Abstract

Novel compounds of the general formula <FORM:1068083/C2/1> wherein R1 represents a hydrogen atom; or a saturated or unsaturated hydrocarbon group which may, if desired be substituted by a phenyl group, by a free or substituted amino group, or by a nitrogen containing hetero cyclic group linked to the said hydrocarbon group via the nitrogen atom; R2 is a saturated or unsaturated acylic hydrocarbon group, an aralkyl or aralkenyl group or an aryl group, and R3 represents an aryl group, and when R1 represents a hydrogen atom, the compound may have the tautomeric formula <FORM:1068083/C2/2> and the non-toxic acid addition salts thereof, are prepared by heating a compound of the general formula <FORM:1068083/C2/3> wherein R1, R2 and R3 are as defined above to effect cyclization with elimination of water, and (when R1 is other than hydrogen) by reacting a compound of general formula <FORM:1068083/C2/4> wherein R2 and R3 are as defined above with a halide of formula R1X wherein R1 is as previously defined and is other than hydrogen, at elevated temperatures. The first process above may be modified in that the substituted urea reactant of the third general formula above is prepared by reacting a hydrazine of the formula R3NHNH2 with a urea of the formula <FORM:1068083/C2/5> N - (a - phenylbutyryl - N1 - phenyl - N1 - amino-ure is prepared by heating a solution of phenylhydrazine and N-(a -phenylbutyryl) urea in toluene. Pharmaceutical compositions having anti-inflammatory, antipyretic and analgesic activity and which may be administered orally, rectally, parenterally or topically in the form of capsules, tablets, coated tablets, suspensions and ointments contain a compound of either of the first two general formulae above as the active ingredient associated with other pharmaceutical carriers.
