Abstract
Description
IL−1F7bのIL−18Rα鎖への報告された結合(Pan 2001, Kumar 2002)に基づいて、IL−1F7b(類似IL−18)が、IL−18α受容体への結合の際、細胞におけるIFNγ産生を刺激するかどうかを評価するために実験を設計した。全長分子(プロIL−1F7b)または推定ICE切断部位でN末端としてE21を用いる成熟分子(成熟IL−1F7b)(図1参照)の両方を以下の実験に使用した。ヒトNKO細胞(Kim 2000)、ヒト全血の培養物、末梢血単核細胞(PBMC)(プレプロテク(Preprotech)製のIL−12 1ng/mlと共刺激された)(PBMCの調製は実施例8参照)またはKG−1細胞(TNFα 10ng/mlで共刺激された)(細胞株はATCCロックビル(Rockville)、MDから入手された)を、100ng/mlの組み換えIL−1F7b(実施例5のプロ型または成熟型)またはIL−18で刺激した。IFNγは、刺激後18時間(KG−1は48時間)の細胞の調整培地中で(Puren 1998記載の液相電気化学発光(ECL)により)測定された。一方、IL−18は、顕著にIFNγ産生を刺激したが(図2A)、プロIL−1F7bおよび成熟IL−1F7bのいずれも、これらの細胞においてはIFNγ産生を全く刺激しなかった。
IL−18はIL−18Rαにその第3の細胞外ドメイン(IL−18Rα:D3)を介して結合することが報告されている(Azam 2002)。IL−1F7bのIL−18Rαへの結合の特徴付けを行なうために、IL−18Rαの第3の細胞外ドメイン(CD3)を個別にE.coliでhis6標識化タンパク質として発現させ、Talonアフィニティーカラムクロマトグラフィーで精製した。ついで、IL−1F7bをそのように精製したIL−18Rα:D3とインキュベートし、化学的に交差結合させた(実施例7)。図3Aに示すように、SDS−RAGEおよびウエスタンブロッティング分析により、交差結合されたIL−1F7bとIL−18Rα:D3に相当する43kDaの複合体が示された。IL−18Rαへの交差結合は、プロIL−1F7bおよび成熟IL−1F7bの両方で観察された。これらの結果から、IL−18RαD3はIL−1F7bへの結合のための重要な部分であり、IL−18に結合するために重要であることが以前に証明されたものとまさに同じドメインであることが示唆される。
IL−18とIL−1F7bとのアミノ酸配列を比較した。図1に示すように、IL−1F7bはIL−18とIL−18においてE42およびK89として保存されている2つのアミノ酸を共有している。E42およびK89は、IL−18の活性とIL−18BP結合にとって重要であることが示されている(Novick 1999)。したがって、IL−1F7bのIL−18との配列類似性に基づき、IL−1F7bがIL−18BPに結合する可能性を調べた。
IL−1F7bがIL−18BPに結合する(前実施例)、おそらくIL−18が結合する正に同じドメインに結合するという知見に鑑み、IL−18BPによるIL−18活性の阻害に対するIL−1F7bの効果を調査した。作業仮説は、IL−1F7bは、IL−18BPへの結合に関してIL−18と競合し得、したがってIL−18はIL−1F7bの存在下でIL−18BPによりほとんど中和されないであろうということである。
IL−1F7bに特異的なIgGをポリクローナルウサギ抗IL−1F7b血清から精製し、ヒトPBMCにおけるIL−1F7bの発現を調べるために使用した。ウサギ抗IL−1F7b血清とIgG沈殿の特異性を、RAW264.7マクロファージ細胞のIL−1F7bcDNAによるトランスフェクションを用いて2つの異なる方法で調べた。第一に、IL−1F7b抗血清は、IL−1F7bトランスフェクトRAW264.7細胞の溶解物においてIL−1F7bを特異的に認識した(図6)。第二に、共焦点デジタル顕微鏡を使用して、アフィニティー精製抗IL−1F7bIgGはトランスフェクトRAW264.7ではIL−1F7b発現を認識したが、模擬コントロール細胞では認識しなかった(示さない)。新たに単離したヒトPBMC(実施例8)を、IL−1F7bについてアフィニティー−精製ポリクローナルウサギ抗−ヒトIL−1F7bIgGを1μg/mlで使用して染色した。共焦点レーザー顕微鏡を使用して、IL−1F7bを発現するヒト血液単球の積層図を作成した。IL−1F7bが、血漿膜の内部表面に局在する細胞質ならびに核の両方で発現されることが観察された(示さない)。リンパ球集団の染色は観察されなかった。
以下のオリゴヌクレオチドプライマーを、ヒト脾臓ライブラリー(クローンテック(Clontech)(登録商標)HLOO11B、BD バイオサイエンス クローンテック(Biosciences Clontech)、パロ アルト、CA)からIL−1F7bcDNAをクローン化するために使用した。センスプライマー5′GTTGAGTAATAAACTCAACG(配列番号1)、リバースプライマー5′GTTCAATGGGGCAGTTTC(配列番号2)(クローンAF200496(ジーンバンク(登録商標))(Kumar 2000)に特異的)。IL−1F7bcDNAは、5′端にEcoRI、3′端にXbaIのための断部位を導入する2番目の対のプライマー(センスプライマー5′−ATATGAATTCATGTCCTTGTGGGGGAG(配列番号3);リバースプライマー5′−TATATCTAGAAGTTTCCTAATCGCTGACC(配列番号4))を用いて再増幅された。TA−クローニングを使用して、IL−1F7bcDNAを、製造元の使用書にしたがってpGEM−T Easy(登録商標)(プロメガ社(Promega Corp.)、マディソン、WI)に移し、正しい配列を確認した。IL−1F7bcDNAを、ついでEcoRIおよびXbaI部位を用いて細菌による発現のためにpPROEXTMHTa(ギブコ−BRL)にライゲートした。pPROEXTMHTaベクターは、発現されるタンパク質のアフィニティー精製のためのN−末端His×6タグを含有する。pPROEXTMHTa/IL−1H4プラスミドをコンピテントE.coli株DH5a(ギブコ−BRL)に形質転換した。一晩培養した培養物(10ml)を、100μg/mlのアンピシリンを含有する200mlのLB培地に添加し、0.6〜1OD600の密度まで増殖させた。
精製タンパク質を30μlのPBSと混合し、氷上で2時間インキュベートした。
PBMCは、健常ドナーの血小板除去残渣白血球(platelet-depleted residual leukocytes)またはヘパリン添加血液のいずれかから精製した。血小板除去白血球または全血を生理食塩水で1:1に希釈し、前述のように、Ficoll−Histopaque(登録商標)グラジエント(シグマ)に適用した(Kim 2001)。遠心分離ののち、界面から細胞を集め、生理食塩水で3回洗浄し、RPMIに再懸濁させた。単離したPBMCを分析を開始するまで氷上に保持した。

Claims (34)
- IL−18BPまたはそのムテイン、融合タンパク質、機能的誘導体もしくはフラグメントに結合することができ、IL−1ファミリーの一員であるサイトカイン−2の受容体を阻害することができるサイトカイン−1またはそのアイソフォーム、ムテイン、融合タンパク質、機能的誘導体もしくはフラグメントの使用であって、該サイトカイン−2の受容体の誘導により発症または悪化する疾患の治療または予防のための医薬の製造における使用。
- サイトカイン−1がIL−1ファミリーに属する請求項1記載の使用。
- サイトカイン−1がIL−1F7bである請求項2記載の使用。
- サイトカイン−2の受容体がIL−18Rである請求項1、2または3記載の使用。
- サイトカイン−1による阻害が、サイトカイン−2の受容体のシグナル鎖へのサイトカイン−1の結合に伴って生じる請求項1、2、3または4記載の使用。
- エンドトキシン致死症(敗血症)、毒素または活性化T細胞またはC型肝炎により引き起こされる肝臓傷害、関節炎、肺障害、乾癬、炎症性腸疾患、脳障害、虚血性傷害、心臓機能障害および神経炎から選択される炎症性疾患の治療または予防のための請求項1、2、3、4または5記載の使用。
- 転移形成を予防するための請求項1、2、3、4または5記載の使用。
- 医薬がさらにIL−18BPまたはそのムテイン、融合タンパク質、機能的誘導体もしくはフラグメントを含有する請求項1、2、3、4、5、6または7記載の使用。
- サイトカイン−1が全身的、皮下的および/または筋肉内的に投与される請求項1、2、3、4、5、6、7または8記載の使用。
- IL−18BPまたはそのムテイン、融合タンパク質、機能的誘導体もしくはフラグメントに結合することができ、IL−1ファミリーの一員であるサイトカイン−2の受容体を阻害することができるサイトカイン−1またはそのアイソフォーム、ムテイン、融合タンパク質、機能的誘導体もしくはフラグメントのコーディング配列を含むベクターの使用であって、該サイトカイン−2の受容体の誘導により発症または悪化する疾患の治療または予防のための医薬の製造における使用。
- サイトカイン−1がIL−1ファミリーに属する請求項10記載の使用。
- サイトカイン−1がIL−1F7bである請求項11記載の使用。
- サイトカイン−2の受容体がIL−18Rである請求項10、11または12記載の使用。
- サイトカイン−1による阻害が、サイトカイン−2の受容体のシグナル鎖へのサイトカイン−1の結合に伴って生じる請求項10、11、12または13記載の使用。

- IL−18BPまたはそのムテイン、融合タンパク質、機能的誘導体もしくはフラグメントに結合することができ、サイトカイン−2、IL−1ファミリーの一員であるサイトカイン−2の受容体を阻害することができるサイトカイン−1の内因性遺伝子活性化のためのベクターの使用であって、該サイトカイン−2の受容体の誘導により発症または悪化する疾患の治療または予防のための使用。
- サイトカイン−1がIL−1ファミリーに属する請求項19記載の使用。
- サイトカイン−1がIL−1F7bである請求項20記載の使用。
- サイトカイン−2の受容体がIL−18Rである請求項19、20または21記載の使用。
- サイトカイン−1による阻害が、サイトカイン−2の受容体のシグナル鎖へのサイトカイン−1の結合に伴って生じる請求項19、20、21または22記載の使用。

- IL−18BPまたはそのアイソフォーム、ムテイン、融合タンパク質もしくはフラグメントに結合することができ、IL−1ファミリーの一員であるサイトカイン−2の受容体を阻害することができるサイトカイン−1の阻害剤の使用であって、該サイトカイン−2の受容体の誘導により予防または軽減される疾患の治療または予防のための使用。
- サイトカイン−1がIL−1ファミリーに属する請求項28記載のサイトカイン−1の阻害剤の使用。
- サイトカイン−1がIL−1F7bである請求項29記載のサイトカイン−1の阻害剤の使用。
- サイトカイン−2の受容体がIL−18Rである請求項28、29または30記載のサイトカイン−1の阻害剤の使用。
- サイトカイン−1による阻害が、サイトカイン−2の受容体のシグナル鎖へのサイトカイン−1の結合に伴って生じる請求項28、29、30または31記載のサイトカイン−1の阻害剤の使用。
- ウイルス性疾患の治療または予防のための請求項28、29、30、31または32記載のサイトカイン−1の阻害剤の使用。
