Claims (1)
- 【特許請求の範囲】 1. カッサバ又は極関連した種の、植物に再生可能のプロトプラストを生産する ための方法であって、 カッサバ又は極関連した種の移植片からフライアブル胚形成カルスを生産する こと、及び 前記フライアブル胚形成カルスからプロトプラストを分離すること、 を含む方法。 2. フライアブル胚形成カルスが、液体培地で培養される、請求項1の方法。 3. プロトプラストを生産するために、セルラーゼ、ペクトリアーゼ、及び/又 はマセロチームのような細胞壁分解酵素の混合物が使用される、請求項1又は2の 方法。 4. 移植片をとる植物がオーキシンで前処理される、請求項1から3のいずれか1 の方法。 5. フライアブル胚形成カルスが、魚雷型の一次又は成熟胚から生産される、請 求項1から4のいずれか1の方法。 6. 胚が、一次の移植片に誘導される、請求項5の方法。 7. 請求項1から6のいずれか1の方法により得られるプロトプラスト。 8. カッサバ又は極関連した種のプロトプラストを形質転換するための方法であ って、 アグロバクテリウム・ツメファシエンスのような付加的な遺伝子情報を含むバ クテリウムによる感染を介するか、前記付加的な遺伝子情報を含むベクターを与 える電気穿孔法又は化学的穿孔法によるか、又は、粒子が前記付加的な遺伝子情 報をコーティングされる粒子射突法により、前記付加的な遺伝子情報をもつ前記 プロトプラストを与える工程、 を含み、 請求項7のプロトプラストが形質転換される、 ところの方法。 9. 請求項8の方法により得られる形質転換したプロトプラスト。 10.付加的な遺伝子情報が対象の遺伝子を含む、請求項9の形質転換したプロト プラスト。 11.付加的な遺伝子情報がアンチセンス構造を含む、請求項9の形質転換したプ ロトプラスト。 12.アンチセンス構造が、アミローゼ合成経路を示すことができる、請求項11の 形質転換したプロトプラスト。 13.胚を生産するために、請求項7、9-12のいずれか1のプロトプラストが誘導さ れ、植物を生産するために、続けて、胚が誘導され る、プロトプラストから植物を再生するための方法。 14.請求項13の方法によって得られる、カッサバ植物又はその極関連した種。 15.その塊茎が本質的にアミローゼを全く含まない、請求項12のプロトプラスト から得られる請求項14の植物。
