1. 2−tert−ブチルオキシカルボニル−アミノヘキサデカノイル酸の合成
1.1. 2−アミノヘキサデカノイル酸の合成
分子式:C16H33NO2
分子量:271.44
方法:0.0298モルの2−ブロモヘキサデカノイル酸(10g)と100mlの28%をNH4OH溶液を、50%硝酸と10%リン酸で前もって洗浄された圧力釜に入れる。攪拌後、圧力釜を60℃で15時間加熱する。発生したアミノ誘導体は反応液中で沈殿する。冷却後、圧力釜を洗浄し、水分散しそしてエタノール除去した。反応液を4号の多孔質ガラスロートで濾過する。エタノール液中での種々の生成物の溶解性の違いに基づいて、洗浄により2−ブロモヘキサデカノイル酸を除去できる。
注意: アミノ誘導体の不溶性は多くの溶媒(水,エタノール,低温酢酸,70%ギ酸,エチルアセテート,トルエン,トルエン/アセトニトリル,エチルアセテート/アセトニトリル)中で試験された。試みられたすべての溶解試験の中で、アミノ誘導体は特別な洗浄剤(水酸化トリメチルベンジルアンモニウム)あるいは煮沸酢酸にだけ溶解した。
純度は85%であった。

−方法:
2−アミノヘキサデカノイル酸の溶解
1.1. 40%“トリトン”(水酸化ベンジルトリメチルアンモニウム)メタノール溶液の1.1当量(2.9ml)と100mlDMFを5mmol(1.655g)のアミノ酸酢酸塩に加えた。反応液が完全に溶解するまで室温で攪拌した。DMFを真空下で蒸発後、残留物をP2O5を入れたデシケーターで乾燥する。
乾燥残留物を36mlの水と8mlの1N KHCO3と30mlのtert−ブチルアルコールの混合液に溶解し、2.5当量のターブチルカーボネート(分子量=218.25)をこの溶液に加える。1N Na2CO3溶液でpHを8から9の間に調製し、反応初期数時間一定になるよう維持した。カップリングの状態は、シリカゲルを用いた薄層クロマトグラフィーで調べた。
1.3. 精製と特性
1.3.1. 精製
再結晶化をくり返すことにより、純度を高めること(融点の上昇)が可能であった。精製により、アミノヘキサデカノイル酸の場合は2%、保護アミノ酸の場合は1%以上の収率の減少は認められなかった。

分子イオンの実験測定値と理論値は同じ分子量を示した。分子イオンは断片化され、Boc基(270のピーク)が遊離された。296.6のピークは、CN基(ニトロセルロース)を含む270の質量をもつイオンを表している。
2.1. コレステリルトシレートの合成
−分子式 :C34H53SO3
−分子量:540.83
−方法:25.86mmolのコレステロール(10g)を最少量のピリジン(5から10ml)に溶解した後、過剰量のトシルクロライド(10g,52.6mmol)を加える。12時間攪拌後、溶液を蒸発乾凅することにより、ピリジンを除いた。その残留物を20mlのアセトンに高温で溶解した(オイル形成を避けるための温度を55℃以下に保つ)。反応混液を濾過し、濾液を結晶化させることによりコレステロールトシレートを回収した。回収した白色結晶を冷アセトンで洗浄し、P2O5の入ったデシケーターで乾燥した。
2.2. 3β−(2´−ハイドロキシエトキシ)−コレスト−5−エンの合成
−分子式 :C29H50O2
−分子量:430.71
−方法:30mlのエチルグリコール(480mmol)を17.5mmolのコレステリルトシレート(10g)の120mlのジオキサン溶液に加えた。この反応混液を120℃で4時間還流し、冷却後150mlの蒸留水に溶解した。形成されたアルコール誘導体をジエチルエーテル(3×200ml)で抽出した。エーテル相を5%NaHCO3(2×200ml)および蒸留水(2×200ml)で洗浄した。無水Na2SO4で乾燥後、エーテル溶液はオイル状になるまで濃縮された。4mlのヘキサンを添加後、冷却チャンバー(4℃)の中でこすりあわせと冷却により、結晶化を開始した。白色結晶は4号の多孔性ガラスロートに回収され、ヘキサンで洗浄後、P2O5を入れたデシケーターで乾燥した。
2.3. 3β−(2´−カルボキシメトキシ)−コレスト−5−エンの合成
−分子式 :C29H48O3
−分子量:444.69
−方法:酸化溶液はあらかじめ次の様に調製した。2.67gの無水クロム酸と2.3mlの濃硫酸に蒸留水を加えて10mlにした。この酸化溶液を4.66mmol(2g)の3β−(2´−ハイドロキシエトキシ)−コレスト−5−エンが溶解した50mlアセトン溶液に滴下した。反応の進行は薄層クロマトグラフィで検査した。一度反応が完結すると、反応液は250mlの蒸留水に混和し、酸性誘導体をエチルアセテートで抽出した(3×200ml)。その有機相を蒸留水で洗浄し(2×200ml)、無水Na2SO4で脱水後、オイル状になるまで濃縮した。4mlの石油エーテルを添加後、酸性誘導体の結晶化は、冷却チャンバー内(4℃)で冷却することにより促進された。白色結晶は4号の多孔性ガラスロートに回収され、石油エーテルで洗浄後、P2O5を入れたデシケーターで乾燥した。
2.4. 精製と特性
2.4.1. 精製
コレステリル P−トルエンスルホン酸をアセトン中でくり返し再結晶することにより精製した。β−(2´−ハイドロキシエトキシ)コレスト−5−エンとその酸性誘導体は、シリカゲル厚層クロマトグラフィーにより精製した。
試料の添加はシリカの厚層上に行われた(メルク社、蛍光指示薬付シリカゲル 60 PF254)。スポットは、紫外線により検出された。

実施例2:リポペプチドの合成
I. 2−アミノヘキサデカノイル酸結合の方法
細胞障害性T細胞の誘導を検討するために、HIV−1 LAV−BRUウイルスのgp120蛋白質の312から327のアミノ酸配列をリポペプチド形成に用いた。このアミノ酸配列を用いて、3回の調製を行った。
1) スクシニル連結を用いたピメラウチド(あるいはトリメキサウチド)のペプチドN末端への結合方法
この方法は、樹脂に固定された固相状態で合成されたペプチドへ無保護のピメラウチド(あるいはトリメキサウチド)を固定するため、また、保護された側鎖に対して適用された。
樹脂上でペプチドN末端を一時的に保護するターブチルオキシカルボニル基は、ジクロロメタン中の40%トリフルオロ酢酸の作用により、20分の攪拌操作で開裂される。
−20mlの5%ジイソプロピルエチルアミンのジクロロメタン溶液で2回
−20mlのジメチルホルムアミドで2回(それぞれの洗浄は3分間)
スクシニル化
スクシニル化溶液中の樹脂を次の試薬と反応することによりカップリング(連結)を3度行った。
−樹脂のアミンモル数に相当するジイソプロピルエチルアミン(20分間攪拌)
活性化
樹脂上のカルボキシ基の活性化は次のように行われた。
−HOBT(ハイドロキシベンゾトリアゾール):カルボキシルの3倍量
−ジイソプロピルエチルアミン:N−メチルピロリドン溶液中カルボキシルの7倍量
洗浄
溶液は次の様に洗浄した。
−30mlのジクロロメタンで3回
カップリング(連結)
樹脂は次のカップリング溶液で処理した。
−ジイソプロピルエチルアミン:活性化エステルの3倍量
−10%ジメチルスルホキサイド
−90%N−メチルピロリドン:ピメラウチド(またはトリメキサウチド)を溶解するための十分量
飽和溶液におけるピメラウチドあるいはトリメキサウチドの濃度は超音波処理と50℃,2分間の保持の後、約4%であった。
a) tert−ブチルオキシカルボニル基によるピメラウチド(またはトリメキサウチド)のN−保護
500ミクロモルのピメラウチド(またはトリメキサウチド)を10mlの0.1モル炭酸緩衝溶液(pH9.5)に溶解した。
合成終了後、ペプチドの遊離は、無水フッ化水素酸を用いて行った。
勾配:溶媒B:0.75 0/00 アセトニトリル0.5% トリフルオロ酢酸
120分間で10から80%のアセトニトリル勾配
215nmの波長で測定
酸加水分解の間、ピメラウチドとトリメキサウチドに含まれるジアミノピメル酸(Dap)は、良い標準結合物質となった。

実施例3
NP147−158ペプチドによる免疫
マウスの免疫は次の様にして行った。
分離したリポペプチドをマウスの腹腔(i.p)あるいは皮下(s.c)にアジュバントと共にまたは単独で投与した。
最終投与の後8から21日後、免疫されたマウスの脾臓を摘出し、脾臓細胞をリポペプチド合成に用いられたペプチドの5μMを含む培養液(DMEM+10%FeS+ピルビン酸+非必須アミノ酸+β−2−メルカプトエタノール)に5×106細胞/2mlの濃度で培養した。
イルス粒子,インフルエンザウイルスNP147−158R蛋白質、さらにNP147−158ペプチドとトリパルミトイル−S−グリセリルシステイニルセリルセリンの結合物であるP3CSS−PEPNP(DERESら、前述)を用いてすでに得られた結果を示してある。
ENV312−327ペプチドによるCB 1 ,CB 2 およびCB 3 での免疫
このペプチドはHIVウイルスのENV遺伝子により記録された蛋白質である。
ENV302−335ペプチドによる免疫
このペプチドはHIVウイルスのENV蛋白質302から335のアミノ酸残基から成るペプチド断片である。
CB 1 ,CB 4 ,CB 5 ,CB 17 ,CB 19 ,CB 21 およびCB 25 でのENV312−327ペプチドによる免疫
この実験方法は実施例3において述べたものとほとんど同じである。
Claims (17)
- 10個〜40個のアミノ酸および少なくとも1つの抗原決定基を有するペプチドと、
ピメラウチド、トリメキサウチド、ヘキサデカン酸および2−アミノ−ヘキサデカン酸からなる群より選ばれた少なくとも1種のものに由来する少なくとも1種の鎖、
ならびに/または上記アミノ酸のα−NH2および/またはε−NH2官能基と結合したステロイド類、を含有することを特徴とする細胞障害性Tリンパ球誘導リポペプチド。 - ステロイド類がN−リジンであるである請求項1に記載のリポペプチド。
- ステロイド類が(コレスト−5−エニル−3−オキシ)酢酸である請求項1に記載のリポペプチド。
- ペプチドがHIV−1またはHIV−2ウイルスの蛋白質断片である請求項1に記載のリポペプチド。
- 温血動物(ただし、ヒトを除く)に十分な量のリポペプチドを投与して細胞障害性Tリンパ球を誘導するに当たり、
上記リポペプチドが10個〜40個のアミノ酸および少なくとも1つの抗原決定基を有するペプチドと、
ピメラウチド、トリメキサウチド、ヘキサデカン酸、2−アミノ−ヘキサデカン酸、ステロイド類ならびに上記アミノ酸のα−NH2および/またはε−NH2官能基と結合したステロイド類の誘導体からなる群より選ばれた少なくとも1種の鎖、を有することを特徴とする細胞障害性Tリンパ球の誘導方法。 - 鎖が2−アミノ−ヘキサデカン酸またはその誘導体である請求項5に記載の細胞障害性Tリンパ球の誘導方法。
- ヘキサデカン酸がヘキサデカン酸またはその誘導体である請求項5に記載の細胞障害性Tリンパ球の誘導方法。
- 鎖がN−パルミトイルリジンである請求項5に記載の細胞障害性Tリンパ球の誘導方法。
- ヘキサデカン酸誘導体がN,N’−ジパルミトイルリジンである請求項5に記載の細胞障害性Tリンパ球の誘導方法。
- 鎖がピメラウチドまたはトリメキサウチドである請求項5に記載の細胞障害性Tリンパ球の誘導方法。
- ステロイド類がN−[(コレスト−5−エニル−3−オキシ)−アセチル]−リジンである請求項5に記載の細胞障害性Tリンパ球の誘導方法。
- ステロイド類が(コレスト−5−エニル−3−オキシ)酢酸である請求項5に記載の細胞障害性Tリンパ球の誘導方法。
- ペプチドがHIV−1またはHIV−2ウイルスの蛋白質断片である請求項5に記載の細胞障害性Tリンパ球の誘導方法。
- ペプチド部分が10個〜20個のアミノ酸を有する請求項5に記載の細胞障害性Tリンパ球の誘導方法。
- 温血動物(ただし、ヒトを除く)に十分な量のリポペプチドを投与して細胞障害性Tリンパ球を誘導するに当たり、
上記リポペプチドが10個〜40個のアミノ酸および少なくとも1つの抗原決定基を有するペプチドと、
ピメラウチド、トリメキサウチド、ヘキサデカン酸、2−アミノ−ヘキサデカン酸、ステロイド類ならびに上記アミノ酸のα−NH2官能基と結合したステロイド類の誘導体から成る群より選ばれた少なくとも1種の鎖、を有することを特徴とする細胞障害性Tリンパ球の誘導方法。 - 温血動物(ただし、ヒトを除く)に十分な量のリポペプチドを投与して細胞障害性Tリンパ球を誘導するに当たり、
上記リポペプチドが10個〜40個のアミノ酸および少なくとも1つの抗原決定基を有するペプチドと、
ピメラウチド、トリメキサウチド、ヘキサデカン酸、2−アミノ−ヘキサデカン酸、ステロイド類ならびに上記アミノ酸のε−NH2官能基と結合したステロイド類の誘導体から成る群より選ばれた少なくとも1種の鎖、を有することを特徴とする細胞障害性Tリンパ球の誘導方法。 - 請求項1〜4のいずれか1つの項に記載のリポペプチドを含有することを特徴とする細胞障害性Tリンパ球誘導用ワクチン組成物。
