Abstract

The invention comprises compounds of the general formula <FORM:0982945/C2/1> (wherein each R1 is a hydrogen atom or a C2-6 alkanoyl radical, R2 is a straight-chain C1-5 alkyl radical and R3 is a hydrogen atom or a C1-5 alkyl radical) and non-toxic acid-addition salts of the compounds in which each R1 is a hydrogen atom, but excluding compounds claimed in Specification 936,074, pharmaceutical preparations containing them, and their preparation by (a) reacting a compound of the formula <FORM:0982945/C2/2> (wherein R4 is a C1-5 alkyl radical) with hydrochlori or hydrobromic acid at an elevated temperature, and reacting the product of the formula <FORM:0982945/C2/3> (wherein X is a chlorine or bromine atom) with a base for a sufficient period of time to liberate the amine and open the coumarin ring, or (b) reacting a compound of the formula <FORM:0982945/C2/4> with concentrated hydrobromic acid at above 90 DEG C. and converting the product to the free amino acid, optionally followed in either case by salt formation, esterification of the carboxyl group or acylation. The preparation of the starting materials is illustrated by the following reaction scheme for the compounds in which R2 and R4 are methyl radicals:- <FORM:0982945/C2/5> <FORM:0982945/C2/6> <FORM:0982945/C2/7> <FORM:0982945/C2/8> <FORM:0982945/C2/9> The compounds of the invention are anti-hypertensive agents and may be administered orally or parenterally in the form of pharmaceutical preparations (e.g. capsules, tablets, solutions and suspensions) containing them together with a diluent or carrier.
