VSESOJUZNY NAUCHNOISSLEDOVATELSKY INSTITUT BIOSINTEZA BELKOVYKH VESCHESTV 4 Nov 1970 52401 /70 Heading C6F [Also in Division C5] A method of preparing a lipid comprises avokically cultivating lipid-producing microorganisms, particularly yeasts of the genus Candida, in a nutrient medium containing mineral salts as sources of nitrogen, phosphorus, potassium and magnesium, and a paraffinic hydrocarbon as a source of carbon, the quantity of paraffinic hydrocarbons not exceeding 75 per cent by volume, maintaining the ratio of nitrogen to carbon no lower than 1:75, and the ratio of phosphorus to carbon, calculating phosphorus as phosphorus pentexide, no lower than 1:60, and maintaining simultaneously a medium dilution co-efficient in the range of 0À5 to 0À07 per hour. The hydrocarbon may be a petroleum paraffin fraction or a synthetic paraffin. Lipids which contain up to 50% by weight phospho-lipids, up to 40 per cent free fatty acids and up to 10 per cent tri-glycerides are obtained when the amount of paraffinic hydrocarbon in the cultivation does not exceed 25 per cent by volume. N:C is from 1:5 to 1:20, P:C is from 1:15 to 1:30 and the medium dilution co-efficient is between 0À1 and 0À07 per hour. Lipids containing approximately up to 75 per cent of tri-glycerides, up to 9 per cent phospho-lipids and up to 5 per cent free fatty acids are obtained when the amount of paraffinic hydrocarbon in the cultivation does not exceed 50 per cent by volume, N:C is from 1:30 to 1:45, P:C is from 1:15 to 1:30 and the medium dilution co-efficient is between 0À1 and 0À07 per hour. Similarly, lipids which contain up to 50 per cent of free fatty acids and have only low contents of phospho-lipids and tri-glycerides are obtained when the amount of paraffinic hydrocarbon is in the range 50 to 70 per cent by volume, N:C is from 1:5 to 1:20, P:C is from 1:15 to 1:30 and the dilution co-efficient is between 0À5 and 0À07 per hour. Examples are directed to the cultivation of Candida tropicalis, Candida pelliculosa and Candida guilliermondii, respectively.
