Abstract
【解決手段】本発明で開示する新規なDNA発現ベクターは、少なくとも酵母Gタンパク質結合受容体の一番端のアミノ末端暗号配列のフラグメントから成る第1セグメントを含む。第2セグメントは第1セグメントの下流(さらに正確な読み取り枠内)に配置し、異種Gタンパク質結合受容体を暗号化するDNA配列から成る。得られる酵母細胞では、第1および第2の異種DNA配列は発現されるが、内因性Gタンパク質αサブユニット(酵母Gα)を発現しないようにすることができる。これらの細胞は細胞内でGαがGβγから解離する速度に影響を与える化合物をスクリーニングするのに有用である。

Description
発明の分野
本発明は異種Gタンパク質結合受容体を発現する酵母細胞、かかる細胞を作るのに有用なベクター、およびこれらのものを用いる方法に関する。
種々の細胞外シグナル (例えば、神経伝達物質、ホルモン、臭気物質、光) の作用は7種のトランスメンブラン領域を有する受容体(Gタンパク質結合受容体)およびヘテロ三量体(heterotrimeric)グアニンヌクレオチド結合性調節タンパク質(Gタンパク質) により媒介される(例えば、非特許文献1及び2参照)。かかるGタンパク質媒介シグナル系は酵母およびヒトのような異なる生物体で確認されている(例えば、非特許文献1、2及び3参照)。β2 アドレナリン受容体(βAR)は哺乳類細胞内のリガンド結合性受容体の7種のトランスメンブランセグメントの原型である。エピネフリンまたはノルエピネフリンに対する応答で、βARは細胞内でアデニル酸シクラーゼおよびサイクリックアデノシン一リン酸産生を交互に刺激するGタンパク質、Gs を活性化する(例えば、非特許文献1及び2参照)。酵母内でGタンパク質結合フェロモン受容体はaおよびα半数体細胞型が接合 (融合) してa/α二倍体を形成するようになる発生プログラムを制御する(例えば、非特許文献3及び4参照)。

本発明の第1の観点は哺乳類Gタンパク質結合受容体の遺伝暗号を指定する第1の異種DNA配列および哺乳類Gタンパク質αサブユニット(哺乳類Gα)の遺伝暗号を指定する第2の異種DNA配列を含む形質転換した酵母細胞である。第1および第2の異種DNA配列はこの細胞内で発現させることができるが、この細胞は内因性Gタンパク質αサブユニット(酵母Gα)を発現することができない。この細胞はフェロモン応答プロモータおよびフェロモン応答プロモータから下流に配置しさらに効果的に結合させた指示遺伝子から成る第3の異種DNA配列を含む。
本発明の第2の観点は細胞においてGαがGβγから解離する速度に影響を与える化合物の能力を試験する方法である。この方法は:上述のように形質転換した酵母細胞を供給し;化合物を細胞に接触させ;さらに次いで細胞においてGαがGβγから解離する速度を検出することから成る。細胞は水溶液中に供給される場合があり、水溶液に化合物を添加することにより接触工程を実施した。
本発明の第4の観点は上述のようなベクターにより形質転換した酵母細胞である。

ムスカリン性コリン作動性受容体(種々のサブタイプ)はイー.ペラルタ(E.Peralta) らの「Nature」 343、 434ページ (1988)およびケイ.フクダ(K.Fukuda)らの「Nature」 327、 623 ページ(1987)に開示されている。
α2 アドレナリン受容体の種々のサブタイプはジェイ.リーガン(J.Regan) らの「Proc.Natl.Acad.Sci.USA」85、6301ページ(1988)およびR.Lefkowitz およびM.Caron の「J.Biol.Chem.」263 、4993ページ(1988)に開示されている。
セロトニン受容体(種々のサブタイプ)はエス.ペロートカ(S.Peroutka)の「Ann.Rev.Neurosci. 」11、45ページ(1988)に開示されている。
カンナビノイド受容体はエル.マツダ(L.Matsuda) らの「Nature」 346、 561ページ(1990)に開示されている。
本発明の実施に有用なベクターにはプラスミド、ウイルス(ファージを含む)、および組み込み可能な(integratable)DNAフラグメント(すなわち、相同組み換えにより宿主ゲノムに組み込み可能なフラグメント)が含まれる。ベクターは、プラスミドの場合、宿主ゲノムとは無関係に再製し機能することができるが、組み込み可能なDNAフラグメントの場合には、ゲノム自体に組み込むことができる。適当なベクターには意図する発現宿主に親和性のある種から誘導した制御配列およびレプリコンが含まれる場合がある。例えば、宿主細胞で操作可能なプロモーターはこの細胞のRNAポリメラーゼに結合するものであり、宿主細胞で操作可能なリボソーム結合部位はこの細胞の内因性リボソームに結合するものである。
DNA領域はこれらが互いに機能的に関連する場合に操作可能に結合される。例えば:プロモーターはこれが配列の転写を制御する場合には暗号配列に操作可能に結合され;リボソーム結合部位はこれを翻訳が可能になるように配置する場合には暗号配列に操作可能に結合される。一般に、操作可能に結合させたとは、先導配列の場合において接触していること、読み取り相(reading phase) で接触していることを意味する。
異種アドレナリン受容体およびこの同種のGタンパク質αサブユニットの発現により酵母フェロモン応答経路を制御する能力は哺乳類Gタンパク質結合受容体の構造上のおよび機能上の特徴づけを促進する可能性を有する。増殖の停止またはβガラクトシダーゼの誘導のような応答の記録により、次に変異受容体の機能的特性を迅速に試験することができる。同様に、推定されている他のGタンパク質結合受容体の遺伝子を単離した際、多数のリガンドを予備選択して以前未確認であった受容体に対して活性を有するリガンドを確認することができる。エフ.リバート(F.Libert)らの「Science 」 244、 569ページ(1989);エム.エス.チー(M.S.Chee)らの「Nature」 344、 774ページ(1990)を参照。さらに、推定されている他のGタンパク質αサブユニットの遺伝暗号を指定している遺伝子を単離し、これらを本発明の細胞中で発現させて種々のGタンパク質結合受容体およびリガンドで予備選抜してこれらのサブユニットを特徴づけることができる。またこれらの細胞を用いて受容体−Gタンパク質相互作用に影響を及ぼす化合物を予備選択することができる。
本発明の細胞はマイクロタイタープレートのウエルに既知の、所定量で配置して上述の種々のスクリーニング方法に従って化合物をスクリーニングするために有用な標準化したキットを提供することができる。
実施例1
ヒトβ 2 アドレナリン発現ベクターpYβAR2の構成および酵母における発現
酵母でヒトβ2 アドレナリン受容体(hβAR)の高レベル発現を達成するため、修飾したhβAR遺伝子を多コピーベクター、YEp24(pYβAR2)のGAL1プロモーターの制御下に配置した。
図1は酵母発現プラスミドpYβAR2の構成を示す。pYβAR2では、hβAR配列の発現はGAL1プロモーターの制御下にある。図1Aは5′非翻訳領域およびpTZNARのhβAR遺伝子の暗号配列の最初の63塩基対(bp)を示し、ビー.オー′ドード(B. O ′Dowd) ら、「J.Biol.Chem.」 263、 15985ページ(1988)、これをAat II 開裂により除去されSTE2遺伝子(SEQ ID NO:1;SEQ ID NO:2) からの11bpの非暗号配列および42bpの暗号配列に対応する合成オリゴヌクレオチドで置換した。エヌ.ナカヤマ(N.Nakayama)らの「EMBOJ.」 4、2643ページ(1985);エイ.ブルクホルダー(A.Burkholder)およびエル.ハートウェル(L.Hartwell)の「Nucleic Acids Res.」13、8463ページ(1985)を参照。得られたプラスミド、 pTZYNARはSEQ ID NO:3 としてここに記載した3′Hind III部位を有する非暗号配列においてHind III部位により側面を接する修飾したhβAR遺伝子を含む。Hind III −Hind IIIフラグメントをpTZYNARから単離し修飾したhβAR遺伝子の3′非翻訳配列に次いで酵母ADH1遺伝子からの終結配列を含む 450bpが続くようにpAAH5に挿入した。ジー.アメラー(G.Ammerer) の「Methods.Enzymol.」 101、 192ページ(1983)を参照。
図1Bに示すように、pyβAR2はYEpG24中にhβARおよびADH1配列を含むBam HI −Bam HI フラグメントを挿入することにより構成した。イー.ウィコフ(E.Wyckoff) およびティー.ヒセー(T.Hsieh) の「Proc.Natl.Acad.Sci. U.S.A.」85、6272ページ(1988)を参照。最大の発現が調査され、細胞をLAC9、GAL1調節転写に必要なS.cerevisiae GAL4トランス作用転写活性タンパク質(transactivater protein)の異種相同物を含むプラスミドpMTL9(Dr.S.Johnston から入手)で共形質転換した。ジェイ.サルメロン(J.Salmeron)らの「Mol.Cell.Biol.」 9、2950ページ(1989)を参照。後期指数期にまで増殖した細胞を3%のガラクトースを含み、約10μMのアルプレノロールを追加した培地中で誘導し、さらに36時間増殖させた。細胞の維持のために標準法を使用した。エフ.シェルマン(F.Sherman) らの「Method in Yeast Genetics(ニューヨーク州コールドスプリングハーバー所在のコールドスプリングハーバー研究所、1986年)」を参照。
最大の発現は(i) トランス作用転写活性タンパク質(LAC9)の発現、(ii)酵母STE2(α因子受容体)遺伝子の対応する領域を有するhβAR遺伝子の5′非翻訳領域および一番端のアミノ末端暗号配列の置換、(iii) 細胞増殖が後期指数期に達した際のガラクトースでの誘導、および(iv)誘導の間の増殖培地にアドレナリン性リガンドを包含することを必要とした。
プラスミドpYβAR2をブタペスト条約の規定に従って米国メリーランド州20852 、ロックビル、パークロウンドライブ12301 所在のアメリカンタイプカルチャーコレクションに1990年9月11日に寄託し、ATCC受諾No.40891が割り当てられた。
pYβAR2で形質転換した酵母のhβARリガンドの結合親和力
細胞表面受容体の本来の機能は他の細胞外刺激中から適当なリガンドだけを認識することである。従って、リガンド結合親和力を決定して酵母で発現されたhβARの機能的な完全性を確立した。詳細を次に記載するように、拮抗薬、125I標識シアノピンドロール(cyanopindolol)( 125I−CYP)を飽和するように、pYβAR2で形質転換した酵母細胞から調製した膜に対して高い親和力で結合した。一連の作動薬で 125I−CYPを置換することによりhβARの期待される効力および立体特異性の程度を観察した。
pYβAR2(URA3)およびpMTL9(LEU2)が含まれるSC261細胞(MATa ura3−52 trpl leu2 prb1−1122 pep4−3 prcl−40) 〔エス.ジョンストン(Dr.S.Johnston) から入手〕を最少グルコースフリー選択培地で後期対数期(OD600=5.0) まで増殖させ、次いで3%のガラクトースおよび40μMのアルプレノロールを添加して誘導した。36時間後、細胞を収穫し記載したようにスフェロプラストを調製した。E.Wyckoff および T.Hsiehの「Proc.Natl.Acad.Sci.U.S.A. 」85、6272ページ(1988)を参照。簡単には、スフェロプラストを50mMのトリス−塩酸pH7.4 、5mMのEDTA中で再懸濁しガラスビーズを用いた渦流混合により4℃で3度1分間溶解した。粗膜を溶解液から調製し 125I−CYPとの結合を以前記載した方法によりアッセイした。H.Dohlman らの「Biochemistry」29、2335ページ(1990)を参照。
図2はpYβAR2で形質転換した酵母細胞由来の膜に対するhβARリガンドの結合を示す。(A) 125I−CYP濃度を変化させること(5〜 400pM) によりBmax (最大リガンド結合) およびKd (リガンド解離定数)値を決定した。特異的結合は全結合(丸で示す)から10μMの(−)アルプレノロールの存在下で測定した非特異的結合(四角で示す)を引いた量として定義する。 125I−CYP結合のために93pMのKd を得て作動薬親和力(次の)を計算するために用いた。(B) 種々の濃度の作動薬を使用した18pMの 125I−CYPの置換を用いて受容体結合のための明らかに低い親和力Ki 値(50μMのGTPの存在下に決定した非Gタンパク質結合) を決定した、四角;(−) イソプロテレノール、丸;(−)エピネフリン、下方に尖った三角;(+)イソプロテレノール、上方に尖った三角;(−)ノルエピネフリン。

図2(A) および(B) の結合データは非線形最小自乗法回帰により分析し、エイ.デリーン(A.DeLean)らの「Mol.Pharmacol.」21、(1982)参照、さらに表Iに示す。記載した値は3回の測定値の平均であり、2〜3度の試験を代表する。酵母の結合親和力は以前哺乳類細胞で発現したhβARについて観察されたものとほとんど同一であった。
酵母発現hβARの接合シグナル伝達による作動薬依存活性化
受容体の第2の主要な機能はシグナル伝達経路での下流成分の作動薬依存調節である。酵母のフェロモン応答エフェクターは知られていないので、間接的な生物学的アッセイが最も有効な受容体機能の指標である。K.BlumerおよびJ.Thorner の「Annu.Rev.Physiol. 」出版中;I.Herskowitzの「Microbiol.Rev.」52、 536ページ(1988)を参照。高濃度のhβARを発現する酵母で、接合シグナル伝達の作動薬依存活性化は任意の代表的なインビボアッセイ;例えば、G1 期停止の賦課物(impositoin)、遺伝子発現の誘導、形態の変化(いわゆる「シュモー」形成)または接合の刺激により検出することができない。応答の欠如に対する適当な解釈はhβARが内因性酵母Gタンパク質と結合することができないことである。
酵母におけるhβARおよび哺乳類G S αサブユニットの同時発現
哺乳類GS αサブユニットの発現はGPA1遺伝子により暗号化された対応する内因性タンパク質を欠如する酵母細胞における増殖の欠陥を修正することができる。シー.ディーツェル(C.Dietzel) およびジェイ.クルジャン(J.Kurjan)の「Cell」50、1001ページ(1987)を参照。さらに、哺乳類細胞の受容体結合の特異性はGタンパク質のαサブユニットにより与えられる。L.StryerおよびH.Bourneの「Ann.Rev.Cell Biol.」 2、 391ページ(1988)を参照。このように、酵母でのhβARおよび哺乳類GS αサブユニット(GS α)の同時発現は酵母をアドレナリン性リガンドに対し感受性にするめに試みられた。従って、銅誘導可能(copper-inducible)CUP1プロモーターの制御下にラットGS αを暗号化するcDNAを第2のプラスミド、pYSK 136Gαsに導入した。C.Dietzel およびJ.Kurjanの 「Cell」50、1001ページ(1987)を参照。hβARおよびラットGS αを同時発現するが、野性型GPA1を含む酵母(NNY19)で、アドレナリン性作動薬誘導シュモー形成は観察されず、接合フェロモンに応じた酵母の特有な形態変化が観察された。
内因性Gタンパク質αサブユニットを欠如する酵母におけるhβARおよび哺乳類G S αサブユニットの同時発現
内因性酵母Gタンパク質αサブユニットによる干渉を防止するため、gpa1突然変異細胞(8c株)を用いた。
酵母株8c(MATa ura3 leu2 his3 trpl gpal::H1S3)、アイ.ミヤジマ(I.Miyajima)らの「Cell」50、1011ページ(1987)、担体プラスミド(carrying plasmid)pYSK 136Gαs(TRP1)、C.Dietzel およびJ.Kurjanの「Cell」50、1001ページ(1987)、pMTL9(LEU2)、J.Salmeronらの「Mol.Cell.Biol.」 9、2950ページ(1989)、およびpYβAR2(URA3)を3%のグリセロール、2%の乳酸、50μMのCuSO4 および3%のガラクトースを含むグルコースフリーの最小選択プレートに維持した。コロニーを0.5mMのアスコルビン酸および望ましいアドレナリン性リガンドを含む同様のプレートに移しかえた。30℃で16〜20時間の後、コロニーを106 〜107 細胞/mlの密度で同様の液体培地に移しかえ位相差顕微鏡により試験した。
pYβAR2、pMTL9、およびpYSK 136Gαsで共形質転換した酵母細胞の形態は(A) アドレナリン性薬品を含まず;(B)100μMの(−)イソプロテレノール;(C)100μMの (−)イソプロテレノールおよび50μMの(−)アルプレノロール;および(D)100μMの(+)イソプロテレノールとともに温置した後試験した。結果はhβARおよびラットGS αを同時発現する8c細胞のβアドレナリン性作動薬イソプロテレノールを用いた処理が実際にシュモー形成を誘導し、この効果が特定の拮抗薬アルプレノロールにより阻害されることを示した。
βガラクトシダーゼのシグナル配列を含む酵母におけるhβARおよび哺乳類G S αサブユニットの同時発現
hβARおよびラットGS αを同時発現する8c細胞をイソプロテレノールで誘導した形態学的応答はこれらの成分が互いにおよび内因性Gαサブユニットを欠如する酵母でフェロモン応答経路の下流成分に結合することができることを示唆している。フェロモンシグナル経路がhβARおよびラットGS αにより活性化されることを確認するため、フェロモン応答FUS1遺伝子プロモーターの作動薬誘導をFUS1−lacZ遺伝子融合がゲノムに確実に組み込まれた8c細胞(8c1)から誘導した酵母の系統で測定した。エス.ノモト(S.Nomoto)らの「EMBO J. 」 9、 691ページ(1990)を参照。
系統8c(図3、凡例)およびNNY19(MATa ura3 leu2 his3 trp1 lys2 FUS1−LacZ::LEU2)をYIpFUS 102(LEU2)での組み込み形質転換により修飾し、S.Nomotoらの「前記」、それぞれ8clおよびNNY19と略記した。これらの系統をpYβAR2、およびpYSK 136Gαsで形質転換しグルコースおよび50μMのCuSO4 を含む最小選択プレートに維持した。コロニーを最小選択培地(3%のグリセロール、2%の乳酸、50μMのCuSO4 )に接種し、初期対数期(OD600=1.0) まで増殖させ、さらに3%のガラクトースの添加により12時間誘導した。細胞を洗浄し0.5mMのアスコルビン酸および望ましいリガンドを含む誘導培地(OD600=5.0) に再び懸濁した。30℃で4時間温置した後、細胞を収穫し1mlのZ緩衝液、ジェイ.ミラー(J.Miller)の「Experiments in Molecular Genetics 」(ニューヨーク州コールドスプリングハーバー所在の、コールドスプリングハーバー研究所、1972)を参照、に0.0075%のSDS を追加して、再び懸濁し、さらにβガラクトシダーゼ活性を以前記載したように3〜4回の独立した試験で測定した。J.Millerの「前記」を参照。
図3にはフェロモン誘導可能な遺伝子活性に及ぼすβアドレナリン性作動薬の影響を比較して示す。α−MF、10μMα接合因子;(−)ISO、50μM(−)イソプロテレノール; (−)ALP、50μM(−)アルプレノロール;(+)ISO、 100μM(+)イソプロテレノールである。hβARおよびラットGS αを同時に発現する8cl(gpa1)細胞において、イソプロテレノールで刺激されたβガラクトシダーゼ活性の劇的な誘導を観察した。作動薬刺激は立体特異的であり特定の拮抗薬を添加することにより阻害された。作動薬応答はhβARおよびラットGS αの発現に依存し、内因性Gタンパク質αサブユニットが分裂した系統を必要とした。形質転換した8cl細胞でイソプロテレノールに応じて達成した最終的βガラクトシダーゼ活性はGPA1(NNY19)を発現する形質転換していない細胞でα因子により誘導したものに匹敵したが、NNY19細胞の基底の(basal) βガラクトシダーゼ活性は8cl細胞よりかなり低かった。総合して、本発明者らの結果はhβARおよびラットGS αの同時発現が酵母の接合シグナル伝達経路のカテコールアミン制御キー観点(control key aspect)の下に配置するのに十分であることを示した。しかし、アドレナリン性作動薬は酵母フェロモン受容体が、結合フェロモンに加え、接合に必要な他の認識事象に関連するという最近の観察結果と一致して、hβARおよびラットGS αを同時に発現する8c細胞またはNNY19細胞のどちらの接合をも刺激しない。エイ・ベンダー(A.Bender)およびジー・スプラーグ(G.Sprague) の「Genetics」 121、 463ページ(1989)を参照。
hβARは動物細胞のアデニル酸シクラーゼをGタンパク質のαサブユニットの作用を介して刺激する。対照的に、酵母の接合因子受容体はこれらのエフェクターをβγサブユニットの作用を介して誘発する。エム.ホワイトウェイ(M.Whiteway)らの「Cell」56、 476ページ(1989)を参照。本発明者らの本結果は酵母のhβARの活性化が酵母のβγから哺乳類GS αを解離させること、およびこれがおそらく応答を誘い出すβγサブユニットであることを示している。
上述の例は本発明の例示であり、制限と解釈すべきではない。本発明は、ここに含まれる請求の範囲と均等なものとともに、次の請求の範囲により明確となる。
Claims (13)
- 酵母細胞の細胞膜中にトランスメンブランタンパク質を発現させることができるDNA発現ベクターであって:
酵母Gタンパク質結合受容体の少なくとも一番端のアミノ末端暗号配列のフラグメントから成る第1セグメント;および
前記第1セグメントから下流でさらに正確な読み取り枠内の第2セグメントであって、異種Gタンパク質結合受容体を暗号化するDNA配列から成る第2セグメントから成り、前記異種Gタンパク質結合受容体がGタンパク質と機能的に相互作用し得ることを特徴とするDNA発現ベクター。 - 前記異種Gタンパク質結合受容体の一番端のアミノ末端暗号配列のフラグメントが欠如していることを特徴とする請求項1記載のDNA発現ベクター。
- 前記第1および第2セグメントが、酵母細胞において操作可能なプロモーターと動作可能に結合していることを特徴とする請求項1記載のDNA発現ベクター。
- 前記プロモーターがGAL1プロモーターであることを特徴とする請求項3記載のDNA発現ベクター。
- 前記第1セグメントが酵母フェロモン受容体の少なくとも一番端のアミノ末端暗号配列のフラグメントから成ることを特徴とする請求項1記載のDNA発現ベクター。
- 前記第1セグメントがSTE2遺伝子およびSTE3遺伝子から成る群より選ばれる酵母フェロモン受容体の少なくとも一番端のアミノ末端暗号配列のフラグメントから成ることを特徴とする請求項1記載のDNA発現ベクター。
- さらに前記第1セグメントの上流に配置され動作可能に結合されている酵母Gタンパク質結合受容体遺伝子の少なくとも5′非翻訳領域のフラグメントを含んでいることを特徴とする請求項1記載のDNA発現ベクター。
- さらに前記第1セグメントの上流に配置され動作可能に結合されている酵母フェロモン受容体遺伝子の少なくとも5′非翻訳領域のフラグメントを含んでいることを特徴とする請求項1記載のDNA発現ベクター。
- 酵母フェロモン受容体遺伝子がSTE2遺伝子およびSTE3遺伝子から成る群より選ばれていることを特徴とする請求項8記載のDNA発現ベクター。
- 前記ベクターがプラスミドから成ることを特徴とする請求項1記載のDNA発現ベクター。
- 前記第2セグメントが哺乳類Gタンパク質結合受容体を暗号化するDNA配列から成ることを特徴とする請求項1記載のDNA発現ベクター。
- 前記第2セグメントが、ドーパミン受容体、ムスカリン性コリン作動性受容体、αアドレナリン受容体、βアドレナリン受容体、オピエート受容体、カンナビノイド受容体、およびセロトニン受容体から成る群より選ばれている哺乳類Gタンパク質結合受容体を暗号化するDNA配列から成ることを特徴とする請求項1記載のDNA発現ベクター。
- 請求項1記載のDNA発現ベクターを有することを特徴とする酵母細胞。
