBSL-2条件下で維持する。それぞれp53およびRbの結合領域を欠失させることにより、E6およびE7を無毒化した(Slebosら, Virol. 1995, 208, 111-120; およびSmahelら, Virol. 2001, 281, 231-238)。PCR断片をPJV7563内にクローニングして、イントロンAを伴わないCMVプロモーターの制御下に該遺伝子を配置した。
E6またはE7 DNAでのB6マウスの免疫化は、有意な数のIFN分泌細胞の誘導を引き起こす(図23)。E6の場合には、CD8特異的エピトープのみが公知である。E7の場合には、CD4特異的エピトープおよびCD8特異的エピトープの両方が同定されており、両方に対する強力な応答がPMED免疫化後に見られる。E6またはE7ワクチンへの大腸菌(E. coli)易熱性毒素(LT)またはコレラ毒素(CT)DNAの添加を試験した。LTはE7ペプチドに対するIFN ELISPOT応答を約2倍増強したが(図23)、いずれの毒素においても腫瘍防御における増強は全く見られなかった。
ICP27単一遺伝子プラスミドを使用して、各群の最終投与が同一日に行われるように、2日間隔の1〜8回の投与をマウスに行った。最終投与の2週間後に、CD8 ELISPOTを測定した。結果を図29に示す。最大に近い効果を得るためには少なくとも3回の投与が必要であり、追加的な投与は改善をほとんどまたは全くもたらさないことを、この結果は示している。
追加免疫ワクチン接種の効果を調べるための実験を行った。初回免疫投与を受けたマウスを、初回免疫投与および追加免疫投与の両方を受けたマウスと比較した。両群のマウスを同時に初回免疫し、同じワクチンをワクチン接種に使用した。第2群には、初回免疫の28日後に追加免疫を行った。
PJV7630をXR1装置により家畜ブタに投与した。各免疫化当たり2用量をブタに投与し、1週間に及ぶ4回の免疫化の集積化スケジュールを行った。すなわち、各ブタには集積化毎に合計8用量のワクチンを投与した。前回の集積化の最後から21日後に、2回の集積免疫化追加免疫を開始した。投与(PB)の開始前および各追加免疫(B1およびB2)の7〜10日後に、血液サンプルを採取した。記載されているとおりにIFN-γ ELISPOTアッセイを行った。数値は10匹の動物に対する平均±SEMである。図32は、得られた結果および追加免疫ワクチン接種の効果を示す。
XR-1によるpPJV7630の1回投与のPMEDの2、3および4日後に採取した皮膚サンプルを凍結し、すり砕き、上清内のサイトカインレベルを試験した。炎症性サイトカインを評価するためにCBAキットを使用して、IL-6、TNF-αおよびMCP-1(単球走化性タンパク質)の著しい増加を見出した。これらは第2日に最高となり、時間経過と共に減少した。いずれの時点においてもIL-10、IL-12またはIFN-γの増加は見出されなかった。サイトカインの増加は一般には創傷治癒において見出される。
記載されている実験に、Balb/cマウスを使用した。腹部領域を剃毛し、粒子媒介免疫治療送達(particle mediated immunotherapeutic delivery (PMID))を用いてDNAの初回免疫化を行った。各動物に合計3μgのDNAを投与した。これは、第0日または第4日の通常の「パルス(pulse)」免疫化(3×1μgのDNA)により、あるいは隔日(第0、2および4日)のそれぞれの日に投与される1μgのDNAでの「集積(cluster)」免疫化により行った。最初のDNA投与の10日後にマウスを選択し、脾臓を集めた。脾臓細胞を薄くそぐことにより脾細胞を回収し、赤血球を細胞溶解した。該脾細胞を洗浄し計数した。専用ELIspotプレート(インターフェロンγ捕捉抗体でコーティングされブロッキングされたもの)を使用した。脾細胞をこれらのプレートに移し、特異的ペプチドの存在下、37℃/5% CO2で一晩インキュベートした。該脾細胞を細胞溶解し、存在するインターフェロンγ分泌細胞の数を示すために標準的な方法により該プレートを現像した。
結果(図33に示す)は、「集積」免疫化を受けた動物からは、通常の「パルス」法を用いて同量のDNAが投与された動物と比べて有意に多い数のIFN-γスポット形成細胞が単離されたことを示した(*は有意差を示す)。
記載されている実験に、Balb/cマウスを使用した。腹部領域を剃毛し、粒子媒介免疫治療送達 (PMID)を用いてDNAの初回免疫化を行った。各動物に合計1μgのDNAを投与した。これは、第0日の通常の「パルス(pulse)」免疫化(2×0.5μgのDNA)により、あるいは第0および7日のそれぞれの日に投与される0.5μgのDNAでの「改変集積(modified cluster)」免疫化により行った。初回免疫化の83日後に1.0μgのDNAの「パルス」を用いて、すべてのマウスを追加免疫した。該追加免疫の7日後(第90日)にマウスを選択し、脾臓を集めた。脾臓細胞を薄くそぐことにより脾細胞を回収し、赤血球を細胞溶解した。該脾細胞を洗浄し計数した。専用ELIspotプレート(インターフェロンγ捕捉抗体でコーティングされブロッキングされたもの)を使用した。脾細胞をこれらのプレートに移し、特異的ペプチドの存在下、37℃/5% CO2で一晩インキュベートした。該脾細胞を細胞溶解し、存在するインターフェロンγ分泌細胞の数を示すために標準的な方法により該プレートを現像した。
結果(図34に示す)は、「改変集積化(modified cluster)」を用いて免疫化された動物からは、通常の「パルス」法を用いて同量のDNAが投与された動物と比べて多い数のIFN-γスポット形成細胞が単離されたことを示した。したがって、HIV由来のGagおよびRT抗原を発現する構築物で「改変集積化」法により免疫化されたマウスの細胞性免疫応答は、通常の「パルス」法により同量のDNAで免疫化された動物のものより有意に高かった。
使用したプラスミドは、HIV抗原であるRT、NefおよびGagを発現した。PMIDのためのカートリッジの調製は、既に記載されているとおり(Eisenbraunら, DNA and Cell Biology, 1993 Vol 12 No 9 pp 791-797; Pertnerら)に行った。簡潔に説明すると、プラスミドDNAを2μm金粒子(DeGussa Corp., South Plainfield, N.J., USA)上にコーティングし、Tefzelチューブ内に充填し、ついでそれを、カートリッジとして使用するために1.27cmの長さに切断し、使用するまで4℃で乾燥保存した。典型的なワクチン接種においては、各カートリッジは、約1μgのDNAでコーティングされた0.5mgの金を含有していた。
結果を図35に示す。初回免疫化後、集積免疫化により初回免疫されたミニブタからのPBMC中のIFN-γ産生スポットの数は、パルス免疫化を受けたものと比べて有意に多かった(それぞれ平均±SEMで311±96および45±31; p<0.05スチューデントt検定)。さらに、パルス追加免疫後のIFN-γ産生スポットの数は、集積化初回免疫を受けた動物においては、パルス初回免疫の場合と比べて有意に多かった(それぞれ平均±SEMで431±60および186±66; p<0.05スチューデントt検定)。要約すると、これらの結果は、集積化初回免疫が通常のパルス初回免疫を上回る利点をもたらすこと、およびこの利点が、免疫応答のその後の追加免疫段階でも維持されることを示している。
記載されている実験に、C57BL/6マウスを使用した。腹部領域を剃毛し、粒子媒介免疫治療送達(PMID)を用いてDNAの初回免疫化を行った。第0日の通常の「パルス」免疫化(1μgのDNAオボアルブミン)により、あるいは隔日(第0、2日)-集積化2× または第0、2および4日-集積化3×、のそれぞれの日に投与される1μgのDNAでの「集積(cluster)」免疫化により、各動物の免疫化を行った。最初のDNA投与の10日後にマウスを選択し、脾臓を集めた。脾臓細胞を薄くそぐことにより脾細胞を回収し、赤血球を細胞溶解した。該脾細胞を洗浄し計数した。専用ELIspotプレート(インターフェロンγまたはIL2捕捉抗体でコーティングされブロッキングされたもの)を使用した。脾細胞をこれらのプレートに移し、特異的ペプチドの存在下、37℃/5% CO2で一晩インキュベートした。既に明らかにされているオボアルブミン特異的CD4およびCD8ペプチドを使用した。脾細胞を細胞溶解し、存在するインターフェロンγまたはIL2分泌細胞の数を示すために標準的な方法により該プレートを現像した。
結果(図36および37に示す)は、「集積」免疫化を受けた動物からは、通常の「パルス」法を用いて同量のDNAが投与された動物と比べて有意に多い数のIFN-γおよびIL2スポット形成細胞が単離されたことを示した。図36においては、各棒グラフは個々のマウスでの応答を表す。
記載されている実験に、C57BL/6マウスを使用した。腹部領域を剃毛し、粒子媒介免疫治療送達(PMID)を用いてDNAの初回免疫化を行った。各動物に合計3μgのDNAを投与した。これは、第0日の通常の「パルス」免疫化(3×1μgのDNA)により、あるいは隔日(第0、2および4日)のそれぞれの日に投与される1μgのDNAでの「集積(cluster)」免疫化により投与された。
図38に示す結果は、集積免疫化により初回免疫された動物が、パルス免疫化により同量のDNAで免疫化されたマウスより強力な回復(recall)応答を示したことを示している。これは、ELISPOTにより、IFNgおよびIL2産生細胞の度数の増加により示された。また、集積免疫化により初回免疫された動物は、パルスDNA追加免疫の後にパルス免疫化により免疫化された動物と比べてより強力なCTL応答を示した。
Claims (28)
- 宿主哺乳類被験体におけるT細胞エピトープに対するT細胞応答を惹起する方法であって、
(i)該T細胞エピトープをコードする目的とするヌクレオチド(NOI)を投与することを各投与が含む、該被験体への1〜14日間隔の少なくとも2回の投与を含む第1の免疫化、および所望により、
(ii)該被験体への(a)該T細胞エピトープをコードするNOIまたは(b)該T細胞エピトープを含むタンパク質の少なくとも1回の投与を含む第2の免疫化、
を含み、かつ、第1の免疫化の第1の投与と第2の免疫化の第1の投与との間が21〜365日間である、方法。 - 第1のおよび/または第2の免疫化の投与を2〜12日間にわたって行う、請求項1記載の方法。
- 第1のおよび/または第2の免疫化において、前記NOIまたはタンパク質を2〜10回投与する、請求項1または2記載の方法。
- 第1のおよび/または第2の免疫化の2、3、4回またはそれ以上の投与が2〜6日間隔である、請求項1〜3のいずれか1項記載の方法。
- 第1の免疫化の第1の投与と第2の免疫化の第1の投与との間が50〜250日間である、請求項1〜4のいずれか1項記載の方法。
- 被験体への(a)前記T細胞エピトープをコードするNOIまたは(b)前記T細胞エピトープを含むタンパク質、の少なくとも1回の投与を含む第3の免疫化を更に含み、かつ、第2の免疫化の第1の投与と第3の免疫化の第1の投与との間が10〜365日間である、請求項1〜5のいずれか1項記載の方法。
- 第3の免疫化において、
・前記NOIまたはタンパク質を2〜5回投与する、および/または
・それらの投与が2〜6日間隔である、および/または
・第2の免疫化の第1の投与と第3の免疫化の第1の投与との間が50〜250日間である、
請求項6記載の方法。 - 前記NOIが、DNA配列を、被験体の細胞内で該DNA配列の発現を誘導しうる調節配列の制御下に含む、請求項1〜7のいずれか1項記載の方法。
- T細胞エピトープがCD4+ヘルパーTリンパ球細胞エピトープおよび/またはCD8+Tリンパ球(CTL)エピトープである、請求項1〜8のいずれか1項記載の方法。
- 1回またはそれ以上のNOIの投与が0.1〜2μgのNOIの投与を含む、請求項1〜9のいずれか1項記載の方法。
- 1回またはそれ以上の投与が皮膚への投与を含む、請求項1〜10のいずれか1項記載の方法。
- 少なくとも1回のNOIまたはタンパク質の投与に関して、該NOIまたはタンパク質が粒子上にコーティングされているかまたは粒子内に取り込まれている、請求項1〜12のいずれか1項記載の方法。
- 粒子を粒子加速装置により被験体に投与する、請求項12記載の方法。
- 少なくとも1回のNOIまたはタンパク質の投与に関して、該NOIまたはタンパク質を、
(i)製薬上許容される担体、賦形剤もしくは希釈剤を含む医薬組成物、または
(ii)免疫学的に許容される担体、賦形剤もしくは希釈剤を含むワクチン組成物、または
(iii)免疫学的に許容される担体、賦形剤もしくは希釈剤を含む免疫治療用組成物
として投与する、請求項1〜13のいずれか1項記載の方法。 - 前記NOIもしくはタンパク質を、アジュバントと共に、もしくは被験体の細胞内でアジュバントを発現しうるポリヌクレオチドと共に投与する、請求項1〜13のいずれか1項記載の方法、または、前記組成物が更にアジュバントを含むかもしくは被験体の細胞内でアジュバントを発現しうるポリヌクレオチドを含む、請求項14記載の方法。
- 前記アジュバントが大腸菌(E. coli)熱不安定性エンテロトキシン(LT)またはビブリオ・コレレ(Vibrio cholerae)コレラ毒素(CT)の非毒性形態である、請求項15記載の方法。
- 前記アジュバントがLTエンテロトキシンのBサブユニット(LTB)またはCTコレラ毒素のBサブユニット(CTB)である、請求項15記載の方法。
- T細胞エピトープが病原体または癌細胞に由来するものである、請求項1〜17のいずれか1項記載の方法。
- T細胞エピトープがHSV、HIVまたはHPVに由来するものである、請求項1〜18のいずれか1項記載の方法。
- 被験体における疾患を予防または治療するために実施する、請求項1〜19のいずれか1項記載の方法。
- 前記NOIが少なくとも2つのHSV、HIVまたはHPV抗原をコードしている、請求項1〜20のいずれか1項記載の方法。
- 前記NOIが、HIV-1 gagタンパク質またはそのgagエピトープを含有する断片、および第2のHIV抗原または該第2のHIV抗原のエピトープをコードする断片をコードしている、請求項21記載の方法。
- 前記第2の抗原が、Nef、RT、またはNefもしくはRTのエピトープを含有する断片よりなる群から選ばれる、請求項22記載の方法。
- 前記NOIが、
・Gag(p17,p24)、Nef末端切断型、
・Gag(p17,p24)(コドン最適化)、Nef(末端切断型)、
・Gag(p17,p24)、RT、Nef(末端切断型)、
・Gag(p17,p24)、コドン最適化RT、Nef(末端切断型)、
・Gag(p17,p24)、コドン最適化RT、コドン最適化Nef末端切断型;
および/または、任意に、アイオワ長(Iowa length)HCMVプロモーター+エキソン1の下流でウサギグロビンポリアデニル化シグナルの上流に、機能しうる形で連結された不活化コドン最適化RT、末端切断型Nef、およびコドン最適化gag遺伝子のp17/p24部分、
よりなる群から選ばれる抗原の組合せをコードしている、請求項22記載の方法。 - 同じエピトープをそれぞれがコードしている少なくとも2つの異なるNOIを投与するか、および/または同じエピトープを含む少なくとも2つの異なるタンパク質を投与する、請求項1〜24のいずれか1項記載の方法。
- T細胞応答の惹起方法の有効性を試験するためのアッセイであって、該方法が
(i)T細胞エピトープをコードする目的とするヌクレオチド(NOI)を投与することを各投与が含む、被験体への1〜14日間隔の少なくとも2回の投与を含む第1の免疫化、および所望により、
(ii)該被験体への(a)該T細胞エピトープをコードするNOIまたは(b)該T細胞エピトープを含むタンパク質の少なくとも1回の投与を含む第2の免疫化を含み、
ここで、第1の免疫化の第1の投与と第2の免疫化の第1の投与との間が21〜365日間であり、かつ
該アッセイが、該方法を哺乳動物被験体に対して行い、次いで該被験体における該エピトープに特異的な活性化または記憶T細胞のレベルを測定することを含んでなる、アッセイ。 - (i)第1の免疫化の投与のすべてが、第1の免疫化の第1の投与と基底レベルへの活性化T細胞のレベルの低下との間の時間内に含まれるのかどうか、および/または
(ii)第2の免疫化の第1の投与が基底レベルへの活性化T細胞のレベルの低下の後に行われるのかどうか、
を判定することを含む、請求項26記載のアッセイ。 - (i)請求項1〜27のいずれか1項において規定されたNOIまたは請求項14記載の組成物、および
(ii)請求項1〜27のいずれか1項記載の方法またはアッセイによる該NOIまたは組成物の投与のための説明書、
を含んでなる、請求項1〜27のいずれか1項記載の方法またはアッセイを行うためのキット。
