Abstract

Данное изобретение обеспечивает способы идентификации биологически активных биомолекул. В одном аспекте биологически активную биомолекулу, такую как РНК или пептид, идентифицируют включением случайных нуклеотидных последовательностей в каркас, состоящий из модулятора активности фермента, трансформацией существенно идентичных клеток полученной таким образом конструкцией и скринингом трансформированных клеток для идентификации клеток, в которых был изменен предварительно выбранный фенотипический признак. Затем из фенотипически измененных клеток выделяют рандомизированную ДНК и определяют пептид и/или РНК, кодируемые этой случайной последовательностью. В свою очередь, идентифицируют партнеры взаимодействия, которые являются возможными лекарственными средствами-мишенями, и выделяют их с использованием указанных пептида и/или РНК в качестве части аффинных реагентов. Предпочтительную каркасную молекулу получают из семейства ингибиторов протеаз ингибиторов I картофеля, примером которых является ингибитор трипсина 2A ячменя (CI-2A). Другой аспект относится к идентификации новых ингибиторов ферментов с использованием, по существу, того же самого подхода, но с проведением специфического скрининга на изменения в активности фермента-мишени. Описаны также способы получения соответствующих трансформирующих и экспрессирующих векторов, а также способы идентификации соединений-примеров и лекарственных средств-мишеней для применения в разработке лекарственных средств.
