Claims (1)
- 【特許請求の範囲】 1. 反復配列を含んで成る標的配列を分析する方法であって、当該反復配列は 式ApCyGaTzを有し、ここで、TはUで置換することができ、A、C、G、T およびUは塩基を表し、p、y、aおよびzは0から5であり、p+y+a+z の合計は少なくとも2であり、反復配列は当該標的配列内で前後して少なくとも 2回繰り返し、そしてA、C、GおよびTまたはUはいずれの順序であっても良 く、当該方法は、 a)標的配列を含んで成る試験試料と標的配列捕獲試薬とを接触させて標的配 列/捕獲オリゴヌクレオチドのハイブリッドを形成させる工程、ここで、当該標 的配列捕獲試薬は、固形支持体に固定した少なくとも第一および第二の捕獲オリ ゴヌクレオチドを含んで成り、ここで、 (i)当該第一捕獲オリゴヌクレオチドは、当該反復配列に相補的な第一 の多重度の配列を含んで成り、 (ii)当該第二捕獲オリゴヌクレオチドは、当該反復配列に相補的な第 二の多重度の配列を含んで成り、そして (iii)当該第一の多重度は、当該第二の多重度と異 なり、 b)当該ハイブリッドの環境のストリンジェンシーを増大して当該ハイブリッ ドの漸進的解離を引き起こす工程、 c)当該ハイブリッドの漸進的解離を検出する工程、および d)当該標的配列内の当該反復配列の多重度を測定する(ここで、当該反復配 列の測定された多重度は、当該捕獲オリゴヌクレオチドに比例する)工程を含ん で成ることを特徴とする、 前記方法。 2. 当該漸進的解離の検出が、当該ハイブリッド漸進的解離中の検出可能なシ グナルの変化を監視することにより達成される、請求項1記載の方法。 3. 当該第一および当該第二捕獲オリゴヌクレオチドが、当該固形支持体に配 置されて固定される、請求項1記載の方法。 4. 当該標的配列/捕獲オリゴヌクレオチドのハイブリッドを形成した後、そ して当該ハイブリッドの環境のストリンジェンシーを増大する前に洗浄工程を更 に含んで成る、請求項1記載の方法。 5. 当該漸進的解離を、光学的導波管装置を用いて検出する、請求項1記載の 方法。 6. 当該試験試料と当該リピート捕獲試薬とを接触させる前に、試験試料前処 理工程を更に含んで成る、請求項1記載の方法。 7. 当該ハイブリッドの環境のストリンジェンシーの当該増大を、当該ハイブ リッドの環境の温度を上昇させることにより達成する、請求項5記載の方法。 8. 標的配列を捕獲するための標的配列捕獲試薬であって、 a)少なくとも2種のオリゴヌクレオチド、ここで、 (i)第一オリゴヌクレオチドは、反復配列に相補的な第一の多重度の配 列を含んで成り、 (ii)第二オリゴヌクレオチドは、当該反復配列に相補的な第二の多重 度の配列を含んで成り、そして (iii)当該標的配列は、当該反復配列を含んで成り、 当該反復配列は、式ApCyGaTzを有し、ここで、TはUで置換することができ 、A、C、G、TおよびUは塩基を表し、p、y、aおよびzは0から5であり 、p+y+a+zの合計は少なくとも2であり、反復配列は当該標的配列内で前 後して少なくとも2回繰り返し、そしてA、C、GおよびTまたはUはいずれの 順序であっても良い、および b)当該少なくとも2種のオリゴヌクレオチドが固定される固形支持体を含んで 成る、前記捕獲試薬。 9. 当該少なくとも2種のオリゴヌクレオチドが、当該固形支持物質に配置 されて固定される、請求項8記載の捕獲試薬。 10. 当該捕獲試薬が、光学的導波管装置の一部を形成する、請求項8記載の リピート捕獲試薬。
