Claims (1)
- 【特許請求の範囲】 1.DNAの鋳型誘導重合を触媒し、3'-5'エキソヌクレアーゼ活性を有し、マ グネシウムイオンの存在下および実質的にマンガンイオンの不存在下で逆転写酵 素活性を有する、カルボキシドテルムス ヒドロゲノホルマンスから取得可能な 精製された熱安定DNAポリメラーゼ。 2.前記ポリメラーゼがMn2+依存性である逆転写酵素活性を示す、請求項1に記 載のポリメラーゼ。 3.前記ポリメラーゼのマグネシウムイオン依存性逆転写酵素活性が、該ポリメ ラーゼのDNAポリメラーゼ活性よりも高い、請求項1〜2のいずれか1項に記 載のポリメラーゼ。 4.前記ポリメラーゼのマグネシウムイオン依存性逆転写酵素活性が、該ポリメ ラーゼのマンガン依存性逆転写酵素活性よりも高い、請求項1〜3のいずれか1 項に記載のポリメラーゼ。 5.前記ポリメラーゼが、SDSポリアクリルアミド電気泳動によって測定した 場合、約100〜105kDaの見掛けの分子量を有する、請求項1〜4のいずれか1項 に記載のポリメラーゼ。 6.前記ポリメラーゼが、AR96と称する株、E.coli BL21(DE3)pUBS520から取得 可能である、請求項1〜5のいずれか1項に記載のポリメラーゼ。 7.カルボキシドテルムス ヒドロゲノホルマンスから取得可能な請求項1〜6 のいずれか1項に記載のポリメラーゼをコードする、単離されたDNA配列。 8.微生物カルボキシドテルムス ヒドロゲノホルマンスのポリメラーゼ活性を コードすることが可能な組換えDNA配列。 9.配列番号7に示された式によって表される単離されたDNA配列。 10.請求項7〜9のいずれか1項に記載された単離されたDNA配列を含むベ クター。 11.前記ベクターが、カルボキシドテルムス ヒドロゲノホルマンスDNAポ リメラーゼ遺伝子を有するプラスミドpDS56であり、pAR4と称するものである、 請求項10に記載のベクター。 12.下記の特徴: (1)転写開始のプロモーターまたは部位 (2)遺伝子発現をオンにしたリオフにしたりするのに用いることができるオ ペレーター (3)翻訳増進のためのリボソーム結合部位 (4)転写または翻訳終結部位 のいくつかまたは全てを付与するものである、請求項10または11に記載のベ クター。 13.請求項10〜12のいずれか1項に記載のベクターで形質転換された宿主 微生物。 14.前記形質転換体が、AR96と称する株、E.coli BL21(DE3)pUBS520由来のも のである、請求項13に記載の宿主微生物。 15.(a)天然株カルボキシドテルムス ヒドロゲノホルマンスを培養する工 程、 (b)天然株の細胞を緩衝液に懸濁する工程、 (c)細胞を破壊する工程、 (d)1以上のセファロースカラムの使用を含むクロマトグラフィー工程によ ってDNAポリメラーゼを精製する工程 を含む、請求項1〜6のいずれか1項に記載のDNAポリメラーゼの調製方法。 16.請求項10〜12のいずれか1項に記載のベクターで形質転換された組換 え体E.coli株を増殖させ、DNAポリメラーゼを精製し、単離する工程を含む、 請求項1〜6のいずれか1項に記載のDNAポリメラーゼの調製方法。 17.請求項1〜6のいずれか1項に記載の熱安定DNAポリメラーゼを使用す ることを特徴とする、DNAの増幅方法。 18.請求項1〜6のいずれか1項に記載の熱安定DNAポリメラーゼを使用す ることを特徴とする、第2のcDNAクローニングおよびDNA配列決定方法。 19.請求項1〜6のいずれか1項に記載の熱安定DNAポリメラーゼを使用す ることを特徴とする、DNA標識方法。 20.請求項1〜6のいずれか1項に記載の熱安定DNAポリメラーゼを使用す ることを特徴とする、逆転写方法。
