本発明は、液状媒体、特に水、タンパク質溶液、又は非経口投与用液体から内毒素を分離するためのミクロ濾過膜に関し、該濾過膜は共有結合した内毒素用のリガンドを特徴とし、その際該リガンドは膜に設けられた一種のポリマーに担持されている。

【発明の詳細な説明】
内毒素に特異性の膜
本発明は、液状媒体から内毒素を分離するためのミクロ濾過膜並びに該ミクロ
濾過膜の利用に関する。
内毒素(エンドトキシン)はグラム陰性菌の細胞壁に存在するリポ多糖で、発
熱物質として作用する。細菌の遍在性により内毒素もどこにでも存在する。しか
し内毒素は細菌と異なり、濾過滅菌又は加圧滅菌のような通常の方法では除去し
たり無害にしたりすることはできない(1)。従って無菌とは内毒素がないという
ことではない。特に問題になるのは注射液や輸液(非経口投与用液体)の中の内
毒素の存在で、静脈内投与の場合体重1kg当たり1ngの量で既に発熱を引き起こ
す。その症候群は投与量の多い場合(例えば多量の非経口投与用液体により)激
しいショックから死亡に及ぶこともある(2,3)。そのため殆ど全ての薬局方には
無菌状態に加えて内毒素の厳しい許容最高値、例えば注射用クロラムフェニコー
ルmg当たり0.2EU(内毒素単位)又はヘパリン単位当たり0.003EUのような記載が
ある(4)。この要求を満たすことは実際にはなかなか容易ではなく、特に生物利
用の薬剤の生産では全ての段階を内毒素なしにすることは不可能で、問題になる
主な内毒素源は次の通りである。
●既に細菌による汚染の可能性があるプラズマ又は組織のような原料、
●組換え生成物では宿主由来の内毒素を考慮しなければならない。
●生産の際の使用機材、フィルター、又は副資材からの細菌による汚染。
耐熱性作用物質の場合によく使用される加熱による汚染除去(250℃で30分)
や酸、アルカリ、又は強酸化剤(過酸化水素)による処理はこれらの製剤に対し
ては何れも適当ではない(1)。
活性炭又は深床フィルター(deep-bed filter)例えばZETA PLUSの使用は生成
物の大きな損失を招くことが多いので、その使用は水の処理に限られる(5、6)。
限外濾過は非常に穏やかな方法であるので内毒素除去の分野で広く使用される
ようになった。この場合、高分子集合体(数百万までの分子量)の他に存在する
モノマーの成分(分子量約14000)を有効に分別するために、普通10000又は5000
の分画分子量で作業するが、それでも同様に発熱を引き起こす低分子量の分解生
成物(例えばリピドA)が常に問題となる。これは特に血液透析の場合に問題で
、透析用緩衝液を限外濾過することは可能ではあるが、それでも例えば米国に於
いて年間400000名の血液透析の患者に敗血症が発生している(7)。―このための
小さい分画分子量の必要性により更に限外濾過の適用が低分子量物質の汚染除去
に限定される(8)。
薬用タンパク質、アルブミン製剤、又はヘパリンのような高分子の製品の問題
は未だに解決されておらず、これらの製品が内毒素で汚染されている場合にFDA,
USP,又はEPの規程に合致させるには、現在の所再処理するしか方法がない。
この面倒な方法によらずに製品をその本来の用途に使用する目的で、汚染した
製品を、内毒素に特異性のあるリガンドを備えたクロマトグラフ用収着媒体によ
り選択的にその汚染を除去しようとする
試みでは、その所期の目的を達成できなかった。リガンドとしてHis,Him,PMBを
備えた従来公知の高親和性収着媒体は、その高い乃至非常に高い会合定数にもか
かわらず内毒素の所期濃度が高い場合には不適当であることが判明した(9)。更
にタンパク質が存在していると競合するタンパク質の収着が認められ、これが内
毒素の除去速度を遅らせ、一部にはタンパク質の大きな損失を招いた(特にBSA
のような酸性タンパク質の場合)。 本発明によれば上述の従来の技術を、液状媒体、特に水、タンパク質溶液、又
は非経口投与用液体から内毒素を分離するためのミク
ロ濾過膜によって改善することができ、このミクロ濾過膜は、膜に取り付けられ
た一種のポリマーに担持された、共有的に結合した内毒素用のリガンドを特徴と
する。
膜の技術と膜の製造に関してはHo & Sirkar編:膜ハンドブック(出版社van No
strand Reinhold,NewYork,1992)を参照されたい。
使用可能の共有結合したリガンドとしては
(a)内毒素に特異性の一種のリガンド、好ましくはヒスタミン、ヒスチジン、ポ
リエチレンイミン、ポリ-L-リシン、又はポリミキシン及び/又は
(b)それ自体は内毒素に非特異性の一種のリガンド、好ましくはジアミノヘキサ
ン、ヂエチルアミノエチル、又はデオキシコール酸塩がある。
本発明のミクロ濾過膜の膜の材料としては、再生セルロース、酢酸セルロース
、ポリスルホン、ポリエチルビニルアルコール、又はポリアミド、特にナイロン
がある。
本発明のミクロ濾過膜に設けられるポリマーとしては、親水性のポリマー、特
にデキストラン、ポリビニルアルコール、又はセルロース誘導体、特にヒドロキ
シエチルセルロースが用いられる。
これらの親水性のポリマーはそれ自体水に可溶、水で膨潤、又は水に不溶であ
ってもよい。
このポリマーは本発明のミクロ濾過膜に一種のスペーサーを用い
て担持させることができる。共有結合したリガンドも一種のスペーサーに担持し
てもよい。この種のスペーサーとしては、ビスオキシラン、グルタルジアルデヒ
ド、エピハロゲンヒドリン、又はジイソシアナートに由来のスペーサーが、場合
によっては酸化により活性化して使用される。
内毒素に特異性のリガンドとスペーサーとに関する活性化と固定化の技術につ
いては例えば、Hermanson,Mallia & Smith著、固定化した親和性リガンドの技術
(出版社Academic Presslnc.,San Diego 1992)を参照されたい。
即ち本発明は、水又は水溶液(緩衝液、タンパク質溶液)から内毒素を除去する
、表面を適当な方法で修飾したミクロ濾過膜を提供する。表面の修飾は二官能性
の共有結合したスペーサを設けて行うことができる。このスペーサは親水性ポリ
マーと反応し、その際膜の、特にタンパク質との非特異的交互作用が少なくなる
。共有結合した親水性ポリマーは、場合によってはもう一つのスペーサを介して
、内毒素に特異性のリガンドと反応することができる。膜の表面修飾の原理に関
しては図1を参照されたい。
内毒素除去の効果はタンパク質の存在のもとで、タンパク質の正味の電荷に関
連して判定できる。条件(pH-値)を最適に設定すれば、酸性のタンパク質(例
えばBSA及びマウスIgG 1)の場合殆ど完全に汚染が除かれ、しかもタンパク質の
損失は認められない。塩基性タンパク質(例えばリゾチーム及びbFGF)の場合に
も同様に高い除去率が達成される。
ポリマーを被覆し、内毒素に特異性のリガンドを共有結合した本
発明のミクロ濾過膜は、一回の通過で内毒素を除去することができ、それも高濃
度の溶液(6000EU/ml)からの除去が可能である。
原理的にはこの膜は図1に相当して構成される。先ず一つのスペーサーの上に
一つの親水性ポリマーを取り付け、更に場合によってはもう一つのスペーサーを
介して、内毒素に特異性のリガンドと反応させる。膜の材料としては特に次のも
のがある。
●セルロース
●ポリスルホン
●PEVA(ポリエチレンビニルアルコール)
●ポリアミド(特にナイロン、例えばN 66)
スペーサとしては二官能性の化合物、特に次の化合物が適している。
●ビスオキシラン
●グルタルジアルデヒド
●エピハロゲンヒドリン
●ジイソシアナート
ビスオキシラン及びエピハロゲンヒドリンの使用の場合に生ずるビシナル・ジ
オール結合の活性化には、場合によっては過ヨウ素酸塩による酸化が用いられ、
その際アルデヒド基が生成する。膜に結合したスペーサーが更に親水性ポリマー
と反応するが、それには好ましくは次のポリマーが用いられる。
●デキストラン
●ポリビニルアルコール(PVA)
●セルロース誘導体、特にヒドロキシエチルセルロース(HEC)
その他の反応を直接内毒素に特異性のリガンドと実施するか或いは上述のスペ
ーサーの一つを介して、場合によってはこれを酸化により活性化してから実施す
る。内毒素に特異性のリガンドとして作用するのは、DAH,Him,His,PEI,PLL,PMB
である(略語の表参照)。DEAE,DOCのような普通は内毒素に非特異性のリガンドも
膜の構成の中では高特異性を示し、同時にタンパク質が大幅に存続する。
この開発した膜の能力は引用した例からも察知されるように、内毒素の除去を
殆ど常に完全に行うことができ、残存内毒素が普通は1EU/ml未満で、LAL-試験
の検出限度未満のこともよくみられる。
内毒素に特異性のリガンドを含まない対照膜(疎水性ポリマーをスペーサー付
き又は無しで設けた修飾なしのナイロン)では内毒素の除去は認められなかった
。
本発明の新規の膜は水及び非経口投与用液体からの内毒素の除去に使用でき、
タンパク質が存在していても好結果が得られる。但し塩基性タンパク質の場合に
は、タンパク質と内毒素との間に相互作用が発生して内毒素を隠蔽する可能性が
あることを考慮する必要がある。タンパク質と結合した内毒素はLAL-試験により
明瞭には検出できなくなる。これと関連して、タンパク質と結合した内毒素が尚
毒性を有るかに就いては、最終的には解明されていないことを付言する。
本発明の膜は次のような多様な用途に利用できる。
医学、薬学の分野では
●血液透析
●懸念のない輸液、注射液(非経口投与用液体)
●安全な診断薬(例えば抗体)
生物工学に於いては
●医薬品の生産
●プロセス用水や原料からの内毒素の除去
●製品の汚染排除(再処理費用の節減)
方法
1.膜の製造
ナイロン系のミクロ濾過膜(好ましくは厚さ0.45μm以上)に親水性ポリマー
、特にデキストラン、ポリビニルアルコール、及びヒドロキシエチルセルロース
を共有結合する。次の工程で結合したポリマーに、内毒素に特異性のリガンドを
固定する。図1に膜の構成を示す。
1.1.膜の被覆、デキストランの例
先ずナイロンの膜をビスオキシランにより活性化した。それにはビスオキシラ
ン9ml、エタノール1ml、25mMの炭酸ナトリウム緩衝液(pH11)1mlの混合液の中
で膜を80℃で16時間振り動かした(図2a)。充分洗浄してから膜1枚毎に分子量40
000のデキストランの20%溶液(pH11)5mlで15分間室温でインキュベートした(図
2b)。次に膜を120℃で14時間乾燥し、非特異的に結合したデキストランを除くた
めに膜を0.1M水酸化ナトリウムで3回、次に水で3回洗浄した。
図3に示すように、この被覆した膜は―ヘモグロビンの吸着量により示される
―非特異性の相互作用が著しく少ないことがその特徴である。
更に図3から、一層のデキストランの被覆ではPVAやHECと同じような効果は得
られないことが判る。第二の層により更に改善されるが、第三の層ではその作用
は僅かである。従ってデキストランは常に二重の被覆層として使用された。
1.2.内毒素に特異性のリガンドの固定
リガンドPLL,PMB,PEIは過ヨウ素酸塩で活性化した被覆ポリマーに直接加える
か、或いは過ヨウ素酸塩で酸化したスペーサー(ビスオキシラン)に合体してか
ら固定化した。この過程を図4に略示する。DEAEはスペーサー無しで直接マトリ
ックスに結合され、他の低分子量のリガンドはエピブロモヒドリンを介して結合
された。
1.2.1.ビスオキシランを介してのPEIの固定化
活性化には親水性ポリマーで被覆した膜を、ホウ水素化ナトリウム100mg、ビ
スオキシラン5ml、1M水酸化ナトリウム45mlの混合液の中で室温で3時間イン
キュベートした。遊離のオキシランリングの加水分解(pH2.5で30分のインキュ
ベート)と得られたビシナル・ジオールの過ヨウ素酸塩による酸化(0.2M過ヨウ
素酸ナトリウムの中で90分のインキュベート)との後で、pH8に調節した0.1Mリ
ン酸塩緩衝液の中で、膜をPEI(分子量50000)0.5gの溶液と2時間室温で反応させ
た所、図1に示した構造が得られた。次に1Mの塩化ナトリウム溶液と水とで洗
浄した。
1.2.2.ヒスチジンの固定化
ヒスチジンをDAHを介して、エピブロモヒドリンで活性化した被覆膜に固定化
した。エピブロモヒドリンの活性化はビスオキシランの場合と同様に実施した。
固定化したDAHを、エピブロモヒドリン5mlと4M水酸化ナトリウム5mlとの混合
溶液と90℃で8分反応させて活性化してから、直ちに80℃でL-ヒスチジンと反応
させた(0.5gL-ヒスチジンを水20mlに溶解、pH12)。得られた膜を1Mの塩化ナト
リウム溶液と水とで洗浄した。
他のポリマーの被覆と内毒素に特異性のその他のリガンドの共有結合とにはそ
れぞれ相当する処方を使用した。
2.除去試験
内毒素の膜への吸着性の試験は全て室温でゆきどまり方式(Dead-end-Modus)
で実施した。
限外濾過用セルの底に膜のディスク(膜の面積13.4cm2)を固定し、0.1M水酸
化ナトリウムの30%エタノール溶液、1.5M塩化ナトリウム溶液、及び発熱物質の
無い水で洗浄して、微量の内毒素を除去した。膜を平衡化してから、それぞれ20
mlの汚染した溶液を2ml/分の流速で膜で濾過し、濾液を集めてLAL-試験で検討
した。
3.内毒素試験
供試溶液と濾液との内毒素の定量には、Limulus Amoebocyt Lysat Test(LAL-
試験、カブトガニの変形細胞の分画上澄液の呈色試験)を使用した。本試験は、
内毒素が色原体であるp-ニトロアニリンを遊離することに基づいており、遊離さ
れたp-ニトロアニリン
と存在する内毒素の濃度との間には、0-1.2EU/mlの範囲で直線関係がある。p-
ニトロアニリンの吸光度測定により検定直線(標準内毒素大腸菌0111:B4)を用
いてプローブの内毒素濃度が求められる。
LAL-試験は欧州に於いて1985年欧州薬局方委員会により内毒素の試験用として
導入され、1989年以降モノグラム『注射用水』の中でもラビット試験を置き換え
た。
適用実施例
1.(図5)高度に汚染された緩衝液からの除去
フィード:20mMリン酸塩緩衝液20ml(pH7)、6000EU/mlの内毒素を添加
-dを付した膜は、スペーサーを合体しなかった膜を表わす。
2.(図6、7)内毒素を蓄積したBSA-溶液からの除去
フィード:BSA1mg/mlを含む20mMリン酸塩緩衝液20ml(pH4.66)、6610EU/ml
の内毒素を添加

除去にはPEI-膜とDAHHis-膜とを使用した。

Claims (1)
- 【特許請求の範囲】 1.液状媒体、特に水、タンパク質溶液、又は非経口投与用液体から内毒素を分 離するためのミクロ濾過膜に於いて、共有結合した内毒素用のリガンドを特徴と し、その際該リガンドは膜に設けられた一種のポリマーに担持されている前記ミ クロ濾過膜。 2.(a)内毒素に特異性のリガンド、好ましくはヒスタミン、ヒスチジン、ポリ エチレンイミン、ポリ-L-リシン、又はポリミキシンB及び/又は (b)それ自体内毒素に非特異性のリガンド、好ましくはジアミノヘキサン、 ジエチルアミノエチル、又はデオキシコール酸塩を特徴とする請求の範囲第1項 に記載のミクロ濾過膜。 3.膜材料として、再生セルロース、酢酸セルロース、ポリスルホン、ポリエチ レンビニルアルコール、又はポリアミド、特にナイロンを特徴とする請求の範囲 第1項又は第2項記載のミクロ濾過膜。 4.親水性ポリマー、特にデキストラン、ポリビニルアルコール、又は修飾セル ロース、好ましくはヒドロキシエチルセルロースを特徴とする前記請求の範囲の 何れか1項記載のミクロ濾過膜。 5.親水性ポリマーがそれ自体水に可溶であるか又は不溶であることを特徴とす る請求の範囲第4項記載のミクロ濾過膜。 6.前記ポリマーが一種のスペーサーを介して膜に担持されていることを特徴と する前記請求の範囲の何れか1項記載のミクロ濾過膜。 7.ビスオキシラン、グルタルジアルデヒド、エピハロゲンヒドリン、又はジイ ソシアナートに由来する一種のスペーサーを特徴とする請求の範囲第6項記載の ミクロ濾過膜。 8.前記リガンドが一種のスペーサーを介して前記ポリマーに担持されているこ とを特徴とする前記請求の範囲の何れか1項記載のミクロ濾過膜。 9.ビスオキシラン、グルタルジアルデヒド、エピハロゲンヒドリン、又はジイ ソシアナートに由来する一種のスペーサーで、場合によっては酸化により活性化 されたスペーサーを特徴とする請求の範囲第8項記載のミクロ濾過膜。 10.液状媒体、特に水、タンパク質溶液、又は非経口投与用液体から内毒素を除 去するための、前記請求の範囲の何れか1項記載のミクロ濾過膜の利用。
