Claims (15)
1. Postupak za proizvodnju rekombinantnog proteina u stanicama biljke koje imaju staničnu stijenku, koji obuhvaća korak izlučivanja rekombinantnog proteina kroz staničnu stijenku ekspresijom proteina u biljnim stanicama u mediju za kultiviranje koji sadrži kombinaciju površinski aktivnog polimera, jednovalentne ione metala i ima osmolarnost od najmanje 0,32 osmol/L.
2. Postupak prema patentnom zahtjevu 1, naznačen time što se izlučivanje u medij odvija iz apoplastičnog prostora stanice.
3. Postupak prema bilo kojem zahtjevu 1 ili zahtjevu 2, naznačen time što biljna stanica je stanica biljke ili biljnog tkiva, posebno je poželjna stanica mahovine.
4. Postupak prema bilo kojem od patentnih zahtjeva 1 do 3, naznačen time što se stanica uzgaja u suspenzijskoj kulturi.
5. Postupak prema bilo kojem od patentnih zahtjeva 1 do 4, naznačen time što je ion metala, ion alkalijskog metala, poželjno natrij, i/ili ion metala je prisutan u mediju u koncentraciji od najmanje 20 mM.

Postupak prema bilo kojem od patentnih zahtjeva 1 do 6, naznačen time što medij sadrži nitratne ione, poželjno u koncentraciji od najmanje 0,2 mM, fosfatne ione, poželjno u koncentraciji od najmanje 0,05 mM, sulfatne ione, poželjno u koncentraciji od najmanje 0,02 mM, kalcijeve ione, poželjno u koncentraciji od najmanje 0,1 mM, kalijeve ione, poželjno u koncentraciji od najmanje 0,1 mM, natrijeve ione, poželjno u koncentraciji od najmanje 20 mM, ili njihove kombinacije.
8. Postupak prema bilo kojem od patentnih zahtjeva 1 do 7, naznačen time što, se prije navedenog koraka izlučivanja rekombinantnog proteina u navedenom mediju, stanice uzgajaju u prethodnom koraku u kultivacijskom mediju koji ima osmolarnost manju od 0,1 osmol/L, naročito bez navedenog izlučivanja, što uzrokuje nakupljanje proteina unutar stanice ili u apoplastičnom prostoru stanica.
9. Postupak prema patentnom zahtjevu 8, naznačen time što kultivacijski medij, u navedenom prethodnom koraku, sadrži nitratne ione, poželjno u koncentraciji od najmanje 0,2 mM, fosfatne ione, poželjno u koncentraciji od najmanje 0,05 mM, sulfatne ione, poželjno u koncentraciji od najmanje 0,02 mM, kalcijeve ione, poželjno u koncentraciji od najmanje 0,1 mM, kalijeve ione, poželjno u koncentraciji od najmanje 0,1 mM, ili njihove kombinacije; i/ili sadrži natrijeve ione u koncentraciji od najviše 20 mM.
10. Postupak prema bilo kojem zahtjevu 8 ili zahtjevu 9, naznačen time što kultivacijski medij, u navedenom prethodnom koraku, ne sadrži površinski aktivan polimer ili ga sadrži u koncentraciji od najviše 0,08 mas. %.
11. Postupak prema bilo kojem od patentnih zahtjeva 8 do 10, naznačen time što se navedeni prethodni korak provodi u periodu od 2 do 30 dana.
12. Postupak prema bilo kojem od patentnih zahtjeva 1 do 11, naznačen time što se navedeni korak izlučivanja provodi kroz vrijeme od najmanje 2 dana, poželjno kroz 3 do 120 dana.
13. Kultivacijski medij pogodan za poticanje izlučivanja pomoću postupka prema bilo kojem od patentnih zahtjeva 1 do 12, naznačen time što sadrži površinski aktivni polimer u količini od najmanje 0,05 mas. % i jednovalentne ione metala u koncentraciji od najmanje 20 mM i ima osmolarnost od najmanje 0,32 osmol/L.
14. Kultivacijski medij prema patentnom zahtjevu 13 naznačen time što medij sadrži nitratne ione, poželjno u koncentraciji od najmanje 0,2 mM, fosfatne ione, poželjno u koncentraciji od najmanje 0,05 mM, sulfatne ione, poželjno u koncentraciji od najmanje 0,02 mM, kalcijeve ione, poželjno u koncentraciji od najmanje 0,1 mM, kalijeve ione, poželjno u koncentraciji od najmanje 0,1 mM, natrijeve ione, poželjno u koncentraciji od najmanje 40 mM, ili njihove kombinacije.
15. Smjesa suhog medija za rast za rekonstituciju kultivacijskog medija prema bilo kojem od zahtjevu 13 ili zahtjevu 14.
