Abstract
A találmány tárgyát képezik biológiai aktivitással rendelkezőbiomolekulák azonosítására szolgáló eljárások, amelyekbenenzimaktivitás-modulátort (vázmolekulaként) kódoló nukleotid-szekvenciába random peptideket illesztve alapvetően azonos sejtekettranszformálnak, és a transzformált sejteket előre kiválasztottfenotípusos elváltozás szempontjából szkrínelik. A randomizált DNS-szekvenciát a sejtekből izolálhatják és a random szekvenciátmeghatározhatják. A sejtben az expresszált fehérjével kölcsönhatásbalépő partnermolekulákat (feltételezett droghatás-célmolekulákként)azonosíthatják és izolálhatják a random peptidet affinitásireagensként alkalmazva. Vázmolekulaként 2A-típusú, árpaeredetűkimotripszin-inhibitort (CI-2A) alkalmaznak. Az eljárássalazonosíthatóak új enzim-inhibitorok és gyógyhatású készítményekbenalkalmazható drog kifejlesztésben alkalmazható lead-vegyületek. Atalálmány tárgyát képezik tehát: - eljárások olyan, biológiailag aktívpolipeptid-fragmens formájú modulátor azonosítására, amely alkalmassejtben fenotípusos sajátosság modulálására in vivo, detektálhatómódon történő modulálására; - eljárások replikációra képes vektorelőállítására - eljárások modulátor azonosítására, izolálására éselőállítására: - eljárások biológiai célmolekula azonosítására,izolálására és előállítására: - eljárások feltételezett drogjelöltkémiai vegyület kiválasztására; - eljárások gyógytermék előállítására.
