Abstract
【解決手段】 骨組織においてγグルタミルトランスペプチダーゼ(γGTPまたはGGT)を発現させた骨組織の脆弱化を伴う疾患のモデル動物が提供された。本発明のモデル動物は、骨粗しょう症症状を、短時間で容易に再現することができる。また骨密度の低下や、尿中の骨吸収マーカーの測定によって、骨組織の脆弱化を容易に追跡することができる。本発明のモデル動物は、骨組織の脆弱化を阻害する化合物のスクリーニングに有用である。

Description
骨組織は、骨基質(Bone Matrix)とそれに沈着する骨塩(Bone Salt)で構成されている。骨基質はコラーゲンなどの蛋白質を中心とする組織である。これに対して骨塩は、ヒドロキシアパタイト(リン酸カルシウム)を中心とする塩類で構成されている。骨基質と骨塩とで構成された骨組織は、一般に骨量(bone mineral density ;BMD)と呼ばれている。

γ-GTPは、ペプチドのN末端のグルタミン酸を他のペプチドまたはアミノ酸に転移する作用を有する酵素である。生体内においてγ-GTPは、抗酸化物質であるグルタチオンなどの生成に関与していることが明らかにされている。グルタチオンは肝ミクロゾームにおける薬物代謝などに重要な役割をもつため、γ-GTPは肝細胞に多量に含まれる。血中γ-GTP活性は様々な肝疾患、あるいは胆道系疾患でも高値となる。したがって、これらの疾患のスクリーニングの指標として、しばしばその血中濃度が測定されている。また、γGTPの発現によって破骨細胞の分化が誘導されることも明らかにされている。しかし、γGTPの発現によって骨粗しょう症のモデル動物を作製できることは知られていなかった。
すなわち本発明は、以下の骨粗しょう症モデル動物、およびその作製方法と用途を提供する。
〔1〕少なくとも骨組織におけるγGTPまたはγGTPと機能的に同等な蛋白質の発現が増強された非ヒト哺乳動物からなる、骨組織の脆弱化および/または破壊をともなう骨疾患のモデル動物。
〔2〕骨疾患が骨粗しょう症である〔1〕に記載のモデル動物。
〔3〕非ヒト哺乳動物が、少なくとも骨組織において、外来性のγGTPまたはγGTPと機能的に同等な蛋白質をコードする遺伝子を発現している動物である、〔1〕に記載のモデル動物。
〔4〕外来性のγGTPまたはγGTPと機能的に同等な蛋白質をコードする遺伝子を導入されたトランスジェニック動物である〔3〕に記載のモデル動物。
〔5〕外来性のγGTPまたはγGTPと機能的に同等な蛋白質をコードする遺伝子が、骨組織特異的な発現制御領域の制御下に発現している〔4〕に記載のモデル動物。
〔6〕骨組織特異的な発現制御領域が、α1(I)-コラーゲンプロモーターである〔5〕に記載のモデル動物。
〔7〕非ヒト哺乳動物がマウスである〔1〕に記載のモデル動物。
〔8〕尿中に排泄される骨吸収マーカーの測定値が正常動物と比較して有意に高い〔7〕に記載のモデル動物。
〔9〕非ヒト哺乳動物の骨組織においてγGTPまたはγGTPと機能的に同等な蛋白質の発現を増強する工程を含む、骨組織の脆弱化および/または破壊をともなう骨疾患のモデル動物の製造方法。

(3)骨組織の脆弱化のレベルが対象と比較して低い場合に、被験化合物が骨組織の脆弱化を阻害する作用を有すると判定される工程

〔19〕骨組織の脆弱化のレベルとして、前記非ヒト哺乳動物の尿中に排泄される骨吸収マーカーのレベルを測定し、当該測定値が対象と比較して低いときに、被験化合物の骨組織の脆弱化を阻害する作用が検出される〔16〕に記載の方法。
〔20〕骨吸収マーカーが、ピリジノリン、またはデオキシピリジノリンである〔19〕に記載の方法。
〔21〕次の工程を含む、骨組織の脆弱化および/または破壊をともなう骨疾患の治療および/または予防に有用な化合物のスクリーニング方法。
(1)〔16〕に記載の方法によって被験化合物の骨組織の脆弱化を阻害する作用を検出する工程、および

X線によって骨組織を撮影し、画像に基いて骨組織の状態を直接観察する方法も確立されている。X線を利用した骨密度の測定方法は、骨組織の状態を直接的に評価することができる方法である。

これらの蛋白質のうち、たとえば(a)マウスγGTPや(b)ヒトγGTPは、本発明におけるγGTPとして好ましい。マウスγGTPを強制発現させたマウスで観察される、骨組織の脆弱化は、ヒトにおける骨粗しょう症の臨床症状との共通点が多い。具体的には、骨組織の病理学的な比較において、高い類似性が確認された。また骨組織の脆弱化に伴って、尿中に骨破壊のマーカーの顕著な上昇が観察される。これらの特徴は、骨粗しょう症の病態の解析や、治療方法の研究においてきわめて有用である。
ヒトやマウスのγGTPの構造は既に明らかにされている。マウスγGTPは配列番号:2に示すアミノ酸配列を有する糖蛋白質である。当該アミノ酸配列をコードするcDNAもクローニングされている(GenBank Acc.#U30509)。またヒトγGTPのアミノ酸配列あるいは塩基配列を、GenBank Acc.#J04131, M24903, J05235で参照することができる。J05235で参照することができるアミノ酸配列を配列番号:4に、そして塩基配列を配列番号:3に示した。

尿中の骨吸収マーカーのみならず、骨塩、骨基質、骨密度、あるいは骨量を指標として、本発明の方法を実施することもできる。この場合には、各指標を同じく対象と比較して、各指標の低下が抑制された場合に、被験化合物の骨の脆弱化を抑制する作用を検出することができる。
現状では、骨粗しょう症の進行を遅らせることはできても、進行してしまった骨組織の脆弱化を治療するための有効な方法は明らかにされていない。したがって、骨組織の脆弱化の治療効果を評価することができる本発明の方法は、骨組織の脆弱化の治療方法を見出すための重要な情報を与えることができると考えられる。
(1)少なくとも骨組織におけるγGTPまたはγGTPと機能的に同等な蛋白質の発現が増強された非ヒト哺乳動物からなる、骨組織の脆弱化および/または破壊をともなう骨疾患のモデル動物に被験化合物を投与する工程、

本発明における骨組織の脆弱化を伴う疾患には、骨粗しょう症が含まれる。本発明のスクリーニング方法に用いられるモデル動物には、たとえば先に述べたような骨疾患のモデル動物を利用することができる。
γGTP(gamma glutamyl transpeptidase)がin vivoで骨吸収を亢進させ骨粗しょう症の病態形成に関わっていることを明らかにするために、γGTPを過剰発現するtransgenic mouseを作成した。γGTPを過剰発現させるにあたって組織特異的に発現させ、その骨吸収に対する効果を観察するためCre-loxP systemを用いた。
Mus musculus γGTP(NCBI U30509)のN末端にkozac配列を配置し、更にその両端にMfeI siteを配置したプライマーを設計した。各プライマーの塩基配列を次に示す。

Mfe1-GGT 3':5'-ggcggggaacctgctggctactgacaattg-3'(配列番号:6)
C57BL6/crSlc(8w male)の両側腎臓を採取し、TRIzol(商標) Reagent(Invitrogen)を用いてtotal RNAを抽出した。このtotal RNAからRT-PCRにより約1.7kbのγGTP cDNA fragment(配列番号:1のCDS 161-1867に相当)を得た。
図1のベクターpCXP-floxed EGFP-GGTは、CMV-IE enhancer + Chicken β-actin Promoterの下流にloxP配列ではさんだEGFP(enhanced green fluorescence protein)+ Rabbit β-globin polyA、(pCXP-floxed EGFP)、さらにその下流に前述のγGTPcDNA fragment + Rabbit β-globin polyAが配置されている(pCXP-floxed EGFP-GGT)。このベクターが導入されるとEGFPが発現する。もしもベクターにCreリコンビナーゼが作用すれば、loxP配列内のEGFPが除去され、その下流にあるγGTPが発現する。

トランスフェクトの48時間後に細胞を回収し、細胞からはノーザンブロッティングアッセイのためのRNAサンプルを抽出した。すなわち、回収した細胞からTRIzol でtotal RNAを回収し、各サンプル とも5μgのRNAをアプライした。また細胞培養液(100μl)はγGTP活性測定に使用した。

GGT 3':5'-gagacacatcgacaaactttggg-3'(配列番号:8)
γGTPを全身に過剰発現させた場合と骨でのみγGTPを過剰発現させた場合の骨吸収を比較検討するため、以下の2種類のCre transgenic miceを入手し、繁殖した。
1) CAG-Cre transgenic Mice
Chicken β-actin(CAG)Promoterの下流にCre geneを配置してある。このマウスにおいては、Creリコンビナーゼが全身に発現する。Floxed EGFP-GGT Transgenic Miceと交配することによって全身でCre-loxP systemが働き、 EGFP配列が除去されγGTPが過剰発現する。この2系統の交配によって得られたマウスはCre geneに設定したプライマーとGGTプライマー(前述)で、必要なコンストラクトを保持していることを確認した。Cre geneに設定したプライマーの塩基配列を次に示す。このプライマーによって、250bpの増幅産物が合成される。

両PCR陽性の個体の尾を1mm採取し、PBS 500μl中で組織を溶解しγGTP活性を測定し、高い活性を有することを確認した。

ColI-Creトランスジェニックマウスは、Osteoblastに発現するα1(I)-collagenのpromoter(proximal fragment 2.3kb)の下流にCre geneを配置してある。このマウスにおいては、Creリコンビナーゼが骨特異的に発現する。floxed EGFP-GGTトランスジェニックマウスと交配することによって骨のみでCre-loxP systemが働く。この2系統の交配によって得られたマウスはCre geneに設定したプライマーとGGTプライマー(前述)で確認した。Cre geneに設定したプライマーの塩基配列を次に示す。このプライマーによって、654bpの増幅産物が合成される。

Claims (27)
- 少なくとも骨組織におけるγGTPまたはγGTPと機能的に同等な蛋白質の発現が増強された非ヒト哺乳動物からなる、骨組織の脆弱化および/または破壊をともなう骨疾患のモデル動物。
- 骨疾患が骨粗しょう症である請求項1に記載のモデル動物。
- 非ヒト哺乳動物が、少なくとも骨組織において、外来性のγGTPまたはγGTPと機能的に同等な蛋白質をコードする遺伝子を発現している動物である、請求項1に記載のモデル動物。
- 外来性のγGTPまたはγGTPと機能的に同等な蛋白質をコードする遺伝子を導入されたトランスジェニック動物である請求項3に記載のモデル動物。
- 外来性のγGTPまたはγGTPと機能的に同等な蛋白質をコードする遺伝子が、骨組織特異的な発現制御領域の制御下に発現している請求項4に記載のモデル動物。
- 骨組織特異的な発現制御領域が、α1(I)-コラーゲンプロモーターである請求項5に記載のモデル動物。
- 非ヒト哺乳動物がマウスである請求項1に記載のモデル動物。
- 尿中に排泄される骨吸収マーカーの測定値が正常動物と比較して有意に高い請求項7に記載のモデル動物。
- 非ヒト哺乳動物の骨組織においてγGTPまたはγGTPと機能的に同等な蛋白質の発現を増強する工程を含む、骨組織の脆弱化および/または破壊をともなう骨疾患のモデル動物の製造方法。
- 外来性のγGTPまたはγGTPと機能的に同等な蛋白質をコードする遺伝子を導入されたトランスジェニック動物を製造する工程を含む、請求項9に記載の製造方法。
- 次の工程を含む、請求項9に記載の製造方法。
(1)i) 発現制御領域、ii) 前記発現制御領域の制御下に発現する不活性蛋白質をコードする遺伝子、およびiii)前記不活性蛋白質をコードする遺伝子の下流に配置された外来性のγGTPまたはγGTPと機能的に同等な蛋白質をコードする遺伝子を含むコンストラクトを導入されたトランスジェニック動物を製造する工程、および
(2)前記コンストラクトのii)不活性蛋白質をコードする遺伝子の除去によって、発現制御領域i)の制御下に外来性のγGTPまたはγGTPと機能的に同等な蛋白質をコードする遺伝子iii)の発現を誘導する工程 - 不活性蛋白質がシグナル生成蛋白質である請求項11に記載の方法。
- 不活性蛋白質をコードする遺伝子がloxP配列によって挟まれており、Creリコンビナーゼによって該遺伝子を除去する工程を含む、請求項11に記載の方法。
- (1)のトランスジェニック動物と、Creリコンビナーゼを発現するトランスジェニック動物を交配させ、(1)に記載のコンストラクトを有する細胞内でCreリコンビナーゼを発現することができる雑種を得る工程を含む、請求項11に記載の方法。
- 次の要素を含むコンストラクトを導入された、骨組織の脆弱化および/または破壊をともなう骨疾患のモデル動物の製造用のトランスジェニック動物。
i) 発現制御領域、
ii) 前記発現制御領域の制御下に発現する不活性蛋白質をコードする遺伝子、および
iii)前記不活性蛋白質の下流に配置された外来性のγGTPまたはγGTPと機能的に同等な蛋白質をコードする遺伝子 - 次の工程を含む、被験化合物の骨組織の脆弱化を阻害する作用を検出する方法。
(1)少なくとも骨組織におけるγGTPまたはγGTPと機能的に同等な蛋白質の発現が増強された非ヒト哺乳動物からなる、骨組織の脆弱化および/または破壊をともなう骨疾患のモデル動物に被験化合物を投与する工程、
(2)骨組織の脆弱化のレベルを測定する工程、および
(3)骨組織の脆弱化のレベルが対象と比較して低い場合に、被験化合物が骨組織の脆弱化を阻害する作用を有すると判定される工程 - 非ヒト哺乳動物が、少なくとも骨組織において、外来性のγGTPまたはγGTPと機能的に同等な蛋白質をコードする遺伝子を発現している動物である、請求項16に記載の方法。
- 外来性のγGTPまたはγGTPと機能的に同等な蛋白質をコードする遺伝子を導入されたトランスジェニック動物である、請求項17に記載の方法。
- 骨組織の脆弱化のレベルとして、前記非ヒト哺乳動物の尿中に排泄される骨吸収マーカーのレベルを測定し、当該測定値が対象と比較して低いときに、被験化合物の骨組織の脆弱化を阻害する作用が検出される請求項16に記載の方法。
- 骨吸収マーカーが、ピリジノリン、またはデオキシピリジノリンである請求項19に記載の方法。
- 次の工程を含む、骨組織の脆弱化および/または破壊をともなう骨疾患の治療および/または予防に有用な化合物のスクリーニング方法。
(1)請求項16に記載の方法によって被験化合物の骨組織の脆弱化を阻害する作用を検出する工程、および
(2)対照と比較して、骨組織の脆弱化を阻害する作用を有する被験化合物を選択する工程 - 骨疾患が、骨粗しょう症である請求項21に記載のスクリーニング方法。
- 請求項21に記載の方法によって得ることができる化合物を有効成分として含有する、骨組織の脆弱化および/または破壊をともなう骨疾患の予防または治療のための医薬組成物。
- 次の工程を含む、外来性遺伝子を導入されたトランスジェニック動物の製造方法。
(1)i) 発現制御領域、ii) 前記発現制御領域の制御下に発現するシグナル生成蛋白質をコードする遺伝子、およびiii)前記シグナル生成蛋白質をコードする遺伝子の下流に配置された導入すべき外来性の遺伝子を含むコンストラクトを導入されたトランスジェニック動物を製造する工程、および
(2)前記コンストラクトのii)シグナル生成蛋白質をコードする遺伝子の除去によって、発現制御領域i)の制御下に外来性の遺伝子iii)の発現を誘導する工程 - シグナル生成蛋白質をコードする遺伝子がloxP配列によって挟まれており、Creリコンビナーゼによって該遺伝子を除去する工程を含む、請求項24に記載の方法。
- (1)のトランスジェニック動物と、Creリコンビナーゼを発現するトランスジェニック動物を交配させ、(1)に記載のコンストラクトを有する細胞内でCreリコンビナーゼを発現することができる雑種を得る工程を含む、請求項24に記載の方法。
- Creリコンビナーゼを発現するトランスジェニック動物が、前記外来性遺伝子の発現を誘導すべき組織においてCreリコンビナーゼを発現するトランスジェニック動物である、請求項26に記載の方法。
