Description
Claims (1)
- 【特許請求の範囲】 1. ヘモグロビン含有赤血球を含む希釈液を処理してヘモグロビンの酸素分配 能力を増大させる方法であって、 (a) 前記希釈液に酸化窒素(NO)を添加して、少なくとも95%がα−ニト ロシル−ヘモグロビンであるヘモグロビンを含む赤血球を有する、NO処理希釈 液を形成し、 (b) 前記NO処理希釈液を回収することを含む、方法。 2. 前記NO処理希釈液中の前記ヘモグロビンの少なくとも99%が前記α− ニトロシル−ヘモグロビンである、請求項1記載の方法。 3. 前記添加工程は、 (i) 前記希釈液を脱酸素化し、次いで、 (ii) 前記赤血球中の前記ヘモグロビンの少なくとも95%がα−ニトロシル ヘモグロビンに転化されるように前記希釈液中にNOを提供することを含む、請 求項1記載の方法。 4. (c) NO処理希釈液を酸素化することをさらに含む請求項4記載の方法 。 5. (d) 哺乳動物への輸血に適当な血液組成物を形成する前記NO処理希釈 液に少なくとも1種の血液成分を添加することをさらに含み、該血液組成物は、 処理されていない前記希釈液に比較して改善された酸素分配能力を有する、請求 項4記載の方法。 6. 前記NOは、化学反応の生成物として生成させることにより前記希釈液に 添加される、請求項1記載の方法。 7. 前記化学反応は、前記添加工程の前に行われる請求項6記載の方法。 8. 前記化学反応は、前記添加工程の最中に行われる請求項6記載の方法。 9. NOは、NOを直接前記希釈液と接触させることにより添加される請求項 1記載の方法。 10. 前記希釈液は全血又は少なくとも1つの血液成分を含む、請求項1記載 の方法。 11. 前記全血又は少なくとも1つの血液成分は、少なくとも3週間貯蔵され たものである、請求項10記載の方法。 12. 請求項1記載の方法により生産された、NO処理希釈液。 13. 少なくとも95%のヘモグロビンがα−ニトロシル−ヘモグロビンであ るヘモグロビンを含む赤血球を含む、NO処理希釈液。 14. 前記ヘモグロビンの少なくとも99%がα−ニトロシル−ヘモグロビン である、請求項13記載のNO処理希釈液。 15. 前記希釈液は全血又は少なくとも1つの血液成分を含む、請求項13記 載のNO処理希釈液。 16. 前記NO処理希釈液中の前記ヘモグロビンの少なくとも80〜99%が 前記α−ニトロシル−ヘモグロビンである、請求項1記載の方法。 17. 前記添加工程は、 (i) 前記希釈液を脱酸素化し、次いで、 (ii) 前記赤血球中の前記ヘモグロビンの少なくとも80ないし99%がα− ニトロシルヘモグロビンに転化されるように前記希釈液中にNOを提供すること を含む、請求項1記載の方法。 18. 少なくとも80〜95%がα−ニトロシル−ヘモグロビンであるヘモグ ロビンを含む赤血球を含む、NO処理希釈液。 19. 前記ヘモグロビンの少なくとも80〜99%が前記α−ニトロシル−ヘ モグロビンである請求項13記載のNO処理希釈液。 20. 請求項1〜19のいずれか1項に記載されたα−ニトロシルヘモグロビ ン。 21. α−ニトロシルHb、リポソーム中に被包されたα−ニトロシルHb、 α−ニトロシルヘモグロビン代替物、又は酸素を運搬することができ、ヘモグロ ビンとして機能し得る、α−ニトロシルヘモグロビン様物質。
