Claims (1)
- 【特許請求の範囲】 1.マイクロサテライト遺伝子座においてイヌを遺伝子型決定する方法であって 、 該遺伝子座に特異的なオリゴヌクレオチドプライマーを用いて染色体DNAの領 域を増幅する工程であって、ここで、DNAの該領域は、AAAまたはTTTからなる反 復テトラヌクレオチドモチーフを含み、第4の残基はG、C、AまたはTのいずれか である、工程; 該増幅の産物をサイズ分画して、該プライマー間の染色体DNAのサイズの測定 を提供する工程を包含し; ここで、該プライマー間の染色体DNAの該サイズは、該遺伝子座の対立遺伝子 マーカーである、方法。 2.前記増幅する工程が、前記産物を検出可能な標識を用いて標識する工程をさ らに包含する、請求項1に記載の方法。 3.前記増幅する工程がポリメラーゼ連鎖反応を使用する、請求項2に記載の方 法。 4.少なくとも約5つの前記マイクロサテライト遺伝子座のパネルが遺伝子型決 定され、ここで該遺伝子座の前記対立遺伝子マーカーが前記イヌの同定のための 遺伝子フィンガープリントを提供する、請求項3に記載の方法。 5.マイクロサテライト遺伝子座においてイヌを遺伝子型決定する方法であって 、 該遺伝子座に特異的なオリゴヌクレオチドプライマーを用いて染色体DNAの領 域を増幅する工程であって、ここで、DNAの該領域は、AAAG、GAAA、AAAT、TTTC 、CTTT、TTTA、AAGG、GAAT、GAAG、GAAAA、AAAAAG、TGCおよびTTCからなる群よ り選択される反復モチーフを含む、工程; 該増幅の産物をサイズ分画して、該プライマー間の染色体DNAのサイズの測定 を提供する工程を包含し; ここで、該プライマー間の染色体DNAの該サイズは、該遺伝子座の対立遺伝子 マーカーである、方法。 6.前記増幅する工程が、前記産物を検出可能な標識を用いて標識する工程をさ らに包含する、請求項5に記載の方法。 7.前記増幅する工程がポリメラーゼ連鎖反応を使用する、請求項6に記載の方 法。 8.少なくとも約5つの前記マイクロサテライト遺伝子座のパネルが遺伝子型決 定され、ここで該遺伝子座の前記対立遺伝子マーカーが前記イヌの同定のための 遺伝子フィンガープリントを提供する、請求項7に記載の方法。 9.前記遺伝子座に特異的な前記オリゴヌクレオチドプライマーが、配列番号21 と41、22と42、23と43、24と44、25と45、26と46、27と47、28と48、29と49、30 と50、31と51、32と52、33と53、34と54、35と55、36と56、37と57、38と58、39 と59、および40と60からなる群より選択される、請求項8に記載の方法。 10.単離されたDNAフラグメントを含む組成物であって、ここで、該DNAの配列 が配列番号1〜配列番号20からなる群より選択される、組成物。 11.長さが少なくとも約18ntのオリゴヌクレオチドプライマーであって、ここ で、該プライマーが、ストリンジェントな条件下で配列番号1〜配列番号20から なる群の隣接する領域の配列を有するDNAにハイブリダイズし得る、オリゴヌク レオチドプライマー。 12.前記プライマーの配列が、配列番号21〜配列番号60からなる群より選択さ れる、請求項11に記載のオリゴヌクレオチドプライマー。 13.イヌを遺伝子型決定するにおいて使用するためのキットであって、オリゴ ヌクレオチド増幅プライマーの対を含み、ここで、該プライマーの配列が配列番 号1〜配列番号20からなる群の隣接する領域より選択される、キット。 14.前記プライマーの少なくとも1つが検出可能な標識に結合される、請求項 13に記載のキット。 15.前記検出可能な標識が蛍光色素である、請求項14に記載のキット。 16.前記キットが、オリゴヌクレオチド増幅プライマーの少なくとも約5つの 前記対のパネルを含む、請求項15に記載のキット。 17.前記オリゴヌクレオチド増幅プライマーの配列が、配列番号21と41、22と 42、23と43、24と44、25と45、26と46、27と47、28と48、29と49、30と50、31と 51、32と52、33と53、34と54、35と55、36と56、37と57、38と58、39と59、およ び40と60からなる群より選択される、請求項16に記載のキット。
