CD11c+DCは、フィコール遠心分離および磁性ビーズを用いたCD3、CD14、CD19、CD56およびグリコフォリンAを発現する細胞のネガティブ除去により、PBMCを分離した後、健康な血液提供志願者(Stanford Medical School Blood Center、Stanford、CA)の成人の血液バフィーコートから精製した。除去した細胞は、抗CD4−TC(Catag、Burlingame、CA)、抗CD11c−PEおよび抗CD3,CD14、CD16−FITC(Becton Dickinson、Franklin Lakes、NJ)を用いてさらに染色した。CD11c+CD4+T細胞は、Vantage FACsorter(登録商標)(Becton Dickinson、Franklin Lakes、NJ)を用いて99%より高い純度になるように単離した。
CD11c+DCは様々な条件で24時間培養した後に回収し、サイトカインを完全に取り除くために2回洗浄し、5×104の新しく精製した非刺激同種異系の非刺激CD4+Tと共に丸底の96ウェル培養プレートで共培養した。共培養は、漸増のDC/T細胞比で三連で行った。DCおよびT細胞単独のものをコントロールとして用いた。5日後、細胞を1mCi3H−チミジン(Amersham Biosciences Corp.、Piscataway、NJ)に16時間曝し、その後回収し、放射能を測定した。
T細胞は共培養6日目に回収し、2回洗浄し、1×106/mlの濃度で、96ウェルまたは48ウェルの平底のプレート中、PMAおよびイオノマイシンで再度刺激した。2.5時間後、ブレフェルディンA(Brefeldin A)を10mg/mlの濃度で添加した。5時間後、細胞を回収し、2%ホルムアルデヒドで固定し、10%のサポニンで透過性にし、IL−4、IL−5,IL−10、IL−13に対してはPE結合型mAbsで、IFN−γに対してはTNFαおよびFITCが結合したmAbで染色した(全てPharmingen、San Diego、CA)。染色した細胞はFACScan(登録商標)フローサイトメーター(Becton Dickinson、Franklin Lakes、NJ)により分析した。
(A.間質細胞)
TSLP/IL−50(配列番号1)の生物学および病態生理学をさらに理解するために、TSLP/IL−50(配列番号1)mRNAの発現を、様々な初代細胞または細胞株からのcDNAライブラリーのパネルおよびFACSで分類した初代細胞のパネル(細胞の純度は99%より高い)でリアルタイム定量PCR(Taqman(登録商標))することにより分析した。TSLP/IL−50(配列番号1)の発現は、B細胞、T細胞、NK細胞、顆粒球、マクロファージ、単球サブセット、およびDCサブセットを含むほとんどのタイプの造血性細胞で見られなかった。興味深いことに、IgEレセプターと高い親和力で架橋するモノクローナル抗体に活性化される肥満細胞は、hTSLP/IL−50(配列番号1)を非常に高レベルで発現する。hTSLP/IL−50(配列番号1)は、培養したヒト初代間質細胞(例えば、皮膚ケラチノサイト、上皮細胞、平滑筋細胞、および肺線維芽細胞)で多量に発現することがわかった。IL−4、IL−13およびTNFα、またはTNFαおよびIL−1β、それぞれにより活性化される気管支平滑筋細胞および皮膚ケラチノサイトは、培地のみコントロールと比較して、hTSLP/IL−50(配列番号1)を多く発現するようである。TSLP/IL−50(配列番号1)の発現は、内皮細胞において見いだされなかった。従って、TSLP/IL−50(配列番号1)mRNAは主に、ほとんどのタイプの間質細胞および肥満細胞で発現するが、ほとんどのタイプの造血細胞および内皮細胞では発現しない。
例えば扁桃腺のような、ヒトの炎症を起こした組織が、hTSLP/IL−50(配列番号1)タンパク質を発現するかどうか調べるために、免疫組織学的に調べた。試料は、mAb 6NE0112F3(hTSLP/IL−50を特異的に認識する)を用いて染色した。ヒトの扁桃腺は、陰窩上皮(この上皮は、陰窩に並び、しばしばウイルスおよびバクテリアを保有し、抗原の侵入および構成的な炎症の部位を示す)扁平上皮(これは扁桃の表面に並ぶ)を含む。5つの異なった扁桃腺のサンプル全ての内、hTSLP/IL−50(配列番号1)は、陰窩上皮細胞によって構成的に発現していることがわかった。この陰窩上皮細胞は、DC−LAMP陽性のリンパ球と密着していて、樹状突起細胞を活性化する。興味深いことに、全ての扁桃腺サンプルで、扁平上皮の先端部分内で、hTSLP/IL−50(配列番号1)のきわめて少ない小さな病変が見いだされた。TSLP/IL−50(配列番号1)の発現は、DC−LAMP陽性の活性化されたDCの浸潤、および同時に起こる、扁平上皮内のランゲリン(langerin)陽性ランゲルハンス細胞の喪失と関係していた。hTSLP/IL−50(配列番号1)は、陰窩上皮内での構成的な炎症および扁平上皮内の散在する炎症に寄与している。
hTSLP/IL−50(配列番号1)の発現が、インビボでTh2タイプのアレルギー性炎症と関係しているかどうか調べるために、hTSLP/IL−50タンパク質の発現を、アトピー性皮膚炎(TH2が介在するアレルギー性疾患)、ニッケル誘導性接触性皮膚炎(IFN−γ産生CD8+T細胞が介在するアレルギー性疾患)および播種性紅斑性狼瘡(TH1が介在する疾患)を含む皮膚の病変で分析した。hTSLP/IL−50は、健康な皮膚およびアトピー性皮膚炎の病変のない皮膚では検出されなかったが、hTSLP/IL−50の高い発現が、急性(4名の患者)および慢性のアトピー性皮膚炎(6名の患者)のケラチノサイトで見いだされた。TSLP/IL−50の発現は、小さな病巣から、急性および慢性のアトピー性皮膚炎の両方における先端の領域全体にいたる、主に上皮の先端層のケラチノサイトで観察された。hTSLP/IL−50は、ニッケル遊動性接触性アレルギー皮膚炎および播種性紅斑性狼瘡の皮膚の病変では見いだされなかった。
hTSLP/IL−50(配列番号1)のアトピー性皮膚炎での発現が、DCの活性化に関係しているかどうか調べるために、hTSLP/IL−50を、ランゲリン(langerin)(ランゲルハンス細胞のマーカー)、またはDC−LAMP(DC活性マーカー)のどちらかで一緒に二重免疫組織学的に染色した。健康な皮膚では、またはアトピー性皮膚炎の病変のない皮膚では、多くのランゲリン(langerin)陽性ランゲルハンス細胞が表皮内のみで見いだされるが、真皮では見つからず、DC−LAMP+DCは、表皮または真皮のどちらにおいても見つからなかった。アトピー性皮膚炎におけるhTSLP/IL−50の強い発現は、表皮中のランゲリン(langerin)陽性ランゲルハンス細胞の消失および同時に起こる真皮中のDC−LAMP+DCの出現と関係していた。真皮中の多量のDC−LAMP+DCは、ランゲリン(langerin)を発現する。このことは表皮のランゲルハンス細胞が活性化され、真皮に移動することを示している。従って、アトピー性皮膚炎のケラチノサイトによるhTSLP/IL−50の発現は、ランゲルハンス細胞の活性化に直接寄与し、このランゲルハンス細胞は、流出するリンパ節に移動し、アレルゲン特異的なTH2応答を刺激する。
TSLP/IL−50の発現を、前述のようにして、Taqman(登録商標)により決定した。示した細胞におけるTSLP/IL−50の相対的な発現は:培養した肺線維芽細胞(++++);培養した気管支平滑筋(++++);前立腺間質細胞(++);乳房間質細胞(+);乳房上皮細胞(+);肝臓線維芽細胞(+);皮膚ケラチノサイト(+)であった。
非刺激CD4+T細胞を、5つの異なる試験条件下で準備した調製した同種異系の樹状突起細胞にさらし、その後、T細胞の増殖を評価した。その5つの条件を、表2に示す。増殖を、3H−チミジン取り込みアッセイにより決定した。同種異系の反応では、TSLP/IL−50(配列番号1)で処理したDCは、T細胞の増殖の最も大きな増大を誘導した一方で、他の薬剤で処理したDCは、T細胞の増殖がより少ないまたは非常に少なかった。
TSLP/IL−50により活性化されたDCを、自己の非刺激CD4+T細胞と混合し、その後、拡大し、増殖しているT細胞のプールにおけるT細胞レセプターの亜種のプロフィールを評価した。アッセイに用いたT細胞レセプターの亜種は、TCRVβ1、TCRVβ2、TCRVβ3、TCRVβ5、TCRVβ8、TCRVβ14、TCRVβ17、TCRVβ22およびTCRVβ23であった。3つのタイプのコントロールのインキュベーションを用いた:(1)未処理T細胞;(2)IL−7で処理したT細胞;および(3)StreptococcusのエンドトキシンBで活性化されたDCで処理したT細胞。T細胞の未処理のコントロール集団を、以下に示されるT細胞レセプターのサブセットを含んでいた:TCRVβ1(約3%)、TCRVβ2(約8%)、TCRVβ3(約6%)、TCRVβ5(約2%)、TCRVβ8(約4%)、TCRVβ14(約2.5%)、TCRVβ17(約7%)、TCRVβ22(約2.5%)、TCRVβ23(約0.2%)。非刺激CD4+T細胞を、TSLP/IL−50(配列番号1)により活性化されたDC(実験的)で処理し、その後、T細胞を増殖させるためにインキュベーションしたところ、培養していないT細胞で見出されたレセプターと非常に類似したT細胞レセプター亜種のプロフィールを示した。培養していないT細胞およびTSLP/IL−50で活性化されたDCで処理したT細胞で見出された類似のプロフィールは、T細胞の多クローン性の増殖が起こしたことを示した。IL−7とのコントロール培養(DCなし)もまた、T細胞の多クローン性の増殖を生じた一方で、エンドトキシン処理DCとのコントロール培養は、TCRVβ3およぼTCRVβ17を持つT細胞の選択的な増殖を生じた。
TSLP/IL−50処理したDCを、自己の非刺激CD4+T細胞とともにインキュベートした後、t=0、6、9、12、15、18、21日で細胞数を評価した。TSLP/IL−50(配列番号1)により活性化されたDCにさらすことで、T細胞数は15日目で10倍に増加し、その後の時点では細胞数は減少した。コントロールインキュベーションは、IL−7に活性化されたDC、LPSに活性化されたDC、polyI:Cに活性化されたDC、CD40Lに活性化されたDCおよび培地処理したDCに曝した非刺激CD4+T細胞を用いた。本質的に全てのコントロール培養は、T細胞の数を少ししか増加しない、または全く増加しなかったが、LPSで活性化したDCでは、6日目に、T細胞の数が3倍に増加したことが見出された。
TSLP/IL−50(配列番号1)により活性化されたDCを処理した前後で、非刺激CD4+T細胞の表現型を決定した。表現型は、T細胞上の次のマーカー:CD45RA;CD45RO;CD25;CD62L;およびCCR7を測定することにより評価した。
TSLP/IL−50(配列番号1)で活性化されたDCを、自己の:(1)非刺激CD4+T細胞;(2)中心記憶4+T細胞;または(3)自己のエフェクター記憶CD4+T細胞とともにインキュベートした;T細胞の増殖を3Hチミジンの取り込みにより評価した。1:1;1:2;1:4;1:8;1:16;1:32;1:64の比のDC/T細胞で、インキュベーションを別々に行った。コントロールのインキュベーションに、T細胞のみおよび培地のみを含めた。増殖の評価により、T細胞の3つの集団のそれぞれの最大増殖は、1:1の比のDC/T細胞で起こることがわかった。非刺激T細胞の増殖は、一般に、中心記憶T細胞の増殖より1.2〜1.8倍大きい一方で、中心記憶T細胞の増殖は、一般に、エフェクターT細胞の増殖より約2倍大きかった。
異なる10人の被験体それぞれからの、健常なヒトの皮膚および乾癬性の皮膚のサンプルを、組織学的方法により分析した。染色を、抗TSLP/IL−50抗体またはコントロールIgG2a抗体(カタログNo.M68178;Pharmingen Inc.,San Diego、CA)(両方ともペルオキシダーゼAEC(Vector Laboratories、Inc.,Burlingame、CA)でタグ化した)で行った。2つの異なるクローンに由来する抗TSLP/IL−50抗体を用いた。ここで、両方の供給源の抗TSLP/IL−50からの結果は、互いに一致した。染色を、ケラチノサイト、毛包、およびエクリン腺で評価した。10人全ての正常な被験体のケラチノサイトの染色は、陰性であった。10人の正常被験体の毛包およびエクリン腺の染色は、陰性であるかまたは低い範囲であった。10人の乾癬被験体のケラチノサイトの染色は高く、毛包およびエクリン腺の染色は、比較的低かった。結果により、TSLP/IL−50の発現と乾癬との間の有意な関係が示された。
配列番号1はヒト胸腺の間質細胞である(hTSLP/IL−50)。
配列番号2はIL−7R−α鎖である。
配列番号3はTSLP受容体(TSLPR)である。
Claims (21)
- T細胞の抗原提示細胞(APC)刺激を調節する方法であって、該方法は、以下の工程:
該APCと、
a)TSLP/IL−50(配列番号1)のアゴニストもしくはTSLP/IL−50レセプター(TSLP/IL−50R)(配列番号2、3)のアゴニスト;または
b)TSLP/IL−50(配列番号1)のアンタゴニストもしくはTSLP/IL−50R(配列番号2、3)のアンタゴニスト、
とを接触させる工程、
を包含する、方法。 - 請求項1に記載の方法であって、前記T細胞は、非刺激CD4+T細胞、中心的記憶T細胞、またはエフェクター記憶T細胞である、方法。
- 請求項1に記載の方法,前記APCは、CD11c+樹状細胞(DC)である、方法。
- 請求項1に記載の方法であって、前記刺激は、前記T細胞の増殖を刺激する、方法。
- 請求項4に記載の方法であって、前記増殖は、ポリクローン性である、方法。
- 請求項1に記載の方法であって、前記APCと前記T細胞との間の前記相互作用は、自己由来であるかまたは同種異系である、方法。
- 請求項6に記載の方法であって、前記相互作用は、自己由来であり、中心的記憶T細胞表現型を生じる、方法。
- 請求項1に記載の方法であって、前記アゴニストまたはアンタゴニストは、以下:
a)ヒト化抗体;
b)モノクローナル抗体;
c)ポリクローナル抗体;
d)Fabフラグメント;
e)F(ab’)2フラグメント;または
f)抗体のペプチド模倣物
を含む、方法。 - 請求項1に記載の方法であって、前記アゴニストは、TSLP/IL−50(配列番号1)またはその抗原性フラグメントを含む、方法。
- 免疫障害に罹患している被験体を処置する方法であって、該方法は、有効量の、以下の(a)または(b):
a)TSLP/IL−50(配列番号1)のアゴニストもしくはTSLP/IL−50R(配列番号2、3)のアゴニスト;または
b)TSLP/IL−50(配列番号1)のアンタゴニストもしくはTSLP/IL−50R(配列番号2、3)のアンタゴニスト
で処置する工程、あるいは該(a)もしくは(b)を投与する工程、
を包含する、方法。 - 請求項10に記載の方法であって、前記免疫障害は、炎症状態であり、前記投与は、有効量のTSLP/IL−50(配列番号1)のアンタゴニストをまたはTSLP/IL−50R(配列番号2、3)のアンタゴニストを含む、方法。
- 請求項11に記載の方法であって、前記免疫障害は、乾癬、乾癬性関節炎、または肺の炎症応答である、方法。
- 請求項12に記載の方法であって、前記肺の炎症疾患は、喘息または慢性閉塞性肺疾患(COPD)である、方法。
- 請求項10に記載の方法であって、前記免疫障害は、免疫不全であり、前記投与は、有効量のTSLP/IL50(配列番号1)のアゴニストを含む、方法。
- 請求項14に記載の方法であって、前記免疫不全は、免疫抑制を引き起こす細胞除去またはウイルス感染の結果である、方法。
- 請求項10に記載の方法であって、前記投与は、自己由来または同種異系の抗原提示細胞(APC)のエキソビボ処置を含む、方法。
- 請求項16に記載の方法であって、前記投与は、有効量のTSLP/IL50(配列番号1)のアゴニストでのAPCのエキソビボ処置を含む、方法。
- 請求項10に記載の方法であって、前記アゴニストまたはアンタゴニストは、以下:
a)ヒト化抗体;
b)モノクローナル抗体;
c)ポリクローナル抗体;
d)Fabフラグメント;
e)F(ab’)2フラグメント;または
f)抗体のペプチド模倣物
を含む、方法。 - 請求項10に記載の方法であって、前記アゴニストは、TSLP/IL−50(配列番号1)またはその抗原性フラグメントを含む、方法。
- T細胞によるIL−4、IL−5、およびIL−13の生成を誘導する方法であって、該方法は、以下の工程:
a)APCと、TSLP/IL−50のアゴニストまたはTSLP/IL−50レセプターのアゴニストとを接触させる工程、および
b)該T細胞を該APCで刺激する工程、
を包含する、方法。 - 被験体におけるTH2応答を調節する方法であって、該方法は、以下の(a)、または(b):
a)TSLP/IL−50(配列番号1)のアゴニストもしくはTSLP/IL−50レセプター(TSLP/IL−50R)(配列番号2、3)のアゴニスト:または
b)TSLP/IL−50(配列番号1)のアンタゴニストもしくはTSLP/IL−50R(配列番号2、3)のアンタゴニスト
の投与を包含する、方法。
