Abstract
The invention comprises basic phenoxazine derivatives of the general formula: <FORM:0965738/C1/1> wherein A is any alkylene radical containing 2 to 4 carbom atoms and acyl is an acyl group derived from an aliphatic, carboxylic acid containing up to 3 carbon atoms and salts thereof, and the preparation thereof (a) by reacting a reactive ester of general formula: <FORM:0965738/C1/2> wherein Y is an acid radical and X is an acyl group derived from an aliphatic carboxylic acid defined above or such an acyl group with a protected carboxyl group and A has the above meaning, with piperidin-4-ol and hydrolysing a protected group X, if necessary, to give the acyl radical; or (b) by reacting a phenoxazine derivative of general formula: <FORM:0965738/C1/3> in which X has the above meaning with a piperidine derivative of general formula: <FORM:0965738/C1/4> in which A and Y have the above meanings and hydrolysing a protected group X, if necessary, to give the acyl radical. The phenoxazine derivatives may be converted to their salts in known manner. In an example 2-acetyl-10-(3-chloropropyl)-phenoxazine-ethylene ketal is first prepared by reacting 2-acetyl-phenoxazine-ethylene ketal with 1-chloro-3-bromopropane in the presence of sodamide in a stirred mixture of ether and liquid ammonia and then converted by method (a) above to 2-acetyl-10-[3-(4-hydroxypiperidyl)-propyl]-phenoxazine-ethylene ketal which is then hydrolysed with 1-N-hydrochloric acid to give 2-acetyl-10-[3-(4-hydroxypiperidyl)-propyl]-phenoxazine. The salts of the basic phenoxazine derivatives have sedative and neuroleptic properties and may find use in psychiatric pharmacotherapy. Specification 850,334 is referred to.
