Claims (1)
- 【特許請求の範囲】 1.サンプル中に含まれる標的生物物質を単離するための方法であって、捕捉相 が与えられ、前記標的生物物質が少なくとも一の捕捉相と接触させられ、捕捉相 に結合した該標的生物物質が検出される方法において、 前記方法は、該捕捉相が、少なくとも一の反応性官能基を含む有機分子及び、特 定的且つ直接的または間接的に、前記標的生物物質を認識または結合可能な少な くとも一のタンパク物質を含み、前記タンパク物質は有機分子の反応性官能基へ の共有結合的な結合のための特定部位を有し、これが少なくとも6の隣接するリ ジンまたはリジンを主成分とする残基を含む少なくとも一のタグからなることを 特徴とする方法。 2.捕捉相が、標識を更に含むことを特徴とする請求項1に記載の方法。 3.捕捉相が、検出相であることを特徴とする請求項2に記載の方法。 4.検出相かさらに与えられ、これが、少なくとも一の反応性官能基を有する有 機分子、特定的且つ直接的または間接的に、前記標的生物物質を認識または結合 可能な少なくとも一のタンパク物質、及び標識を含み、前記タンパク物質が有機 分子の反応性官能基への共有結合的な結合のための特定部位を有し、これが少な くとも6の隣接するリジンまたはリジンを主成分とする残基を含む少なくとも一 のタグからなることを特徴とする請求項1に記載の方法。 5.検出相中の有機分子及びタンパク物質が、捕捉相中の有機分子及びタンパク 物質と、それぞれ同一及び/または相違することを特徴とする請求項4に記載の 方法。 6.有機分子が、粒子状または直線状であることを特徴とする請求項1及び/ま たは4に記載の方法。 7.ポリマーが、ホモポリマー、例えばポリリジンまたはポリチロシン;及びコ ポリマー、例えばマレイン酸無水物コポリマー、N-ビニルピロリドンコポリマ ー、天然または合成の多糖類、ポリヌクレオチド及びアミノ酸コポリマー、例え ば酵素より選択されるポリマーであることを特徴とする請求項6に記載の方法。 8.ポリマーが、マレイン酸無水物/メチルビニルエーテルコポリマー、N-ビ ニルピロリドン/N-アクリルオキシスクシンイミドコポリマー及びポリ-6-ア ミノ グルコースより選択されることを特徴とする請求項7に記載の方法。 9.有機分子が、ハプテンであることを特徴とする請求項1及び/または4に記 載の方法。 10.有機分子が、坦体分子に結合することを特徴とする請求項9に記載の方法 。 11.有機分子がビオチンであり、坦体分子がアビジンであることを特徴とする 請求項9に記載の方法。 12.検出相中の有機分子が、標識であることを特徴とする請求項4に記載の方 法。 13.タグが、タンパク物質のN-またはC-末端に配されることを特徴とする請 求項1及び/または4に記載の方法。 14.有機分子の反応性官能基が、エステル、酸、ハロカルボニル、スルフヒド リル、ジスルフィド、エポキシド、ハロアルキル及びアルデヒド官能基より選択 されることを特徴とする請求項1及び/または4に記載の方法。 15.タンパク物質が抗原を含み、生物物質が前記抗原によって特異的に認識さ れる抗体であることを特徴とする請求項1から14のいずれか一項に記載の方法 。 16.タンパク物質が、抗体を含み、生物物質が前記抗体によって特異的に認識 される抗原であることを特徴とする請求項1から14のいずれか一項に記載の方 法。 17.標的生物物質のための捕捉相であって、少なくとも一の反応性官能基を含 む有機分子及び、特定的且つ直接的または間接的に、前記標的生物物質を認識ま たは結合可能な少なくとも一のタンパク物質を含み、前記タンパク物質は有機分 子の反応性官能基への共有結合的な結合のための特定部位を有し、これが少なく とも6の隣接するリジンまたはリジンを主成分とする残基を含む少なくとも一の タグからなることを特徴とする捕捉相。 18.標的生物物質のための検出相であって、少なくとも一の反応性官能基を含 む有機分子、特定的且つ直接的または間接的に、前記標的生物物質を認識または 結合可能な少なくとも一のタンパク物質、及び標識を含み、前記タンパク物質は 有機分子の反応性官能基への共有結合的な結合のための特定部位を有し、これが 少なくとも6の隣接するリジンまたはリジンを主成分とする残基を含む少なくと も一のタグからなることを特徴とする検出相。 19.有機分子が、粒子状または直線状であることを特徴とする請求項17また は18に記載の捕捉または検出相。 20.ポリマーが、ホモポリマー、例えばポリリジンまたはポリチロシン;及び コポリマー、例えばマレイン酸無水物コポリマー、N-ビニルピロリドンコポリ マー、天然または合成の多糖類及びポリヌクレオチド及びアミノ酸コポリマー、 例えば酵素より選択されるポリマーであることを特徴とする請求項19に記載の 捕捉または検出相。 21.ポリマーが、マレイン酸無水物/メチルビニルエーテルコポリマー、N- ビニルピロリドン/N-アクリルオキシスクシンイミドコポリマー、ポリ-6-ア ミノグルコース、ホースラディッシュペルオキシド(HRP)及びアルカリ性ホ スファターゼより選択されることを特徴とする請求項20に記載の捕捉または検 出相。 22.有機分子が、ハプテン、例えばビオチンであることを特徴とする請求項2 1に記載の捕捉相。 23.タグが、タンパク物質のN-またはC-末端に配されることを特徴とする請 求項17または18に記載の捕捉または検出相。 24.有機分子の反応性官能基が、エステル、酸、ハロカルボニル、スルフヒド リル、ジスルフィド、エポキシド、ハロアルキル及びアルデヒド官能基より選択 されることを特徴とする請求項17または18に記載の捕捉または検出相。 25.有機分子が、標識であることを特徴とする請求項18に記載の検出相。 26.標識が、酵素、タンパク質、ペプチド、抗体、ハプテン、例えばビオチン またはイミノビオチン、蛍光化合物、例えばローダミン、放射性化合物、化学発 光化合物、電子密度成分、磁性成分及び類似物からなる群より選択されることを 特徴とする請求項18または25に記載の検出相。 27.請求項17及び請求項19から24のいずれか一項に記載の捕捉相及び/ または請求項18から26のいずれか一項に記載の検出相を含む、標的生物物質 を単離するための試薬。 28.捕捉相が、直接または間接的に受動吸収または共有結合により固体サポー トに結合することを特徴とする請求項27に記載の試薬。
