Claims (1)
- 【特許請求の範囲】 1.ビオチンに対するよりもビオチン置換体に対してより高い親和性を有するス トレプトアビジン変異体をコードする核酸配列。 2.前記ストレプトアビジン変異体がビオチンに対するよりも2−イミノビオチ ンに対してより高い親和性を有する、請求項1に記載の核酸配列。 3.前記配列が、159固のアミノ酸からなる天然のストレプトアビジンの16〜133 番目のアミノ酸からなるストレプトアビジン変異体をコードし、該変異体が1以 上のアミノ酸置換を含むように、該配列が1以上のコドン置換を含む、請求項1 に記載の核酸配列。 4.前記159個のアミノ酸からなる天然のストレプトアビジンの23位のAsnに対す るコドンが、Alaに対するコドンによって置換されている、請求項3に記載の核酸 配列。 5.前記159個のアミノ酸からなる天然のストレプトアビジンの27位のSerに対す るコドンがGluに対するコドンによって置換されている、請求項4に記載の核酸配 列。 6.前記159個のアミノ酸からなる天然のストレプトアビジンの27位のSerに対す るコドンがAspに対するコドンによって置換されている、請求項4に記載の核酸配 列。 7.ストレプトアビジン変異体が1以上のアミノ酸置換を含み、且つビオチンに 対するよりもビオチン置換体に対してより高い親和性を有するように、該配列が 1以上のコドン置換を含む、159個のアミノ酸からなる天然のストレプトアビジン の16〜133番目のアミノ酸からなるストレプトアビジン変異体をコードする核 酸配列。 8.前記ストレプトアビジン変異体がビオチンに対するよりも2−イミノビオチ ンに対してより高い親和性を有する、請求項7に記載の核酸配列。 9.前記159個のアミノ酸からなる天然のストレプトアビジンの23位のAsnに対す るコドンがAlaに対するコドンによって置換されている、請求項8に記載の核酸配 列。 10.前記159個のアミノ酸からなる天然のストレプトアビジンの27位のSerに対す るコドンがGluに対するコドンによって置換されている、請求項9に記載の核酸配 列。 11.前記159個のアミノ酸からなる天然のストレプトアビジンの27位のSerに対す るコドンがAspに対するコドンによって置換されている、請求項9に記載の核酸配 列。 12.ビオチンに対するよりもビオチン置換体に対してより高い親和性を有するス トレプトアビジン変異体。 13.前記変異体がビオチンに対するよりも2−イミノビオチンに対してより高い 親和性を有する、請求項12に記載のストレプトアビジン変異体。 14.前記変異体が1以上のアミノ酸置換を含む、159個のアミノ酸からなる天然の ストレプトアビジンの16〜133番目のアミノ酸からなる、請求項13に記載のスト レプトアビジン変異体。 15.前記159個のアミノ酸からなる天然のストレプトアビジンの23位のAsnがAla によって置換されている、請求項14に記載のストレプトアビジン変異体。 16.前記159個のアミノ酸からなる天然のストレプトアビジンの27位のSerがGlu によって置換されている、請求項15に記載のストレプトアビジン変異体。 17.前記159個のアミノ酸からなる天然のストレプトアビジンの27位のSerがAsp によって置換されている、請求項15に記載のストレプトアビジン変異体。 18.ストレプトアビジン変異体が1以上のアミノ酸置換を含み、且つビオチンに 対するよりも2−イミノビオチンに対してより高い親和性を有する、159個のア ミノ酸からなる天然のストレプトアビジンの16〜133番目のアミノ酸からなるス トレプトアビジン変異体。 19.前記159個のアミノ酸からなる天然のストレプトアビジンの23位のAsnがAla によって置換されている、請求項18に記載のストレプトアビジン変異体。 20.前記159個のアミノ酸からなる天然のストレプトアビジンの27位のSerがGlu によって置換されている、請求項19に記載のストレプトアビジン変異体。 21.前記159個のアミノ酸からなる天然のストレプトアビジンの27位のSerがAsp によって置換されている、請求項19に記載のストレプトアビジン変異体。
