Abstract
The invention comprises: (a) lysergic acid pyrazole derivatives of formula <FORM:0933082/IV(a)/1> (wherein R1 is hydrogen, C1-4 alkyl or C2-4 alkenyl, R3 and R4 are C1-4 alkyl and x y signifies -CH=C< or <FORM:0933082/IV(a)/2> ; (b) a process for producing compounds of formula <FORM:0933082/IV(a)/3> (wherein R2 is hydroxy, C1-4 alkylamino or C1-4 hydroxyalkylamino) or their addition salts by reacting a pyrazole derivative (a) with a compound HR2, the reaction being effected in the presence of alkali or acid when R2 is hydroxy, and, if desired, salifying the product with an organic or inorganic acid; and (c) the combination of process (b) with the preparation of the pyrazoles (a) by reacting the corresponding lysergic acid or dihydrolysergic acid hydrazide with a diketone R3-CO-CH2-CO-R4 in the presence of slightly more than the equivalent quantity of inorganic acid. In a modification to the process, when R2 is a hydroxy group, the pyrazole is not isolated, the carboxylic acid being formed directly by reacting the hydrazide with the diketone in the presence of at least two equivalents of an aqueous organic acid. Apart from water, the reactants H-R2 utilized in the examples are ethylamine and (+)-butanolamine-(21).
