Abstract
Description
本発明は、親和性リガンド、ならびに水、水溶液、血液、血漿、医薬製剤、抗生物質、ワクチン、タンパク質、核酸および他の生物学的産物のような種々の液体から内毒素を除去するためのそれらの使用に関する。
内毒素は、イー・コリ(E. coli)のようなグラム陰性細菌の外膜にて見出されるリポ多糖(LPS)である(Raetz, Ann. Rev. Biochem., 1990, p.129, Vol.59)。該LPS分子は、リピドA領域、中心多糖領域およびO−抗原領域の3つの異なる化学的領域を含有している。リピドA領域は、リン酸基を含有するグルコサミン二糖からなり、長鎖脂肪酸で高度に置換されており、内毒素の毒作用の原因であると考えられる(Rietschel et al., Cellular and Molecular Aspects of Endotoxin Reaction, Ed. Nowotny, A., Spitzner, J.J. and Ziegler, E.J., 1990, p.15)。
本発明は、生物学的治療製剤を含有する溶液から内毒素を選択的に捕獲および除去するための新規クラスの合成親和性リガンド構造の発見に関する。本発明の合成親和性リガンド−マトリックス複合体は、下記一般構造式(構造1および2)で示すことができる:
Aは、置換されていてもよい直鎖状、分枝鎖状または環状の飽和炭化水素鎖を表し、それぞれの反復単位における各Aは、同一であっても異なっていてもよく;
X1、X2およびX3は、各々、独立して、水素原子またはアルキル置換基を表し;
nは2またはそれ以上であり;
mは1またはそれ以上であり;
該リガンドは、任意のスペーサーアームを介して支持マトリックスZに結合している。
特に、構造式2で示される複合体は新しい。
構造1または2を有するポリアミンリガンドは、少なくとも3個の塩基性窒素原子を有しており、これらの窒素原子は、好ましくは、互いに少なくとも2個の炭素原子によって隔てられている。窒素原子は、第1、第2、第3または第4アミン基の形態であってよく、対になっている窒素原子間に介在する炭素原子は、直鎖状、分枝鎖状または環状の炭化水素鎖の形態であってよい。

ここで、Zおよび(Spacer)は、上記と同じ意味を有する。
保存剤を含まないPuraBeadR 6XL(沈殿した重量200g)、RO水(128ml)および10M水酸化ナトリウム(3.2ml)のスラリーをエピクロロヒドリン(14.4ml)と40℃で1時間反応させた。エポキシ活性化PuraBeadR 6XLをRO水(10×200mlアリコート)で徹底的に洗浄して過剰の反応体を除去し、すぐに、工程2に使用した。
工程1からのエポキシ活性化PuraBeadR 6XL(沈殿した重量200g)をアリコートに分けて、トリス(2−アミノエチル)アミンの水溶液(RO水80ml中15.97g)に添加し、反応混合物を40℃で16時間撹拌した。得られたアミノ化吸着剤をRO水(10×200mlアリコート)で洗浄し、次に使用するまで、20%(v/v)の水性エタノール中にて貯蔵した。
保存剤を含まないPuraBeadR 6XL(沈殿した重量200g)、RO水(128ml)および10M水酸化ナトリウム(3.2ml)のスラリーをエピクロロヒドリン(14.4ml)と40℃で1時間反応させた。エポキシド活性化PuraBeadR 6XLをRO水(10×200mlアリコート)で徹底的に洗浄し、すぐに工程2に使用した。
工程1からのエポキシ活性化PuraBeadR 6XL(沈殿した重量200g)をアリコートに分けて、トリエチレンテトラアミンの水溶液(RO水60ml中8.04g)に添加し、得られたスラリーを40℃で16時間攪拌した。アミノ化吸着剤を多量のRO水(10×200ml部分)で洗浄して過剰のポリアミンを除去した。
Claims (12)
- 水性系における内毒素の単離、分離、精製、特徴付け、同定または定量化のための、構造式:
Z−Spacer−[NX−A]m−NY−A−NX2
[式中、
mは、少なくとも1の整数であり;
各Aは、独立して、炭素数1〜6の、置換されていてもよい直鎖状、分枝鎖状、または環状の飽和炭化水素鎖を表し;
各Xは、独立して、水素またはアルキルを表し;
YはXまたはA−NX2であり;
Zは、任意のスペーサーアーム(Spacer)を介してリガンドと結合している支持マトリックスである]
で示される親和性リガンド−マトリックス複合体の使用。 - 水性系がバッファー、タンパク質、ワクチン、抗生物質、血液、血漿、血清、核酸、医薬製剤、および他の生物学的化合物から選択される、請求項1記載の使用。
- 各Aが独立して二価のC1-4アルキル基を表す、請求項1または2記載の使用。
- Spacerが存在し、それが構造:

[式中、Tは、O、SまたはN−R2を表し、R2は、水素原子または炭素数1〜6のアルキル基を表し;
Lは、2〜20個の原子を含有する、置換されていてもよいアルキル、アルキルエーテル、アルキルチオエーテル、アルキルエステルまたはアミド結合である]
を有する、請求項1〜3いずれか1項記載の使用。 - Spacerが存在しない、請求項1〜3いずれか1項記載の使用。
- Zが多孔質アガロースビーズ、パーフルオロカーボン粒子および機能的な膜から選択される、請求項1〜5いずれか1項記載の使用。
- リガンドがジエチレントリアミン、トリエチレンテトラアミン、N,N−ビス(3−アミノプロピル)エチレンジアミンおよびN−(2−アミノエチル)−1,3−プロパンジアミンから選択される、請求項1〜6いずれか1項記載の使用。
- リガンドがトリス(2−アミノエチル)アミンである、請求項1〜6いずれか1項記載の使用。
- 水性系が3〜10の範囲のpHを有する、請求項1〜8いずれか1項記載の使用。
- 水性系を複合体の懸濁液または充填床と接触させる、請求項1〜9いずれか1項記載の使用。
- 全血または血漿から内毒素を除去するための体外装置における、請求項1〜10いずれか1項記載の使用。
- リガンドを内毒素のインビボ捕獲のために可溶性の高分子担体または非高分子担体と結合させる、請求項1〜11いずれか1項記載の使用。
