Abstract
【解決手段】TLR2又はMyD88遺伝子の細胞内領域等を含むエクソン部位の全部または一部の遺伝子フラグメントを、ポリAシグナルとマーカー遺伝子をもつプラスミドに置換することにより構築したターゲッティングベクターを用いた相同組み換え法により作製する。

Description
他方、動物やヒトの病原性細菌として知られるマイコプラズマは細胞壁を欠如しているが、マクロファージを活性化する能力を備えている。これらマクロファージ活性化物質がリポタンパク/リポペプチドであるとの数多くの報告が現在までになされており、かかるリポペプチドのうちの1つであるマイコプラズマ・ファーメンタンス(M.fermentans)由来の2kDマクロファージ活性化リポペプチドMALP−2は、その生化学的特性が明らかになっており、かつ合成されたものも利用することができる(J.Exp.Med.185:1951,1997)。この脂質種は2つの不斉炭素原子をもち、S,Rラセミ体からなる合成MALP−2はインビトロでピコモル濃度で天然化合物作用と類似の特異活性を示すことが知られている。そして、MALP−2がNF−κBを活性化することを除いては、MALP−2のシグナル経路や細胞表面レセプターについては殆ど知られていない。
また本発明は、(10)上記(1)〜(9)のいずれか記載の細菌菌体成分に対して不応答性である非ヒト動物から得られるマクロファージ又は脾臓細胞と被検物質とをあらかじめインビトロで接触せしめた後、該マクロファージ又は脾臓細胞を細菌菌体成分の存在下で培養し、該マクロファージ又は脾臓細胞のマクロファージ活性又は脾臓細胞活性の程度を測定・評価することを特徴とする細菌菌体成分に対する応答性の抑制物質又は促進物質のスクリーニング方法や、(11)上記(1)〜(9)のいずれか記載の細菌菌体成分に対して不応答性である非ヒト動物から得られるマクロファージ又は脾臓細胞と細菌菌体成分とをあらかじめインビトロで接触せしめた後、該マクロファージ又は脾臓細胞を被検物質の存在下で培養し、該マクロファージ又は脾臓細胞のマクロファージ活性又は脾臓細胞活性の程度を測定・評価することを特徴とする細菌菌体成分に対する応答性の抑制物質又は促進物質のスクリーニング方法や、(12)上記(1)〜(9)のいずれか記載の細菌菌体成分に対して不応答性である非ヒト動物にあらかじめ被検物質を投与した後、該非ヒト動物から得られるマクロファージ又は脾臓細胞を細菌菌体成分の存在下で培養し、該マクロファージ又は脾臓細胞のマクロファージ活性又は脾臓細胞活性の程度を測定・評価することを特徴とする細菌菌体成分に対する応答性の抑制物質又は促進物質のスクリーニング方法や、(13)上記(1)〜(9)のいずれか記載の細菌菌体成分に対して不応答性である非ヒト動物にあらかじめ被検物質を投与した後、該非ヒト動物を細菌により感染させ、該非ヒト動物から得られるマクロファージ又は脾臓細胞のマクロファージ活性又は脾臓細胞活性の程度を測定・評価することを特徴とする細菌菌体成分に対する応答性の抑制物質又は促進物質のスクリーニング方法や、(14)上記(1)〜(9)のいずれか記載の細菌菌体成分に対して不応答性である非ヒト動物をあらかじめ細菌により感染させた後、該非ヒト動物から得られるマクロファージ又は脾臓細胞を被検物質の存在下で培養し、該マクロファージ又は脾臓細胞のマクロファージ活性又は脾臓細胞活性の程度を測定・評価することを特徴とする細菌菌体成分に対する応答性の抑制物質又は促進物質のスクリーニング方法や、(15)上記(1)〜(9)のいずれか記載の細菌菌体成分に対して不応答性である非ヒト動物をあらかじめ細菌により感染させた後、該非ヒト動物に被検物質を投与し、該非ヒト動物から得られるマクロファージ又は脾臓細胞のマクロファージ活性又は脾臓細胞活性の程度を測定・評価することを特徴とする細菌菌体成分に対する応答性の抑制物質又は促進物質のスクリーニング方法や、(16)上記(1)〜(9)のいずれか記載の細菌菌体成分に対して不応答性である非ヒト動物にあらかじめ被検物質を投与した後、該非ヒト動物を細菌により感染させ、該非ヒト動物におけるマクロファージ活性又は脾臓細胞活性の程度を測定・評価することを特徴とする細菌菌体成分に対する応答性の抑制物質又は促進物質のスクリーニング方法や、(17)上記(1)〜(9)のいずれか記載の細菌菌体成分に対して不応答性である非ヒト動物をあらかじめ細菌により感染させた後、該非ヒト動物に被検物質を投与し、該非ヒト動物におけるマクロファージ活性又は脾臓細胞活性の程度を測定・評価することを特徴とする細菌菌体成分に対する応答性の抑制物質又は促進物質のスクリーニング方法や、(18)マクロファージ活性又は脾臓細胞活性の程度を測定・評価するに際し、対照として細菌菌体成分に対して不応答性である非ヒト動物と同種の野生型非ヒト動物の測定値と比較・評価することを特徴とする上記(10)〜(17)のいずれか記載の細菌菌体成分に対する応答性の抑制物質又は促進物質のスクリーニング方法や、(19)マクロファージ活性の程度の測定・評価が、該マクロファージにおけるサイトカイン及び/又は亜硝酸イオンの産生量の測定・評価であることを特徴とする上記(10)〜(18)のいずれか記載の細菌菌体成分に対する応答性の抑制物質又は促進物質のスクリーニング方法や、(20)脾臓細胞活性の程度の測定・評価が、該脾臓細胞におけるMHCクラスIIの発現量の測定・評価であることを特徴とする上記(10)〜(18)のいずれか記載の細菌菌体成分に対する応答性の抑制物質又は促進物質のスクリーニング方法や、(21)細菌菌体成分が、リポタンパク/リポペプチドであることを特徴とする上記(10)〜(20)のいずれか記載の細菌菌体成分に対する応答性の抑制物質又は促進物質のスクリーニング方法や、(22)リポタンパク/リポペプチドが、マイコプラズマ属、スピロヘータ属又はエセリシア属に属する細菌の菌体成分由来のリポタンパク/リポペプチドであることを特徴とする上記(21)記載の細菌菌体成分に対する応答性の抑制物質又は促進物質のスクリーニング方法や、(23)細菌菌体成分が、ペプチドグリカンであることを特徴とする上記(10)〜(20)のいずれか記載の細菌菌体成分に対する応答性の抑制物質又は促進物質のスクリーニング方法や、(24)細菌菌体成分が、エンドトキシンであることを特徴とする上記(10)〜(20)のいずれか記載の細菌菌体成分に対する応答性の抑制物質又は促進物質のスクリーニング方法や、(25)細菌菌体成分が、リポテイコ酸であることを特徴とする上記(10)〜(20)のいずれか記載の細菌菌体成分に対する応答性の抑制物質又は促進物質のスクリーニング方法や、(26)細菌菌体成分が、結核菌溶解物であることを特徴とする上記(10)〜(20)のいずれか記載の細菌菌体成分に対する応答性の抑制物質又は促進物質のスクリーニング方法や、(27)細菌菌体成分に対する応答性の抑制物質又は促進物質が、細菌感染症に対する抑制物質又は促進物質であることを特徴とする上記(10)〜(26)のいずれか記載の細菌菌体成分に対する応答性の抑制物質又は促進物質のスクリーニング方法や、(28)マウス遺伝子ライブラリーからマウスESTクローン由来のプローブを用いてスクリーニングすることにより得られたMyD88遺伝子領域のC末端部分をコードした2つのエクソン領域の全部又は一部の遺伝子フラグメントを、ポリAシグナルとマーカー遺伝子をもつプラスミドに置換してターゲッティングベクターを構築し、該ターゲッティングベクターを線状化した後胚幹細胞に導入し、MyD88遺伝子機能を欠損した標的胚幹細胞を、マウスの胚盤胞中にマイクロインジェクションしキメラマウスを作製し、このキメラマウスと野生型マウスとを交配させてヘテロ接合体マウスを作製し、かかるヘテロ接合体マウスをインタークロスすることによって得られることを特徴とするMyD88ノックアウトマウスに関する。
MyD88遺伝子を129/SvJマウス遺伝子ライブラリー(Stratagene社製)からスクリーニングし、pBluescriptベクター(Stratagene社製)中でサブクローンし、制限酵素マッピング及びDNAシーケンシングにより特定した。ターゲッティングベクター(標的ベクター)は、pMC1−neo(Stratagene社製)からのネオマイシン耐性遺伝子で、野生型アレルの1.0kb遺伝子断片を置換することにより構築された。置換された遺伝子断片は、IL−1RAcP(受容体補助タンパク)の細胞質ドメインに似ているドメインをコードする2つのエクソンを含んでいた。ネオマイシン耐性遺伝子は、1.1kbの5′遺伝子断片と5.2kbの3′遺伝子断片をフランキング配列として有していた。次いで、HSV−tkカセットを遺伝子断片の3′端に導入した。線状化された標識ベクターをES細胞E14.1にトランスフェクションし、G418及びガンシクロヴィアで選択した。両者に抵抗性を示す176個のクローンを、PCRによる相同組換えのためにスクリーニングし、図1に示すプローブを用いるサザンブロット分析により33個を確かめた。
本発明のMyD88ノックアウトマウス10匹に、大腸菌(O55:B5)由来のLPSを1mg投与し、その生存率によりエンドトキシン不応答性を調べた。対照として同腹の野生型マウス10匹を用いた。結果を図2に示す。図2より、野生型マウスはLSPに応答し、投与後4日ですべて死亡したが、本発明のMyD88ノックアウトマウスは、LPS投与後4日では死亡するものはなく、エンドトキシン不応答性であることを確認することができた。
本発明のMyD88ノックアウトマウスの胸腺細胞1×105を、T細胞増殖についてのIL−1との共刺激物であるフィトヘマグルチニン(PHA)2μg/ml、コンカナバリンA(ConA)2.5μg/ml又は2ng/mlのIL−2のそれぞれと、IL−1β(Genzyme社)100U/mlとの混合物と共に96ウェルの培養皿で72時間培養し、T細胞を増殖させた。T細胞の増殖は、細胞内に取り込まれた[3H]チミジンの[3H]量を測定することにより求めた。その結果、PHA、ConA又はIL−2とIL−1βとの共存下で培養したとき、同腹子の野生型マウスの胸腺細胞は大いに増殖したが、本発明のMyD88ノックアウトマウスの胸腺細胞は細胞増殖の増加がほとんど見られなかった(図3参照)。また、胸腺細胞に代えて脾臓B細胞を用いても、同じような結果が得られることがわかった。
IL−18がNK細胞の溶解活性を増強することはよく知られている。本発明のMyD88ノックアウトマウス及び同腹子の野生型マウスの脾臓B細胞を、IL−18(林原生化学研究所株式会社)20ng/mlの存在下又は不存在下、51Crでラベルされたマウスリンホーマ細胞(以下「YAC−1」という)標的細胞といっしょに24時間培養し、4時間後ガンマーカウンターを用いて上清中の遊離した51Crを測定した。その結果、インビトロで脾臓B細胞をIL−18の存在下培養したとき、野生型マウスにおけるYAC−1標的細胞に対する溶解活性は劇的に増強したが、MyD88ノックアウトマウスにおいては増強されることはなかった。なお、IL−18に代えてIL−2を用いた場合には、本発明のMyD88ノックアウトマウスの脾臓B細胞においても溶解活性が増強した(図5参照)。
5−1(TLR4欠損マウスの作製)
最近、C3H/HeJマウスがToll様受容体(TLR)4遺伝子のミスセンス点突然変異によりLPSに対して低応答性であることが報告され(Science 282,2085−8,1998、J.Exp.Med.189,615−625,1999、J.Immunol.162,3749−3752,1999)、また本発明者らによって、TLR4欠損マウスのマクロファージと脾臓B細胞がLPSに低応答性であり、TLR4遺伝子がLPSシグナル伝達に不可欠であることが判明した(J.Immunol.162,3749−3752,1999)。そこで、TLR4欠損マウスとMyD88欠損マウスとのマクロファージと脾臓B細胞の細菌細胞壁成分に対する応答性を比較するために、TLR4欠損マウス(129/OlaXC57BL/6から交配したF2)を文献記載(J.Immunol.162,1999,3749−3752)のようにジーンターゲティング法により作製した。また、以下の実施例には、年齢が一致する野生型マウス、TLR4欠損マウス及びMyD88欠損マウスを使用した。
フェノール抽出し、ゲル濾過法によって精製されたエセリシア・コリ(Escherichia coli)血清型O55:B5(シグマ社製)、クレブシェラ・ニューモニエ(Klebsiella pneumoniae)(シグマ社製)、シュードモナス・アエルギノサ(Pseudomonas aeruginosa)血清型10(シグマ社製)、サルモネラ・チフィムリウム(Salmonella typhimurium)(シグマ社製)、セラチア・マルセッセンス(Serratia marcescens)(シグマ社製)、フレクスナー赤痢菌(Shigella flexneri)血清型1A(シグマ社製)、ビブリオ・コレレエ(Vibrio cholerae)血清型イナバ569B(シグマ社製)等のLPSを購入した。フェノールクロロフォルム石油エーテル抽出法により調製されたサルモネラ・ミネソタ(Salmonella minnesota)Re−595のLPSは購入した(シグマ社製)。また、文献(FEBS Lett.332,1994,197−201)記載の方法で、ポルフィロモナス・ジンジバリス(Porphyromonas gingivalis)381のLPS及びリピドAを調製した。結核菌の全細胞溶解物は、デュボス培地(ディフコ社製)で結核菌アオヤマB株(NIHJ1635)を1ヶ月間培養した後、細胞を回収してリン酸緩衝生理食塩水(PBS)で再懸濁し、細胞を超音波処理して調製した。
上記作製した野生型マウス、TLR4欠損マウス及びMyD88欠損マウスのそれぞれの腹腔内に4%のチオグリコール酸塩を2mlずつ注入し、3日後に腹膜腔から腹膜滲出細胞を単離し、氷温のハンクス緩衝液(Hank’s buffered salt solution:HBSS)で洗浄することによって腹膜細胞を得た。この細胞をRPMI1640培地に浮遊させ、プラスチックシャーレに分注し、37℃で2時間培養し、その後、ハンクス緩衝液で洗浄することにより非付着細胞を取り除き、付着細胞を腹膜マクロファージとして以下の実験に使用した。
上記野生型マウス(野生型)、TLR4欠損マウス(TLR4−/−)、MyD88欠損マウス(MyD88−/−)等のそれぞれの腹膜マクロファージのLPSに対する応答性をサルモネラ・ミネソタRe−595のLPSを用いて調べた。それぞれのマウスの腹膜マクロファージを、種々の濃度(0、0.01、0.1、1、10又は100μg/ml)のLPSの存在下で24時間培養して刺激し、LPS応答性のマクロファージから放出される腫瘍壊死因子(TNF−α)の濃度をELISAにより測定した(図8A参照)。この結果から、野生型マウスのマクロファージでのTNF−αの産生は、LPSの投与量に応じて増加するのに対して、TLR4欠損マウスやMyD88欠損マウスでは、100μg/mlの濃度のLPS刺激においてもTNF−αを産生せず、これらがLPS不応答性であることがわかった。
TLR4欠損及びMyD88欠損マウスの脾臓B細胞が全ての刺激物に対して不応答性であるかどうかを調べるため、TLR4欠損マウス及びMyD88欠損マウスの脾臓B細胞の他の刺激物に対する応答性を検討したところ、以下に示すように応答性は損なわれておらず、これらのマウスはLPSに対する応答性が特異的に欠損していることがわかった。
野生型マウス、MyD88欠損マウス及びTLR4欠損マウスのマクロファージに0.025%の蛍光ラテックスビーズ(0.75μm)(ポリサイエンス社製)を加え、37℃で2時間、CO2インキュベーター中で培養した。その後、貧食されなかったビーズを取り除くため、この培養物をPBSで3回勢いよく洗浄し、20分間、2.5%のホルムアルデヒドを含むPBSでインキュベートし、培養物をホルムアルデヒドで固定した。これらの固定細胞を、Axiophoto顕微鏡(Carl Zeiss社製)で視覚化したところ、TLR4欠損マウス及びMyD88欠損マウスの腹膜マクロファージは共に、ラテックス粒子を貧食していることがわかった。したがって、これらの他の刺激によるTLR4欠損マウス及びMyD88欠損マウスのマクロファージの貧食能は損なわれていないことがわかった。
ポルフィロモナス・ジンジバリスのLPSは、LPS低応答性であるC3H/HeJマウスの細胞の活性化能において、ある程度の反応を示すことから(J.Immunol.158,1997,4430−6)、それぞれのマウスのポルフィロモナス・ジンジバリスのLPSに対する応答性を前記サルモネラ・ミネソタRe−595の場合と同様に調べた。野生型マウスのマクロファージでは、ポルフィロモナス・ジンジバリスのLPSの投与量に依存してTNF−αを誘発していたが、TLR4欠損マウスのマクロファージでは、C3H/HeJマウスのマクロファージと同様に低応答性であり、野生型マウスのマクロファージの3分の1程度のTNF−α産生能を示したに過ぎなかった。これに対し、MyD88欠損マウスのマクロファージでは、高濃度のLPSで刺激しても、検出しうる程のTNF−αを産生しなかった(図10A参照)。
大腸菌(O55:B5)のLPSに対する応答性についても、前記サルモネラ・ミネソタRe−595の場合と同様に調べた。TLR4欠損マウス及びMyD88欠損マウスの腹膜マクロファージでは、野生型マウスのマクロファージと比較して、大腸菌(O55:B5)のLPSに対する応答性を損なっていた(図11A)。しかし、高濃度のLPSで刺激した場合、TLR4欠損マウスのマクロファージは少量のTNF−αを産生したが、それに対してMyD88欠損マウスのマクロファージは、高濃度のLPS刺激においてもTNF−αを産生しなかった。
グラム陽性菌の主要な細胞壁成分であるペプチドグリカン(PGN)がマクロファージを活性化することが報告されている(J.Immunol.155,1995,2620−30、Infect.Immun.62,1994,2715−21)。そこで、スタフィロコッカス・アウレウスのPGN(Fluka社製)に対する応答性を前記サルモネラ・ミネソタRe−595の場合と同様に調べた。PGNで刺激した場合、TLR4欠損マウスの腹膜マクロファージは、投与量に依存して野生型マウスのマクロファージとほぼ同程度のTNF−αを産生したが、MyD88欠損マウスのマクロファージは、高濃度のPGN刺激に対してもにTNF−αを産生しなかった(図12A参照)。
リポテイコ酸(LTA)は、グラム陽性菌の細胞壁成分であって、単球とマクロファージの活性化を誘導する(Infect.Immun.62,1994,2715−21)ことから、ストレプトコッカス・ニューモニアのLTAに対する応答性を前記サルモネラ・ミネソタRe−595の場合と同様に調べた。野生型マウスの腹膜マクロファージは、LTAの投与量に応じてTNF−αの産生を増大していた。これに対して、MyD88欠損マウスのマクロファージでは、高濃度のLTA刺激に対してもTNF−αを産生しなかった。また、TLR4欠損マウスも野生型マウスと比較するとTNF−αの産生が損なわれていたが、100μg/mlのLTA刺激ではTNF−αを誘発していた(図13A参照)。
結核菌細胞壁成分、特にリポアラビノマンナンは、骨髄性細胞の活性化を誘導することで知られていることから(J.Immunol.149,1992,541−7、J.Clin.Invest.91,1993,2076−83)、結核菌全細胞溶解物に対する応答性を前記サルモネラ・ミネソタRe−595の場合と同様に調べた。野生型マウスのマクロファージは、全細胞溶解物の投与量に依存してTNF−αを産生していた。また、TLR4欠損マウスのマクロファージも、野生型マウスのものと比較するとわずかであるがTNF−αを産生していた。しかし、MyD88欠損マウスのマクロファージは、高濃度の全細胞溶解物に対してもTNF−αを産生しなかった(図14A参照)。
野生型マウス、TLR4欠損マウス及びMyD88欠損マウスの応答性について、他の菌体細胞壁成分[クレブシェラ・ニューモニエ、シュードモナス・アウリギノサ10、サルモネラ・チフィムリウム、フレクスナー赤痢菌、ビブリオ・コレラ等のLPS、及び大日本製薬株式会社のカワタシゲオ氏から提供されたスタフィロコッカス・エピデルミディス(Staphylococcus eqidermidis)のPGN]に対する応答性についても上記と同様に調べた。結果を表1に示す。この表1から、全ての菌体の成分において、MyD88欠損マウスが不応答性であることがわかった。
129/SvJマウス遺伝子ライブラリー(ストラタジーン社製)から、ヒトTLR2遺伝子と類似したマウスESTクローン(登録番号D77677)由来のプローブを用いて、TLR2遺伝子をスクリーニングし、pBluescriptベクター(ストラタジーン社製)中でサブクローンし、制限酵素マッピング及びDNA配列決定により特定した。ターゲッティングベクターは、TLR2遺伝子の細胞内領域を含むエクソン部位1.3kbの遺伝子フラグメントを、ポリAシグナルをもつpMC1−neo(ストラタジーン社製)に置換することにより構築した。かかるターゲッティングベクターは、4.8kbの5′遺伝子フラグメントと1.0kbの3′遺伝子フラグメントとをフランキング配列として有し、HSV−tkカセットを5′末端に含んでいる。このターゲッティングベクターをSalIにより線状化し、胎生14.1日目の胚幹細胞(ES細胞)にエレクトポレーションし、G418及びガンシクロビアに抵抗性を示す120個のクローンを、PCRによる相同組換えのためにスクリーニングし、図15Aに示すプローブを用いるサザンブロット分析により9個を確かめた。
本発明のTLR2ノックアウトマウス(5匹)、TLR4ノックアウトマウス(5匹)及び野生型マウス(5匹)に、それぞれ大腸菌(O55:B5)由来のLPSを1mg投与し、その生存率によりLPS不応答性を調べた。結果を図16に示す。図16より、本発明のTLR2ノックアウトマウス(TLR2−/−)及び野生型マウスはLPSに応答し、投与後4日でほとんどが死亡したのに対して、TLR4ノックアウトマウス(TLR4−/−)は、LPS投与後6日目においても死亡するものはなく、エンドトキシン不応答性であることを確認することができた。
TLR2ノックアウトマウス(TLR2−/−)、TLR4ノックアウトマウス(TLR4−/−)及び野生型マウス(野生型)のそれぞれの腹腔内に4%のチオグリコール酸培地(DIFCO社製)を2mlずつ注入し、3日後に各マウスの腹腔内から腹膜滲出細胞を単離し、これらの細胞を10%のウシ胎仔血清(GIBCO社製)を添加したRPMI1640培地(GIBCO社製)中で37℃にて2時間培養し、氷温のハンクス緩衝液(Hank’s buffered salt solution:HBSS;GIBCO社製)で洗浄することにより非付着細胞を取り除き、付着細胞を腹膜マクロファージとして以下の実験に使用した。
サルモネラ・ミネソタRe−595のLPSに対する各種マウス(野生型、TLR2−/−、TLR4−/−)の脾臓細胞の応答性について調べた。それぞれのマウスの脾臓細胞(1×105)を単離し、図18Aに示す各種濃度のLPSにより96ウェルプレート内で培養して刺激した。培養から40時間後に1μCiの[3H]−チミジン(デュポント社製)を添加して更に8時間培養し、[3H]の摂取量をβシンチレーションカウンター(パッカード社製)で測定した(図18A)。この結果から、野生型マウス及びTLR2ノックアウトマウスの脾臓細胞では、LPSの投与量に依存して同様に細胞増殖反応を促進していたが、TLR4欠損マウスの脾臓細胞では、いかなる濃度のLPS刺激においてもLPSによる細胞増殖反応は見られなかった。
上記野生型マウス(野生型)、TLR2ノックアウトマウス(TLR2−/−)、TLR4ノックアウトマウス(TLR4−/−)等のそれぞれの腹膜マクロファージのグラム陽性菌由来の細胞壁成分に対する応答性を、スタフィロコッカス・アウレウス(S.aureus)、コリネバクテリウム・ジフテリア(C.diphtheriae)及びノカルジア・コエリアカ(N.coeliaca)の細胞壁調製物を用いて調べた。細胞調製物は、文献(Biken J.18,77−92,1975、Infect.Immun.38,817−824、1982)記載の方法と同様に、適切な培養条件下で培養した細胞菌体を、ブラウン・メカニカル・セル・ホモジナイザー(MSKモデル;B.Braun Apparatebau社製)又はDyno−Mill(タイプKDL;Willy A,Biochofen Manufactureing Engineers社製)のいずれかで破壊した。破壊した細胞懸濁液の分画遠心法により得た粗細胞壁画分をプロテアーゼで処理し、細胞壁に元々存在していない成分を除去することにより精製調製した。
次に、グラム陽性菌のどの細胞壁成分がTLR2を介してマクロファージを活性化するのかを調べてみた。従来、グラム陽性菌の細胞壁成分であるペプチドグリカン(PGN)及びリポテイコ酸(LTA)が単球/マクロファージを活性化するとの報告がある(Infect.Immun.60,3664−3672,1992、Immunity 1,509−516,1994、J.Biol.Chem.271,23310−23316,1996、Infect.Immun.64,1906−1912,1996)ことから、10μg/mlのスタフィロコッカス・アウレウスのPGN(Fluka社製;図20A)又は10μg/mlのスタフィロコッカス・アウレウスのLTA(シグマ社製;図20C)を用いて、実施例8と同様の方法により、各種マウスの腹膜マクロファージに対する応答でのIL−6及びNO2 −の産生量を測定した。また、実施例10と同様に、各種マウスの腹膜マクロファージのPGN(図20B)又はLTA(図20D)に対する応答でのTNF−αの産生を測定した。
TLRファミリーメンバーは、アダプタータンパク質MyD88を介してセリン/トレオニンキナーゼであるIRAKを活性化し、次いでrel型転写因子であるNF−κBを活性化する、細胞内シグナル伝達分子として知られている(Mol.Cell 2,253−258,1998、J.Exp.Med.187,2097−2101,1998、Immunity 11,115−122,1999)。LPS及びPGNが、かかる細胞内シグナル伝達分子を活性化するかどうかを次のようにして調べた。各種マウスの腹腔マクロファージ(1×106)を、1ng/mlのサルモネラ・ミネソタRe−595のLPS又は10μg/mlのスタフィロコッカス・アウレウスのPGNで図21に示された時間刺激し、これらの細胞菌体を、溶解緩衝液(最終濃度で1.0%のトリトンX−100、137mMのNaCl、20mMのトリス−HCl、5mMのEDTA、10%のグリセロール、1mMのPMSF、20μg/mlのアプロチニン、20μg/mlのロイペプチン、1mMのNa3VO4及び10mMのβ−グリセロリン酸を含有する緩衝液:pH8.0)中にて溶解し、抗IRAK抗体(林原生化学研究所株式会社)で免疫沈降して、文献(Biochem.Biophys.Res.Commun.234,183−196,1998、Immunity 11,115−122,1999)記載のように、インビトロキナーゼアッセイを行い、IRAKの自己リン酸化を測定した(図21A,BにおけるAuto)。
また、上記溶解物を、SDS−ポリアクリルアミドゲル電気泳動により分離させ、ニトロセルロース膜に移し、この膜を抗IRAK抗体(Transduction Laboratories社製)でブロットして、エンハンスド・ケミルミネッセンス装置(デュポント社製)を使用して視覚化した(図21A,BにおけるWB)。以上の結果から、LPSに対する応答でのIRAK活性化は、野生型マウス(野生型)及びTLR2ノックアウトマウス(TLR2−/−)において観察できたが、TLR4ノックアウトマウス(TLR4−/−)では観察できなかった。一方、PGNに対する応答でのIRAK活性化は、野生型マウス及びTLR4ノックアウトマウスのみにおいて確認することができた。これらのことから、LPSはTLR4を、PGNはTLR2をそれぞれ介し認識されることがわかった。
出発原料として、各々99%以上の純粋なエナンチオマーを含有する(S)−(−)−グリシドール及び(R)−(+)−グリシドール(シグマ−アルドリッチ社製)の2つの試薬を用いて文献(Int.J.Peptide Protein.Res.38545,1991)記載の方法により、S−(2,3−ジハイドロオキシプロピル)−L−システインの立体異性体を合成した。これら立体異性体からNα−フルオレニルメトキシカルボニル基で保護されたS−[2(S),3−ビス(パルミトイルオキシ)プロピル]−L−システインとS−[2(R),3−ビス(パルミトイルオキシ)プロピル]−L−システインとの異性体をそれぞれ合成し、上記文献記載の方法でカップリングし、担体に結合したフルオレニルメトキシカルボニル基で保護されたペプチドを得た。10mgの粗精製MALP−2を、SP250/10 Nucleosil 300−7 C8 column(Macherey & Nagel社製)を用いて逆相HPLCによりバッチ処理してさらに精製し、0.1%のトリフルオロ酢酸を含んだ水/2−プロパノールの直線的グラジエントにより40℃で溶出し、活性溶出画分をNO解離分析によりモニターし、最終生成物は質量分析、及び、正確なペプチド含量を決定するためのをアミノ酸分析により特徴づけられた。これらMALP−2を水/2−プロパノール1:1(容積比)の溶液を用い1mg/mlの濃度に調整し、4℃で保存した。
CH3/HeJ由来のエンドトキシン低応答性マウスからPEC(腹腔滲出細胞)を単離し、これらPEC(6×105)を5%のFCSと25μMの2−メルカプトエタノールを含んだダルベッコMEM培地(DMEM)1.25mlの入った24穴細胞培養プレート中で37℃にて一晩培養した。この培養物から非付着細胞を取り除き、新しい培養液に交換して腹膜マクロファージを調製した。各種濃度(0.1、1、10、102、103、104、105又は106pg/ml)の実施例8の方法により得られたR−MALP−2又はS−MALP−2と、濃度30unit/mlの組換えインターフェロン−γ(rIFN−γ)の共存下に、上記腹膜マクロファージを培養し、培養上清中のNO2 −、TNF−α及びIL−6の産生量を測定した(図22)。TNF−αは培養から3時間後にELISA(Genzyme社製)によって、IL−6は培養から21時間後にELISA(ENDOGEN社製)によって、NO2 −は培養から46時間後にNO2/NO3アッセイキット(同仁科学研究所社製)を使用したGreiss法によってそれぞれ測定した。これらの結果から、S−MALP−2よりR−MALP−2の方が、腹膜マクロファージに対してより高い特異活性を示すことがわかった。
健康なヒトから得られた単球を洗浄後、実験に用いた。実施例13の方法により得られたR−MALP−2又はS−MALP−2の各種濃度(0.1、1、10、102、103、104又は105pg/ml)下、ヒト単球(7.5×105)を20時間刺激した。刺激後、IL−8、MCP−1及びTNF−αの産生量をELISAにより測定した(図23)。この結果から、実施例14のマウス由来のマクロファージの場合と同様に、S−MALP−2よりR−MALP−2の方がマクロファージ等に分化する前のヒト単球に対してより高い特異活性を示すことがわかった。
野生型マウス(野生型)、TLR2ノックアウトマウス(TLR2−/−)、TLR4ノックアウトマウス(TLR4−/−)、MyD88ノックアウトマウス(MyD88−/−)のそれぞれの腹膜マクロファージのリポタンパク/リポペプチドに対する応答性を、マイコプラズマ由来のMALP−2を用いて調べた。それぞれのマウスの腹膜マクロファージを実施例14と同様の方法により単離し、各腹膜マクロファージをrINFγ(30unit/ml)の存在下(図24B及びD)又は非存在下(図24A及びC)において、実施例13より得られた各種濃度(0、0.1、1、10、102、103又は104pg/ml)のR−MALP−2又はS−MALP−2といっしょに24時間培養した。培養後、培養上清中のTNF−α及びNO2 −の産生量を測定した(図24)。
実施例16の結果から、リポタンパク/リポペプチドが細胞内シグナル伝達分子を活性化するかどうかを調べるために、上記4種のマウスの腹腔マクロファージ(1×106)を、0.3ng/mlのR−MALP−2で10分間刺激し、実施例12と同様に抗IRAK抗体を用いてインビトロキナーゼアッセイ(図25AのAuto)及びウエスタンブロット分析(図25AのWB)や電気泳動移動度シフト分析(図25B)を行った。また、抗JNK1抗体を用いたインビトロキナーゼアッセイ(図25CのAuto)及びウエスタンブロット分析(図25CのWB)を行った。これらの結果から、TLR2ノックアウトマウス及びMyD88ノックアウトマウスのマクロファージにおいて、MALPに対するIRAK、NF−κB及びJNKの活性化が確認できなかった。以上のことから、マイコプラズマ由来のリポタンパク/リポペプチドは、TLR2及びMyD88シグナル伝達経路を介して生体反応を引き起こしていることが明かとなった。

- 請求項1〜9のいずれか記載の細菌菌体成分に対して不応答性である非ヒト動物をあらかじめ細菌により感染させた後、該非ヒト動物に被検物質を投与し、該非ヒト動物から得られるマクロファージ又は脾臓細胞のマクロファージ活性又は脾臓細胞活性の程度を測定・評価することを特徴とする細菌菌体成分に対する応答性の抑制物質又は促進物質のスクリーニング方法。
- 請求項1〜9のいずれか記載の細菌菌体成分に対して不応答性である非ヒト動物にあらかじめ被検物質を投与した後、該非ヒト動物を細菌により感染させ、該非ヒト動物におけるマクロファージ活性又は脾臓細胞活性の程度を測定・評価することを特徴とする細菌菌体成分に対する応答性の抑制物質又は促進物質のスクリーニング方法。
- 請求項1〜9のいずれか記載の細菌菌体成分に対して不応答性である非ヒト動物をあらかじめ細菌により感染させた後、該非ヒト動物に被検物質を投与し、該非ヒト動物におけるマクロファージ活性又は脾臓細胞活性の程度を測定・評価することを特徴とする細菌菌体成分に対する応答性の抑制物質又は促進物質のスクリーニング方法。
- マクロファージ活性又は脾臓細胞活性の程度を測定・評価するに際し、対照として細菌菌体成分に対して不応答性である非ヒト動物と同種の野生型非ヒト動物の測定値と比較・評価することを特徴とする請求項10〜17のいずれか記載の細菌菌体成分に対する応答性の抑制物質又は促進物質のスクリーニング方法。
- マクロファージ活性の程度の測定・評価が、該マクロファージにおけるサイトカイン及び/又は亜硝酸イオンの産生量の測定・評価であることを特徴とする請求項10〜18のいずれか記載の細菌菌体成分に対する応答性の抑制物質又は促進物質のスクリーニング方法。
- 脾臓細胞活性の程度の測定・評価が、該脾臓細胞におけるMHCクラスIIの発現量の測定・評価であることを特徴とする請求項10〜18のいずれか記載の細菌菌体成分に対する応答性の抑制物質又は促進物質のスクリーニング方法。
- 細菌菌体成分が、リポタンパク/リポペプチドであることを特徴とする請求項10〜20のいずれか記載の細菌菌体成分に対する応答性の抑制物質又は促進物質のスクリーニング方法。
- リポタンパク/リポペプチドが、マイコプラズマ属、スピロヘータ属又はエセリシア属に属する細菌の菌体成分由来のリポタンパク/リポペプチドであることを特徴とする請求項21記載の細菌菌体成分に対する応答性の抑制物質又は促進物質のスクリーニング方法。
- 細菌菌体成分が、ペプチドグリカンであることを特徴とする請求項10〜20のいずれか記載の細菌菌体成分に対する応答性の抑制物質又は促進物質のスクリーニング方法。
- 細菌菌体成分が、エンドトキシンであることを特徴とする請求項10〜20のいずれか記載の細菌菌体成分に対する応答性の抑制物質又は促進物質のスクリーニング方法。
- 細菌菌体成分が、リポテイコ酸であることを特徴とする請求項10〜20のいずれか記載の細菌菌体成分に対する応答性の抑制物質又は促進物質のスクリーニング方法。
- 細菌菌体成分が、結核菌溶解物であることを特徴とする請求項10〜20のいずれか記載の細菌菌体成分に対する応答性の抑制物質又は促進物質のスクリーニング方法。
- 細菌菌体成分に対する応答性の抑制物質又は促進物質が、細菌感染症に対する抑制物質又は促進物質であることを特徴とする請求項10〜26のいずれか記載の細菌菌体成分に対する応答性の抑制物質又は促進物質のスクリーニング方法。
- マウス遺伝子ライブラリーからマウスESTクローン由来のプローブを用いてスクリーニングすることにより得られたMyD88遺伝子領域のC末端部分をコードした2つのエクソン領域の全部又は一部の遺伝子フラグメントを、ポリAシグナルとマーカー遺伝子をもつプラスミドに置換してターゲッティングベクターを構築し、該ターゲッティングベクターを線状化した後胚幹細胞に導入し、MyD88遺伝子機能を欠損した標的胚幹細胞を、マウスの胚盤胞中にマイクロインジェクションしキメラマウスを作製し、このキメラマウスと野生型マウスとを交配させてヘテロ接合体マウスを作製し、かかるヘテロ接合体マウスをインタークロスすることによって得られることを特徴とするMyD88ノックアウトマウス。
