Claims (15)

1. Postupak ekstrahiranja lipopolisaharida (LPS) iz gram negativnih bakterijskih stanica, naznačen time što se sastoji u koraku: ekstrahiranja LPS iz stanica u smjesi za ekstrahiranje LPS koja sadrži kloroform, metanol i vodu, gdje je smjesa za ekstrahiranje LPS jednofazna, te gdje je količina vode u smjesi za ekstrahiranje LPS između 0,4 i 1,5% (vol./vol.), postotak metanola u smjesi za ekstrahiranje LPS je između 5% i 40% (vol./vol.), te gdje je postotak kloroforma u smjesi za ekstrahiranje LPS između 60% (vol./vol.) i 95% (vol./vol.).
2. Postupak u skladu s patentnim zahtjevom 1, naznačen time što je količina vode 0,8 do 1,2% (vol./vol.).
3. Postupak u skladu s patentnim zahtjevom 2, naznačen time što je količina vode 1% (vol./vol.).
4. Postupak u skladu s bilo kojim od patentnih zahtjeva 1 do 3, naznačen time što je postotak metanola u smjesi za ekstrahiranje LPS između 10% i 30% (vol./vol.).
5. Postupak u skladu s bilo kojim od patentnih zahtjeva 1 do 4, naznačen time što je postotak kloroforma u smjesi za ekstrahiranje LPS između 75% (vol./vol.) i 90% (vol./vol.).
6. Postupak u skladu s bilo kojim od patentnih zahtjeva 1 do 5, naznačen time što se ekstrahiranje LPS provodi na temperaturi između otprilike 35 °C i otprilike 65 °C.
7. Postupak u skladu s bilo kojim od patentnih zahtjeva 1 do 6, naznačen time što se LPS ekstrahiranje provodi na pH između 7,8 i 9.
8. Postupak u skladu s bilo kojim od patentnih zahtjeva 1 do 7, naznačen time što se LPS ekstrahiranje provodi u trajanju od otprilike 0,5 do otprilike 20 sati.
9. Postupak u skladu s bilo kojim od patentnih zahtjeva 1 do 8, naznačen time što dodatno uključuje korake: i. ispiranja stanica otopinom etanola ili etanolom; i izborno ii. ispiranja stanica po drugi put etanolom ili metanolom.
10. Postupak u skladu s bilo kojim od patentnih zahtjeva 1 do 9, naznačen time što dodatno uključuje korak: isparavanja vode, alkohola i dodatnog organskog otapala iz otopine LPS, čime se dobije suhi ostatak LPS.
11. Postupak u skladu s bilo kojim od patentnih zahtjeva 1 do 10, naznačen time što je bakterijska stanica ona duboko grubog mutantnog bakterijskog soja iz roda Salmonella ili Escherichia.
12. Postupak u skladu s patentnim zahtjevom 11, naznačen time što su bakterijske stanice one od bakterije Escherichia coli.
13. Postupak u skladu s patentnim zahtjevom 11, naznačen time što su bakterijske stanice one od bakterije Salmonella minnesota.
14. Postupak u skladu s patentnim zahtjevom 11, naznačen time što su bakterijske stanice one od bakterije Salmonella minnesota R595.
15. Postupak u skladu s bilo kojim od patentnih zahtjeva 1 do 14, naznačen time što dodatno uključuje korak: podvrgavanja LPS uzastopno kiseloj hidrolizi i bazičnoj hidrolizi, kako bi se dobilo 3D-MPL.
