Claims (1)
- 【特許請求の範囲】 1.(a)配列番号2のアミノ酸1から352を含むポリペプチドをコードす るポリヌクレオチドに対して少なくとも70%の同一性を有するポリヌクレオチ ド; (b)(a)のポリヌクレオチドに相補的なポリヌクレオチド;および (c)(a)または(b)のポリヌクレオチドの少なくとも15個の連続した 塩基を含むポリヌクレオチド からなる群から選択されるポリヌクレオチド配列を含んでなる単離ポリヌクレオ チド。 2.ポリヌクレオチドがDNAである請求項1記載のポリヌクレオチド。 3.ポリヌクレオチドがRNAである請求項1記載のポリヌクレオチド。 4.配列番号1に示すヌクレオチド1から1056を含む請求項2記載のポリ ヌクレオチド。 5.配列番号1に示すヌクレオチド1から1056を含む請求項2記載のポリ ヌクレオチド。 6.配列番号2のアミノ酸1から352を含むポリペプチドをコードする請求 項2記載のポリヌクレオチド。 7.(a)NCIMB受託番号40771に含まれるtRNAシンセターゼ遺 伝子により発現される同一の成熟ポリペプチドをコードするポリヌクレオチドに 対して少なくとも70%の同一性を有するポリヌクレオチド; (b)(a)のポリヌクレオチドに相補的なポリヌクレオチド;および (c)(a)または(b)のポリヌクレオチドの少なくとも15個の塩基を含 むポリヌクレオチド からなる群から選択されるものを含んでなる単離ポリヌクレオチド。 8.請求項2記載のDNAを含んでなるベクター。 9.請求項8記載のベクターを含んでなる宿主細胞。 10.該DNAにコードされるポリペプチドを請求項9記載の宿主細胞から発 現させることを特徴とするポリペプチドの製造方法。 11.細胞が請求項8記載のベクターに含まれるcDNAによりコードされる ポリペプチドを発現するように、該ベクターで細胞を形質転換またはトランスフ ェクトすることを特徴とする、ポリペプチドを発現する細胞の製造方法。 12.tRNAシンセターゼポリペプチドまたはフラグメントを製造するのに 充分な条件下で、請求項9記載の宿主を培養することを特徴とする、tRNAシ ンセターゼポリペプチドまたはフラグメントの製造方法。 13.配列番号2のアミノ酸1から352に対して少なくとも70%同一であ るアミノ酸配列を含んでなるポリペプチド。 14.配列番号2に示すアミノ酸を含んでなるポリペプチド。 15.請求項13記載のポリペプチドに対する抗体。 16.請求項13記載のポリペプチドの活性を阻害するアンタゴニスト。 17.治療上有効量の請求項13記載のポリペプチドを個体に投与することを 特徴とする、tRNAシンセターゼを必要とする個体の処置方法。 18.該ポリペプチドをコードするDNAを個体に提供し、該ポリペプチドを インビボで発現させることにより、治療上有効量の該ポリペプチドが投与される 請求項16記載の方法。 19.治療上有効量の請求項16記載のアンタゴニストを個体に投与すること を特徴とする、tRNAシンセターゼポリペプチドを阻害することを必要とする 個体の処置方法。 20.請求項13記載のポリペプチドをコードする核酸配列を決定することを 特徴とする、該ポリペプチドの発現に関与する疾患の診断方法。 21.宿主由来のサンプル中の請求項13記載のポリペプチドの存在を分析す ることを特徴とする診断方法。 22.請求項13記載のポリペプチドに結合し、該ポリペプチドの活性を阻害 する化合物の同定方法であって; 結合部位との結合を可能にする条件下、ポリペプチドの結合部位を表面に発現 する細胞を、スクリーニングしようとする化合物と接触させること(ここで、該 結合部位は、化合物の該結合部位への結合に反応して検出可能なシグナルを提供 する能力を有する第2の成分に関連するものである);次いで、 該化合物と該結合部位との相互作用により生じるシグナルの存在または不在を 検出することにより、該化合物が該結合部位に結合して該結合部位を活性化する かまたは阻害するかを決定すること; を特徴とする方法。 23.哺乳動物を疾患から防御する抗体を産生させるに充分なtRNAシンセ ターゼまたはそのフラグメントもしくは変種を哺乳動物に接種することを特徴と する、哺乳動物における免疫学的応答を誘起する方法。 24.免疫学的応答を誘起させて哺乳動物を疾患から防御する抗体を産生する ために、インビボでtRNAシンセターゼ、またはそのフラグメントもしくは変 種を発現させるために、遺伝子治療によりtRNAシンセターゼフラグメントま たはその変種をコードする遺伝子を送達することを特徴とする、哺乳動物におけ る免疫学的応答を誘起する方法。 25.tRNAシンセターゼポリヌクレオチドまたはそれによりコードされる タンパク質をコードし、発現するDNAを含んでなる免疫学的組成物であって、 哺乳動物に導入されると、哺乳動物において所定のtRNAシンセターゼポリヌ クレオチドまたはそれらによりコードされるタンパク質に対する免疫学的応答を 誘起する免疫学的組成物。 26.厳密なハイブリダイゼーション条件下で、配列番号1に示すポリヌクレ オチド配列の完全遺伝子を含有する適当なライブラリーを、配列番号1に示す該 ポリヌクレオチド配列またはそのフラグメントの配列を有するプローブでスクリ ーニングし、次いで、DNA配列を単離することにより得られるDNA配列を必 須としてなるポリヌクレオチド。 27.tRNAシンセターゼの相互作用を妨害する薬物を同定するための、薬 物のスクリーニング方法であって、全アミノアシル化反応または部分PPi/A TP交換反応による酵素活性の測定を特徴とするスクリーニング方法。
