Abstract
The invention comprises compounds of general formula: <FORM:0961980/C1/1> wherein R1 is alkyl (1-6C), phenyl or benzyl, R2 is hydrogen, alkyl (1-6C), cycloakyl, phenyl or benzyl, X is alkyl (1-6C), phenyl, tolyl or benzyl, and Z is hydrogen or a cation, salts being formed with inorganic or organic bases; and their tautomers; and their preparation (1) by condensing a compound of formula: <FORM:0961980/C1/2> or its tautomer with a compound of formula halogen-SO2X in the presence of an acid binding agent and, in the case where an intermediate substituted by two sulphonyl radicals is formed, hydrolysing off one of the radicals, or (2) by condensing a compound of formula: <FORM:0961980/C1/3> wherein R3 is a halogen atom, with an alkali metal salt of a sulphonamide of formula H2N-SO2-X in the presence of a solvent. Amines II can be prepared from the corresponding 3-halogen compounds by treatment with ammonia in the presence of a cuprous salt. Halo-compounds III are prepared in examples either by the action on corresponding 1,4-substituted -3- chloro -2- pyrazolin -5- ones (obtained from 3,5-pyrazolidindiones using phosphorus oxychloride) of dimethyl sulphate to give the compounds wherein R1 is methyl, or of appropriate esters of benzene sulphonic acid to give the compounds wherein R1 is butyl or benzyl. Specification 563,279 is referred to.ALSO:Pharmaceutical compositions contain sulphonamides of general formula <FORM:0961980/A5-A6/1> wherein R1 is alkyl (1-6 C), phenyl or benzyl, R2 is hydrogen, alkyl (1-6 C), cycloalkyl, phenyl or benzyl, X is alkyl (1-6 C), phenyl, tolyl or benzyl, and Z is hydrogen or a cation (salts being formed with inorganic and organic bases), and the tautomers thereof, together with pharmaceutical vehicles. Forms (tablets, dragees, ointments, creams, solutions, suspensions, emulsions) are specified for enteric, parenteral and topical administration in human and veterinary medicine. The compounds have antiphlogistic and analgesic properties. The incorporation of, for example, local anaesthetics and use of salts with tetraethyl ammonium hydroxide are mentioned. Specification 563,279 is referred to.
