Claims (1)
- 【特許請求の範囲】 1.生体液の循環のための区画を備えた生体液の特異的浄化用モジュールであっ て、この区画が少なくとも一部、機能化担体、又は表面が機能化基を有する壁か ら画定されており、 −少なくとも約15℃〜約45℃の温度において安定であり、 −医療装置に用いることができる滅菌方法の少なくとも1つの型に対して、特に ガンマ線に対して安定であり、 −活性化物質の存在下に、次の意味において、官能基を発生するのに適している 、すなわち下記の基を有するリガンドと共有結合を形成するのに適している、 生体液の循環のための区画を備えた生体液の特異的浄化用モジュール。 2.生体液の循環のための少なくとも1つの区画を備えた生体液の体外浄化用モ ジュールであって、この区画は少なくとも一部、機能化担体、又は表面が電荷を 示す壁によって画定されており、これらを介してイオン結合によって第一の型の 分子又は巨大分子が結合され、これらの分子は一度結合されると遊離アミン基を 示し、これらの遊離アミン基上には共有結合によって、第二の型の分子が固定さ れ、これらの第二の型の分子は一度結合されると、アミド、エステル、チオエス テル、及びグアニジンの誘導体結合を直接的又は間接的に形成するのに適した遊 離カルボン酸基を示す体外浄化用モジュール。 3.共有結合によって、カルボン酸基上に第三の型の分子が固定され、この第三 の型の分子は、立体障害を減少させるため、且つリガンド及び/又は分子又は浄 化される有形成分の、その後の固定を促進するために、スペーサー機能を有する ことを特徴とする、請求項2に記載の体外浄 化用モジュール。 4.共有結合によって、カルボン酸基上に第四の型の分子が固定され、この第四 の型の分子は、非特異的相互作用を制限するために、担体の親水性を増す機能を 有することを特徴とする、請求項2又は3に記載の体外浄化用モジュール。 5.共有結合によって、カルボン酸基上に第五の型の分子が固定され、この第五 の型の分子は一度結合されると、もう1つの種類の基を示し、特に基−CHO又 は−COOHを有するリガンドともう1つの型の共有結合カップリングを可能に することを特徴とする、請求項2から4のうちのいずれか1つに記載の体外浄化 用モジュール。 6.膜が、アクリロニトリルコポリマー及びメタリルスルホン酸ナトリウムから 製造される半透膜であることを特徴とする、請求項2から5のうちのいずれか1 つに記載の体外浄化用モジュール。 7.第一の型の分子が、分子量10,000〜2,000,000ダルトンのポ リエチレンイミン(PEI)であり、このPEIは置換又は非置換であることを 特徴とする、請求項2から6のうちのいずれか1つに記載の体外浄化用モジュー ル。 8.第二の型の分子が、式: (ここにおいてX=[CH2]nであり、nは2又は3であるか、又はX=−CH =CH−である) のジカルボン酸の無水物であることを特徴とする、請求項2〜7のうちの1つに 記載のモジュール。 9.第二の型の分子は、式: のモノカルボン酸の無水物であることを特徴とする、請求項2から7のうちのい ずれか1つに記載の体外浄化用モジュール。 10.単一の分子が、第三、第四、及び第五の型の分子の機能を果たし、この分 子は式: NH2−NH−CO−Y−CO−NH−NH2 (ここにおいてYは好ましくは基(CH2)mであり、ここにおいてmは2〜6で ある) のジヒドラジンであることを特徴とする、請求項2から9のうちのいずれか1つ に記載の体外浄化用モジュール。 11.有機基、特に: と共有結合を形成するのに適した基を有する生物学的適合性を有する機能化担体 の調製方法であって、下記工程を備える方法: a)固定アニオン基を有する担体と、分子量10,000〜2,000,000 のポリエチレンイミン(PEI)であって、このPEIは置換又は非置換である ものとを接触させる工程; b)a)で処理された担体と、式: (ここにおいて、X=[CH2]nであり、nは2又は3であるか、又はX=−C H=CH−である)、 又は式: のカルボン酸の無水物の溶液とを接触させる工程; c)場合によって、工程a)及びb)によって修飾された担体と、式: NH2−NH−CO−Y−CO−NH−NH2 (ここにおいてYは好ましくは基(CH2)mであり、ここにおいてmは2〜6で ある) のジヒドラジドとを接触させる工程。 12.有機基、特に: と共有結合を形成することができる基を有する生物学的適合性を有する機能化担 体の調製方法であって、 前記担体は、 a)置換又は非置換の、分子量10,000〜2,000,000のポリエチレ ンイミン(PEI); b)式: (ここにおいて、X=[CH2]nであり、nは1〜4であるか、又はX=−CH =CH−である)、 又は式: のカルボン酸の無水物の溶液、 との反応生成物と、イオン性を有する担体との接触によって得ることができ、 このように修飾された担体が、場合によっては式: NH2−NH−CO−Y−CO−NH−NH2 (Yは、補体を活性化しない親水基を有するという特性と、γ線に対して安定で あるという特性の二重の特性を有する基から選ばれ、好ましくは基[CH2]m( ここにおいてnは2〜6である)) のヒドラジドと接触させられる機能化担体の調整方法。 13.イオン性を有する担体が、アクリロニトリルコポリマー及びメタリルスル ホン酸ナトリウムである、請求項11又は12に記載の方法。 14.カルボン酸の無水物が無水コハク酸であることを特徴とする、請求項11 から13に記載の方法。 15.ジヒドラジンがADH(アジピン酸ジヒドラジン)である、請求項11か ら14のうちのいずれか1つに記載の方法。 16.請求項11〜15のうちのいずれか1つによる方法の実施によって得るこ とができる生物学的適合性を有する機能化担体。 17.抗体、抗原、ペプチド、蛋白質、又は糖蛋白質、ホルモン、酵素、これら の補助因子、これらの基質又は阻害物質、多糖類、レクチン、毒素又は抗毒素、 核酸又はポリヌクレオチド酸、ハプテン、又はハプテンのリガンド、顔料又は色 素から成る群から選ばれるリガンドと共有結合的にカップリングされうることを 特徴とする、請求項16に記載の担体。 18.多孔質又は非多孔質、中実又は中空の平面フィルム又は繊維の形態、多孔 質又は非多孔質の微小球、又はこれらの組合わせの形態の膜、 特に透析膜の成分中に入っていることを特徴とする、請求項17に記載の担体。 19.腎透析用又は血液濾過用の透析膜であることを特徴とする、請求項16に 記載の担体。 20.生体液の体外浄化のための、多目的に使用しうるモジュールの製造におけ る請求項15から19のうちのいずれか1つに記載の担体の使用であって、前記 モジュールが請求項1から9のうちのいずれか1つの定義に当てはまる使用。 21.生体液中に存在し、除去が求められている生化学種又は細胞が、このモジ ュールによって浄化される請求項20に記載の使用。 22.請求項16から18のうちのいずれか1つに定義されている担体と、抗体 、抗原、ペプチド、蛋白質、又は糖蛋白質、ホルモン、酵素、これらの補助因子 、これらの基質又は阻害物質、多糖類、レクチン、毒素又は抗毒素、核酸又はポ リヌクレオチド酸、ハプテン、又はハプテンのリガンド、顔料又は色素から成る 群から選ばれるリガンドとのカップリング生成物。 23.請求項16から19のうちのいずれか1つに記載の担体を組込んでいる、 血液の体外循環のための装置。 24.透析装置であることを特徴とする、請求項23に記載の装置。 25.次のものを含むキット: −請求項1から11のうちのいずれか1つに記載の多目的に使用しうるモジュー ル; −機能化担体とリガンドとの反応の活性化溶液を含む滅菌容器又は滅菌袋; −場合によっては選ばれたリガンドの溶液を含む滅菌容器又は滅菌袋; −場合によってはリガンドの活性化及び/又は共有結合カップリングに必要な緩 衝溶液又はリンス溶液。 26.活性化溶液がカルボジオミドであり、リガンドがリポ酸であることを特徴 とする、請求項25に記載のキット。 27.活性化溶液が、過ヨウ素酸ナトリウムであり、リガンドが抗体であること を特徴とする、請求項25又は26に記載のキット。 28.生体液の体外浄化における、請求項25から27のうちのいずれか1つに 記載のキットの使用。
