こで用いられる「医薬的に受容可能な担体」という言葉は、医薬投与に適合するあらゆるすべての溶剤、分散媒、被覆、抗バクテリアおよび抗真菌剤、等張および吸収遅延剤などを含むことが意図される。医薬的活性物質に対してこうした媒質および薬剤を用いることは当該技術分野において周知である。あらゆる従来の媒質または薬剤が抗−TSG101抗体に適合しない場合を除き、組成物におけるその使用が予期される。組成物に追加の抗−TSG101抗体が取込まれてもよい。

ウイルス感染に対する抗−TSG101抗体の影響を定めるために、レトロウイルス感染アッセイが開発された。末端反復配列(LTR)プロモータから発現される大腸菌lacZ遺伝子を含むネズミ白血病ウイルス(MLV)由来ベクター(pBMN−Z−I−Neo)が、293細胞(フェニックスA、ATCC)由来の両栄養性ネズミ白血病レトロウイルスパッケージング細胞系にトランスフェクションされた。フェニックスAヘルパー細胞によって生成されるレトロウイルスが回収されて、マウスN2A細胞(ATCC)の感染に用いられた。MLVベクターのトランスフェクションの24時間後に、抗−TSG101抗体が293ヘルパー細胞に加えられた。ウイルス生成に対するTSG101抗体の効果は、ウイルス上清が標的細胞(N2A)に感染する効率によって定められた。次に、β−ガラクトシダーゼ活性の細胞染色(正の細胞は青く染色され、図2における暗い点のように示される)によってN2A細胞の感染が定められた。
この実施例は、HIV放出の際にTSG101のドメインが細胞表面に露出され、抗−TSG101抗体がHIV放出および感染を阻害したことを示す。
TSG101が形質膜におけるウイルス放出に積極的にかかわっていることを示すために、GFP−TSG101融合タンパク質の発現ベクターが構築されて、M−MuLVウイルスを積極的に生成するフェニックス細胞(スタンフォード大学のノラン(Nolan)らにより開発されたレトロウイルスヘルパー細胞系)にトランスフェクションされた。トランスフェクションの24時間後、共焦点顕微鏡(ウルトラビュー(Ultraview)、パーキン−エルマー)の下でGFP−TSG101融合タンパク質のトラフィックが観察された。図3A−Eは、GFP−TSG101タンパク質が細胞質から細胞表面へ移動してウイルス生成細胞から「出芽」する画像の時間経過を示す。
TSG101がHIV出芽にも積極的にかかわっているかどうかを定めるために、抗−TSG101抗体を用いて、HIVでトランスフェクションされたヒトCD4+ヒトT細胞系H9(H9ΔBglと示す)における細胞表面TSG101を直接検出し、対照としてトランスフェクションしていないH9細胞を用いた。2つのウサギ抗−TSG101ポリクローナル抗体、1つはN−末端に対するもの(抗−TSG101“C”と示す)で、1つはC−末端TSG101に対するもの(抗−TSG101“E”と示す)、がこの研究に対して用いられた。どちらの抗体もよく特徴付けされたものである(リら、2001, Proc Natl Acad Sci USA 98(4): 1619-24)。どちらの抗体も、HIVを生成するH9ΔBgl細胞においてのみTSG101の細胞表面局在性を特異的に検出し、対照H9細胞においては細胞表面TSG101は検出されなかった(図4)。興味深いことに、抗−TSG101抗体は、抗−HIV抗体によって観察されるような「キャップ形成」様出芽構造を検出した(リー(Lee)ら、1999, J Virol. 73:5654-62)。
次いで、HIV生成細胞(H9ΔBgl)および対照H9細胞におけるTSG101の細胞表面局在性が蛍光活性化細胞選別器(FACS)によって調べられた。H9ΔBglおよびH9細胞はいずれも固定され、抗−TSG101抗体で染色され、蛍光ラベルされた二次抗体によって検出された。免疫染色された細胞はFACSで分析された。6回を超える独立した実験により、約70−85%のH9ΔBgl細胞が表面TSG101に対して正に染色されたのに対し、約0.1%未満のH9対照細胞が表面TSG101に対して正に染色されたことが示された(図5)。これらの結果は、共焦点顕微鏡によるH9ΔBglおよびH9対照細胞の両方の直接調査によってさらに確認された。少数(0.1%未満)のH9対照細胞は、共焦点顕微鏡分析後の免疫染色手順に関連する弱いバックグラウンド蛍光シグナルによってもたらされた。H9ΔBgl細胞の正の集団は明るい蛍光を示した。
HIV−1ウイルス生成アッセイを用いて、レトロウイルス生成に対するTSG101の阻害的効果をさらに調べた。HIV−1ベクターpNL4−3が293T細胞にトランスフェクションされた。トランスフェクションの24時間後、2つの抗−TSG101抗体(10ug/ml)、すなわち抗−TSG101抗体“E”および抗−TSG101抗体“B”、ならびに非特異的対照抗体(10ug/ml)がそれぞれ細胞培養物に加えられた。抗−TSG101抗体“B”はネズミTSG101N−末端断片に対して増やされ
たもので、ヒトTSG101タンパク質にあまり結合しない。抗−TSG101抗体“B”は対照として用いられた。追加の24時間インキュベーションの後、細胞溶解物が抽出され、細胞培養上清が回収されて、スクロース勾配によってHIV−1ビリオンが精製された。細胞溶解物および精製ビリオンの両方が、2つの抗−HIV−1抗体(抗−p55および抗−p24)を用いたウェスタンブロットにより分析された。図6に示されるとおり、抗−TSG101抗体“E”処理はHIV−1ビリオン放出の顕著な阻害を示した(ウェスタンブロットの密度追跡による70%を超える阻害、レーン8)のに対し、抗−TSG101抗体“B”(レーン7)および対照抗体(レーン6)はHIV−1放出の顕著な阻害を示さなかった。
HIV放出に対する抗体の効果を特定的に調べるために、H9ΔBgl細胞に基づくHIV放出アッセイが開発された。H9ΔBgl細胞は、エンベロープ欠損HIV構成(HIVゲノムのBglII断片の削除)によってトランスフェクションされたヒトCD4+Tリンパ球である。安定にトランスフェクションされたH9ΔBgl細胞は、非感染性の形のHIV(HIVエンベロープ欠損のために培養物中の他のH9ΔBgl細胞に感染できない)を生成し、H9ΔBgl細胞からのHIV放出は細胞培養上清のHIVp24ELISAによって直接測定できる。いくつかの濃度のTSG101抗体“E”および対照抗体(同じ濃度のウサギIgG)を用いて、H9ΔBgl細胞とともにインキュベートした。抗体を加えて48時間後に、培養上清を回収してHIVp24ELISAに供した。80ug/mlにおいてウイルス放出の顕著な抗体阻害が観察された(図7)。
抗−TSG101抗体がウイルス放出に続くHIV伝染性に対する付加的な影響を有するかどうかを定めるために、ジャーカット細胞からのHIV上清を用いて、抗−TSG101抗体“E”および対照としてのウサギIgG(40ug/ml)の存在下でMAGI細胞を感染した。抗−TSG101抗体はHIV伝染性の顕著な阻害を示し(図8)、TSG101がウイルス放出に続く標的細胞のHIV成熟および/または感染における役割を有することを示唆した。
この実施例は、TSG101がEBOV VP40と相互作用し、TSG101がEBOV VLPに取込まれ、また抗−TSG101抗体がEBOVウイルス様粒子(VLP)の放出を阻害することを示す。

いくつかのウイルス様粒子は、ウイルスマトリックスタンパク質の単なる発現によって生成できる(ジョンソン(Johnson)ら、2000, Curr Opin Struct Biol 10, 229-235)。EBOVおよびMARVマトリックスタンパク質(VP40)は形質膜およびウイルス封入体の両方に局在化することが示されており(コレスニコバら、2002, J Virol 76(4): 1825-38; マーティン−セラーノら、2001, Nature Medicine 7:1313-19)、VP40は成熟ビリオンの集合および放出を行なうことが示唆されている。しかし、VP40のみの発現によってVLPを効率的に生成する試みは全く実を結ばず、アモルファスVP40含有材料の非効率的な放出が示された(ババリ(Bavari)ら、2002, J Exp Med 195, 593-602)。レトロウイルスにおいて、集合複合体のラフト(raft)局在化はN−末端アシル化Gagタンパク質の会合によって制御される(キャンベル(Campbell)ら、2001, J Clin Virol 22, 217-227)のに対し、VP40などのフィロウイルスタンパク質のラフトターゲッティングは主にウイルス糖タンパク質(GP)によって制御されているようである(ババリら、2002, J Exp Med 195, 593-602)。したがって、フィロウイルスVLPの生成にはGPおよびVP40の両方の共発現が必要なのではないかという仮説が立てられた。GPおよびVP40が培養上清に放出されるかどうかがまず調べられた。GPまたはVP40のいずれかのみを発現する細胞においては、いずれのタンパク質も細胞と上清との両方において検出された(図9A)。しかし、両方のタンパク質の共発現によって、細胞からの放出がかなり増加した(図9A)。もし放出されたGPおよびVP40が粒子中で会合しているなら、VP40は抗−GPmAbによって共免疫沈降するはずだと推論された。図9Bに示されるとおり、EBOVのGPおよびVP40の両方でトランスフェクションした細胞の上清からの抗GP−免疫沈降物からVP40は容易に検出された。VP40のみを発現する細胞の上清からはVP40は落とされず、この共免疫沈降(co-IP)が特異的であることを示した。
共免疫沈降実験は、上清に放出されたGPおよびVP40が何らかの形で互いに会合していることを示した。これらの複合体がウイルス様粒子(VLP)を表わすかどうかを確認するために、培養上清からの粒状材料がスクロース勾配超遠心分離によって精製され(ババリら、2002, J Exp Med 195, 593-602)、電子顕微鏡を用いて分析された。興味深いことに、GPおよびVP40によって共トランスフェクションされた細胞の上清から得られた粒子のほとんどは、フィロウイルスに著しく類似した線状形態を示した(図10AおよびB)(ゲイスバート(Geisbert)ら、1995, Virus Res 39,129-150; マーフィ(Murphy)ら、1978, エボラおよびマールブルグウイルス形態および分類(Ebola and Marburg virus morphology and taxonomy), 初版(アムステルダム、Elsvier))。これに対し、単独でトランスフェクションされた細胞から得られた材料は、細胞死の際に放出されたと思われる少量の膜断片しか含んでいなかった。VLPは直径が50−70nmであり、長さが1−2μmである(図10)。これは、インビトロ感染後の細胞培養上清において見られるエボラビリオンの長さの範囲に類似している(ゲイスバートら、1995, Virus Res 39,129-150)。VLPの直径が(EBOVの80nmに比べて)より小さいのは、おそらくリボ核タンパク質複合物の欠如によるものであろう。分岐形、棒形、U形および6形など、フィロウイルスに対して記載されるあらゆる種類の形態(フェルドマン(Feldmann)ら、1996, Adv Virus Res 47, 1-52; ゲイスバートら、1995, Virus Res 39,129-150)がこれらの粒子において観察された(図10)。また、VLPは5−10nmの表面突起または「スパイク」で覆われており(図10)、これはEBOVの特徴である(フェルドマンら、1996, Adv Virus Res 47, 1-52; ゲイスバートら、1995, Virus Res 39,129-150)。抗−エボラGP抗体によるVLPの免疫金染色は、粒子の表面上のスパイクのエボラ糖
タンパク質としての同一性を示した(図10B)。マールブルグウイルスに対するVLPも類似の態様で生成された。
我々は、EBOV集合および放出における液胞タンパク質選別(vps)タンパク質TSG101のVP40の後期ドメインとの相互作用のかかわりを調べるために一連の生化学的研究を行なった。これらの研究には、C−末端がMycエピトープで標識された1組の短くしたTSG101が用いられた。全長TSG101ならびに140、250および300の位置で短くされた変異体と、EBOV VP40とによって293T細胞がトランスフェクションされた。48時間後に細胞が溶解され、抗−Myc抗体による免疫沈降を受けた。図11に示されるとおり、VP40は、1−140の短くされた変異体を除くすべてのTSG101タンパク質とともに共沈した。この変異体と会合しないことは、残基141−145がHIV Gag由来PTAPペプチドとの重要な接触を行なうことを示す構造的データ(ポルニロスら、2002, Nat Struct Biol 9, 812-7)に一致している。興味深いことに、TSG101の1−300変異体とのVP40の会合は、全長または1−250変異体とのものよりも顕著に強かった(図11)。全長TSG101の会合の方が低かったのは、TSG101のUEVドメインとの分子間または分子内会合を形成し得る、この分子のC−末端領域中のPTAPモチーフの存在のためだろう。アミノ酸250−300の削除による相互作用の劇的な減少は、この領域における残基がウイルスマトリックスタンパク質との結合に寄与している可能性を示唆する。
SPR測定を用いて、TSG101と相互作用するエボラVP40の定量分析を行なった。エボラVP40のN末端からの11アミノ酸残基を含むビオチン化ペプチド(Bio−ILPTAPPEYME)をストレプトアビジンチップ上に固定した。UEVドメインのみを含む精製TSG101タンパク質を異なる濃度(1、2、5、20uM)で連続的に注入した。図13に示されるように、ペプチドおよびタンパク質間の中程度の親和性の相互作用が検出できる。このSPRデータに基づいて、我々はこの相互作用に対する約2μMのKD値を算出した。
TSG101がEBOV VLPに取込まれているかどうかを定めるために、TSG101(1−312)をGPまたはGP+VP40とともに293T細胞中で発現した。抗GP抗体を用いてVLPを上清から免疫沈降した。TSG101(1−312)の発現の結果、VLP放出が顕著に増加し、VLP出芽におけるTSG101の正の役割が示唆された(図14A、レーン4)。加えて、TSG101(1−312)は、GPおよびVP40とともに発現されたときに培養上清から抗−GP抗体によって共免疫沈降した(図14A、レーン4)。VP40不在下で発現されたときにはGPと会合したTSG101は見出されず(図14A、レーン3)、そのGPとの会合はVLPの形成に依存することが示唆された。全長TSG101についても同様の結果が得られた。これらのデータはTSG101がVLPに取込まれることを強く示唆するものであり、TSG101がウイルス集合および/または出芽における役割を果たすという仮説を支持するものである。この結果をさらに実証するために、我々はまた、TSG101の存在について、不活性化しバンド精製したEBOV(iEBOV)を分析した。TSG101の存在について、5μgのiEBOVを免疫ブロットにより分析した。図6Bに示されるとおり、我々はiEBOV中に容易に検出可能なレベルのTSG101を見出し、エボラウイルス中へのTSG101の取込みを明らかに示した。
生化学的データおよびVLP放出データは、エボラウイルスの出現にTSG101が重要であることを示唆した。したがって、エボラザイール−95ウイルスに感染したヒーラ細胞におけるウイルス生成に対するポリクローナル抗−TSG101抗体“C”および“E”(HIVに対する阻害的影響を示した、実施例1および2を参照)の影響をテストした。ヒーラ細胞の単分子層を1MOIのウイルスとともに50分間インキュベートし、洗浄し、抗−TSG101抗体または対照ウサギ抗マウス抗体を含む培地を5μg/mlにて加えた。24時間後に上清を集め、放出されたウイルスを以前に述べたように(ババリら、2002, J Exp Med 195, 593-602)プラークアッセイによって数えた。図15に示すとおり、これらの抗体はヒーラ細胞上清へのビリオンの放出を部分的に阻害した。

- 前記流体は血液または血清である、請求項34に記載の方法。
- 前記哺乳動物はヒトである、請求項34または35に記載の方法。
- 前記TSG101抗体は、VRETVNVITLYKDLKPVL(SEQ ID NO:2)およびQLRALMQKARKTAGLSDLY(SEQ ID NO:3)からなる群より選択されるアミノ酸領域に含まれるエピトープに結合する、請求項36に記載の方法。
- 前記エンベロープウイルスは、ヒト免疫不全ウイルスI型(HIV−I)、ヒト免疫不全ウイルスII型(HIV−II)、マールブルグウイルス、およびエボラウイルスからなる群より選択される、請求項37に記載の方法。

- 前記TSG101抗体に結合された前記細胞は、前記TSG101抗体に結合する抗体を含むカラムを用いて除去される、請求項34または35に記載の方法。
- 哺乳動物におけるエンベロープウイルスによる感染を処置または防止する方法であって、前記哺乳動物に治療上または予防上十分な量のワクチン組成物を投与するステップを含み、前記ワクチン組成物はTSG101タンパク質を含むポリペプチドを含む、方法。
- 前記ポリペプチドは前記TSG101タンパク質のN−末端またはC−末端領域を含む、請求項41に記載の方法。
- 前記哺乳動物はヒトである、請求項42に記載の方法。
- 前記ポリペプチドは、VRETVNVITLYKDLKPVL(SEQ ID NO:2)およびQLRALMQKARKTAGLSDLY(SEQ ID NO:3)からなる群より選択されるアミノ酸配列、またはその少なくとも5アミノ酸の断片を含む、請求項43に

- 前記ポリヌクレオチド分子は、前記TSG101タンパク質のN−末端またはC−末端領域を含むポリペプチドをコードする、請求項46に記載の方法。
- 前記哺乳動物はヒトである、請求項47に記載の方法。
