본 발명에 따른 공정을 다이아그람식으로 도식한 것임.
Claims (10)
- a) 목적 단백질 암호인 DNA서열, 짧은 사슬펩티드 암호인 DNA서열 및 생체 특이성 폴리펩티드 암호인 DNA서열을 짧은 사슬 펩티드용 DNA서열이 다른 두 DNA서열 사이에 위치하도록 서로 결합시키고, b) 생성된 새로운 DNA 단편은 생체특이성 폴리펩티드, 짧은 사슬 펩티드 및 목적 단백질로된 혼성화 단백질의 전사, 해독 및 표현과정이 일어나도록 적당한 유전공학기법에 의하여 숙주 미생물의 DNA에 도입시키고, c) 숙주 미생물이 혼성화 단백질을 생성하도록 배양시키고, d) 혼성화 단백질을 함유한 배양계부분을 필요한다면 다른 구성성분이 제거후에 적당한 담체물질로 이루어진 고정계와 접촉시키고, 여기에 생체 특이성 폴리펩티드에 상보성(相補性)이고, 소정의 반응조건에서 그 폴리펩티드에 강력하게 결합되는 물질을 결합시키고, e) 혼성화 단백질이 결합되어 있는 고정계를 적당한 방법으로 다른 구성성분으로부터 분리시켜서 그 결합된 혼성화단백질로부터 짧은 사슬펩티드를 특이적으로 절달시키는 시약으로 처리하고, f) 이와 같이 하여 유리시킨 목적 단백질을 다른 구성 성분으로부터 분리해 내어 적당한 방법으로 단리시키는 것을 특징으로 하는 단백질 제조방법.
- 제1항에 있어서, 생체 특이성 폴리펩티드가 높은 비율로 보체(補體)를 결합시킨 담체에 높은 비율로 결합됨을 특징으로 하는 방법.
- 제2항에 있어서, 결합이 +2℃에서 실온까지의 온도에서 형성됨을 특징으로 하는 방법.
- 제1 내지 제3항에 있어서, 생체 특이성 폴리펩티드와 상보성의 담체 결합 물질이 효소 및 관련 억제제로 이루어진 계임을 특징으로 하는 방법.
- 제1 내지 제4항에 있어서, 생체 특이성 폴리펩티드가 아프로티닌이고, 상보성의 담체 결합 물질이 트립신임을 특징으로 하는 방법.
- 제1 내지 제5항에 있어서, 짧은 사슬 펩티드가 Lys-Arg-Pro-Pro-Gly-Phe-Ser-Pro-Phe-Arg 데카펩티드임을 특징으로 하는 방법.
- 제1 내지 제6항에 있어서, 결합된 혼성화 단백질로부터 특이적으로 짧은 사슬펩티드를 절단시키는 시약이 효소임을 특징으로 하는 방법.
- 제6 내지 제7항에 있어서, 효소가 칼리크레인(E.C. 2.4.21.8)임을 특징으로 하는 방법.
- 제1 내지 제8항에 있어서, 제조될 단백질이 호르몬, 플라스마단백질, 응고화 인자, 효소, 항체 또는 백신류임을 특징으로 하는 방법.
