Claims (10)

1. Metoda za nedestruktivno utvrđivanje obilježja embrija Gallus Gallus domesticus in ovo, metoda koja obuhvaća: (a) dobivanje uzorka materijala povezanog s jajetom koje sadrži zametak, i (b) mjerenje bodovne vrijednosti za prisutnost i koncentraciju najmanje prvog biomarkera u uzorku koji ukazuje na karakteristike embrija, i (c) primjena praga na vrijednost rezultata i koncentraciju dobivenu u (b) radi identificiranja karakteristike embrija povezane s prisutnošću i koncentracijom prvog biomarkera, pri čemu prvi biomarker sadrži amino spoj molekularne težine u rasponu od 150 do 170 g / mol; pri čemu korak (c) nadalje obuhvaća: (i) korelaciju svakog relevantnog signala biomarkera s referentnim biomarkerom usklađivanjem spektra svakog korelacijskog signala s očekivanim spektrom korelirajućeg referentnog biomarkera pomoću mjere sličnosti, kako bi se definirao barem jedan pozitivno korelirajući signal; (ii) mjerenje intenziteta svakog pozitivno korelirajućeg signala i bodovanje njegovog apsolutnog i/ili relativnog intenziteta signala; i (iii) primjenu praga na vrijednost rezultata dobivenu funkcijom sličnosti za određivanje korelirane karakteristike embrija; pri čemu je prvi biomarker 3-[(2-aminoetil)sulfanil]butanska kiselina, a koncentracija prvog biomarkera u količini od 50 ng/ml ili više u alantoičnoj tekućini embrija 7, 8 ili 9 dan korelira sa ženskim zametkom, dok prisutnost prvog biomarkera prisutnog u manje od 50 ng/ml korelira s muškim embrijem.
2. Postupak u skladu s patentnim zahtjevom 1, naznačen time što se otkrivaju i analiziraju prvi i drugi biomarker, i pri čemu se apsolutne i relativne količine najmanje prvog i drugog markera koriste za određivanje jedne ili više karakteristika.
3. Postupak prema zahtjevu 1 ili 2, naznačen time što obuhvaća primjenu jedne ili više metoda magnetske rezonancije; metoda spektralne rezonancije; analitička kromatografska metoda povezana s jednim ili više prikladnih detektora; fluorescentna spektroskopija; i/ili metode ispitivanja koje uključuju reagense selektivne za biomarker, za mjerenje prisutnosti i koncentracije jednog ili više biomarkera, po mogućnosti gdje masena spektrometrijska analiza obuhvaća masenu spektroskopiju ionskom raspršivanjem (ESI), lasersku ionsku jonizaciju uz pomoć matrice - vrijeme leta (MALDI- TOF) masena spektroskopija ili površinski pojačana laserska desorpcijska ionizacija - masena spektroskopija vremena leta (SELDI-TOF), izvor iona SPE/MS/MS, LDTD (termalna desorpcija laserske diode) i/ili uporaba senzora pokretljivosti iona (IMS).
4. Metoda prema bilo kojem od prethodnih zahtjeva, naznačena time što se testni uzorak obrađuje prije analize, po mogućnosti gdje obrada uzorka uključuje odvajanje uzorka ekstrakcijom u čvrstoj fazi (SPE), plinskom kromatografijom, jednofaznom ili višefaznom visokotlačnom tekućinskom kromatografijom (HPLC).
5. Postupak prema bilo kojem od zahtjeva 3 ili 4, naznačen time, što se jedan ili više internih standarda referentnih biomarkera dodaje uzorku prije analize masenom spektrometrijom.
6. Postupak prema bilo kojem od prethodnih zahtjeva, naznačen time što se apsolutni intenzitet signala ocjenjuje mjerenjem intenziteta signala biomarkera i usporedbom s intenzitetom signala jednog ili više poznatih internih standarda.
7. Metoda prema bilo kojem od prethodnih zahtjeva, naznačena time što se mnoštvo jajašaca ispituje na jednu ili više embrionalnih karakteristika; i po mogućnosti dalje obuhvaća utvrđivanje je li embrij u jajetu održiv i muški, ili održiv i ženski, ili neživ, i odvajanje mnoštva održivih muških jajašaca od mnoštva održivih ženskih jajašaca i jedne ili više neživih jajnih stanica.
8. Metoda prema bilo kojem prethodnom zahtjevu, naznačena time što je metoda potpuno automatizirana.
9. Postupak u skladu s patentnim zahtjevom 8, naznačen time što nadalje uključuje sortiranje održivih jajašaca prema predviđenom vremenu dok se vjerojatnost da se ne izlegne.
10. Postupak selektivne inkubacije mladunaca vrsta peradi koje nesu jaja sa specifičnim karakteristikama, koji obuhvaća a. pružanje mnoštva jaja iz vrste, i b. podvrgavanje jaja postupku prema bilo kojem zahtjevu od 1 do 9 za određivanje karakteristike embrija, i c. odabir jaja sa željenom karakteristikom za formiranje odabranog mnoštva jaja, i d. inkubiranje odabranih jaja dok se ne izleže jedno ili više mladunaca.
