Abstract
【解決手段】 ガラス表面を有する粒子の調製物であって、これらの粒子の75重量%超が0.5μm〜15μmの粒子サイズを有し、且つガラス表面が2〜6モル%の酸化SiO2, B2O3, K2O, CaO, Al2O3及びZnOから構成される粒子を含む調製物が核酸の精製のための方法において極めて有利であることが証明された。これは特に核酸収率の上昇をもたらす。

Description
B2 O3 (0〜30%)、Al2 O3 (0〜20%)、CaO(0〜20%)、BaO(0〜10%)、K2 O(0〜20%)、Na2 O(0〜20%)、MgO(0〜18%)、Pb2 O3 (0〜15%)、ZnO(0〜6%)。
亜鉛非含有層の調製のためのゾル(74SiO2 ・15B2 O3 ・4K2 O・2CaO・5Al2 O3 )
1750mlのテトラエチルオルトシリケート(製造者:Wacker Burghausen )を5リットルの丸底フラスコに入れ、そして以下の化合物を室温にて撹拌しながらすばやく添加する(500rpm ):
541mlのトリエチルボレート(製造者:Aldrich, Steinheim)
250mlのメタノール酸カリウム(メタノール中25%;製造者:Fluka, Deisenhofen)
261mgの第二ブチル酸アルミニウム(製造者:Aldrich, Steinheim)
292mlのエタノール及び
8.49gのカルシウム(製造者:Fluka, Deisenhofen)
ガラスコート化顔料の調製物(MGP)
直径0.75m、高さ2.5m及び蒸発容量(水で換算)1〜3リットル/時間のNubilosa Company, Konstanz由来のスプレータワーでスプレーイングを実施する。空気入口温度は270℃、出口温度は約130℃とする。空気の流速は7.2m3 /min とする。2bar のスプレー圧を有する二流体ノズルをスプレーイングのために用いる。ボールバルブ膜ポンプの輸送能力は60gのゾル/min とする。
亜鉛含有層を調製するためのゾル(70.67SiO2 ・14.33B2 O3 ・4K2 O・2CaO・5Al2 O3 ・4ZnO)
亜鉛を含むゾルを実施例1と同様にして調製する。このために、以下の重量の抽出物を加え、そして同様にして処理する:
1258mlのテトラエチルオルトシリケート(製造者:Wacker Burghansen )
387mlのトリエチルボレート(製造者:Aldrich, Steinheim)
188mlのメタノール酸カリウム(メタノール中25%;製造者:Fluka, Deisenhofen)
196gの第二ブチル酸アルミニウム(製造者:Aldrich, Steinheim)
1285mlのエタノール
6.39gのカルシウム(製造者:Fluka, Deisenhofen)
58.5gの酢酸亜鉛脱水・二水和物(製造者:Fluka, Deisenhofen)
亜鉛含有コート化顔料の調製物
実施例3由来の顔料含有ゾルを実施例3に類似して処理する。しかしながら、圧縮温度は750℃とする。
改良式後処理を利用した亜鉛含有ガラスコート化顔料の調製物(MGP、磁性ガラス粒子)
実施例3由来の顔料含有ゾルを実施例2と似たようにして処理する。しかしながら、圧縮温度は750℃とし、そして酸素内での処理温度は200℃とする。
放射性32Pを利用するDNA又はRNAの収率の決定
1.4kbの32P−ラベル化HIV gag RNA標準品又は3kbの32P−ラベル化ラムダアンプリコンを結合した又は未結合のDNA又はRNAの直接検出のために用いる。各々109 コピーを含む陰性血漿(ヒト)を試料として用いる。
109 コピーの32P−ラベル化ラムダアンプリコンを含む500μlの陰性血漿を2mlのEppendorf 容器に入れる。480μlの結合バッファー/プロテイナーゼ・K溶液(5:1)をピペクトにより加え、ボルテックスにかけ、そして70℃で10分インキュベーションする。室温にまで冷却後、全部で3mgのMGPを含む400μlのイソプロパノールMGP懸濁物をピペクトにより加える。その直後にそれをボルテックスにより混合する。次にそのサンプルをミキサー、例えばEppendorf 由来のサーモミキサー5436で15分インキュベーションする。

顔料ベースとしての黒雲母(対照例)
顔料を黒雲母(BM)とするバッチを実施例1に従って製造する。
顔料ベースとしてのマイクロマットブラック(MM13)
顔料をMMB(マイクロマットブラック(製造者:Merck, Darmstadt))とするバッチを実施例1に従って製造する。
試料を様々な顔料を含むMGPを用いて調整する、増幅後のシグナルレベル
実施例7及び8に記載のMGPを試料の調製のために利用する。100コピー/mlのHCVウィルスを含むヒト血漿を試料として用いる。この試料調製物の溶出液を増幅にかけ、そして増幅結果を電気化学発光法により検出する。追加の実験において、試料は600コピー/mlのHBVウィルスを含むヒト血漿とした。

RNAの結合の決定
1.4kbの32P−ラベル化HIV gag標準品を実施例6に従って実施例10の粒子に結合させる。放射能測定は>80%の結合を示す。
沈降速度の比較
実施例10の粒子3mgを2mlの容量を有する2本のEppendorf 容器に移し、そして各々1.5mlのH2 Oに懸濁する。
Claims (11)
- ガラス表面を有する粒子を含む調製物であって、当該粒子の75重量%超が0.5〜15μmの粒子サイズを有し、ここで当該ガラス表面がSiO2, B2O3, K2O, CaO, Al2O3及びZnOから構成される、調製物。
- 前記0.5〜15μmの粒子サイズを有する粒子の95重量%超が磁性粒子である、請求項1記載の調製物。
- 前記粒子の半分未満が2μm未満の粒子サイズを有する、請求項1又は2記載の調製物。
- 前記粒子の2%未満が0.5μm未満の粒子サイズを有する、請求項1,2及び3のいずれか1項記載の調製物。
- 前記磁性粒子がガラスでコーティングされた磁性コアを有する、請求項2記載の調製物。
- 前記粒子が2〜6モル%の酸化亜鉛を含むガラスコートを有する、請求項1〜5のいずれか1項記載の調製物。
- コアのない粒子5重量%未満を含むゲル層でコーティングされたコアを有し、当該ゲル層がSiO2, B2O3, K2O, CaO, Al2O3及びZnOから構成される粒子の調製物の製造方法であって、
・コア粒子調製物を利用してゾル中でコア粒子を懸濁し、
・この懸濁物をスプレードライしてゲルを形成する、
工程を含んで成り、ここで当該粒子調製物は75重量%の0.5〜15μmの粒子サイズを有する粒子を含んで成る、方法。 - 核酸を精製する方法であって、核酸を試料からZnOを含有するガラス表面を有する粒子へと非共有式に結合させ、未結合の試料成分を除去し、そして結合した核酸を当該ガラス表面から溶出させることによる方法。
- 前記試料を請求項1〜6のいずれか1項記載の調製物と接触させるものとする、請求項8記載の方法。
- コアのない粒子5重量%未満を含むガラス層でコーティングされたコアを有し、当該ガラス層がSiO2, B2O3, K2O, CaO, Al2O3及びZnOから構成される粒子の調製物の製造のための方法であって、
コア粒子調製物を利用してゾル中にコア粒子を懸濁し、
当該懸濁物をスプレードライしてゲルを形成し、
当該ゲルを圧縮してガラスを形成する、
工程を含んで成り、ここで当該コア粒子調製物を0.5〜15μmの粒子サイズを有する粒子75重量%を含んで成る、方法。 - 核酸のためのガラス表面の結合能力を増大させるため、ゾル/ゲル法により作製されるガラス層内で酸化亜鉛ガラス表面を利用する方法。
