Abstract
【解決手段】宿主脊椎動物によるこの転写ユニットの取込みの結果として、目的の抗原(単数または複数)の発現が生じ、これにより体液性もしくは細胞性免疫応答、又は体液性及び細胞性免疫応答の両方が誘導される。誘導された体液性及び細胞性免疫応答は病原体による感染に対する防御を提供し、抗腫瘍応答を提供し、又は避妊を提供する。宿主は脊椎動物、鳥類、ヒトを含む哺乳動物のいずれでもよい。

Description
ロタウイルス又は免疫不全ウイルスに対する防御的免疫応答を誘導することによる脊椎動物免疫化に使用するための医薬の製造における、プロモータ領域のDNAに機能的に連結しかつ目的抗原をコードしているDNAを含んでなるDNA転写ユニットの使用であって、この防御的免疫応答が目的抗原に対して誘導される体液性免疫応答及び/又は細胞性免疫応答であるDNA転写ユニットの使用
に関する。
(1)サブグループA〜FなどのHIV−1の異なるサブグループを表すEnv。これらは広い地理的防御を与えるために使用される(マイヤース(Myers) ら、Human Retrovirusesand AIDS, Los Alamos National Laboratory, Los Alamos NM (1992))。
(2)感染の異なる相の指標となる生育特徴を示す又は異なる組織屈性(tissue tropisms) を有するサブグループ内からのEnv。これらは風土性の感染に存在するウイルスのスペクトラムに対する防御のために用いられる。
(3)異なる伝達経路に好ましいものの代表的なEnv。これらには、同性愛的伝達、異性愛的伝達、静脈内薬物使用による伝達、胎盤経由伝達又は新生児感染が挙げられる。
(4)目的の免疫応答を生起させるのに特に有効なEnvの突然変異型。このようなEnvの1例はヨゼフ ソドロスキー(Joseph Sodroski) 博士(ダナ・ファーバー・キャンサー・インスティテュート、ボストン、MA)によるV1、V2及びV3を欠失したHXB−2 Envである。このEnvはCD−4結合ドメインと関連するコンホーメーションを持つエピトープを保持しており、広い中和活性を持つ抗体を発生させるのに有効である(ワイアット(Wyatt) ら、1993、J. Virol. 67 4557-4565) 。
インフルエンザウイルスに対するニワトリの免疫化
インフルエンザウイルス血球凝集素タイプ7(H7)遺伝子を発現する複製可能なトリ白血症ウイルスをコードする、pP1/H7(図1)と呼ぶDNA転写ユニットをハント (Hunt)ら、J. of Virology, 62(8), 3014-3019 (1988) の記載のように構築した。H7を発現するが鳥ウイルスベクターポリメラーゼ及びエンベロープ蛋白質を欠損しているpP1/H7の複製欠損誘導体をコードするDNAユニットp188(図2)を、pP1/H7からXbaI断片を欠失させることにより構築した。トリ白血症ウイルスベクターをコードし、インフルエンザウイルス挿入物を持たないDNAユニットpRCAS(図3)を、ヒューズ(Hughes)ら、J. of Virology, 61, 3004 (1987) に記載されたように構築した。DNAユットは接種のために0.2ml当たり100μgの濃度に食塩水で希釈した。

複製欠損H7を発現するDNAで免疫化することにより誘導される防御の再現性を評価するために、p188及びpRCASのDNAのみを接種に用いて上述の実験を3回繰り返した。実験条件の相違は、最初の実験におけるブースト後1週間の代わりにブースト後2週間目にチャレンジしたことであった。3週齢のSPAFASニワトリに、3ルート(iv、ip及びsc)のそれぞれから100μgのDNAを接種した。4週間後にiv、ip及びsc投与により各100μgのDNAを接種してブーストした。2週間後に、ニワトリに鼻孔経由でCk/Vic/85(H7N7)の50%致死量の100倍量(100 lethal doses 50)をチャレンジした。

ワクチン投与された動物と投与されない動物のH7に対する抗体応答の分析
ワクチン投与されたニワトリとワクチン投与されないニワトリのH7に対する抗体応答の比較をするために、実施例1の実験2(表2を見よ)に抗インフルエンザAウイルス薬であるアマンタディン塩酸塩(ウエブスター(Webster), R. G.ら、J. Virol. 55, 173-176 (1985)) を投与したワクチン非投与群を含めた。5羽のアマンタディン処理トリのすべてが発病した。これらのうちの4羽は生き残り、それらから免疫化ニワトリと非免疫化ニワトリのH7に対する抗体応答を比較するために使用できる血清が得られた。
非レトロウイルス転写ユニットを用いるニワトリの免疫化
この実験は、レトロウイルスDNAを欠くDNA転写ユニットを本明細書に記載の方法により防御的免疫応答を形成させるために使用しても成功することを証明するために行われた。ニワトリにワクチン投与するためこの実験に用いたベクターを図4及び図5に示す。図4はサイトメガロウイルス(CMV)即時型プロモーターの転写制御の下にインフルエンザウイルスH7サブタイプ血球凝集素を発現することができるプラスミドである、pCMV/H7の図式的表示である。図5はインフルエンザ抗原を発現することができない対照のプラスミドである、pCMV/対照を示す。これらのプラスミドは、ブライアン カレン(Bryan Cullen)博士、デューク大学、ダーハム、ノースキャロライナ(Cullen, B.
R., Cell 45, 973-982 (1986)) のpBC12/CMVベクターの誘導体である。
pCMV/H1DNAを用いるワクチン投与によるマウスの免疫化:種々の接種ルートの分析
pCMV/H1と呼ぶDNA転写ユニットは、マウス適応A/PR/8/34H1N1インフルエンザウイルスでの致死量チャレンジに対しマウスを免疫化するために使用して成功した。この転写ユニットはCMV即時型プロモーターの転写調節の下にインフルエンザタイプH1血球凝集素をコードする。この構築に用いられたH1インフルエンザウイルス血球凝集素遺伝子は、ウインターズ(Winters) ら、Nature 292, 72 (1981) により詳細に記載されており、そしてチャレンジウイルスにおけるH1インフルエンザウイルス血球凝集素遺伝子と同一である。ベクターはH7の発現のために実施例3で使用したものと同一である(図5を見よ)。pCMV/H1転写ユニットは図6に図示してある。
ニワトリのインフルエンザモデルにおけるpCMV/H7の異種経路接種による防御的免疫化
DNAワクチン投与に対する接種経路の影響を、トリのインフルエンザウイルスモデルでさらに評価した。実施例1に記載したように、ニワトリにおける接種経路についてDNAワクチン投与実験を行った。接種経路としては次のものが含まれる。静脈内(羽根静脈)、筋肉内(胸筋肉)、気管内(DNA滴を気管内に投与)、皮下(うなじ)、粘液嚢内(ニワトリの肛門(vent)の直ぐ上に注射)、及び眼窩内(DNA滴を眼に投与)。一般に、100又は200μgのDNAを200μlの食塩水に溶かして投与した。結果を下記の表7に示す。p188DNA及びpRCASDNAのデータは3回の独立の実験の集計である。pCMV/H7DNA及びpCMV/対照DNAのデータは2回の独立の実験の集計である。確率は対照群に対するワクチン投与群の生存率及び死亡率を比較してフィッシャーの直接両側検定を用いて計算した。示された経路は、静脈内(iv)、腹腔内(ip)、皮下(sc)、筋肉内(im)、気管内(it)、粘液嚢内(ib)、及び眼窩内(io)である。

マウス表皮へのDNAの遺伝子銃送達(gene gun delivery) による防御的免疫化
表皮へDNAを送達するためにDNA銃を使用することができるか否かをテストするために、アクセル・パーティクル・ボムバードメント・デバイス(アグラセツス、ミドルトン、WI)を用いて、マウスの表皮にDNA被覆金ビーズを送達した。これらの実験はアグラセツス,インクのジョエル R.ヘインズ(Joel R. Haynes)博士と共同で行った。
pCMV/H1のワクチン投与を受けたマウスにおける抗体応答
実施例4及び6に上記したネズミの実験において、各DNAの接種の直前と、チャレンジの直前と、及びチャレンジ後に2回、それぞれ血清を採取した。麻酔したマウスの眼の静脈から40μlの非ヘパリン化ミクロヘマトクリット管に採血した。同一グループ内のメンバーから得た血清をプールした。ニワトリの赤血球細胞及びカオリンで前処理してバックグラウンド活性を除去したマウスの血清を用いて血球凝集阻止試験を行った(ノバック(Novak M.)ら、Vaccine 11, 55-60 (1993)) 。血球凝集阻止タイターは血球凝集の完全阻止を与える最大血清希釈度の逆数である。マウス抗体のイソタイプは、標準プロトコールと精製A/PR/8/34(H1N1)インフルエンザウイルスで被覆されたミクロウエルプレートを用いる酵素結合イムノソルベント検定(ELISA)により決定した。これらの検定は、タイターで類似の活性を持つイソタイプ特異的ペルオキシダーゼ結合抗体(シグマ・イムノケミカルズ)の1:1000希釈を使用した。血清分析のデータは下記の表9に示す。データは所与の条件に対し陽性のスコアであった血清プールの最終希釈の逆数の幾何平均である。
* 3種の血清プールの一つのみについてHI活性を試験した。ワクチン投与前=DNAワクチン投与前の採血、10d PB=第2回目DNA接種後10日目に採血(チャレンジ前)、4d PC=チャレンジ後4日目に採血。#テスト=血清プールが試験された群の数(そのプールに寄与する動物の血清の数)。NT=試験しなかった。<=この試験で用いられた最小希釈度の血清でも活性は検出されなかった。
A/PR/8/34(H1N1)インフルエンザチャレンジに対するフェレット獣を防御するためのpCMV/H1・DNA転写ユニットの使用
フェレット獣のインフルエンザモデルはヒトのインフルエザ感染に多くの類似性を有するので、このインフルエンザモデルにおけるpCMV/H1・DNA免疫化に関する研究を行った。最初の実験では、生理食塩水中の精製pCMV/H1のDNAを用い、1ヶ月間隔で筋肉内接種によりフェレットを免疫化した。若い雌成獣フェレットを前採血しついで後足それぞれに125μlずつ2回注射し総量500μlを接種することにより、食塩水中の500μgのpCMV/H1・DNA又はpCMV/対照DNAを用いてワクチン投与した。1頭のフェレットは1ヶ月間隔で500μgのpCMV/H1・DNAの筋肉内接種を3回受け、一方第2の動物は1ヶ月間隔で500μgのpCMV/H1・DNAの筋肉内接種を2回受けた。対照の動物は1ヶ月間隔で500μgのpCMV/対照DNAの筋肉内接種を3回受けた。
免疫誘導の効率を高めるために、フェレットでアクセル遺伝子銃を用いてDNA被覆金ビーズをフェレットの皮膚内に送達する第2の実験を行った。腹部表皮を遺伝子銃送達DNAの標的として用い、1ヶ月間隔で2回の遺伝子銃DNA投与をフェレットに行った。遺伝子銃接種をケタミンで麻酔した若い雌成獣フェレットに施した。皮膚は剃り、脱毛剤ネール(NAIR)(カーター−ワラス、ニューヨーク)で処理した。前述のように、DNAビーズ(1〜3ミクロン)を接種用に調製した(ファイナン(Fynan) ら、Proc. Natl. Acad. Sci. USA 90, 11478-11482 (1993))。接種には、15kVの送達電圧を用いた。フェレットを2μg又は0.4μgのDNAで接種した。2μgのDNAを接種されたフェレットは0.2μgのDNAを被覆された0.8mgのビーズからなるショットを10回受けた。0.4μgのDNAを受けたフェレットは同様のショットを2回受けた。
粘膜投与のための微小体カプセル化DNA
致死量のインフルエンザウイルスチャレンジに対する防御的応答を発生させる微小体カプセル化DNAの能力を試験することにより、DNA接種のための粘膜経路をさらに発展させた。ネズミインフルエンザウイルスモデルをこれらの研究に使用した。ウイルス・リサーチ・インスティチュート,インク,ケムブリッジ,MAにおいて、pCMV/H1DNA及びpCMV/対照DNAをアルギン酸塩の微小体中にカプセル化した。各群が一次接種及びブーストを受ける実験を行った。A群は一次接種として遺伝子銃送達DNAの0.4μgを受け、ブーストは受けなかった。B群は一次接種及びブーストとして0.4μgの遺伝子銃送達DNAを受けた。C群は一次接種として0.4μgの遺伝子銃送達DNAを受けそしてブーストとしてアルギン酸塩カプセル化DNAの100μgを受けた。D群は一次接種及びブースト接種の両方において100μgのアルギン酸塩カプセル化DNAを受けた。アルギン酸塩カプセル化DNAの各投与はメトファンで麻酔されたマウスの鼻孔に100μlの水に懸濁して送達することにより行った。第2のDNA接種後10日目に、メトファンで麻酔したマウスの鼻孔にA/PR/8/34(H1N1)インフルエンザウイルスの500pfuを投与して致死量チャレンジを行った。4頭の対照群は同量のpCMV/対照DNAを受けた後pCMV/H1DNAを受けた群と同様の処理を受けた。この実験のデータを表13に示す。

ロタウイルス蛋白質をコードするDNA転写ユニットを用いるマウスの免疫化
マウスを免疫化する能力についてロタウイルスDNA転写ユニットを試験した。マウスについてロタウイルスに対するワクチン投与のためにこの実験で用いたpCMV/VP7ベクターは、プラスミドpCMV/VP7がネズミロタウイルス中和キャプシド蛋白質VP7を発現することができるものである点を除き、図4及び6に示すものと類似している。VP7DNAはハリー グリーンバーグ(Harry Greenberg)博士、スタンフォード大学、Palo Alto, CA, USAから入手した。
*抗VP7抗体は全マウスEDIMロタウイルスに対しELISAにより検定した。
HIV−1に対する免疫化のためのDNA構築物
HIV−1に対する免疫化のために2系列のDNA転写ユニットを調製する。その第一はpBC12/CMVベクター(上記及び図5を見よ)を用いてHIV−1配列のための転写制御要素を提供する。pBC12/CMVベクターにおいては、HIV−1蛋白質の発現はRev依存性である。第二の系列はJames I. Mullinsラボラトリー(スタンフォード大学)(Palo Alto, CA)で開発されたJW4303ベクターを用いる(図8を見よ)。これらのベクターはEnvのRev非依存性発現を支持する。JW4303ベクター及びそれに伴うオリゴヌクレオチドはHIV−1単離物すべてのEnvのPCR増幅断片のクローニングを容易にするように設計される。
pBC12/CMVに基づくベクター中へのクローニングはすべてpBC12/CMV/IL−2中で行った。具体的には、挿入断片は、IL−2のcDNAのBamHIからHinDIII までの断片と置換された。これらのクローニングには3種の挿入物が用いられた(図10〜12)。
図9は、HIV−1−NL4−3(NL4−3)プロウイルスDNA及びその会合した長末端反復(LTR)配列及び読み取り枠(open reading frames) を図示する。pNL4−3はマルコーム(Malcolm, A.) 博士、マーチンス・ラボラトリー(National Institutes of Health, Bethesda, MD) から提供された( アダチ(Adachi)ら、J. Virol. 59, 284-291 (1986)) 。HIV.NL4−3株のジーンバンク受託番号はM1991である。NL4−3.dpol挿入物は非感染性HIV−1粒子をコードし、生のしかし非感染性の感染を真似るように構築された。非感染性の粒子をコードする挿入物を得るために、それぞれHaeII及びBanII消化を用いて5’−LTRのすべて及び3’−LTRの大部分をpNL4−3から欠失させた。1932bpの内部BalI欠失によりpol遺伝子を非機能性とした。トランスフェクトしたコス細胞のウエスタンブロット分析を用いてGag及びEnvの発現が証明された。Gag蛋白質とEnv蛋白質は細胞内及び培養基の中の両方に存在した。このことは、Gagが粒子形成に要求される唯一のHIV−1蛋白質であることからも予想された。
HXB−2.envは完全なHXB−2Env及びRevを発現するように設計された。HIV.HXB2株のジーンバンク受託番号はK03455及びM38432である。正常なHIV−1のEnvの発現はRev依存性であるから、Revはこの構築物中に含まれる。この構築物はヨゼフ ソドロスキー(Joseph Sodroski) 博士(ダナ・ファーバー・キャンサー・インスティチュート,ボストン,MA)(ヘルゼート(Helseth) ら、J. Virol. 64, 2416-2420 (1990)) のpSVIII.env 構築物のSalIからXhoIまでの断片をpBC12/CMV/IL−2のIL−2のBamHIからHindIII 断片と置換することにより得られた。トランスフェクトしたコス細胞のウエスタンブロット分析により、Envの発現が証明された。
HIV−1Env及びRevを発現する構築物の第二の例であるNL4−3.envはHXB−2/NL4−3Env融合蛋白質及びRevを発現した。この構築物においては、HXB−2envの末端付近のユニークな制限部位であるKpnI及びBamHIを用いてNL4−3配列をpCMV/HXB−2.env内の相同的なHXB−2env配列と置換した。トランスフェクトしたコス細胞のウエスタンブロット分析によりEnvの発現が証明された。
JW4303プラスミドは、CMV即時型プロモーター由来の約2000bpとウシ成長ホルモン由来の配列を挿入物発現のために使用する(図8)。CMV即時型プロモーター由来の配列はCMVイントロンAをコードする配列を含む。このイントロンは挿入された遺伝子の発現を高めることができる(チャップマン(Chapman) ら、Nucleic Acids Research14, 3979-3986 (1991))。JW4303ベクターは組織プラスミノーゲン活性化因子(TPA) 蛋白質に対する合成リーダー配列を含む。この合成リーダーはEnv発現の開始部位を提供する。組織プラスミノーゲン活性化因子のリーダーはグリコシル化蛋白質の合成及び分泌を加速する(ヘイグウッド(Haigwood)ら、Prot. Eng. 2, 611-620 (1989)) 。デザイナーオリゴヌクレオチドからのPCR増幅を用い、TPAリーダーとフレームを合わせて挿入されたenv断片を作成した。HIV−1の異なるサブグループの配列に共通な5’−オリゴヌクレオチドをHIV−1env配列のどれかと成熟Envの正常末端の又はその付近のTPAリーダーに融合させる。このオリゴヌクレオチドはJW4303中へのサブクローニングのために合成TPAリーダー内のユニークな制限部位を使用できるように設計される。例えば、5’−オリゴヌクレオチドJApcr503は、HIV−1のサブグループB単離物に対する成熟Envのアミノ酸6の直前でTPAリーダーの融合を可能とするXbaI部位を含む。
HXB−2.sgp120(図14を見よ)は、JApcr503(gtcgctcctctagattgtgggtcacagtctattatggggtacc)(配列表の配列番号:1)とJApcr504(ggtcggatccttactgcaccactcttctctttgcc)(配列表の配列番号:2)を用いてpCMV/HXB−2.envからの配列を増幅して合成した。この増幅された断片をXbaIとBamHIで消化し、NheIとBamHIで消化したJW4303中にサブクローニングした。トランスフェクトしたコス細胞のウエスタンブロット分析により、トランスフェクトした細胞内及び培養基内の両方にsg120の存在が明らかにされた。
HXB−2.sgp140(図15を見よ)は、JApcr503(配列表の配列番号:1)とJApcr502(cgacggatccttatgttatgtcaaaccaattccac)(配列表の配列番号:3)を用いてpCMV/HXB−2.envからの配列を増幅することにより構築した。増幅された断片をXbaIとBamHIで消化し、NheIとBamHIで消化したJW4303中にサブクローニングした。トランスフェクトしたコス細胞のウエスタンブロット分析により、トランスフェクトした細胞内及び培養基内の両方にsg120の存在が明らかにされた。
HIV−1−NL4−3 pBC12CMVに基づくベクターの免疫原性テスト
pBC12/CMVに基づくベクターで免疫化する試みをBALB/cマウスで行った。6〜8週齢のマウスを静脈内(iv)及び筋肉内(im)経路の両方でDNA200μgの一連の注射により免疫化した(免疫化のスケジュールは図17に示す)。6尾ずつのマウスからなる4実験群にDNAを投与した。A群は挿入物のないpCMV/対照DNAを投与された。B群はpCMV/NL4−3.envDNAを受けた。C群はpCMV/NL4−3.dpolDNAを受けた。D群はpCMV/NL4−3.envDNA100μgとpCMV/NL4−3.dpolDNA100μgの混合物を受けた。マウスを免疫化に先立って採血し、免疫化後の種々の時点で採血した。各採血時に同一テスト群のマウスからの血清をプールした。実験の終わりにマウスを屠殺し脾臓を採取し、BALB/cマウスのNL4−3Env内の既知の細胞障害性T細胞(CTL)エピトープに対するCTL活性について試験した。
SIVmac に対する免疫化のためのDNA構築物
SIV構築物
HIV−1の場合と同様に、SIVmac に対する免疫化のために2系列のDNA転写ユニットを調製した。これらの第1は、ブライアン R.クレン(Cullen)博士のpBC12/CMVベクターを使用する(上記及び図5を見よ)。第2の系列はジェームス I.ムリンス ラボラトリー(上記及び図8を見よ)で開発されたJW4303ベクターを用いた。
pBC12/CMVに基づくベクターへのクローニングはpBC12/CMV/IL−2中で行われた。SIV239挿入物はSIV239プロウイルスDNAをコードするプラスミドから調製された(図26)。これらのプラスミド(p239SpSp5’及びp239SpE3’)は、ロナルド C.デスロジアース(Desrosiers)博士、ニューイングランド・リージョナル・プライメート・リサーチ・センター(サウスボロー、MA)により提供された。具体的には、pCMV/SIV239.dpol(239.dpol)挿入物(図27)をpBC12/CMV/IL−2内のIL−2cDNAのBamHIからHindIII までの断片と置換した。この239.dpol挿入物は、NarI欠失により5’LTR非機能性とし、内部のBstEII欠失によりpol非機能性とし、そしてStuI消化でLTRの大部分を除去することにより構築した。p239SpE3’は、点彩により示された欠陥nef遺伝子をコードする。トランスフェクトしたコス細胞のウエスタンブロット分析を用いてGag及びEnvの発現が証明された。Gag及びEnv蛋白質は細胞内及び培養基内の両方に存在した。このことはGagが粒子形成のために要求される唯一のSIV−1蛋白質であることから予期されたとおりであった。
SIVに対するJW4303・DNA転写ユニットはSIVenv配列のPCR増幅断片を用いて構築した(図26−29)。5’センスプライマーはTPAリーダーとの融合蛋白質をコードするDNAの構築において役に立った。3’アンチセンス オリゴヌクレオチドを用いてsgp120、sgp140、及び全長SIVenv断片を作成した。
239.sgp120は、p239spE3’からの配列を増幅するためにオリゴヌクレオチドJApcr19及びオリゴヌクレオチドJWpcr8を用いて合成した。増幅された断片をNheI及びBamHIで消化し、NheI及びBamHIで消化したpJW4303中にサブクローニングした。トランスフェクトしたコス細胞のウエスタンブロット分析により、トランスフェクト細胞内及び培養基内の両方にsg120が見出された。SIV239は赤毛猿(macaques)におけるワクチン投与実験のための確立されたモデルであるから、この構築に239配列を使用した。SIV239はSIV251の突然変異体であり、後者も構築物の作成に使用される(以下を見よ)。SIV239株に対するジーンバンク受託番号はM33262、M61062、及びM61093である。SIV251株に対するジーンバンク受託番号はM19499及びX06393である。
239.sgp140は、p239SpE3’からの配列を増幅するためにオリゴヌクレオチドJApcr19とオリゴヌクレオチドHKpcr2を用いて構築された。増幅された断片をNheIとBamHIで消化し、NheIとBamHIで消化したpJW4303中にサブクローニングした。トランスフェクトしたコス細胞のウエスタンブロット分析により、トランスフェクトした細胞内及び培養基内の両方にsg140が見出された。上に指摘したように、239配列が用いられたのは、239ウイルスが赤毛猿のワクチン投与実験のためのモデルとして確立されているからである。
251.sgp140は、pM40KSIV251env(ロナルド C.デスロジアース(Desrosiers)博士、ニューイングランド・プライメート・リサーチ・センター、サウスボロー、MAから入手した)からの配列を増幅するためにオリゴヌクレオチドJapcr19とオリゴヌクレオチドHkpcr2を用いて構築した。増幅された断片をNheI及びBamHIで消化し、NheI及びBamHIで消化したJW4303中にサブクローニングした。トランスフェクトしたコス細胞のウエスタンブロット分析により、トランスフェクト細胞内及び培養基内の両方にsg140が見出された。
316.sgp140は、ロナルド C.デスロジアース (Desrosiers)博士、ニューイングランド・プライメート・リサーチ・センターから入手したPCRenvクローン316−3からの配列を増幅するためオリゴヌクレオチドJapcr19とオリゴヌクレオチドHkpcr2を用いて構築した。増幅された断片をNheI及びBamHIで消化し、NheI及びBamHIで消化したpJW4303中にサブクローニングした。トランスフェクトしたコス細胞のウエスタンブロット分析により、トランスフェクト細胞内及び培養基内の両方にsg120が見出された。SIV316はSIV239の突然変異体であり、後者は単球/マクロファージ屈性 (tropism) を有する(モリ(Mori) K. ら、J. Virology 66(4), 2067-2075 (1992))。
SIV DNAワクチン投与実験の設計
赤毛猿を免疫化するためにSIVをコードするDNAを用いてワクチン投与実験を試みた。雄及び雌の若い免疫能力を有する動物をこの実験に用いる。三回のDNA接種が実験の1週と3週、11週と13週、及び21週と23週に行われる。最終DNA接種の2週間後に致死量チャレンジが投与される。このチャレンジは、静脈内接種により投与されるSIV239の10猿感染ユニットからなる。
ワクチン転写ユニットに使用するための患者単離物からのHIV−1env配列の分子クローニング
健康なセロポジティブな感染相に特徴的なゆっくりとした/低い、非シンシチウム誘導性の単球/マクロファージ屈性ウイルスを表すサブグループB HIV−1単離物に対するenv配列、並びにAIDS患者に見出される急速な/高い、シンシチウム誘導性のT細胞株屈性ウイルスを表すenv配列を得るために、2系列の一連の患者単離物をエバ マリア フェンヨ(Fenyo) 博士、カロリンスカ・インスティチュート、スエーデンから入手した(Von Gegerfelt, A. ら、Virol. 185, 162-168 (1991)( 表16を見よ)。これらの単離物は感染の健康なセロポジティブな相からAIDS相までの進行の期間の2〜3年間にわたって得られたものであった。一系列は患者5からのものであり、第2は患者6からのものであった。
当技術分野に熟練せる者は単なる日常的実験を用いて本明細書に記載された発明の具体的態様に均等な多くの事を認識し又は確認することができるであろう。これらのそしてその他のこのような均等物は以下の請求の範囲に包含されるものである。
[1] ロタウイルス又は免疫不全ウイルスに対する防御的免疫応答を誘導することによる脊椎動物免疫化に使用するための医薬の製造における、プロモータ領域のDNAに機能的に連結しかつ目的抗原をコードしているDNAを含んでなるDNA転写ユニットの使用であって、この防御的免疫応答が目的抗原に対して誘導される体液性免疫応答及び/又は細胞性免疫応答であるDNA転写ユニットの使用。
[2] ロタウイルス又は免疫不全ウイルスに対する防御的免疫応答を誘導することにより脊椎動物を免疫化する方法であって、該方法はプロモータ領域のDNAに機能的に連結しかつ目的抗原をコードしているDNAを含んでなるDNA転写ユニットを脊椎動物に投与することを含んでなる方法であり、これによる防御的免疫応答が目的の抗原に対して誘導される体液性免疫応答及び/又は細胞性免疫応答である方法。
[3] DNA転写ユニットを含んでなる生産物であって、この転写ユニットがプロモータ領域のDNAに機能的に連結しかつ目的抗原をコードしているDNAを含んでなるものであり、このDNAが微小体でカプセル化されている(microsphere encapsulated)ものである生産物。
[4] 目的の抗原に対して誘導される体液性免疫応答及び/又は細胞性免疫応答を誘導することによる脊椎動物免疫化における使用などの治療における使用のための、微小体でカプセル化されているDNA転写ユニットを含んでなる生産物であって、該転写ユニットがプロモーター領域のDNAに機能的に連結しかつ目的抗原をコードしているDNAを含んでなるものである生産物。
[5] 目的の抗原に対して誘導される体液性免疫応答及び/又は細胞性免疫応答を誘導することによる脊椎動物免疫化に使用するための医薬の製造のための、微小体でカプセル化されているDNA転写ユニットの使用であって、該転写ユニットがプロモーター領域のDNAに機能的に連結しかつ目的抗原をコードしているDNAを含んでなるものである、使用。
[6] 脊椎動物を免疫化する方法であって、該方法が微小体でカプセル化されたDNA転写ユニットを脊椎動物に投与することを含み、このDNA転写ユニットがプロモーター領域のDNAに機能的に連結しかつ目的抗原をコードしているDNAを含んでなるものであり、これによる体液性免疫応答及び/又は細胞性免疫応答が目的抗原に対するもの/又は目的抗原に対して誘導されるものである、方法。
[7] 1個以上のDNA転写ユニットを含んでなる生産物であって、転写ユニットのそれぞれがプロモーター領域のDNAに機能的に連結しかつ目的抗原をコードしているDNAを含んでなるものであり、これによる体液性免疫応答及び/又は細胞性免疫応答が目的抗原に対するもの/又は目的抗原に対して誘導されるものであり、一つの転写ユニットに対する目的抗原が他の転写ユニットの目的抗原もしくは複数の他の転写ユニットそれぞれの目的抗原とは異なるものである、生産物。
[8] 治療に使用するための1個以上のDNA転写ユニットを含んでなる生産物であって、転写ユニットのそれぞれがプロモーター領域のDNAに機能的に連結しかつ目的抗原をコードしているDNAを含んでなるものであり、一つの転写ユニットに対する目的抗原が他の転写ユニットの目的抗原もしくは複数の他の転写ユニットそれぞれの目的抗原とは異なっており、該治療における使用が、例えば該生産物を投与することにより脊椎動物を免疫化する方法であって、これによる体液性免疫応答及び/又は細胞性免疫応答が該目的抗原に対するものであり/又は該目的抗原に対して誘導されるものである、生産物。
[9] 脊椎動物に生産物を投与することによる該脊椎動物を免疫化するための医薬の製造における1個以上のDNA転写ユニットを含んでなる生産物の使用であって、該転写ユニットのそれぞれがプロモーター領域のDNAに機能的に連結しかつ目的抗原をコードしているDNAを含んでなるものであり、これによる体液性免疫応答及び/又は細胞性免疫応答が目的抗原に対するもの/又は目的抗原に対して誘導されるものであり、一つの転写ユニットに対する目的抗原が他の転写ユニットの目的抗原もしくは複数の他の転写ユニットそれぞれの目的抗原とは異なっているものである、使用。
[10] 脊椎動物を免疫化する方法であって、該方法は1個以上のDNA転写ユニットを脊椎動物に投与することを含んでなる方法であり、該転写ユニットはそれぞれプロモーター領域のDNAに機能的に連結しかつ目的抗原をコードしているDNAを含んでなるものであり、これによる体液性免疫応答及び/又は細胞性免疫応答が目的抗原に対するもの/又は目的抗原に対して誘導されるものであり、一つの転写ユニットに対する目的抗原が他の転写ユニットの目的抗原もしくは複数の他の転写ユニットそれぞれの目的抗原とは異なっているものである、方法。
[11] 異なる抗原がインフルエンザウイルス由来の防御的応答を誘導するものである、前記[7]、[8]、[9]又は[10]いずれか記載の生産物、使用又は方法。
[12] 異なる抗原がインフルエンザの異なるサブタイプ由来のものである前記[11]記載の生産物、使用又は方法。
[13] 異なる抗原がインフルエンザの異なるサブグループ由来のものである前記[11]記載の生産物、使用又は方法。
[14] 異なる抗原がインフルエンザの異なるサブタイプ由来のもの及びインフルエンザの異なるサブグループ由来のものである前記[11]記載の生産物、使用又は方法。
[15] 異なる抗原が免疫不全ウイルス由来の防御的応答を誘導するものである、前記[7]、[8]、[9]又は[10]いずれか記載の生産物、使用又は方法。
[16] 異なる抗原が免疫不全ウイルスの異なるサブグループを表すものである、前記[15]記載の生産物、使用又は方法。
[17] 異なる抗原が免疫不全ウイルスの異なる感染相(phases of infection)を表すものである、前記[15]記載の生産物、使用又は方法。
[18] 異なる抗原が免疫不全ウイルスの異なる組織屈性(tissue tropisms) を表すものである、前記[15]記載の生産物、使用又は方法。
[19] 異なる抗原が免疫不全ウイルスの異なる伝達ルートを表すものである、前記[15]記載の生産物、使用又は方法。
[20] 転写ユニットが微小体でカプセル化されているものである、前記[1]、[2]、又は[7]〜[19]いずれか記載の生産物、使用又は方法。
[21] 1個以上の転写ユニットが脊椎動物に投与されるものであり、一つの転写ユニットに対する目的抗原が他の転写ユニットの目的抗原又は複数の他の転写ユニットそれぞれの目的抗原とは異なっているものである、前記[1]〜[6]いずれか記載の生産物、使用又は方法。
[22] 前記[11]〜[19]のいずれかの特徴をさらに含む前記[21]記載の生産物、使用又は方法。
[23] 静脈内、筋肉内、腹腔内、皮内、及び皮下内からなる群より選択される投与経路を介して脊椎動物に投与するための先行する前記[1]〜[22]いずれか記載の生産物、使用又は方法。
[24] 粘膜表面への接触により脊椎動物へ投与するための前記[1]〜[22]いずれか記載の生産物、使用又は方法。
[25] 該粘膜表面が鼻粘膜表面又は気管粘膜表面などの呼吸器系粘膜表面である、前記[24]記載の生産物、使用又は方法。
[26] 転写ユニットのプロモーター領域がレトロウイルス起源のもの又は非レトロウイルス起源のものである、先行する前記[1]〜[25]いずれか記載の生産物、使用又は方法。
[27] 該脊椎動物がヒトなどの哺乳動物である、先行する前記[1]〜[26]いずれか記載の生産物、使用又は方法。

Claims (1)
- ロタウイルス又は免疫不全ウイルスに対する防御的免疫応答を誘導することによる脊椎動物免疫化に使用するための医薬の製造における、プロモータ領域のDNAに機能的に連結しかつ目的抗原をコードしているDNAを含んでなるDNA転写ユニットの使用であって、この防御的免疫応答が目的抗原に対して誘導される体液性免疫応答及び/又は細胞性免疫応答であるDNA転写ユニットの使用。
