Abstract
【解決手段】 染色体上のTANK結合キナーゼ1(TBK1)遺伝子の一部もしくは全部が欠損し、野生型において発現されるTBK1を発現する機能が失われているマウス、又は該マウス由来の組織若しくは細胞と、被検物質と、TLR4が認識するリガンド又はこれらの含有物とを用いて、前記マウス又は該マウス由来の組織若しくは細胞における、TLR4が認識するリガンド又はこれらの含有物に対するIFN−β等の抗ウイルスタンパク質の誘導の程度を測定・評価し、LPS刺激やウイルス感染に対する応答の促進物質をスクリーニングする。

4%のチオグリコール酸2mlをマウス腹腔内に投与し、3日後にチオグリコール酸を誘発した腹膜細胞を回収した。胚繊維芽細胞(EF)は、胎生12.5日目の胚より前記非特許文献3記載の通りに調製した。組換え水疱性口内炎ウイルス(VSV)は、Dr. Yoshiharu Matsuura及びDr. Takayuki Abe(大阪大学)より提供された。センダイウイルス(SeV)は、Dr. Tatsuo Shiota(大阪大学)より提供された。EFを、1×109のVSVのRNAコピー/ml又は感染効率(MOI)10のSeVウイルスで感染させた。サルモネラ・ミネソタ(S. minnesota)Re−595のLPSは、Sigma-Aldrich社より購入した。合成Pam3CSK4(細菌リポペプチド、BLP)は、Boehringer Mannheim社より入手した。TNF−α及びIL−1βは、Genzyme社より購入した。
野生型IKK−i及びIKK−i変異アレル由来の変異IKK−iの発現ベクターを構築するため、野生型EF及びIKK−i-/-EFから得たIKK−i cDNAのRT−PCR産物を配列決定し、pFLAG-CMV2ベクター(Sigma-Aldrich社製)にクローニングした。ヒトIRF−3の発現ベクターは、ヒトIRF−3 cDNAをpFLAG-CMV2ベクターに連結させることにより構築した。ヒトTBK1(hTBK1)の発現ベクターは、ヒト胎盤cDNAライブラリーより得たTBK1 cDNAのRT−PCR産物を配列決定し、pEF-BOSベクターに連結させることにより構築した。
IKK−i及びTBK1をコードするゲノムDNA断片を129/Svマウスゲノムライブラリーからスクリーニングし、制限酵素マッピング及びシーケンシング分析で特徴づけを行った。IKK−i遺伝子の1.0kbp断片とTBK1遺伝子の1.5kbp断片の両方をネオマイシン耐性遺伝子カセットで置換し、ターゲティングベクターを設計した。MC1プロモーターにより作動する単純ヘルペスウイルスチミジンキナーゼ遺伝子をネガティブ選択のために用いた。これらのターゲティングベクターを、胎生14.1日目のES細胞にトランスフェクションした。G418及びガンシクロビア二重耐性クローンから、PCR及びサザンブロットによりターゲティングしたES細胞を同定し、その後、C57BL/6胚盤胞中に注入した。得られた雄キメラマウスとC57BL/6雌マウスとを交配した。その後、F1子孫同士をインタークロスさせ、各変異アレルのホモ接合体を作製した。
Sepazol-RNA I(Nacalai Tesque社製)を用いて全RNAを単離し、電気泳動にかけ、ナイロン膜に移した。前記非特許文献3記載のcDNAプローブを用いて、ハイブリダイゼーションを行った。cDNAプローブは全て、前記非特許文献3記載のサブトラクティブスクリーニングより得られた。
野生型胚、IKK−i-/-胚及びTBK1-/-胚から得たEF(1×106細胞)を、1.0μg/mlのLPSで、所定の時間刺激した。核抽出物を調製し、NF−κB結合部位又はIFN刺激制御要素(ISRE)を含む放射能標識したオリゴヌクレオチドプローブを用いてインキュベーションを行い、前記非特許文献3記載の通りオートラジオグラフィーで視覚化した。
核抽出物をSDS−PAGEで分離し、PVDF膜に移した。抗IRF−3抗体(前記非特許文献8参照)及び抗NF−κBp65抗体(Santa Cruz Biotechnology社製)を用いて、かかる膜にブロットした。かかる膜結合抗体は、ECLシステム(PerkinElmer Life Sciences社製)を用いて、ウサギIgG(Amersham Bioscience社製)に対する西洋わさびペルオキシダーゼ標識抗体により視覚化した。
ヒト胎児腎臓293細胞を24ウエルプレートに播種し、Lipofectoamin 2000試薬(Invitrogen社製)を用いて、レポータープラスミドと共に、所定の発現ベクターでトランジェントにトランスフェクションした。6ウエルプレートに播種したEFを、Fu GENE6トランスフェクション試薬(Roche Diagnostics社製)を用いて、IFN−βプロモーターレポータープラスミドと共に、空のpEF-BOSベクター(MOCK)又はpEF-BOSヒトTBK1でトランジェントにトランスフェクションした。トランスフェクションから24時間後、かかる細胞を10μg/mlのLPSでさらに12時間刺激した。全細胞溶解液のルシフェラーゼ活性は、デュアルルシフェラーゼレポーターアッセイシステム(Dual-luciferase reporter assay system)(Promega社製)を用いて測定した。内部標準として、レニラルシフェラーゼレポーター遺伝子(Renilla-luciferase reporter gene)(Promega社製)を使用した。
LPSで2時間若しくは4時間刺激したEF又は刺激していないEFから全RNAを抽出し、cRNAプローブの合成を行った。製造者の指示に従い、cRNAの調製、ハイブリダイゼーション及びマイクロアレイのスキャニング(scanning)を行った。アフィメトリックスマイクロアレイMG U74Aバージョン2(Affymetrix社製)を用いて分析を行った。Microarray Suite version 5.0 software(Affymetrix社製)及びGeneSpring software(Silicon Genetics社製)を用いてデータ分析を行った。

IKK−i及びTBK1の生理学的役割を検討するため、ジーンターゲティングによりIKK−i-/-マウス及びTBK1-/-マウスを作製した。IKK−i-/-マウスを作製するためのターゲティングベクターは、第2のキナーゼドメインの一部をコードするIKK−i遺伝子のエキソン7及びエキソン8を置換するために、ネオマイシン耐性遺伝子カセットを用いて構築した(図1A)。IKK−i-/-マウスは、予想されたメンデル比で生まれ、繁殖性であり、健康そうであった(図1B)。IKK−i遺伝子のN末端断片をプローブとして使用し、ホモ接合体変異マウスのチオグリコール酸誘発腹膜細胞中のIKK−i転写物を検出した(図1C)。しかしながら、シーケンシング分析を行ったところ、変異アレルから得たIKK−i mRNAはターゲティングしたエキソンを欠き、早発性(premature)のストップコドンを含み、2つのキナーゼドメインのうち1つのみを含む切断タンパク質を生成することが明らかになった(データは図示せず)。トランジェントなトランスフェクションアッセイを用いて、野生型IKK−iタンパク質及び変異IKK−iタンパク質の生物活性を評価した(図1D)。野生型IKK−iタンパク質は、IFN−βプロモーターの活性を投与量依存的に誘導したが、変異IKK−iタンパク質は誘導しなかった。したがって、変異IKK−iタンパク質が、少なくともIFN−βの誘導に関しては、生物活性を有さないと結論づけた。
IKK−i及びTBK1は、NF−κBの活性化に関係している。IL−6産生はNF−κBに依存するため、本発明者らは、NF−κBを活性化することで知られている様々な刺激に応答して、EFにおけるIL−6の産生を測定した。EFを、TNFα、IL−1β、TLR2リガンドであるPam3CSK4(細菌性リポペプチド、BLP)及びTLR4リガンドであるLPSで刺激した。図2Aに示されるように、IKK−i-/-EF及びTBK1-/-EFは、調べた全ての刺激剤に応答して、野生型細胞と比較して同様のIL−6量を産生した。
LPS刺激は、IRF−3の活性化を誘導し、IFN−β及びIFN誘導性遺伝子セット(a set of IFN-inducible genes)の誘導を引き起こす(前記非特許文献3、Nakaya, T, M. Sato, N. Hata, M. Asagiri, H. Suemori, S. Noguchi, N. Tanaka, and T. Taniguchi. Gene induction pathways mediated by distinct IRFs during viral infection. 2001. Biochem. Biophys. Res. Commun. 283:1150-1156.)。そこで、ノーザンブロット分析によりこれらIRF−3制御遺伝子の発現を調べた(図2B)。IKK−i-/-EFにおいては、LPSに応答して、IFN−β及びIL−6遺伝子の発現の増大を示した。さらに、LPSで刺激したIKK−i-/-EFにおいては、ISG54、IP−10、IRG1、RANTES等のIFN誘導性遺伝子の発現もアップレギュレートした。これらの応答は、IKK−i-/-マウスから得た骨髄由来樹状細胞及びチオグリコール酸誘発腹膜細胞においても、インタクトであった(データは図示せず)。一方、TBK1-/-EFにおいては、IFN−β、ISG54、IP−10及びIRG1のmRNA誘導が著しく損なわれたのに対し、RANTES及びIL−6のmRNA誘導は野生型細胞と類似していた(図2B)。
ウイルス感染は、IRF−3のリン酸化及び二量体化を誘導し、IFN−β及びその他の抗ウイルス性分子の誘導を引き起こす(Lin, R, C. Heylbroeck, P. Genin, P. M. Pitha, J. Hiscott. 1999. Essential role of interferon regulatory factor 3 in direct activation of RANTES chemokine transcription. Mol. Cell Biol. 19:959-966.、Genin, P., M. Algarte, P. Roof, R. Lin, and J. Hiscott. 2000. Regulation of RANTES chemokine gene expression requires cooperativity between NF-kB and IFN-regulatory factor transcription factors. J. Immunol. 164:5352-5361.)。そこで、IKK−i又はTBK1のどちらが、ウイルスを誘導した遺伝子の発現を仲介するのかを調べた(図4A)。EFを水疱性口内炎ウイルス(VSV)又はセンダイウイルス(SeV)に感染させ、ノーザンブロット分析によりmRNAの発現を調べた。IKK−i-/-EFでは、VSV又はSeVのウイルス感染に応答して、IFN−β、RANTES及びIP−10に対するmRNAが、野生型細胞と同様に誘導された。一方、TBK1-/-細胞では、IFN−βのmRNA発現は観察できず、RANTES及びIP−10のmRNA発現は野生型細胞と比較して著しく減少した。IRF−3の活性化を調べるため、VSVに感染した野生型EF及びTBK1-/-EFから核タンパク質を調製し、免疫ブロットを行ってIRF−3を検出した。図4Bに示されるように、野生型細胞では、核のIRF−3及びNF−κBp65の蓄積が認められた。TBK1-/-細胞では、IRF−3の蓄積が損なわれたが、NF−κBp65の移行には影響を与えなかった。これらの結果は、TBK1がIRF−3の活性化に必須の分子であり、それがウイルス感染においてIFN−β及びその他の抗ウイルス性分子のmRNA誘導を引き起こすことを示唆する。ウイルス感染したTBK1-/-EFにおいて、RANTES転写物の誘導は損なわれたが、これに対して、LPSで刺激した場合は、RANTESのmRNAは明らかに誘導されたことに注目すべきである。
上記実施例2により、IFN−βを含むIFN誘導遺伝子セットの誘導にTBK1が必須であることが証明された。これらの遺伝子は、MyD88非依存的に、しかしTRIF依存的に制御されており、転写因子IRF−3により活性化される(前記非特許文献3、8、9参照)。さらに、TBK1はTRIFと相互作用し、IRF−3をリン酸化すると報告されており(前記非特許文献17、18参照)、TBK1がTLR4−TRIF−IRF3シグナル伝達経路に必須の分子であることを示唆している。これらのデータと一致して、TBK1-/-細胞では、ISRE結合活性の誘導が消滅した。また、ウイルス感染における、IFN−β、RANTES及びIP−10のmRNA誘導は、TBK1-/-細胞では消滅した。これに対して、IKK−i-/-細胞は、LPS及びウイルス感染の両方に応答して、正常なレベルの遺伝子誘導を示した。したがって、インビトロ研究においてIKK−i及びTBK1は同様の機能を示したが(前記非特許文献17〜20、22参照)、上記実施例2におけるデータは、これら2つのキナーゼが、LPS刺激及びウイルス感染においては異なる機能を有することを示している。
Claims (13)
- 染色体上のTANK結合キナーゼ1(TBK1)遺伝子の一部もしくは全部が欠損し、野生型において発現されるTBK1を発現する機能が失われているマウス、又は該マウス由来の組織若しくは細胞と、被検物質と、TLR4が認識するリガンド又はこれらの含有物とを用いて、前記マウス又は該マウス由来の組織若しくは細胞における、TLR4が認識するリガンド又はこれらの含有物に対する応答の程度を測定・評価することを特徴とするTLR4が認識するリガンド又はこれらの含有物に対する応答の促進物質のスクリーニング方法。
- TLR4が認識するリガンド又はこれらの含有物に対する応答の程度を測定・評価するに際し、対照としての同腹の野生型マウスとの比較・評価を行うことを特徴とする請求項1記載のTLR4が認識するリガンド又はこれらの含有物に対する応答の促進物質のスクリーニング方法。
- マウス由来の細胞が、TBK1ノックアウトマウス由来の胚線維芽細胞であることを特徴とする請求項1又は2記載のTLR4が認識するリガンド又はこれらの含有物に対する応答の促進物質のスクリーニング方法。
- TLR4が認識するリガンド又はこれらの含有物が、リポポリサッカライド(LPS)であることを特徴とする請求項1〜3のいずれか記載のTLR4が認識するリガンド又はこれらの含有物に対する応答の促進物質のスクリーニング方法。
- LPS刺激に対する抗ウイルスタンパク質の誘導の程度を測定・評価することを特徴とする請求項4記載のTLR4が認識するリガンド又はこれらの含有物に対する応答の促進物質のスクリーニング方法。
- 抗ウイルスタンパク質が、インターフェロンβ(IFN−β)であることを特徴とする請求項5記載のTLR4が認識するリガンド又はこれらの含有物に対する応答の促進物質のスクリーニング方法。
- 抗ウイルスタンパク質が、ISG54、IP−10又はIRG1であることを特徴とする請求項5記載のTLR4が認識するリガンド又はこれらの含有物に対する応答の促進物質のスクリーニング方法。
- 抗ウイルスタンパク質が、Mx2、IRF−7、インターフェロン誘導性GTPase、ISG15であることを特徴とする請求項5記載のTLR4が認識するリガンド又はこれらの含有物に対する応答の促進物質のスクリーニング方法。
- LPS刺激に対するISRE結合活性の誘導の程度を測定・評価することを特徴とする請求項4記載のTLR4が認識するリガンド又はこれらの含有物に対する応答の促進物質のスクリーニング方法。
- TLR4が認識するリガンド又はこれらの含有物が、ウイルスであることを特徴とする請求項1〜3のいずれか記載のTLR4が認識するリガンド又はこれらの含有物に対する応答の促進物質のスクリーニング方法。
- ウイルス感染に対する抗ウイルスタンパク質の誘導の程度を測定・評価することを特徴とする請求項10記載のTLR4が認識するリガンド又はこれらの含有物に対する応答の促進物質のスクリーニング方法。
- 抗ウイルスタンパク質が、インターフェロンβ(IFN−β)であることを特徴とする請求項11記載のTLR4が認識するリガンド又はこれらの含有物に対する応答の促進物質のスクリーニング方法。
