Abstract

A micro-organism is cultivated in the presence of a petroleum fraction consisting wholly or in part of straight chain hydrocarbons, an aqueous nutrient medium and a gas containing free oxygen, the petroleum fraction and aqueous medium being continuously fed to a fermenter containing the micro-organism wherein takes place a first stage of cultivation of the micro-organism, a product stream comprising the micro-organism is continuously passed to a second stage fermenter wherein the micro-organism is treated with a second stage aqueous nutrient medium which consists of a mixture of all or part of the aqueous nutrient medium withdrawn from the first stage fermenter together with additional water, and a product stream comprising micro-organism is continuously removed from the second stage fermenter and treated to recover the microorganism, and in the first stage of cultivation the amount of essential nutrients in the aqueous phase in the first stage fermenter is such that the rate of growth of the micro-organism is not impeded by shortage of nutrients and in the second stage the amount of at least one essential nutrient in the aqueous phase in the second stage fermenter is less than the amount required for unimpeded growth of the micro-organism in the first stage of cultivation and/or at least one essential nutrient which is present in the aqueous phase of the first stage fermenter is absent from the aqueous phase of the second stage fermenter. The second stage aqueous nutrient medium may consist of a mixture of all or part of the aqueous nutrient medium withdrawn from the first stage fermenter together with 1-8 times the volume of additional water, sufficient to reduce any toxin produced in the first stage aqueous nutrient medium to a level insufficient to substantially impede growth in the second stage. The dilution rate used in the operation in the second stage fermenter should not be lower than that used in the first stage fermenter. An example is directed to a two-stage cultivation of Candida tropicalis upon a gas-oil, in the second stage of which the aqueous nutrient medium was deficient in potassium and phosphorus (as phosphate).
