Claims (1)
- 【特許請求の範囲】 1.可溶性植物脂肪酸デサチュラーゼの鎖長及び二重結合位置特異性を改変する ための方法であり、脂肪酸と接触する可溶性脂肪酸デサチュラーゼの基質結合チ ャンネル内のアミノ酸接触残基を改変することを含む方法。 2.可溶性植物脂肪酸デサチュラーゼがアシル−ACPデサチュラーゼである請 求の範囲第1項の方法。 3.アシル−ACPデサチュラーゼがΔ9デサチュラーゼである請求の範囲第2 項の方法。 4.Δ9デサチュラーゼをチュンベルギア・アラタ又はリシヌス・コムニスから 成る群より選ばれる植物により生産する請求の範囲第3項の方法。 5.アミノ酸接触残基が残基114、115、117、118、179、181 、188及び189から成る群より選ばれる請求の範囲第4項の方法。 6.アシル−ACPデサチュラーゼの鎖長及び二重結合位置特異性を改変するた めの方法であり: a)アシル−ACPデサチュラーゼの一次アミノ酸配列を得; b)アシル−ACPデサチュラーゼの一次アミノ酸配列をリシヌス・コムニス Δ9デサチュラーゼの一次アミノ酸配列と最大配列保存に関して整列させ; c)リシヌス・コムニスΔ9デサチュラーゼとの配列保存に基づいてアシル− ACPデサチュラーゼに関する3次元モデルを構築し; d)段階c)においてモデル化された構造の基質結合チャンネル内の アミノ酸接触残基を同定し; e)段階d)で同定されたアミノ酸接触残基の1つ又はそれ以上を他のアミノ 酸残基で置換することにより、改変された鎖長及び二重結合位置特異性を有する 突然変異アシル−ACPデサチュラーゼを生成させることを含む方法。 7.第1の脂肪酸及び第2の脂肪酸の脱飽和を触媒する能力を特徴とする突然変 異アシル−ACPデサチュラーゼであり、第1及び第2の脂肪酸はその鎖長が異 なっており、第1及び第2の脂肪酸の両方の脱飽和速度の差が約4倍以下である 突然変異アシル−ACPデサチュラーゼ。 8.基質結合チャンネル内のアミノ酸接触残基における点突然変異を含有する請 求の範囲第7項の突然変異アシル−ACPデサチュラーゼ。 9.第1の脂肪酸が16:0の鎖長を有し、第2の脂肪酸が18:0の鎖長を有 している請求の範囲第7項の突然変異アシル−ACPデサチュラーゼ。 10.基質結合チャンネル内の接触残基においてアミノ酸置換を有する突然変異 アシル−ACPデサチュラーゼ。 11.相手の野生型アシル−ACPデサチュラーゼと比較して鎖長及び二重結合 位置特異性の変化を特徴としている請求の範囲第10項の突然変異アシル−AC Pデサチュラーゼ。 12.アシル−ACPデサチュラーゼがΔ9アシル−ACPデサチュラーゼであ り、基質結合チャンネル内の接触残基が114、115、117、118、17 9、181、188及び189から成る群より選ばれる請求の範囲第11項の突 然変異アシル−ACPデサチュラーゼ。 13.Δ9アシル−ACPデサチュラーゼをチュンベルギア・アラタ又 はリシヌス・コムニスからクローニングされる核酸を突然変異させることにより 生産する請求の範囲第12項の突然変異体。 14.第1の脂肪酸及び第2の脂肪酸の脱飽和を触媒する能力を特徴とする突然 変異アシル−ACPデサチュラーゼをコードする核酸配列であり、第1及び第2 の脂肪酸はその鎖長が異なっており、第1及び第2の脂肪酸の両方の脱飽和速度 の差が約4倍以下である核酸配列。 15.突然変異アシル−ACPデサチュラーゼが基質結合チャンネル内のアミノ 酸接触残基における点突然変異を含有する請求の範囲第14項の核酸配列。 16.アシル−ACPデサチュラーゼがΔ9デサチュラーゼである請求の範囲第 15項の核酸配列。 17.アミノ酸接触残基が残基114、115、117、118、179、18 1、188及び189から成る群より選ばれる請求の範囲第16項の核酸配列。 18.第1の脂肪酸及び第2の脂肪酸の脱飽和を触媒する能力を特徴とする突然 変異アシル−ACPデサチュラーゼをコードする核酸配列を発現可能な形態で含 むDNA発現構築物であり、第1及び第2の脂肪酸はその鎖長が異なっており、 第1及び第2の脂肪酸の両方の脱飽和速度の差が約4倍以下であるDNS発現構 築物。 19.突然変異アシル−ACPデサチュラーゼが基質結合チャンネル内のアミノ 酸接触残基における点突然変異を含有する請求の範囲第18項のDNA発現構築 物。 20.アシル−ACPデサチュラーゼがΔ9デサチュラーゼである請求の範囲第 19項のDNA発現構築物。 21.アミノ酸接触残基が残基114、115、117、118、179、18 1、188及び189から成る群より選ばれる請求の範囲第20項のDNA発現 構築物。 22.第1の脂肪酸及び第2の脂肪酸の脱飽和を触媒する能力を特徴とする突然 変異アシル−ACPデサチュラーゼをコードする核酸配列を発現可能な形態で含 むDNA発現構築物を用いて形質転換された細胞であり、第1及び第2の脂肪酸 はその鎖長が異なっており、第1及び第2の脂肪酸の両方の脱飽和速度の差が約 4倍以下である細胞。 23.突然変異アシル−ACPデサチュラーゼが基質結合チャンネル内のアミノ 酸接触残基における点突然変異を含有する請求の範囲第22項の細胞。 24.アシル−ACPデサチュラーゼがΔ9デサチュラーゼである請求の範囲第 23項の細胞。 25.アミノ酸接触残基が残基114、115、117、118、179、18 1、188及び189から成る群より選ばれる請求の範囲第24項の細胞。 26.原核細胞である請求の範囲第22項の細胞。 27.真核細胞である請求の範囲第22項の細胞。 28.植物細胞である請求の範囲第27項の細胞。 29.第1の脂肪酸及び第2の脂肪酸の脱飽和を触媒する能力を特徴とするキメ ラアシル−ACPデサチュラーゼであり、第1及び第2の脂肪酸はその鎖長が異 なっており、第1及び第2の脂肪酸の両方の脱飽和速度の差が約4倍以下である キメラアシル−ACPデサチュラーゼ。 30.アミノ酸172〜201がΔ9−18:0 ACPデサチュラー ゼからのアミノ酸178〜207で置換されているΔ6−16:0を含む請求の 範囲第29項のキメラアシル−ACPデサチュラーゼ。 31.アミノ酸172〜196がΔ9−18:0 ACPデサチュラーゼからの アミノ酸178〜202で置換されているΔ6−16:0を含む請求の範囲第2 9項のキメラアシル−ACPデサチュラーゼ。 32.アミノ酸176、183、184、200、201及び202がそれぞれ Δ9−18:0 ACPデサチュラーゼからのアミノ酸181、188、189 、205、206及び207で置換されているΔ6−16:0を含む請求の範囲 第29項のキメラアシル−ACPデサチュラーゼ。 33.アミノ酸183及び184がそれぞれΔ9−18:0 ACPデサチュラ ーゼからのアミノ酸188及び189で置換されているΔ6−16:0を含む請 求の範囲第29項のキメラアシル−ACPデサチュラーゼ。 34.アミノ酸176及び195がそれぞれΔ9−18:0 ACPデサチュラ ーゼからのアミノ酸181及び200で置換されているΔ6−16:0を含む請 求の範囲第32項のキメラアシル−ACPデサチュラーゼ。 35.アミノ酸176、195、200、201及び202がそれぞれΔ9−1 8:0 ACPデサチュラーゼからのアミノ酸181、200、205、206 及び207で置換されているΔ6−16:0を含む請求の範囲第32項のキメラ アシル−ACPデサチュラーゼ。
