Abstract
Description
図1の注入器(及び図2の相当する注入器100)の構成要素と異なる形態が、調査された。要求された仕様で運転するために、注入器は、以下の特性を有すべきである。すなわち、1. 流体が受入れ端部に供給された時に、乾いた状態からの再現可能な毛管現象による充填、2. 電気浸透を駆動するための印加される電力が無い場合に、流出端部を越える流れが無いこと、及び3. 電力が一体化電極に印加される場合に、流出端部を越える有効な流速の再現可能な流れである。注入器の流路要素は、その構成物、すなわち材料、表面処理、空隙率、及びポアサイズに関して、且つその形状及び大きさに関して調査された。一体化電極は、それらの接点位置と接触面積に関して調査された。空気チャンバは、キャビティの大きさ、空気ギャプの大きさ、通気の形態に関して調査された。ポンプの始動の間の初期の毛管現象の流体充填速度における上記の構造パラメータの結果と、始動段階の間のポンプ要素の流出端部での流れの抑止の有効性と、ポンプ輸送している要素の流れ抵抗に依存する場合の電気浸透ポンプ輸送特性とが、調査された。
流出端部で通気される空気導管を備える流体工学的負荷の無い注入器を用い、その他の流体受容要素を備えていない、注入器ポンプの輸送特性を調査することが行われた。この形態は、略図2Eに示される。注入器は、最初に乾いた注入器の流体受入れ端部に、水様の流体を先ず供給することによって始動される。次に、電圧が一体化電極の間に印加され、体積流量率が、異なる時間で、既知の断面積の通気導管の流体の長さを測定することによって計測された。これから、電気浸透の移動度(EOM)が得られた。
最初に乾燥した注入器の微小多孔性流路要素は、注入器の流体が注入器の流体受入れ端部に供給される時に始動される。流体は、毛管現象で輸送することによって、流出端部まで要素を充填する。所望される流路材料、すなわち、0.11μmのポア径を有する微小多孔性硝酸/酢酸セルロースを使用して、長さ5mmの流路要素を備える注入器において、満杯にする時間はおよそ50s以内である。
アノードが流体受入れ端部の近くにある時はいつでも、概ね、良好な性能を得ることが出来た。最良の性能は、アノードが、流体受入れ端部を越えて注入器の微小多孔性流路の外側の、しかし流路と電気的な接触を保つ流体の中に埋められた時に得ることができた。カソードの配置は、注入器の微小多孔性流路の長手方向に沿った流出端部までのどこでもよいが、最善は、長手方向に沿って流出端部に向かう、およそ半分から四分の三のところである。これは、固体試薬の配置を可能とするために、流出端部でカソードの向こう側に電界がゼロの領域を残した。カソードが流体受入れ端部で、アノードに近づき過ぎた場合に、電流は高くなり過ぎて、低電圧に、及び低ポンプ速度運転に装置を制限した。電極接点の一般的な面積は、アノードが0.5×5mm、カソードが0.5×1mmであった。
矩形と台形の両方の注入器流路が調査された。一般的な矩形流路要素は、長さ約4.25mm×幅1mm×厚さ150μmの、空隙率0.7及び0.11μmポア径を有する硝酸/酢酸セルロースであった。この流路で組立てられ、流体受入れ端部の向こう側にアノード及び流体受入れ端部から3mm(流出端部から1.25mm)のカソードを備える注入器は、2mmol/LのDEA注入器流体を用いて運転される。印加された電圧に線形であるポンプ速度は、0.5nL/s/Vであった。公称運転電圧40Vで、ポンプ速度は20nL/sであった。一般的な台形の流路は、長さ約4.25mm、流体受入れ端部で幅4mm、及び流出端部で幅1.5mmであった。同一の電極配置及び注入器流体を用いた運転の場合に、電圧に線形であるポンプ速度は、1.1nL/s/Vであった。公称運転電圧40Vで、ポンプ速度は45nL/sであった。矩形の注入器と同様な流出端部の外形を有するが、より高いポンプ速度のゆえに、我々は、台形の注入器を使用することを所望した。流出端部の大きさは、受容する流体工学的要素の大きさによって制約される。
0.11μmのポア径を有する微小多孔性硝酸/酢酸セルロース(ミリポアMF膜GSWP)は、2mmol/LのDEA注入器流体を使用する場合に、約2.5×10-8m2/Vsの優れていて矛盾のないEOMを有することが見出された。これは、台形の(矩形の)注入器の1.1(0.5)nL/s/Vに相当する。その他の調査された材料は、もっと低いか又はゼロのEOMを有する。低EOM材料の表面の前処理、例えば、乾燥した後にドデシルスルホン酸アンモニウムのようなアニオン界面活性剤に予め浸すことは、表面の変化をもたらし、EOMを高くすることが出来る。しかしながら、界面活性剤が、流体受容要素とそれに接続される流体工学的回路に、注入された流体と共に排出されて、潜在的にそこで行われる生化学反応に有害な結果を引き起こす、このような処理を回避することが所望される。これは、後に述べるルシフェラーゼ反応とは、特に顕著であった。従って、先に引用された硝酸/酢酸セルロースは、表面の改良無しに使用できるから、注入器の流路のために所望される。
別の流体受容要素を備えるのではなく、流出端部に閉ざされた空気チャンバを備えた注入器は、無限の流体荷重を有する注入器ポンプの輸送特性を調査するために構成される。この配置は、略図2Aに示される。初めに、注入器は、最初に乾燥した注入器の流体受入れ端部に水様の流体を供給することによって始動された。次に、電圧が、一体化電極の間に印加された。流体は、注入器の流出端部から、当初の体積がP1=1atmでV1の閉じられたチャンバに移動された。空気は、流体がチャンバを満たした時に、流体の流動が停止した定常状態まで、圧縮された。空気の新しい体積は、V2<V1であった。流れを停止した結果の圧力は、P2=V1/V2で与えられるボイルの法則から計算される。0.11μmのポア半径を有する微小多孔性硝酸/酢酸セルロースが、使用される。
0.75から0.85の空隙率及び0.11から2.5μmの範囲で変化するポア径を有する、微小多孔性硝酸/酢酸セルロース材料から、台形の注入器(流入端部幅4mm、流出端部幅1.5mm、長さ4.25mm、厚さ0.15mm)が組立てられた。注入器は、流出端部に閉じられた空気チャンバを備えて組立てられた。0から100Vの様々なポンプ電圧で流れを停止するための圧力が、計測された。流れを停止するために必要な圧力は、電圧に伴ってほぼ線形に増加した。小さなポア径のためにより大きな背圧が、より大きなポア径の材料と比較して、流れを停止するために要求された。ポア径0.11μmを有する注入器は、0.17atm/Vの背圧に対してポンプ輸送が出来た。40Vの典型的な作動電圧で、注入器の流れを停止するための背圧は、7atmだった。2.5μmのポア径材料に対し、注入器の流れを停止するための背圧は、0.01atm/Vだった。40Vの典型的な作動電圧で、注入器の流れを停止するための背圧は、たったの0.4atmであった。
注入器の外辺部の密閉の品質は、良好な注入器の流速を得るために重要である。不適切な密閉の場合に、長手方向に沿った注入器の流路の外辺部で空気導管は、電気浸透ポンプ輸送中に、注入器の流出端部と流体受入れ端部との間の圧力差によって駆動される、導管を通過する逆流を発生するだろう。結果は、安定性が低下し、期待された電気浸透ポンプ速度よりも遅くなる。
流体抵抗を有する流体受容要素に接続される注入器ポンプの特性を調査するために、長手方向に沿った流体受容場所で流体受容の細長い領域要素に接続される流出端部に、閉じられた空気チャンバを有する注入器が、組立てられた。矩形と台形の両方の注入器が、調査された。注入器と流体受容要素の形態が、略図2Dに示される。この形態の注入器の運転において、様々な段階が、図3Aから3Eに示される。第一流体は、最初に乾燥した細長い領域(図3A)の流体受入れ端部に供給された。細長い領域は、側方毛管現象流れ(図3B)によって、第一流体で満たされた。次に、最初に乾燥した注入器は、流体受入れ端部(図3C)に水様の流体(2mmol/LのDEA溶液)を供給することによって始動された。注入器は、毛管現象流れ(図3D)によって流出端部まで満たされた。電圧は、一体化電極間に印加された。流体は、注入器の流出端部から、P1=1atmで当初の容積V1の閉じられたチャンバに移動した。閉じられたチャンバの空気は、流体がチャンバを充填した時に、圧縮が止まって(図3E)定常状態になるまで圧縮された。この定常状態で、図3Fに示したような、流体受容細長い領域に沿ってその両端部に向かう流体の流れ(図2Cの領域105と106に向かって流れる流体)があった。チャンバの中の空気の新しい体積は、V2<V1となった。結果として定常状態の圧力は、空気チャンバの圧力P2=V1/V2を与える、ボイルの法則から算出された。流体注入段階の後で、電圧は停止され、空気チャンバの中の圧縮された空気は、注入器の流出端部でその位置を回復し、従って、流体工学的及び電気的に、注入器の流体を流体受容要素の中の流体から遮断する(図3G)。
注入器の性能が、如何に注入器の構造に依存するかをより良く理解するために、パラメータとしては、注入器の受入れ端部から流出端部まで、毛管現象流れによって流体で始動される注入器の流路を含むモデル注入器を考慮する。注入器の流路は、長さL、流出端部で幅wと流体受入れ端部で幅W、及び高さhの、空隙率Ψ、ポア導管ねじれτ、及びポア径aの微小多孔性材料の台形スラブを包含する。注入器の流体受入れ端部に、(又は流体受入れ端部を越えるがそれと流体工学的に接続する流体中に)第一電極がある。入力から距離lに、注入器の流路の長手方向に沿って第二電極があり、結果として、電界がゼロの流出端部で長さL-lの領域がある。粘性ηの流体の流速Qは、方程式1によって与えられ、
図5は、図4の形態と寸法の、台形注入器及び矩形の流体受容要素の合計のポンプデータを示す。速度に対する無負荷(開放運転)の電圧は、三角形のデータ点として示される。流れを停止する圧力に対する無限負荷(閉じられた流出チャンバ)の電圧は、菱形のデータ点として示される。負荷を取って注入する間の圧力に対する電圧は、四角形の点である。
本発明の注入器は、二つの状態の、すなわちポンプ電力が付加されていない場合のオフ状態、及びポンプ電力が一体化電極に付加されている場合のオン状態の、一つの状態にあるとして特徴付けられる。最初のオフ状態において、注入器は、流出端部で空気ギャプ遮断手段によってその他の流体工学的要素から遮断される。理想的な最初のオフ状態において、空気ギャプ遮断手段を通過する漏洩流れは無い。オン状態において、注入器の流出端部を越える流体流れがある。理想的なオン状態において、流体流速は、付加ポンプ電力のみに依存すべきであり、ポンプの通常運転中に発生するかもしれないような、注入器の流入端部及び流出端部にわたる圧力差に依存するべきでない。ポンプ作動後の理想的なオフ状態において、微小反応器のような下流の流体工学的要素中の注入された流体の位置が、オフ状態の継続時間中に安定であるために、注入器への又は注入器からのさらなる漏洩流れは、あるべきでない。
一体化電極を備える注入器及びそれに接続される流体工学的回路を包含する、本発明の流体工学的モジュールは、注入器を始動する流体を収納する一体化密閉された貯槽をまた包含する、プラスチックのカード筐体の中に組込むことができる。流体工学的モジュール及び一体化流体貯槽を備える、カード筐体は、単一の一体化ユニット内に収納された評価分析のために必要な全ての試薬を備える一段階装置を包含する。本発明の流体工学的モジュールは、図1から4の略配置で述べられたような標準の印刷された回路基板で組立てられる。この場合に、外部の接触する手段と一体化電極の電気的な接触場所は、流体工学として同一側のモジュールの基板上にある。流体工学的モジュールは、二つの面を有する可撓性回路基板にまた組立てることもでき、基板は、基板を貫通する電気的な接続管を有し、その結果、流体工学的回路要素は、可撓性の基板の上部面に組立てることができ、外部の接触手段と接触する場所が下部面にある。これは、図6及び6Aで略図的に示された、流体工学的要素をクレジットカードの大きさのカード筐体に組込む場合に、所望される組立である。
この実験において、通気口を有する空気チャンバを備えていることを除いて、図2Qに示されたものと同一の注入器の形態が、使用される。この装置において、注入器は、流出開口部で1mm、流入開口部で4mm、及び長さ4.25mm×厚さ0.15mmの寸法の台形要素であり、空隙率0.7及びポア径0.11の微小多孔性硝酸/酢酸セルロースを包含する。通気口を有する空気チャンバがあり、それは、流出端部を第一流体受容要素から分離する長さ0.5mmの空気ギャプを含む注入器の流出端部で、幅1mmの導管であった。第一流体受容要素は、中央に配置された流体受容領域、試料受入れ端部、及び流出端部を備える側方流動の細長い領域であった。この要素は、厚さ0.15mm×幅1mm×長さ8mmの、空隙率0.7及びポア径0.25μmを有する微小多孔性ポリエーテルサルフォンであった。別の0.5mm空気ギャプによって第一から分離される第二流体受容要素があった。第二流体受容要素は、厚さ0.15mm×2mm角のポリエーテルサルフォンパッドを包含する反応領域であって、ATP、発光酵素、マグネシウムイオン及び緩衝剤を含む溶液で含浸され、乾燥されている。評価分析試薬は、シグマ社から得られる。

これは、ルミノジェニック基質を供給するための注入器を備えた、側方流動の細長い領域で行われるリガンド結合評価分析の例である。この実験において、装置の形態は、図21に示されたものと同一である。注入器は、流出開口部で1mm、流入開口部で4mm、及び長さ4.25mmで厚さ0.15mmの寸法の台形要素であり、空隙率0.7及びポア径0.11の微小多孔性硝酸/酢酸セルロースを包含する。第一流体受容要素は、アルカリ性ホスファターゼ(トロピックス社から得られるCDP-star)のためのルミノジェニックジオキセタン基質を含有する、乾燥した試薬受入れ領域であった。別の0.5mmの空気ギャプによって最初は分離された、第二流体受容要素があった。第二流体受容要素は、中央に配置された流体受容領域、試料受入れ端部、及び流出端部を備える側方流動の細長い領域だった。この要素は厚さ0.15mm、幅1mm、長さ8mmで、空隙率0.7及びポア径0.25μmの微小多孔性ナイロンであった。要素は、(10mg/Lを含有する溶液の600nLを注入することによって、)細長い領域の長手方向に沿って中央に配置された長さ1mmの捕縛場所にストレプトアビジンを供給することによって最初に処理され、それから、装置に組込む前に、製造者の推奨手順に従ってスーパーブロック(ピアースバイオテクノロジー社製)を用いてブロッキングすることによって処理された。

Claims (43)
- 流体受容装置の流体受容場所に流体を輸送するための注入器ポンプであって、
流体を受け入れるための流体受入れ端部及び受容場所に流体を輸送するための流出端部部を有する、最初は乾燥している流路で、前記受入れ端部に流体の供給があると、前記流出端部まで自動的に流体で充填される前記流路と、
前記流体受容場所へ流路要素の前記流出端部から、流体を電気浸透的にポンプ輸送するための駆動配置と、
電気浸透のポンプ輸送をする間に、外辺部で前記流路からの流体流れを予防するために、前記流路の外辺部に沿って前記流路を密閉するための密閉要素と、
を具備する注入ポンプ。 - 前記流路が流体を含んでいる時に、前記流出端部からの受動的な流体流れを防止するために、前記受容場所から、前記流出端部を流体工学的に遮断するための遮断器をさらに具備する、請求項1に記載の注入ポンプ。
- 前記遮断器が、前記流出端部に近接する空気ギャプである、請求項2に記載の注入ポンプ。

- 前記流路が、可動性の試薬を含有し、流体が前記受入れ端部に供給される時に、前記試薬は移動性を持って、毛管現象の流れによって前記微小多孔性流路の長手方向に沿って輸送され、好ましくは試薬が、発光性の基質、蛍光性の基質、起電性の基質、及び化学ルミネッセンスの基質、並びにその組合せのグループから選択される、請求項1から11のいずれか一項に記載の注入器ポンプ。
- 前記微小多孔性流路が、1μmより小さい、好ましくは、0.2μmより小さい半径のポアを有する、請求項3に記載の注入器ポンプ。
- 前記電気浸透的なポンプ輸送される流体が、10mmol/Lより低い電解質濃度を有する、請求項1から13のいずれか一項に記載の注入器ポンプ。
- 前記流路が、その流出端部よりも幅広いその流体受入れ端部を備える台形の形状である、請求項1から14のいずれか一項に記載の注入器ポンプ。
- 前記流体で充填された流路の流れコンダクタンスが、前記流体受容デバイスの受容場所での流れコンダクの少なくとも二十分の一である、請求項1から15のいずれか一項に記載の注入器ポンプ。
- 前記流体受容場所で通気口を有する空気チャンバに、又は前記流体受容場所で閉じられた空気チャンバに、液体を供給するための、請求項1から16のいずれか一項に記載の注入器ポンプ。
- 前記流体受容デバイスが、その長手方向に沿って流体受容場所を備える微小多孔性側方流動の細長い領域であり、好ましくは、試料受入れ端部と流出端部とを備える側方流動の細長い領域である、請求項1から17のいずれか一項に記載の注入器ポンプ。
- 前記最初に乾燥した流路の中に、その受入れ端部で導入される流体が、一体化した流体貯槽から前記受入れ端部に供給される、請求項1から18のいずれか一項に記載の注入器ポンプ。
- 微小評価分析装置であって、
微小反応器と、
前記微小反応器の中に試料を導入するための第一流体工学的要素と、
請求項1から19のいずれか一項に画定されるような注入器ポンプと、
を具備する微小評価分析装置。 - 前記微小反応器の中に試料を導入するための前記第一流体工学的要素が、微小流路化された微小多孔性要素であって、それは、最初に乾燥していて、可動性の試薬を含有する、請求項20に記載の微小評価分析装置。
- 前記微小反応器が、前記第一流体工学的要素の長手方向に沿って配置される、請求項20に記載の微小評価分析装置。
- 前記第一流体工学的要素及び前記注入器ポンプの前記流路要素が、平面基板上に微細加工される、請求項20に記載の微小評価分析装置。

前記少なくとも一つのN、M流路が、請求項1から19のいずれか一項に画定されるような注入器ポンプである、微小評価分析装置。 - 前記注入器ポンプの前記流路の前記流出端部、前記空気ギャプ、及び前記微小反応器に試料を導入するための前記第一流体工学的要素が、空気を収納し、周囲から密閉された閉じられたチャンバの中に密閉され、又は空気を収納し、空気の通気導管を介して放出される閉じられたチャンバの中に密閉される、請求項20又は25に記載の微小評価分析装置。
- 前記流体受容デバイスが、その長手方向に沿って流体受容場所を備える、微小多孔性側方流動の細長い領域である、請求項25又は26に記載の微小評価分析装置。
- 一つ又はそれ以上の、前記N、M流路が、最初に乾燥していて、可動性の試薬、好ましくは、発光性の基質、蛍光性の基質、起電性の基質、及び化学ルミネッセンスの基質、並びにその組合せのグループから選択される試薬を含有する、請求項25から27のいずれか一項に記載の微小評価分析装置。
- 一つ又はそれ以上の前記微小反応器が、導管であって、それが流体工学的に、第一流体工学的手段の領域に、又は前記注入器ポンプの流路の領域に接続される、請求項25から28のいずれか一項に記載の微小評価分析装置。

- 一つ又はそれ以上の、前記M流出流路又は前記N流入流路が、微小多孔性要素であって、それが最初に乾燥していて、可動性の試薬を含有している、請求項25から30のいずれか一項に記載の微小評価分析装置。

化学反応を行うための、前記側方流動要素の長手方向に沿う少なくとも一つの微小反応器と、
前記微小反応器に流体を選択的に供給するための、請求項1から17のいずれか一項に画定されるような注入器ポンプと、
を具備する診断装置。 - 少なくとも一つの側方流動要素が、可動性の化学試薬を備える最初に乾燥した微小多孔性要素である、請求項33に記載の診断装置。
- 可動性の試薬が、標識を付けられたコンジュゲイトである、請求項34に記載の診断装置。
- 前記注入ポンプの前記流体通路要素の前記流出端部、前記空気ギャプ、及び前記側方流動要素の前記流体受容場所が、空気を収納する閉じられたチャンバに密閉される、請求項33に記載の診断装置。
- 試料流体中の検体を検出するために、肝要な流体工学を備えた診断装置であって、
電気的に絶縁された基板と、
前記基板上の微小反応器と、
前記微小反応器に、検体コンジュゲイト複合体を形成するためのレポータコンジュゲイトを供給するための少なくとも一つの側方流動要素と、
前記微小反応器に試料を供給するための流動工学的要素と、
前記微小反応器の流体受容場所に試薬を供給するための、請求項1から17のいずれか一項に画定されるような注入器ポンプと、
を具備する診断装置。 - 検体コンジュゲイト複合体構造を検出するための手段をさらに具備する、請求項37に記載の診断装置。
- 試料流体中の検体の濃度を検査するための一体化した診断装置であって、
検体コンジュゲイト複合体を捕縛するための微小反応器と、
前記微小反応器へ試料流体中の検体の輸送のための、主たる側方流動要素を備える基板と、
前記微小反応器へ試薬を供給するための少なくとも一つの補足の流路において、請求項1から17のいずれか一項に画定されるような注入器ポンプである補足の流路と、
を具備する診断装置。 - 診断装置であって、
カード本体と、
流体受入れ端部及び流出端部を備えるカード本体に装着された、最初に乾燥した微小多孔性流路と
前記カード本体上に同様に装着された、密閉された流体貯槽と、
前記貯槽から貯蔵された流体を放出するために、前記密閉された流体貯槽を選択的に開放するためのバルブと、
前記微小多孔性流路の前記受入れ端部へ、前記流体チャンバによって放出される前記流体を供給するための導管と、
を具備する診断装置。 - 診断装置であって、
試料受入れ端部及び流出端部を含む側方流動の細長い領域と、
機器制御された流体注入器から流体を受容するための、長手方向に沿った少なくとも一つの流体受容場所と、
前記流体受容場所の閉じられた空気チャンバと
を具備する診断装置。 - 少なくとも一つの電気浸透ポンプを備える微小流体工学装置であって、
流路が、その表面でゼータ電位を有し、二つの離間する電極の場所で、一対の離間する電極と電気的に接続される流路をさらに具備していて、離間する電極が、絶縁基板の一方の側に形成され、電極を横切って電位を発生するための外部機器と接続するために、絶縁基板の他方の側に形成される二つの離間する接点位置に、基板を介して電気的に接続される、微小流体工学装置。 - 絶縁基板が可撓性ホイルである、請求項42に記載の装置。
