Abstract

The invention comprises 3H - pyrimido-[5,4-c]-1,2,5-oxadiazines of the formula <FORM:0993595/C2/1> wherein R1 and R2, which may be the same or different, represent hydrogen atoms or straight or branched chain alkyl radicals and R3 and R4, which may be the same or different, are alkyl radicals or R3 and R4 may together form a cyclic radical and the preparation thereof by reaction of a compound of the formula <FORM:0993595/C2/2> with nitrous acid or an ester thereof, preferably at an elevated temperature or by a modification of the above process in which a 4-aminouracil of the formula <FORM:0993595/C2/3> is nitrosated with excess nitrosation agent to give the 5-nitrose compound which without isolation is cyclized by the action of excess nitrosation agent. 1,3 - Dimethyl - 8,8 - pentamethylene - 8H - xanthine - 7 - N - oxide is formed as an intermediate by the action of isoamyl nitrite on 1,3 - dimethyl - 4 - cyclohexylamino - 5 - nitrouracil or on 1,3-dimethyl-4-cyclohexylaminouracil. 5-Nitrosouracils of the Formula II where stable may be prepared by nitrosation of uracils of the Formula III. Uracils of the Formula III may be prepared by reaction of the appropriately 1,3-substituted 4-chlorouracils with amines of the formula <FORM:0993595/C2/4> or by transemination of the appropriately substituted 4-aminouracils with amine hydrochlorides of the formula <FORM:0993595/C2/5>
