Abstract
Hydroxocobalamin which has been treated chromatographically is purified by dissolving in a water-soluble organic solvent and treating with a strong acid to form an acid hydroxocobalamin salt which is passed in solution successively through two ion exchanger resins, the first being a neutral resin of the snake-cage type which retains the salt impurities while allowing the hydroxocobalamin to pass and the second a basic resin which frees the acid and regenerates the hydroxocobalamin. Chromatography may be effected in an alumina column using a solution in acetone, dioxan or tetrahydrofuran, the solution is then treated with hydrochloric or sulphuric acid, and the resultant salt separated by crystallisation. The crystals are dissolved in water and passed through "Retardion" resin and then through "Dowex" 2 or "Amberlite" IRA 400 resins (Registered Trade Marks). Specification 916,186 is referred to.
