Abstract
Description
非晶相、ナノ結晶相、または結晶相の存在は、示差走査熱量分析装置(DSC)によって検出される。薬剤結晶の鋭い融解ピークに比べ、ナノ結晶は広いピークを示し、同時に温度の最高値が顕著に低くなる(I.コロンボ(Colombo)ら、4th Int.Conf.Pharm.Technol.、1986年、F.カーリ(Carli)ら、Acta Pharm.Jugosl.、第38巻、361、1988)。非晶相は、いかなる熱事象も示すことがない。
長さ50cm、直径0.6cmの円筒形の反応器中に装入された架橋ポリビニルピロリドン5gを、8時間、ニメスリドで飽和した二酸化炭素(CO2)の形態をとる、450mL/分の超臨界流と接触させる。温度および圧力はそれぞれ、40℃および130barである。

Claims (22)
- 薬剤を架橋ポリマーに組込むプロセスであって、
a.前記架橋ポリマーを超臨界流体で前処理するステップと、
b.前記前処理された架橋ポリマーを溶解された薬剤を含有する超臨界流体と接触させるステップと、
c.該超臨界流体を除去し、それによって該架橋ポリマー内部に該薬剤を凝結させるステップと
を含んで成るプロセス。 - ステップa.において、該架橋ポリマーが1分間〜6時間にわたり該超臨界流体と接触した状態で維持される、請求項1に記載のプロセス。
- ステップa.において、該架橋ポリマーが5分間〜4時間にわたり該超臨界流体と接触した状態で維持される、請求項1〜2のいずれかに記載のプロセス。
- ステップb.において、該前処理された架橋ポリマーが2分間〜48時間にわたり該超臨界流体と接触した状態で維持される、請求項1〜3のいずれかに記載のプロセス。
- ステップb.において、該前処理された架橋ポリマーが10分間〜12時間にわたり該超臨界流体と接触した状態で維持される、請求項1〜4のいずれかに記載のプロセス。
- 該架橋ポリマーと該超臨界流体との接触が、静的および/または動的条件下に実行される、請求項1〜5のいずれかに記載のプロセス。
- 前記超臨界流体が、二酸化炭素、エチレン、プロピレン、クロロフルオロカーボン、亜酸化窒素、およびその混合物の中から選択される、請求項1〜6のいずれかに記載のプロセス。

- 5分間〜4時間にわたり該超臨界流体で該架橋ポリマーを処理することによって得られる、請求項16〜17のいずれかに記載の変性架橋ポリマー。
- 該超臨界流体が、二酸化炭素、エチレン、プロピレン、クロロフルオロカーボン、亜酸化窒素、およびその混合物の中から選択される、請求項16〜18のいずれかに記載の変性架橋ポリマー。
- 変性すべき該架橋ポリマーが、架橋ポリビニルピロリドン、架橋セルロース誘導体、でんぷんおよびその誘導体、シクロデキストリン類およびその誘導体、架橋ポリスチレン、架橋アクリルポリマー、およびその混合物の中から選択される、請求項16〜19のいずれかに記載の変性架橋ポリマー。
- 薬剤が組込まれた、請求項16〜20のいずれかに記載の変性架橋ポリマー。
- 請求項21に記載された該変性架橋ポリマーを含有する医薬組成物。
