Claims (11)
1. Metoda za odabir i umnožavanje polinukleotida na čvrstoj podlozi, obuhvaća:
a) omogućavanje mnoštva oligonukleotida za umnožavanje imobiliziranih na čvrstoj podlozi;
b) hibridiziranje populacije oligonukleotidnih sondi na podskup navedenih oligonukleotida za umnožavanje, svaka od navedenih oligonukleotidnih sondi sadrži prvi dio koji je komplementaran oligonukleotidima za umnožavanje i drugi dio koji sadržislijed iz odabranog područja polinukleotidnog kalupa;
c) izvođenje reakcije produljivanja zbog produljivanja hibridiziranih oligonukleotida za umnožavanje u svrhu stvaranja populacije potporno učvršćenih veznih nukleotida, svaki vezni oligonukleotid u navedenoj populaciji sadrži slijed koji je komplementaran odabranom području polinukleotidnog kalupa, stoga omogućuje čvrstu podlogu koja obuhvaća populaciju potporno učvršćenih veznih nukleotida i populaciju neproduljenih oligonukleotida za umnožavanje;
d) primjenu populacije polinukleotida kalupa na čvrstu podlogu pod uvjetima selektivne hibridizacije polinukleotida kalupa na potporno učvršćene vezne nukleotide, pri čemu polinukleotidi kalupa sadrže adapterski slijed na 5' kraju, pri čemu je adapterski slijed isti kao slijed neproduljenih oligonukleotida za umnožavanje;
e) produljivanje potporno učvršćenih veznih nukleotida koji su hibridizirani za polinukleotide kalupa, stoga stvaraju produljene produkte, produljeni produkti imaju dio komplementaran polinukleotidima kalupa i dio komplementaran s neproduljenim oligonukleotidima za umnožavanje; i
f) umnožavanje produljenih produkata, pri čemu umnožavanje obuhvaća nalijeganje jednog ili više neproduljenih oligonukleotida za umnožavanje na jedan ili više produljenih produkata, stoga stvarajući produkt umnožavanja na čvrstoj fazi.
2. Metoda iz patentnog zahtjeva 1, nadalje obuhvaća:
g) sekvenciranje prvog lanca produkta umnožavanja na čvrstoj fazi iz koraka f) zbog dobivanja barem dijela slijeda nukleotida polinukleotida kalupa.
3. Metoda iz patentnog zahtjeva 2, pri čemu produkti umnožavanja na čvrstoj fazi obuhvaćaju vezno mjesto za univerzalnu početnicu za sekvenciranje.
4. Metoda iz patentnog zahtjeva 1, pri čemu vezni oligonukleotidi sadrže najmanje 10 različitih veznih slijedova.
5. Metoda iz patentnog zahtjeva 1, pri čemu su oligonukleotidi za umnožavanje, koji nisu hibridizirani s oligonukleotidnom sondom u koraku b) patentnog zahtjeva 1, reverzibilno blokirani za vrijeme produljivanja veznih nukleotida.
6. Metoda iz patentnog zahtjeva 5, pri čemu je reverzibilno blokiranje pomoću kemijskih vrsta pričvršćenih na 3' kraju oligonukleotida za umnožavanje.
7. Metoda iz patentnog zahtjeva 6, pri čemu je kemijska vrsta fosfatna grupa.
8. Metoda iz patentnog zahtjeva 1, pri čemu je umnožavanje izotermalno.
9. Metoda iz patentnog zahtjeva 1, pri čemu dio produljenih produkata komplementaran s neproduljenim oligonukleotidima za umnožavanje obuhvaća adapterski slijed.
10. Metoda iz patentnog zahtjeva 9, pri čemu polinukleotidi kalupa u uzorku nukleinske kiseline obuhvaćaju različite sljedove i adapterski slijed je isti za svaki polinukleotid kalupa.
11. Metoda iz patentnog zahtjeva 10, pri čemu svako mnoštvo oligonukleotida za umnožavanje obuhvaća zajednički slijed koji je komplementaran adapterskom slijedu koji je isti za svaki polinukleotidni kalup.
