1336334 Dibenzocycloheptenylidene-piperidine derivatives MERCK & CO Inc 15 Jan 1971 [19 Jan 1970 5 Feb 1970] 2168/71 Heading C2C The invention comprises compounds of the Formula (I): in which Z is -CO- or -CH(OH)-, R 2 is C 1-6 alkyl or C 2-6 alkenyl, R is CH<SP>3</SP> or C 2 H 5 and replaces one or more of the H atoms in positions 2, 3, 5 or 6 of the piperidine ring, provided that only one of positions 3 and 5 is substituted in any one compound, n is 0, 1 or 2, each of X and X<SP>1</SP>, which are similar or dissimilar, is H or halogen, or a C 1-6 alkyl, C 1-4 perfluoroalkyl, phenyl, substituted phenyl, C 1-4 acyl, C 1-4 perfluoroaeyl, amino, C 1-4 alkylamino, di-(C 1-4 alkyl)amino, C 1-4 acylamino, C 1-4 perfluoroacylamino, C 1-4 alkylsulphonylamino, hydroxy- C 1-4 alkoxy, C 1-4 perfluoroalkoxy, cyano, carb, oxy, formyl, carbamoyl, C 2-5 alkylcarbamoyl, di-(C 1-4 alkyl)-carbamoyl, C 2-6 alkoxycarbonyl, mercapto, C 1-4 alkylthio, C 1-4 perfluoroalkylthio, aralkylthio, C 1-4 alkylsulphonyl, C 1-4 perfluoroalkylsulphonyl, C 1-4 alkylsulphinyl, sulphamoyl, C 1-4 alkylsulphamoyl, or di-(C 1-4 alkyl)sulphamoyl group, and more than one of these substituents may be on each benzenoid ring, but with the proviso that when Z is carbonyl, n is 0 and X and X<SP>1</SP> are hydrogen, R 2 contains at least two carbon atoms. Compounds of Formula (I) in which R 2 is replaced by R 1 which has the same value as R 2 but with no provisos, may be prepared by hydrolysing an enamine compound of the formula to form a compound of Formula (I) in which Z is -CO- and optionally reducing this 10-oxo compound to the corresponding 10-hydroxy compound where Z is -CH(OH)-. The enamine compound may be obtained as described in U.K. Specification 1,336,336. Pharmaceutical compositions useful for the treatment of allergic conditions comprise compounds of Formula (I) together with a non- toxic diluent, carrier or coating, in forms suitable for administration orally or parenterally.
