方法
微生物学的技術に着手するために一般的な分子生物学的方法を用いた。分子生物学的技術はSambrookら、1989に一般的に記載されているとおりである。
菌株およびプラスミド
groEL2のクローニングおよび配列決定のためにR.エクイATCC 6939を用いた。大腸菌BL21(DE3)におけるヒスチジン(His)標識GroEL2の産生のためにベクターpET−28a(+)(Novagen)を用いた。
細菌を蛋白質発現のためにL−ブロス中で培養した。形質転換体の増殖のためにコロンビア寒天を用いた。groEL2のクローニングのために組み替えプラスミドの宿主として大腸菌DH5αを用い、His標識GroELの発現のために大腸菌BL21(DE3)を用いた。
R.エクイgroEL2遺伝子の断片を増幅するために、オリゴヌクレオチドプライマーを設計した。これらのプライマーの配列は、公知の結核菌H37 RvのgroEL2の配列(GenBank寄託番号:AL021932)とツカムレラチロシノソルベンスのgroELの配列(GenBank寄託番号:U90204)との間の相同領域に基づいている。(これらの遺伝子は、ツカムレラおよびミコバクテリア属がR.エクイに密接に関連しており(Ruimyら、1995)、したがってR.エクイgroEL2遺伝子と高度に類似したgroEL2遺伝子を有する可能性があることを示す16S rRNAの研究に基づいて選択した。)
R.エクイの10mlアリコートの2つをL−ブロス中、30℃で振盪しながら一晩培養した。翌朝、それぞれのアリコートの培養物を30℃または42℃で2時間振盪培養した。次いで、培養物を17,000gで15分間遠心沈降し、ペレットを1mlの1×PBS中に再懸濁し、1分間超音波処理し、トラコーマ病原体Hsp60モノクローナル抗体(Affinity Bioreagents Inc.、米国コロラド州)を用いたウエスタンイムノブロット分析のために調製した。この抗体はトラコーマ病原体HSP60アミノ酸配列の517から522のアミノ酸に特異的で、大腸菌HSP60とは交差反応しないことが知られている。モノクローナル抗体の免疫反応性エピトープの最初の5残基(LTTEAL)[配列番号6]はR.エクイGroEL2配列のアミノ酸残基512から516(図1)と同一で、したがってウエスタンイムノブロット分析で用いた時、この蛋白質を検出することが予想された。
pET−28a(+)(Novagen)発現ベクター中にクローニングするために、groEL2遺伝子を、導入されたNcoI部位(下線)を含む順方向プライマー5'−ACGGTACCATGGCCAAGATCATCGC−3'[配列番号7]および導入されたHindIII部位(下線)を含む逆方向プライマー5'−CGTCAAGCTTGAAGTCCATGCCGC−3'[配列番号8]を用いてPCR増幅した。PCRはDyNAzyme(登録商標) EXT DNAポリメラーゼを用い、アニーリング温度61℃で、鋳型としてCsCl勾配法で精製したpIMVS−Re1の調製物を用いて標準的条件下で実施した。PCR生成物およびベクターはNcoIおよびHindIIIで別々に消化し、連結して、構築体pIMVS−Re2を作製した(図2)。
プラスミドpIMVS−Re2を大腸菌BL21(DE3)に形質転換し、下記の方法によりHis標識GroEL2蛋白質を発現させ、Ni2+−NTAアガロース(Qiagen)を用いて精製した。
ホールセルおよび蛋白質調製物を10%SDS PAGEで分離した。
精製したHis標識GroEL2蛋白質100μlを10%SDS−PAGEで分離し、二フッ化ポリビニリデン膜(Immobilon−P、Millipore、米国マサチューセッツ州)に転写した。次いで、蛋白質をエドマン分解法によるN末端アミノ酸配列決定にかけた(配列決定はAustralian Proteome Analysis Facility、Macquarie University、オーストラリア、ニューサウスウェールズ州によって実施された)。
オリゴヌクレオチドプライマーにより、R.エクイ由来の402塩基対(bp)PCR生成物を増幅した。このPCR生成物はトリ結核菌およびパラ結核菌のgroEL2配列と部分的に相同である(P=6×10−61)ことが判明し、推定groEL2遺伝子を含むR.エクイゲノムの断片を有する形質転換体を同定するためにサザンハイブリダイゼーションにおけるプローブとして用いた。
推測R.エクイGroEL2蛋白質は、結核菌、ライ菌、トリ結核菌およびツカムレラチロシノソルベンスのGroEL2蛋白質に、約90%の同一性で非常に密接に関連していることが判明した。同様に、ストレプトミセスアルブス、ストレプトミセスリビダンスおよびストレプトミセスコエリコルなどの他のグラム陽性放線菌由来GroEL2様蛋白質にも関連していた(表2)。R.エクイGroEL2は他の放線菌のGroEL1配列に対しては相同性が低く(約60〜69%の同一性)、大腸菌およびヘリコバクターピロリなどの生物のGroEL配列とは約60%の同一性を有することが明らかとなった。
ミコバクテリアおよびストレプトミセス属菌種などのR.エクイ関連生物には2つのgroEL遺伝子がある(Rinke de Witら、1992)。これらのうちgroEL1はgroEオペロンの一部であると考えられるが、groEL2は通常は染色体の異なる位置で見いだされる(Ducheneら、1994)。配列決定したR.エクイ遺伝子は下記の理由からgroEL2遺伝子と同定された。まず、他の放線菌groEL2遺伝子と相同である(90%の同一性)ことが判明した。さらに、上流にgroES様遺伝子を持たないようで、groEオペロンの一部ではないことが示唆された。ミコバクテリアおよびストレプトミセスなどの他のR.エクイ関連細菌属菌種に関する過去の研究から、groEL遺伝子の同様の配列が明らかにされている(Ducheneら、1994、Rinke de Witら、1992)。R.エクイはGroELをコードする少なくとも2つの遺伝子を含むようで、その1つは本研究で配列決定されたモノシストロニックなgroEL2である。
R.エクイに対するワクチン候補の開発
groEL2を組み込んだDNAワクチンの作製
groEL2遺伝子をPCR増幅し、ベクターpcDNA3(Invitrogen)(Boshartら、1985)にクローニングした。開始コドンを含む順方向オリゴヌクレオチドプライマーは5'−ACGGTACCATGGCCAAGATCATCGC−3'[配列番号7](KpnI部位に下線;開始コドンは太字)で、逆方向オリゴヌクレオチドプライマーは5'−CTTCTAGACGGCGGATGCGAAATGC−3'[配列番号8](XbaI部位に下線)であった。順方向プライマーはKozak配列CCATGG(開始コドンに下線)(Kozak、1982)も含んでいた。
vapA遺伝子をPCR増幅し、ベクターpcDNA3(Invitrogen)(Boshartら、1985)にクローニングした。順方向オリゴヌクレオチドプライマーは5'−GAGGATCCATGGAGACTCTTCACAAGACG−3'[配列番号9](BamHI部位に下線;開始コドンは太字)で、逆方向オリゴヌクレオチドプライマーは5'−GATGAATTCTAACAACCGAGGCTGAGCG−3'[配列番号10](EcoRI部位に下線)であった。順方向プライマーはKozak配列CCATGG(開始コドンに下線)(Kozak、1982)も含んでいた。PCRは、DyNAzyme(登録商標) EXT DNAポリメラーゼ(Finnzymes、フィンランド)、および鋳型としてR.エクイATCC 33701の小規模プラスミド抽出物を用い、アニーリング温度65℃の標準的条件下で実施した。PCR生成物およびpcDNA3ベクターはBamHI/EcoRIで別々に消化し、連結した。ワクチン調製のために、pcDNA3−Re2と命名した構築体を大腸菌DH5αにクローニングした(図4)。
ワクチンとしてのVapAの免疫原性を高めるために、VapA B細胞エピトープをコードする遺伝子配列をgroEL2に挿入して、キメラgroEL2/vapA構築体を作製した。このアプローチは他の研究者らも用いており、彼らは、キメラ遺伝子構築体において、アジュバントとして作用する担体遺伝子は挿入エピトープに対する免疫応答を顕著に増強し、したがって通常のアジュバントが必要なくなることを明らかにしている(Fomsgaardら、1998)。
キメラgroEL2/vapAワクチン構築体を、VapAの免疫原性エピトープNLQKDEPNGRA[配列番号3]をGroELの親水性領域(HoppおよびWoodsの疎水性プロットによって示されるとおり)および予想した免疫原性エピトープ(Panchanathanによりチフス菌GroELで行われた試験に基づく)(Panchanathanら、1998)に挿入することによって調製した。VapAエピトープNLQKDEPNGRA[配列番号3]をコードするDNA配列をgroEL2に、オーバーラップ伸長PCR(overlap extention PCR)突然変異生成を用いて挿入した(Hoら、1989)(図6および図7)。最初の(2回の)PCR反応における鋳型として構築体pcDNA3−Re1を用いた。
100μg/mlのアンピシリンを含むL−ブロスの10ml量中、37℃で6時間振盪しながら、ワクチン構築体を含む単コロニーを培養することにより、ワクチン構築体を増殖させた。この培養物を、100μg/mlのアンピシリンを含むL−ブロス500mlに加え、37℃で振盪しながら一晩培養した。翌日、大規模プラスミド抽出を実施した。プラスミド抽出物を、CsCl勾配遠心法で2回精製し、次いで1×TEに対し一晩の透析を2回行った。
Cos−7細胞を、L−グルタミン(CSL、米国カンザス州)および10%ウシ胎仔血清(Sigma Chemical Co.)を含むRPMI−1640細胞培地中で維持した。トランスフェクションの24時間前に、細胞を確実に対数増殖期とするように継代培養した。約3×105細胞をNunc(登録商標)の35mm細胞培養皿に加え、下記の方法を用いてpcDNA3、pcDNA3−hsp1、pcDNA3−vap1、pcDNA3−Re1、pcDNA3−Re2またはpcDNA3−Re3ベクターで一時的にトランスフェクトした:Fugene(登録商標)トランスフェクション試薬(Boehringer Mannheim)の15μl量を無血清細胞培地85μlで希釈し、室温で5分間インキュベートした。各ワクチン構築体調製物およびpcDNA3ベクター(CsCl勾配法で精製したプラスミド調製物5μg)をFugene(登録商標)混合物に別々に加え、室温で15分間インキュベートした。次いで、混合物を新鮮培地中の細胞に加え、5%CO2存在下、37℃のインキュベーター内で48時間インキュベートした。回収前に、細胞をコンフルエンスであるかチェックした。培地を遠沈管に移し、1×PBS 1mlを細胞に加えた後、細胞培養皿の底から細胞を回収した。細胞を同じ遠沈管に加えた。次いで、遠沈管を10,000gで遠心沈降して、細胞をペレットとした。ペレットを1×PBSを加えて洗浄し、10,000gで遠心沈降した。最後に、ペレットを1×PBS 50μlに再懸濁し、同量の試料緩衝液と混合し、SDS−PAGE分析およびウエスタンイムノブロットに用いた。
vapA遺伝子をpET−28a(+)(Novagen)発現ベクター中にクローニングするために、導入されたNcol部位(下線)を含む順方向プライマー5'−GAGGATCCATGGAGACTCTTCACAAGACG−3'[配列番号17]および導入されたXhoI部位(下線)を含む逆方向プライマー5'−GCCTCGAGGGCGTTGTGCCAGCTACC−3'[配列番号18]によるPCRを用いて増幅した。PCRは、鋳型としてR.エクイATCC 33701のプラスミド抽出物およびDyNAzyme(登録商標) EXT DNAポリメラーゼ(Finnzymes、フィンランド)を用い、アニーリング温度65℃の標準的条件下で実施した。PCR生成物およびベクターをNcoIおよびXhoIで別々に消化し、連結して、構築体pIMVS−Re3を作製した(図5)。
pIMVS−Re3からのHis標識VapAは、基本的にはHis標識GroEL2の産生について記載した方法を用い、下記の変更を加えて調製した:蛋白質をNi−NTAアガロースから100mM EDTA(Ni−NTAアガロースに結合した蛋白質をうまく溶出するためにイミダゾールを用いることはできないため、EDTAを用いた)を用いて溶出した。蛋白質溶出液を1×PBSで2回透析したが、TX−114を用いたエンドトキシン除去は行わなかった。この処理は、おそらくはこの蛋白質が親油性であるために、VapA蛋白質調製物からのエンドトキシン除去に効果的でないことが判明したためである。使用前に、蛋白質調製物中のエンドトキシンレベルを、QCL−1000 Limulus Amoebocyte Lysate Kit(BioWhittaker、米国メリーランド州ウォーカーズビル)を用いて定量すると(試験はMedia production Unit of the IMVSによって実施された)、100〜500pg/mlの間で変動した。His標識蛋白質を15%SDS PAGEゲルで分離し、VapA特異的モノクローナル抗体(Takaiら、1993a)を用いたウエスタンイムノブロットで検出した。
His標識GroEL2/VapA蛋白質を発現するキメラ遺伝子の調製法は、基本的にはpcDNA3−Re3におけるgroEL2/vapA産生に用いたものに、下記の変更を加えて行った:GVORの代わりに下記のプライマー5'−CGTCAAGCTTGAAGTCCATGCCGC−3'[配列番号21](HindIII部位に下線)を用い、その後His標識GroEL2/VapA構築体(pIMVS−Re4)をpIMVS−Re2について記載したように生成した。
His標識GroEL2/VapA蛋白質の調製法は、基本的にはHis標識GroEL2生成について記載したとおりであった。精製蛋白質をSDS PAGEゲルで分離し、GroEL2特異的モノクローナル抗体を用いてウエスタンイムノブロットで検出した。
groEL2を組み込んだDNAワクチンのCos−7細胞における発現
pcDNA3−Re1でトランスフェクトした細胞でのみ約60kDaの大きい蛋白質バンドが観察され、GroEL2の産生が示された。この蛋白質バンドはトラコーマ病原体Hsp60特異的モノクローナル抗体を用いたウエスタンイムノブロット分析で検出した。
pcDNA3−Re2でトランスフェクトした細胞において、約19kDaの蛋白質バンド、および約15kDaのより大きく拡散したバンドが発現された。VapA特異的モノクローナル抗体(Takaiら、1993a)を用いたウエスタンイムノブロット分析で検出した。pcDNA3−hsp2(理想的なKozak配列を含まない構築体)でトランスフェクトしたCos−7細胞またはpcDNA3ベクターでトランスフェクトした細胞では蛋白質発現は認められなかった。pcDNA3−Re2による15および19kDaの蛋白質の発現は、クーマシーブリリアントブルー染色したSDS PAGEでは観察されなかったが、ウエスタンイムノブロットでのみ観察された。これは、他の研究者らがクーマシーブリリアントブルー染色を用いたSDS PAGE上でのVapAの可視化において同様の困難を報告している(S.Takai、personal communication)ため、驚くことではなかった。
pcDNA3−Re3(キメラgroEL2/vapAワクチン構築体)でトランスフェクトした細胞で、約60kDaの大きい蛋白質バンドが発現された。この蛋白質はCos−7細胞で発現されたGroEL2蛋白質よりもわずかに大きかった。
ワクチンを使用する前に、蛋白質調製物をインビボでの使用に適するように下記のとおりに処理した。すべての調製物を1×PBSで2回透析した。TX−114を用いてGroEL2およびキメラGroEL2/VapA蛋白質調製物からエンドトキシンを除去した。His標識VapA調製物からはエンドトキシンを除去しなかった。TX−114による方法でこの調製物からエンドトキシンをうまく除去することができなかったためである。
R.エクイ菌株ATCC 33701を、過去に報告されている方法(Takaiら、1995a、Takaiら、1991a)を用いてマウスの感染のために調製した。動物試験での使用前に、R.エクイ菌株をvapA遺伝子の存在についてPCRで、VapAの発現についてウエスタンイムノブロットで確認した。菌株を−70℃で保存した一定量から、BHIブロス中、37℃で撹拌しながら48時間培養した。細菌を10,000gで10分間遠心沈降してペレットとし、1×PBSで1回洗浄し、滅菌食塩水で希釈して、550nmでのODが約0.6の懸濁液を得た。この懸濁液を滅菌食塩水によって50%希釈し、100μl中に約1.5×107の生物を含む最終接種材料を得た。懸濁液を滅菌食塩水でさらに希釈して、生ワクチンとして用いるために約105の菌濃度とした。マウスに接種する直前に接種材料の一定量をHBAに播種し、37℃で48時間インキュベートした後にコロニーを計数して、細菌の概数を遡及的に確認した。
6〜8週齢の雌BALB/cマウスの群(各群5匹)を用いた。動物はVeterinary Services Division of the IMVS(Gilles Plains、南オーストラリア、アデレード)から入手した。これらは特定病原体不在(SPF)であることが証明されていた。マウスの各群を免疫後、別々の上部にフィルターがついたケージに入れた。
マウスの各群をpcDNA3−Re1、pcDNA3−Re2、pcDNA3−Re3またはpcDNA3ベクター(対照群)でワクチン接種した。DNA 50μg(50μl)をそれぞれの大腿四頭筋に注射した。
100μgの濃度の蛋白質を含む蛋白質調製物50μlを、同量の1.3%水酸化アルミニウムゲル(Alhydrogel、Asia Pacific Specialty Chemicals Ltd、オーストラリア、ニューサウスウェールズ)と混合して100μlとし、各動物に投与した。マウスの各群にHis標識GroEL2、キメラGroEL2/VapAまたはVapA蛋白質調製物でワクチン接種した。対照群のマウスには、1×PBS 100μlでワクチン接種した。2週間の間隔で3回腹腔内にワクチン接種し、各追加免疫前と投与の直前に採血した。
マウス群を、他のワクチンとの比較のために致死量以下の生R.エクイで免疫した。動物に、約105の生R.エクイ菌株ATCC 33701を腹腔内経路で投与してワクチン接種した。2週間の間隔で3回ワクチン接種し、各追加免疫前と投与の直前に採血した。ワクチンに用いた生物の数は過去の試験(Takaiら、1999a)に基づいて選択した。ワクチン接種前に、調製物の一定量をHBAに播種し、調製物中の生菌数を遡及的に定量した。
マウスサイトカインIL−12を発現するプラスミドpORF−mIL12(InvivoGen、米国カリフォルニア州)(製造者によりこのプラスミドはマウスIL−12を分泌すると報告されており、いかなる改変もせずに用いた)を大腸菌DH5αに電気穿孔法により導入し、DNAワクチンを筋肉内注射用に前述(前述のDNAワクチン調製の項を参照されたい)のとおりに調製した。IL−12挿入断片は単一のIL−12の開いた読み枠を生成し、両サブユニットの同レベルの発現を確実にする、2つのウシエラスチンモチーフ(10アミノ酸の長さ)で連結された2つのネズミIL−12サブユニット(p35およびp40)をコードする遺伝子を含んでいた(Leeら、1998)。この調製物5μgを前述の抗原と同時に注射(筋肉内)した。
各免疫の2週間後と投与の直前に、マウスの眼窩後方から血液試料を採取した。採血前に、Fluothane(ハロタン)(登録商標)(Zeneca、英国チェシャー州)の吸入により軽く麻酔した。マウス各群の血液試料を保存した(血液を含む試験管を室温で30分間、次いで−4℃で1時間インキュベートし、最後に1000gで遠心沈降して血清を除去し、必要時まで−20℃で保存した)。
免疫応答中に産生されるIgGサブクラスのパターンは、その応答で産生されるサイトカインのタイプの信頼できる指標であることが広く認められている。一般に、IgG2aはIFN−γ応答(細胞性応答に関連)を反映すると考えられるが、IgG1アイソタイプスイッチングはIL−4(液性免疫に関連するサイトカイン)によって促進される(MosmannおよびCoffman、1989)。
R.エクイ菌株ATCC 33701をBHIブロス500ml中37℃で撹拌しながら48時間培養した。培養物を10,000gで10分間遠心沈降してペレットとし、1×PBSで2回洗浄した。ペレットを1×PBS 200〜500μlに再懸濁した。懸濁液を氷上で30秒間超音波処理し、10分間煮沸した。蛋白質濃度を定量し、1×PBS中で100μg/mlに調節した。この調製物を必要時まで−20℃で保存した。
後足蹠のDTH応答を測定するために、2つの別々の試験(前述の異なるワクチン候補で免疫した3匹のマウス群に対して)を実施した。各マウスの右後足蹠に抗原20μlを注射し、対応する左後足蹠に1×PBS 20μlを注射した。足蹠の厚みを24、48および72時間後にVernierノギスを用いて測定し(3回の測定値の平均を取った)、24時間の時点での反応が対照に比べて最も有意であったため、この時点のDTHをすべての解析で用いた。腫脹のパーセンテージを下記の式を用いて計算した:
データをウィルコクソンの(順位和)2標本検定を用い、有意レベルP≦0.05で解析した。データは正規分布していないことが判明したため、非パラメーター検定を用いた。データをSASバージョン8.01(SAS Institute,Inc.米国ノースカロライナ州)を用いて解析した。
R.エクイによる投与後にマウスで観察された症状
生R.エクイワクチンで免疫したものを除くすべての動物が、投与の24時間後に軽度の疾患の症状を発症した。これらのマウスで有意な体重減少は認められず、感染で死亡した動物はなかった。マウスは投与後4または5日目までに完全に正常のようであった。
生R.エクイでワクチン接種したマウスは、中等度に高いIgG2aレベルおよび低いIgG1レベルによって示されるとおり、Th1に偏った免疫応答を示した。IgG1、IgG2aおよびIgG2b応答は追加免疫のたびに増大した。興味深いことに、VapA特異的抗体応答はGroEL2特異的応答よりも高かった(表6.1)。重要なことに、有意なDTH応答もこれらのマウスで検出された。さらに、ワクチン接種したマウスは静脈内投与後にR.エクイのクリアランス促進を示した。
His標識GroEL2蛋白質およびDNAワクチン(pcDNA3−Re1)ワクチン接種したマウスの双方で、R.エクイGroEL2蛋白質に対するIgG2a抗体の有意なレベルが検出されたが、2回の追加免疫後、DNAワクチンは蛋白質ワクチンよりも高いIgG2a応答を誘発することが判明した(図10B)。IgG1(図10A)およびIgG2a抗体(図10B)応答はいずれも、追加免疫毎に漸増的に高まった。最後の追加免疫後、DNAワクチンに対してIgG2b応答(図11A)はIgG2a応答よりも低く、IgG1応答よりも高かった。
VapAを組み込んだDNAワクチン(pcDNA3−Re2)でワクチン接種したマウスにおいて、His標識VapA蛋白質に対する中等度のレベルのIgG2a抗体が検出された。IgG1抗体のレベルはIgG2aのレベルよりもはるかに低く(図12Aおよび図12B)、Th1に偏った免疫応答を示していた。His標識VapAワクチンに対する免疫応答は、DNAワクチンに対するよりもIgG2aでは高く、IgG1応答でははるかに高く、免疫応答におけるTh1型への偏りがDNAワクチンで観察されるよりも弱いことを示していると考えられる。試験したDNAおよび蛋白質ワクチンではいずれも、IgG2b応答(図13A)はIgG2aよりも低く、IgG1応答に比べるとほぼ同等またはそれよりも高く、ここでも免疫応答のTh1への偏りが示された。これらの結果は、VapAを組み込んだワクチンによって免疫応答におけるTh1への偏りが誘発されたことを示すものである。pORF−mIL12の同時投与は、pcDNA3−Re2へのIgG1応答を実質的に高め、同じくIgG2a応答も高めたが、最後の追加免疫までにすぎなかった。pORF−mIL12の同時投与はIgG2b応答を有意に変えることはなかった。
pcDNA3−Re3(groEL2/vapAキメラDNAワクチン)およびキメラGroEL2/VapA蛋白質ワクチンでワクチン接種したマウスにおいて、R.エクイGroEL2蛋白質に対する有意なレベルのIgG2a抗体が検出された。IgG1およびIgG2a抗体応答はいずれも、追加免疫毎に漸増的に高まった。IgG1抗体のレベルはIgG2aのレベルよりもはるかに低く(図14A、図14B)、免疫応答におけるTh1への偏りを示していた。試験したDNAおよび蛋白質ワクチンではいずれも、IgG2b応答(図14A)はIgG2aよりも低く、IgG1応答よりも高く、免疫応答のTh1への偏りが示唆された。
VapA B細胞エピトープNLQKDEPNGRA[配列番号3]への抗体を検出するために、キメラgroEL2/vapA DNAワクチン(pcDNA3−Re3)で免疫したマウスからの血清をアッセイした。これは、標的抗原としてビオチン化ペプチドNLQKDEPNGRA[配列番号3]によるELISAを用いて実施した。加えて、別のELISAでHis標識VapAを標的抗原として用いた。キメラgroEL2/vapAワクチンで免疫したマウスからの血清で得られたOD値は、対照マウスからの血清で得られた値と有意差はなかった(結果は示していない)。これは、GroEL2に挿入したVapAエピトープがマウスにおいて検出可能なIgG応答を誘発しなかったことを示唆している。
groEL2(pcDNA3−Re1)およびキメラgroEL2/vapA(pcDNA3−Re3)ワクチンはいずれも、有意な全IgG、特に高いIgG2aおよびDTH応答を生じた。IgG1およびIgG2b応答は軽度から中等度であった。一般に、IgG1およびIgG2a応答はいずれもpORF−mIL12存在下で増大する。pcDNA3−Re1およびpcDNA3−Re3によって生じた応答は一般にほぼ同等であったが、IgG1応答は例外でpcDNA3−Re1の方が高かった。驚くことではないが、IgG1およびIgG2a抗体応答はいずれも、追加免疫毎に漸増的に高まった。
一般に、3つのHis標識蛋白質ワクチンはすべて主にIgG2a応答を誘発したが、生じたIgG1応答も対応するDNAワクチン候補で観察された応答よりも実質的に高かった。これは、DNAワクチンと比較した場合に、蛋白質ワクチンによって誘発された免疫応答でTh1への偏りが小さいことを示していた。

- 表面に露出する外因性アミノ酸配列が挿入されたGroEL蛋白質、その修飾体または類縁体であり、前記外因性アミノ酸配列が抗原決定基に特異的に反応する免疫応答を誘発するよう構成されているキメラ蛋白質。
- 前記外因性アミノ酸配列が前記GroEL蛋白質の親水性領域に挿入されている、請求項1記載のキメラ蛋白質。
- 前記外因性アミノ酸配列がGroEL抗原決定基を含む前記GroEL蛋白質の位置に挿入されている、請求項1記載のキメラ蛋白質。
- 前記GroELがR.エクイ(R.equii)由来である、請求項2記載のキメラ蛋白質。
- 前記親水性領域がV26〜S54、V73〜T90、G109〜A155、M191〜L246、R270〜I290、G342〜A197、およびV415〜N468からなる疎水性領域の群より選択される、請求項4記載のキメラ蛋白質。
- 前記親水性領域がM191〜L246である、請求項4記載のキメラ蛋白質。
- 前記外因性アミノ酸配列が3から25アミノ酸の範囲の長さを有する、請求項1記載のキメラ蛋白質。
- 前記外因性アミノ酸配列が約11アミノ酸の長さを有する、請求項1記載のキメラ蛋白質。
- 前記外因性アミノ酸配列が病原性細菌種の主要抗原決定基を含む、請求項1記載のキメラ蛋白質。
- 前記GroEL蛋白質が前記病原性細菌種由来である、請求項9記載のキメラ蛋白質。
- 前記病原性の種がロドコッカスエクイ(Rhodococcus equii)であり、前記主要抗原決定基がVapA蛋白質由来である、請求項9記載のキメラ蛋白質。
- 前記抗原決定基が配列番号2の中に存在するものである、請求項11記載のキメラ蛋白質。
- 前記抗原決定基が配列番号3の中に存在するものである、請求項11記載のキメラ蛋白質。
- キメラ蛋白質をコードする配列および前記キメラ蛋白質発現のために配置される制御因子を含む核酸分子であって、前記キメラ蛋白質は表面に露出する外因性アミノ酸配列が挿入されたGroEL蛋白質、その修飾体または類縁体であり、前記外因性アミノ酸配列は抗原決定基に特異的に反応する免疫応答を誘発するよう構成されている核酸分子。
- 前記外因性アミノ酸配列が前記GroEL蛋白質の親水性領域に挿入されている、請求項14記載の核酸分子。
- 前記外因性アミノ酸配列がGroEL抗原決定基を含むGroEL蛋白質の位置に挿入されている、請求項14記載の核酸分子。
- 前記GroEL蛋白質がR.エクイ由来である、請求項15記載の核酸分子。
- 前記外因性アミノ酸配列が3から25アミノ酸の範囲の長さを有する、請求項14記載の核酸分子。
- 前記外因性アミノ酸配列が病原性細菌種の主要抗原決定基を含む、請求項14記載の核酸分子。
- 前記GroEL蛋白質が前記病原性細菌種由来である、請求項19記載の核酸分子。
- 前記病原性の種がロドコッカスエクイであり、前記主要抗原決定基が前記VapA蛋白質由来である、請求項19記載の核酸分子。
- 前記抗原決定基が配列番号2の中に存在するものである、請求項21記載の核酸分子。
- 前記キメラ蛋白質が前記蛋白質の精製を助ける非GroEL配列も含む、請求項14記載の核酸分子。
- 複数のヒスチジン残基が前記キメラ蛋白質のC末端に付加されている、請求項23記載の核酸分子。
- 免疫応答を誘発するための宿主細胞における発現のプロモーターを含む、請求項14記載の核酸分子。
- 前記DNA分子ベクターが共刺激分子をコードし、前記共刺激分子が宿主の前記免疫応答を刺激することができる、請求項25記載の核酸分子。
- 哺乳動物において抗原決定基に対する免疫応答を誘発する方法であって、前記哺乳動物にキメラ蛋白質を投与する工程を含むとともに、前記キメラ蛋白質が表面に露出する外因性アミノ酸配列が挿入されたGroEL蛋白質、その修飾体または類縁体であり、前記外因性アミノ酸配列が抗原決定基に特異的に反応する免疫応答を誘発するよう構成されている方法。
- 前記外因性アミノ酸配列が前記GroEL蛋白質の親水性領域に挿入されている、請求項27記載の免疫応答誘発法。
- 前記外因性アミノ酸配列がGroEL抗原決定基を含む前記GroEL蛋白質の位置に挿入されている、請求項27記載の免疫応答誘発法。
- 前記外因性アミノ酸配列が3から25アミノ酸の範囲の長さを有する、請求項27記載の免疫応答誘発法。
- 前記外因性アミノ酸配列が病原性細菌種の主要抗原決定基を含む、請求項27記載の免疫応答誘発法。
- 前記GroEL蛋白質が前記病原性細菌種由来である、請求項31記載の免疫応答誘発法。
- 前記病原性の種がロドコッカスエクイであり、前記主要抗原決定基が前記VapA蛋白質由来である、請求項31記載の免疫応答誘発法。
- 前記抗原決定基が配列番号2の中に存在するものである、請求項33記載の免疫応答誘発法。
- 前記GroEL蛋白質がR.エクイ由来である、請求項28記載の免疫応答誘発法。
- 前記免疫応答が前記抗原決定基に特異的な抗体応答を含む、請求項27記載の免疫応答誘発法。
- 前記抗体応答がIgG1に比べてIgG2aで比例的に大きい、請求項36に記載の免疫応答誘発法。
- 前記キメラ蛋白質を薬学的に許容される担体中の精製された形で投与する、請求項27記載の免疫応答誘発法。
- アジュバントを同時投与する、請求項38記載の免疫応答誘発法。
- 哺乳動物の細胞に挿入されるとキメラ蛋白質が細胞内で発現されるように、キメラ蛋白質を発現可能な核酸分子を哺乳動物に投与する、請求項27記載の免疫応答誘発法。
- 前記核酸が前記細胞内で免疫刺激分子も発現する、請求項40記載の免疫応答誘発法。
