Abstract
Description
本出願は、2001年11月16日付けで提出した表題が「Methods for Monitoring and Treating Amyotrophic Lateral Sclerosis」の仮出願USSN 60/333,263の優先権を主張するものである。
本発明は、神経変性障害を軽減、特に筋委縮性測索硬化症(ALS)の症状または発症を軽減する手段としてポリアミン経路活性(polyamine pathway activity)を調節する)方法および組成物を提供するものである。ALSはポリアミンの濃度が高くなることを伴う。特に、本発明は、ポリアミン経路に関与する遺伝子および蛋白質生成物を調節、例えば酵素であるオルニチンデカルボキシラーゼ(ODC)を阻害してポリアミン類、例えばプトレシンなどの濃度を制限することなどで、現在のALS治療の限界を回避する手掛かりを提供することにある。ポリアミン経路に関与していて調節可能な他のポリアミンには、これらに限定するものでないが、スペルミジンおよびスペルミンが含まれる。本発明の方法および組成物はALSにかかっている被験体におけるポリアミンの濃度またはレベルを変えることで保護効果を与えるものである。このような保護効果は、前記被験体の寿命が長くなるばかりでなく病気の進行が遅くなるか或は阻止されることで明らかになる。

本発明の理解をより明瞭にする目的で下記の用語を定義する。
で記述される}およびこれらの薬学的に受け入れられる塩および異性体も包含させることを意図する。本技術分野で公知の他のODC阻害剤が米国特許第4,499,072号、米国特許第5,002,879号そしてBey他の研究[「Inhibition of Basic Amino Acid Decarboxylases Involved in Polyamine Biosynthesis」、Inhibition of Metabolism Biological Significance and Basis for New Therapies、McCann他編集、Academic Press、(1987)、1−32]に記述されている。
I. 神経変性疾患
いろいろな神経変性疾患、例えばクルツフェルトヤコブ病(CJD)、ハンチントン病(HD)、発作(Stroke)およびアルツハイマー病(AD)などでポリアミンおよびポリアミン代謝の局所的変化が認められている。加うるに、ALSの動物モデルの1つであるWobblerマウスでは基本的条件下でオルニチンデカルボキシラーゼ(ODC)が過剰発現することが観察される。ヒトALS患者では、脊髄組織の中のオルニチン(ポリアミン前駆体)の濃度が高くかつ脳脊髄液(CSF)の中のアルギナーゼ(アルギニンをオルニチンに変化させる酵素)の濃度が高いことが分かっている。ポリアミンの調節不良がいろいろな神経変性状態の原因である可能性があり、それらのいくつかを以下に記述する。
(a)ハンチントン病
ハンチントン病は、脳の特定領域の中のニューロンが変性を起こすことで引き起こされる遺伝性障害である。そのような変性は脳の特定領域で起こるように遺伝的にプログラムされており、そのような領域には、動きの調整の責任を負っている構造である脳幹神経節の細胞が含まれる。ハンチントン病では、脳幹神経節の中の線条の中の神経細胞ばかりでなく脳の皮質、即ち外側表面の細胞(これは思考、知覚および記憶を制御する)が特に標的になっている。ハンチントン病によってニューロンの変性が起こると、結果として、動きが制御不能になり、知的能力および機能が失われ、かつ感情の混乱、例えば気分の動揺または非特徴的な興奮または鬱などがもたらされる可能性がある。
(b)多発性硬化症
多発性硬化症(MS)は、「攻撃(attacks)」[この間に中枢神経系の白色物の領域(プラークとして知られる)が炎症を受ける]を受けることを特徴とする慢性病である。そのような領域のプラークが炎症を起こした後、ミエリン、即ち脳および脊髄の中の神経細胞繊維を絶縁している鞘または覆いを形成している脂肪物質が破壊される。ミエリンは、脳、脊髄および体の残りの部分の間の電気化学的メッセージの高速で滑らかな伝達を助長するものである。ミエリンの鞘が損傷を受けると、そのような電気化学的メッセージの伝達速度が遅くなるか或は完全に遮断され、その結果として、体の機能が減退するか或は完全に失われる可能性がある。
(c)アルツハイマー病
アルツハイマー病は、思考、記憶および言語を制御している脳部分に影響を与える進行性の神経変性疾患である。この病気は、痴呆が進行して最終的に知覚および挙動の両方が実質的に悪化することを特徴とする。このような病気は、これ自身、脳組織の中に異常な細胞外蛋白質付着物(「アミロイドプラーク」として知られる)が存在しかつニューロンの中に絡み合った繊維の束[「神経原線維変化として知られる」が蓄積しかつニューロン細胞が失われることで現れる。アルツハイマー病による影響を受ける脳領域は多様であり得るが、最も一般的に影響を受ける領域には、関連皮質および縁領域が含まれる。アルツハイマー病の症状には、記憶の損失、言語術の悪化、視覚空間スキルの悪化および判断の悪化が含まれるが、アルツハイマー病に苦しんでいる人は運動機能を保持している。
(d)パーキンソン病
パーキンソン病は、中脳の黒質領域に見られる神経細胞、即ちニューロンが失われることが原因で起こる運動系の障害である。前記ニューロンは、脳の中に存在していて筋肉の活動を制御または支配するに役立つ化学的メッセンジャー分子であるドーパミンを産生する。黒質の細胞がドーパミンを他の神経細胞に信号を送る神経伝達物質として利用する。パーキンソン病は、そのようなニューロンが死滅または悪化することで脳内のドーパミン濃度が低下した時に起こる。ドーパミンが失われるとそのようなニューロンが制御不能な様式で興奮することで、その患者は体の動きを正常な様式で支配することも制御することもできなくなってしまう。パーキンソン病の主な4症状は、手、腕、足、顎および顔の震え、四肢および/または体幹の硬直、動きの遅さ(運動緩徐と呼ばれる)および均衡および/または調整の悪化である。パーキンソン病は慢性、即ち長期間に渡って持続する病気、および進行性、即ち症状が経時的に悪くなって行く病気の両方である。
(e)筋委縮性測索硬化症
筋委縮性測索硬化症(ALS)は、一般に致命的な神経変性状態であり、その患者は進行的にあらゆる運動機能を失う、即ち自分自身で歩くことも話すことも呼吸することもできなくなり、ALS患者は診断されて2から5年以内に死亡する。ALSの罹病率は実質的に50歳代が高い。正常な老化過程の結果として変異を起こしたか或は折り畳みが誤った蛋白質を充分に分解させる能力を体が次第に失って行くと言った証拠が集まってきている。プロテアソームは、細胞の中で起こる蛋白質の最も正常な分解の責任を負っている生物学的機能を果たす部分である。現在では、プロテアソームの機能が年齢に関連して失われるか或は前記機能が変化することがいろいろな神経変性状態(アルツハイマー病、パーキンソン病、ハンチントン病およびALSを包含)の心臓部であると考えられている。
(f)プリオン関連病
プリオン蛋白質(PrP)はある群の致命的神経変性疾患に密に関連している[Ma,J.およびLindquist,S.、「Wild−type PrP and a mutant associated with prion disease are subject to retrograde transport and proteasome degredation」、Proc.Natl.Acad Sci.、98(26):14955−14960(2001)]。この群の障害は、脳の灰色物の空胞化[また海綿状変化としても知られる]を特徴とする。このような病気はいろいろな形態を取り得る。例えば、このような病気は散発性、主に遺伝性ばかりでなく伝播性障害であり得る。人に関して最も流行している形態のプリオン病はクルツフェルトヤコブ病である一方、動物に関して最も一般的な形態はスクレイピーとして知られる形態である。この群の他の障害には、クールー、Gerstmann−Straussler−Scheinker病および胎児家族性不眠病が含まれる。あらゆる障害が一様に致命的である。
(g)旧小脳運動失調
運動失調は、人が随意筋収縮中に筋肉の活動を調整する能力を失い、従って体の滑らかな動きを調整する能力を失う病気である。旧小脳運動失調は、遺伝性運動失調の最も一般的な形態である。旧小脳運動失調の開始の症状には、四肢の運動失調、眼振(動揺または反射運動のいずれかで眼球がリズミカルに振動)、脊柱後側湾症(過度に屈曲することで特徴づけられる脊椎変形)および凹足(足の短縮または足の正常な弧の誇張)が含まれる。この病気の状態の時に起こる主な病理学的変化は脊髄の後柱に生じる。旧小脳運動失調は最も頻繁に常染色体の劣性遺伝障害である。
(h)脊髄筋萎縮症
脊髄筋萎縮症(SMA)は、脊髄の前角細胞の病気である。異なる数種類のSMAが存在し、それにはI型または急性(重度)SMA[これはまたWerdnig−Hoffmann Diseaseとしても知られる]、II型(慢性)SMA、III型(軽度)SMA[しばしばKugelberg−WelanderまたはJuvinile SMAと呼ばれる]、IV型(成人になって発病)SMA、および成人発病X関連SMA(Adult Onset X−Linked SMA)[またKennedy’s SyndromeまたはBulbo−Spinal Muscular Atrophyとしても知られており、これはオスに発生するが、メスはキャリアーであり得る]が含まれる。SMAは、活動の責任を負っている随意筋に影響を与え、例えばほふく、歩行、頭および首の制御および飲み込みに影響を与える。SMAは主に人の体の幹の基部の筋肉または前記幹に近い筋肉に影響を与える。症状には足および腕の弱化が含まれ、足の弱化の方が腕の弱化よりも大きい。他の症状には、舌の収縮または舌の異常な動きが含まれ得る。しかしながら、SMAの過程中、人の知覚、感覚および知的活動は影響を受けないままである。
II. ポリアミン経路を調節する薬理学的作用剤
いろいろな神経変性疾患、例えばクルツフェルトヤコブ病(CJD)、ハンチントン病(HD)、発作およびアルツハイマー病(AD)などでポリアミン代謝の局所的変化が注目されている。運動ニューロン病モデルとして用いられるWobblerマウスではポリアミン生合成における律速酵素であるオルニチンデカルボキシラーゼ(ODC)が脊髄に多く発現することが分かっている。ヒトALS患者では脊髄組織の中のオルニチン、即ちODCの基質が実質的に高くなる(300%)ことが分かっている。オルニチン合成の責任を負っている酵素であるアルギナーゼがヒト患者のCSFの中に2から8倍過剰発現する。本発明は、ポリアミンの調節不良を神経学的障害、例えばALSなどに関係付ける方法および検定を提供するものである。加うるに、本発明では、ポリアミン調節不良の度合を軽減し得る薬理学的作用剤を選別する検定も提供する。
(a)ポリアミン経路
ポリアミンであるプトレシン、スペルミジンおよびスペルミンは、生きているあらゆる細胞に存在する群の多価有機カチオン性細胞成分である。このようなポリアミンはいろいろな基本的細胞過程で重要な役割を果たし、そのような細胞過程には、細胞の増殖、細胞の分化、シグナルを送る免疫細胞の活性化、形質転換および細胞消滅の調節が含まれる。シグナル導入経路は大部分がポリアミンの生合成経路および/または細胞内のポリアミン濃度を調節する経路と交差している。それらがそのような過程で果たす正確な役割はまだ探求中ではあるが、推定により、そのような過程は前記カチオン性ポリアミンが有するユニークな電荷分布および柔軟性と関係している。それらは細胞内シグナル、クロマチン構造、複製、転写および翻訳を調節することを通して前記過程に影響を与える。より最近になって、それらはイオンチャンネルさえ調節すると考えられている。ポリアミンはDNAと直接結合してDNA−蛋白質の相互作用を調節し得る。そのような機能はポリアミンが細胞増殖で果たす役割ばかりでなく細胞の死滅にも直接関係している可能性がある。ポリアミン異化作用中に過酸化水素が生じることが細胞の死滅に直接的な影響を与えている可能性がある[Thomas他、Cell.Mol.Life Sci.58:244−258(2001)]。細胞は、ポリアミンが細胞機能の点で果たす基本的な役割が理由で複数の合成経路から余分な細胞を吸収する機構に及ぶ範囲の数多くの還元機構をポリアミンの調節の目的で開発してきた。主要な3種類のポリアミンであるプトレシン、スペルミジンおよびスペルミンの合成、吸収および定常状態は転写、翻訳および分解のレベルで密に調節される。
(b)ポリアミン経路の調節
いろいろな病気でポリアミンの調節不良が観察される。ポリアミン代謝の律速酵素であるオルニチンデカルボキシラーゼは転写および後翻訳的に調節される。ODCの翻訳後の安定性は抗酵素によってODCの分解が調節されることを通して26Sプロテアソームによって調節される。ODCはまたユニークな蛋白質でもあり、これは大部分の蛋白質が分解を起こす時に見られるユビキチン化経路では分解を受けない。この上に記述した神経変性疾患の多くおよびALSにプロテアソームの機能不全が関係していると思われる。酵母菌を用いた研究により、プロテアソームの変異体では野生型の細胞に比べてODCの半減期が長いことが分かっている。このことは、欠陥のある細胞ではプロテアソームの機能不全によってODCの半減期が長くなりかつポリアミンのプールが増加する可能性があることを示唆している。発癌および過剰増殖病ではODCの増幅またはアップレギュレーションが観察される。炎症性クローン病ではODCの発現が少ない。ポリアミンの阻害はある種の自己免疫全身性エリトマトーデス(SLE)病および炎症モデルに有益である。ポリアミン生合成の抑制剤、ポリアミン類似物およびオリゴヌクレオチド/ポリアミン類似物の組み合わせは癌の軽減、治療および予防で肯定的な効果を示すことが分かっている。
(c)ポリアミン合成抑制剤
i)ポリアミン類似物
ポリアミンの構造に類似しているが前記経路を抑制する化合物を本発明で用いて神経学的障害にかかっている被験体のポリアミン濃度を低くすることができる。ポリアミン類似物の電荷分布が変化するように前記類似物のメチレンバックボーンを変えてポリアミン類似物を設計することができる。カチオンとカチオンの間のメチレンブリッジ(bridges)の電荷および長さがポリアミンの生物学的機能にとって重要であることが分かった。従って、そのような変更を受けさせたポリアミン類似物はポリアミン経路を遮断することでそれの産生量を低下させる可能性がある。ポリアミン類似物の例が米国特許第6,235,794号および6,172,261号に記述されている。ポリアミン類似物はまた抗増殖剤としても有効であることが分かっている[Bergeron他、J Med Chem 44(15):2451−2459(2001)]。

ポリアミン類は細胞の成長および増殖に関してある役割を果たしており、細胞の成長および増殖の誘発とODC活性の向上および相当してプトレシンの濃度ばかりでなく他のポリアミンの濃度も高くなることが関連していることが分かっている。ODC阻害剤が癌の治療で用いられており、この場合、それらは恐らくはポリアミンの生成を遮断して癌細胞の増殖および転移を遅らせるか、妨げるか、抑制するか或は停止させることを通して治療効果を及ぼすであろう(米国特許第6,277,411号および4,499,072号を参照)。
(iii)酵素阻害剤
ポリアミン経路の少なくとも1種の酵素の阻害剤との非共有結合を生じさせることを通して前記経路の酵素を可逆的に阻害することができる。このような阻害は、天然の基質が結合しないように阻害剤が当該酵素の活性部位と可逆的に結合するようにするならば競合的であり得る。加うるに、阻害剤と基質が同じ結合部位で競合することなく同時に酵素と結合するならば、そのような阻害は非競合的であり得る。阻害剤がポリアミン経路を可逆的に抑制するならば、そのような阻害剤は前記経路におけるいろいろな酵素を阻害することで、プトレシン、スペルミジンおよびスペルミンから成る群から選択されるポリアミンの中の少なくとも1種を減少させる可能性がある。可逆的ODC阻害剤の例はα−メチルオルニチンである[T.Thomas他、Cell.Mol.Life Sci.58:244−258(2001)]。
で記述される}およびこれらの薬学的に受け入れられる塩および異性体も包含させることを意図する。本技術分野で公知の他のODC阻害剤が米国特許第4,499,072号、米国特許第5,002,879号そしてBey他の研究[「Inhibition of Basic Amino Acid Decarboxylases Involved in Polyamine Biosynthesis」、Inhibition of Metabolism Biological Significance and Basis for New Therapies、McCann他編集、Academic Press、(1987)、1−32]に記述されている。S−アデノシル−L−メチオニンデカルボキシラーゼの阻害剤をPegg[「Polyamine Metabolism and Its Importance in Neoplastic Growth and as a Target for Chemotherapy」、Cancer Res.48:759−774(1998)]およびWilliams−Ashman他[「Methylgly−oxal Bis(guanylhydrazone) as a Potent Inhibitor of Mammalian and Yeast S−Adenosylmethionine Decarboxylases」、Biochem Biophys.Red.Commun.46:288−295(1972)]が記述している。
III. ポリアミン調節用薬剤を用いた神経変性障害の治療
1つの面において、本発明の方法を用いて神経変性障害を軽減(modulate
or ameliorate)することができる。そのような神経変性疾患をALS、クルツフェルトヤコブ病(CJD)、ハンチントン病(HD)、発作およびアルツハイマー病(AD)から成る群から選択する。好適な態様における神経変性障害はALSである。別の面において、本発明の方法を用いてポリアミン、例えばプトレシン、スペルミジンおよびスペルミンなどの調節不良または異常な濃度を選別することができる。好適な態様におけるポリアミンはプトレシンである。
IV. 薬理学的作用剤の選別検定
本発明の方法を用いて多種多様な候補組成物を選別にかけることである被験体におけるポリアミン濃度を調節し得る薬理学的作用剤を見つけだすことができる。本発明の選別検定方法は好適には細胞検定であり、この検定に、本分野の通常の技術を有する技術者に公知の標準的な細胞培養技術を用いて安定に培養することができる細胞系を含める。例えば、本発明の選別検定方法に下記の段階を含めてもよい:(i)ある基質の中のポリアミン濃度を測定し、(ii)前記基質に薬理学的作用剤を加え、そして(iii)その加えた薬理学的作用剤に対する反応によって変化するポリアミン濃度の変化を測定する。
相当物
本分野の技術者は、ほんの常規実験を用いることでこの上に記述した態様を基にして本発明のさらなる特徴および利点を理解するか或は確かめることができるであろう。従って、添付請求の範囲に示す如き事項を除き、本発明を個々に示しかつ記述した事項に限定するものでない。出版物および文献は全部引用することによって全体が明らかに本明細書に組み入れられる。
以下の実施例では、本発明の薬理学的作用剤、例えばDFMOおよびDENSPMなどがオスSOD1 G93Aマウスの病気の進行を遅らせて寿命を長くすることを例証する。DFMOがSOD1 G93Aマウスモデルで有益な効果を示すことは、ポリアミン生合成経路がALSの薬剤を見つけだすための新規な標的であることを示している。以下の実施例は単に本発明の説明であり、本発明の範囲を限定する目的であるとして解釈されるべきでない。
(i)モデルの説明
SOD1 G93A(高コピー)マウスモデルをALSモデルとして用いた。このマウスモデルはヒトG93A SOD変異のコピーを25個持ち、これは内因性プロモーターによって駆動される。このマウスの生存率はコピー依存的である(copy dependent)。このような高コピーG93Aは約128日の中間的な生存率を示す。変異SOD蛋白質の高分子量複合体が約30日目から脊髄の中に見られる。60日目に反応性(GFAP反応性)星状細胞の増加が観察され、活性化されたミクログリア細胞が90日目から観察された。Gurney他による研究により、反応性星状細胞の増加は90日目に統計学的有意さを失うがミクログリア細胞の活性化は有意に向上して病気の最終段階を通して高くなり続けることが示された。
(ii)マウスFALSモデル:
ヒトSOD−1(G93A)遺伝子を担持している異型接合形質転換マウスをJackson Laboratory(Bar Harbor、Maine、米国)から入手した。
(iii)薬理学的作用剤:
ジフルオロメチルオルニチン(DFMO)は、ポリアミン生合成における律速酵素であるオルニチンデカルボキシラーゼ(ODC)酵素の不可逆的阻害剤である。DFMOはアフリカ睡眠病治療の目的でオルニジルとして認可されている。DFMOをILEX Oncology,Inc.(San Antonio、TX)から経口形態で入手した。
(iv)方法:
Jackson Laboratory(Bar Harbor、ME)から入手した40匹(n=40)のSOD1 G93A高コピーマウスを、各群のオスの数(n=10)とメスの数(n=10)が等しくなるように20匹(n=20)の2群に分けた。群の変動が低くなるように前記群にまた同腹子適合も受けさせた(litter matched)。オスの動物を1匹づつケージに入れそして空間を保存する目的でメスを群れの状態でケージに入れた。動物に標準的な齧歯類用食物と水を与えると同時に自身で餌を食べることができるようにした。動物が移動して餌を取るのが困難な時には食物と水を敷きわらの状態の食物ペレットと交換しかつ水に関してはJell−Oと交換した。
(v)薬理学的作用剤の搬送:
あらゆる薬理学的作用剤をSOD1 G93Aマウスに投与した。ヒトグリア芽腫試験に適用された長期搬送プロトコル(ALSのUCSF臨床試験プロトコル)に従ってDFMOを搬送した。
(vi)神経学的スコア:
四肢各々の神経学的スコアを監視して、以下に明記する限定した4点スケールに従って記録した:
0=後ろ足が正常な反射作用を示す(尾を用いて動物を持ち上げた時に後ろ足を広げる)
1=後ろ足が異常な反射作用を示す(尾を用いて動物を持ち上げた時に後ろ足を広げない)
2=四肢が異常な反射作用を示しかつ麻痺の証拠がある
3=反射作用を示さずかつ完全な麻痺
4=横にした時に30秒間で立ち上がることができないか或は死亡を確認(この段階で生きている時には動物を屠殺)。
(vii)エンドポイントおよび統計:
一次エンドポイントは生存率であり、二次エンドポイントは神経学的スコアおよび体重である。神経学的スコアの観察および体重の測定を週に5日行って記録する。
(viii)統計学的解析:
混合線形モデル反復測定(mixed linear model repeated measure)ANOVAを用いて体重および神経学的スコアに関する統計学的解析を実施した。性および同腹子補正に関してフレイルティーモデル(frailty model)を用いたカプランメイヤー(Kaplan−Meier)解析を用いて生存率の解析を実施した。
この研究中、実施例1に記述した方法に従って、DFMOを飲料水に入れて3200mg/Kg/日の用量で21日間に渡って長期間搬送した後、7日間の薬剤休日(薬剤なし)を設けた。結果を図2および3に示し、以下に説明する。
生存パーセントの解析:
メスの処置群では動物の70パーセントが薬剤休日期間中または休日直後の数日間で死亡した。しかしながら、対照群では同じ期間の間に死亡したパーセントは30%のみであった(図2)。
神経学的スコアの解析:
組み合わせた結果にもオスマウスの結果にも効果が全く見られなかった(データは示していない)。しかしながら、メスSOD1 G93Aマウスでは、DFMOを搬送している期間の間にDFMOの搬送を断続的にすると保護が見られた。図3に示すように、メス全体の効果は3.5%であった。メスのマウスはDFMOに薬剤休日を設けない選別を再び受けさせるに正当な理由があるに充分なほどの肯定的な傾向を示した(実施例3に示すように)。この研究でオスマウスには効果が観察されなかったことは、性が異なるとポリアミンの量も異なることが反映されている可能性がある。例えば、ポリアミンの基本的濃度はオスの方が高いことから、薬理学的作用剤が肯定的な効果を示すことを観察することができるようにするにはDFMOの投薬量をより多くするか或は投薬を長くする必要があり得る。このような仮定を次の研究で探求する。
この研究中、実施例1に記述した方法に従って、DFMOを飲料水に入れて3200mg/Kg/日の用量で薬剤休日なしに死亡するまで21日間に渡って長期間搬送した。結果を図4−9に示し、以下に説明する。
生存パーセントの解析:
DFMOを用いると図4、6および8に示すようにSOD1 G93Aマウスの寿命が長くなった。オルニチンデカルボキシラーゼの阻害剤に類似した物質であるDFMOは病気の発病を遅らせかつSOD1 G93Aマウスの生存を長くした。
神経学的スコアの解析:
メスとオスSOD1 G93Aを組み合わせた結果(図5)ばかりでなくオス(図7)およびメス(図9)のSOD1 G93A集団でもDFMOを連続的に搬送すると保護が見られた。フレイルティー項として同腹子効果を用いたCox比例ハザード(proportional hazard)を用いて解析した時、全体を組み合わせた効果は9.69%(p=0.02)であった。オスのマウスが示した効果は8.21%である一方、メスのマウスが示した効果は11.48%であった。このように、DFMOを連続的に搬送すると、性に関係なく肯定的な傾向が観察された。
この実施例では、薬理学的作用剤を添加した後に細胞の増殖が低下することをどのように監視して選別することができるかを示す。実施可能な増殖検定の1つの非限定的実施例を記述する。細胞を培養皿に接種し、増殖を誘発し、薬理学的作用剤またはPBSのいずれかで処置し、そして細胞サイクルを進行させてもよい。決めた時間が経過した後、その培地に[3H]チミジンを添加する。37℃で1時間後、細胞をPBSで洗浄し、細胞のDNAに取り込まれた放射性チミジンを液体シンチレーション計数で量化した。別法として、増殖マーカー1種または2種以上を検出しそして/または基質、例えば[3H]チミジン、BrdUおよびテトラゾリウム塩(例えばMTTおよびXTT)の吸収を検出することで細胞の増殖を測定することも可能である。
この実施例では、薬理学的作用剤を添加した後に細胞のポリアミン濃度が低下することをどのように監視することができるかを示す。下記が実施可能な非限定的ポリアミン濃度測定検定である。
この実施例では、SOD−1(G93A)マウスを用いてヒドロキシ尿素がALSに対してインビボで示す保護効果を立証する。この研究中、実施例1に記述したように、ヒドロキシ尿素を腹腔内注射で250mg/Kg/日の用量で50日から110日まで日に1回の投薬で週7日与えた。ヒドロキシ尿素による毒性が理由で処置を110日後に止めた。ヒドロキシ尿素に関連した毒性にも拘らず、ヒドロキシ尿素は動物の寿命を長くする意味で保護効果を与え続けた。この薬剤に由来する細胞毒性は、ニュートロフェニア、血小板減少および溶血などの如き毒性の兆候を観察することを通して、この薬剤が示す保護効果から容易に識別可能である。ヒドロキシ尿素を動物に投与する用量を変えて最適にすることで、この薬剤が示す細胞毒性効果を低くすることができる。その結果を図10および11に示し、以下に説明する。
生存パーセントの解析:
図10に示すように、ヒドロキシ尿素はSOD1 G93Aマウスの寿命を長くした。この効果は、DFMOで処置した動物で観察したそれと統計学的および定量的に類似していた(図2および3を参照)。ヒドロキシ尿素はALS病の発病を遅らせかつSOD1 G93Aマウスの生存を長くした。ヒドロキシ尿素は抗増殖剤、リボヌクレオチド還元酵素阻害剤であり、また、ポリアミン経路に関与して恐らくはODCを阻害する作用を示すであろう。
神経学的スコアの解析:
ヒドロキシ尿素を用いた搬送および治療はメス(図11)SOD1 G93A集団に保護をもたらした。このような結果は、ヒドロキシ尿素はニューロンの増殖を抑制し、特に星状細胞およびミクログリア細胞の増殖を減少させることでニューロンの増殖を抑制する可能性があることを立証している。オスSOD1 G93Aマウスにも同様な結果が期待される。
この実施例では、SOD−1(G93A)マウスを用いてDENSPMがALSに対してインビボで示す保護効果を立証する。この研究中、実施例1に記述した方法に従って、DENSPMを腹腔内注射で24mg/Kg/日の用量で死亡するまで日に2回の投薬で週7日搬送した。その結果を図12および13に示し、以下に説明する。
生存パーセントの解析:
図12に示すように、抗増殖活性を示すポリアミン類似物であるDENSPMは病気の発病を遅らせかつメスSOD1 G93Aマウスの生存を長くした。DENSPMはポリアミンを模擬することで、細胞を騙してポリアミンが充分な濃度で存在すると細胞が「考える」ようにする負のフィードバック機構で機能することで、ポリアミン合成経路を遮断する。オスSOD1 G93Aマウスにも同様な結果が期待される。
神経学的スコアの解析:
DENSPMの搬送はメスSOD1 G93A集団に保護をもたらした(図13)。オスSOD1 G93Aマウスにも同様な結果が期待される。
Claims (17)
- ある被験体におけるポリアミン経路活性を調節するか或は筋委縮性測索硬化症(ALS)の進行を軽減する薬剤を製造する目的で調節剤を用いる使用であって、前記ポリアミン経路における少なくとも1種の酵素を調節する調節剤を前記調節によってALSの進行の軽減がもたらされるような治療有効量で前記被験体に投与することを含んで成る方法。
- 前記調節剤がポリアミン類似物、ポリアミン経路における少なくとも1種の酵素を標的にする阻害剤および抗酵素の活性化剤から成る群から選択される請求項1記載の使用。
- 前記調節剤がポリアミン類似物である請求項1記載の使用。
