- 【特許請求の範囲】 1. 下記式(5a) (ここで、Rは、t-ブチルジフェニルシリルと、トリフェニルシリル、t-ブチル ジメチルシリル、トリチル、メトキシトリル、そしてピバロイルからなる群から 選ばれるヒドロキシルの保護基(A)である。) のラクトンのジアステレオ選択性の製造方法であって、下記ステップ(1)と( 2)を含んでなることを特徴とする製造方法。 (1) 下記式(4) のヒドロキシル基を保護されたラクトンに有効量の強塩基を反応させ、続いて( 第2段階として)シリルブロッキング基(B)を導入して、シリルで保護された エノールエーテル生成物を製造するステップと、 (2) ステップ(1)で得られたシリルで保護したエノールエーテル生成物に 式ArSeX(ここで、Arはフェニルあるいは置換フェニルであり、Xは、-0Acと、-O C(O)CF3、-SCN、-SO2Ar、NR2、そしてN-フタルイミドからなる群から選ばれる) のフェニルセレノ化合物を反応させて、式5aの化合物を製造するステップ。 2. 上記塩基がブチルリチウムと、リチウムジエチルアミド、リチウム(ビス) トリメチルシリルアミド、ナトリウム(ビス)トリメチルシリルアミド、そしてカ リウム(ビス)トリメチルシリルアミドからなる群から選ばれることを特徴とする 、請求項1に記載の製造方法。 3. 上記シリルブロッキング基(B)がトリメチルシリル基であることを特徴 とする、請求項1に記載の製造方法。 4. 下記式(I) (ここで、Yはプリンあるいはピリミジン塩基であり、XはCHあるいはCH2であ る。) のL-ヌクレオシドのジアステレオ選択性の製造方法であって、下記ステップ(1 )と(4)を含んでなることを特徴とする製造方法。 (1) 下記式(5a) (ここで、Rはt-ブチルジフェニルシリルと、トリフェニルシリル、t-ブチルジ メチルシリルトリル、メトキシトリル、そしてピバロイルからなる群から選ばれ るかさ高なヒドロキシルの保護基である。) の化合物を、下記式(4)の化合物から、(第1段階として)上記化合物(4)に有効量の強塩基を反応させ、続いて(第2 段階として)シリルブロック基を導入して、シリルで保護したエノールエーテル 生成物を製造し、続いて、上記シリルで保護したエノールエーテルに式ArSeX(こ こで、Arはフェニルあるいは置換フェニルであり、Xは、-OAc、-OC(O)CF3、-SC N、-SA2Ar、NR2、とN-フタルイミドからなる群から選ばれる)のフェニルセレノ 化合物を反応させて、式(5a)のフェニルセレノ化ラクトンを製造するステップと 、 (2) 式(5a)の化合物のラクトン部分を還元して、下記ヘミアセタール(6) を製造するステップと、 (3) ヘミアセタール(6)をエステル化して、下記式(7) (ここで、Tはエステル基である。) のエステル化された化合物を製造するステップと、 (4) 上記エステル化された化合物(7)にプリンあるいはピリミジン塩基を縮 合させて、下記式(8) (ここで、Yはプリンあるいはピリミジン塩基であり、更に酸化あるいは還元状 態にさらされて式(I)のヌクレオシドを生成する。) のヌクレオシドを製造するステップ。 5. 上記式(8)のヌクレオシドが酸化状態にさらされ、続いてヒドロキシル基 が脱保護されて、下記式(10) の2',3'-ジデオキシ-2',3'-ジデヒドロヌクレオシドを生成することを特徴とす る、請求項4に記載の方法。 6. Yがピリミジン塩基であることを特徴とする、請求項4に記載の製造方法 。 7. 上記ピリミジン塩基がシトシンであることを特徴とする、請求項5に記載 の製造方法。 8. 上記ピリミジン塩基が5−フルオロシトシンであることを特徴とする、請 求項5に記載の製造方法。 9. 更に式(8)のヌクレオシドを還元し、下記式(12) の2',3-ジデオキシヌクレオシド誘導体を製造することからなることを特徴とす る4項に記載の製造方法。 10. Yがピリミジン塩基であることを特徴とする、請求項9に記載の製造方 法。 11. 上記ピリミジン塩基がシトシンであることを特徴とする、請求項10に 記載の製造方法。 12. 上記ピリミジン塩基が5-フルオロシトシンであることを特徴とする、請 求項10に記載の製造方法。 13. XがN-フタルイミドであることを特徴とする、請求項9に記載の製造方 法。 14. 強塩基がブチルリチウム、リチウムジエチルアミド、リチウム(ビス)ト リメチルシリルアミド、ナトリウム(ビス)トリメチルシリルアミドとカリウム( ビス)トリメチルシリルアミドからなる群から選ばれることを特徴とする、請求 項9に記載の製造方法。 15. シリルブロック基がトリメチルシリルであることを特徴とする、請求項 9に記載の製造方法。 16. 還元ステップ(2)がボロハイドライド還元剤あるいはハイドライド還 元剤の存在下で行なわれることを特徴とする、請求項9に記載の製造方法。 17. 下記式(5c)(ここで、Rは、t-ブチルジフェニルシリルと、トリフェニルシリル、t-ブチル ジメチルシリル、トリチル、メトキシトリル、そしてピバロイルからなる群から 選ばれるヒドロキシルの保護基(A)である。) のラクトンのジアステレオ選択性の製造方法であって、下記(1)と(2)ステ ップを含んでなることを特徴とする製造方法。 (1)下記式(4a) のヒドロキシル基を保護されたラクトン(4a)に有効量の強塩基を反応させ、続い て(第2段階として)シリルブロッキング基(B)を導入して、シリルで保護し たエノールエーテルを製造するステップと、 (2) ステップ(1)から得られたシリルで保護したエノールエーテルに式Ar SeX(ここで、Arはフェニルあるいは置換フェニルであり、Xは、-OAcと、-OC(O) CF3、-SCN、-SO2Ar、NR2、そしてN-フタルイミドからなる群から選ばれる。)の フェニルセレノ化合物を反応させて、式5cの化合物を製造するステップ。 18. 塩基がブチルリチウムと、リチウムジエチルアミド、リチウム(ビス)ト リメチルシリルアミド、ナトリウム(ビス)トリメチルシリルアミド、そしてカリ ウム(ビス)トリメチルシリルアミドからなる群から選ばれることを特徴とする、 請求項17に記載の製造方法。 19. シリルブロッキング基(B)がトリメチルシリル基であることを特徴と する、請求項17に記載の製造方法。
